Conditioned Medium of Soybean Extract Treated Osteoblasts Inhibits RANKL Induced Differentiation of Osteoclasts (대두추출물을 처리한 조골세포 조건배양액은 RANKL에 의해 유도된 파골세포 분화를 억제)

  • Soybean is of particular interest as a food supplement of isoflavones for inhibiting bone resorption in postmenopausal woman. These beneficial effects of isoflavones are caused by functioning as partial agonists or antagonists of estrogen, of which anti-resorptive effect is mediated indirectly through paracrine factors produced by osteoblasts that act on osteoclasts. In this study, the indirect effect of soybean on osteoclastic differentiation of RAW264.7 cells were investigated. The conditioned medium was collected from MC3T3-E1 osbeoblasts treated with 0.001 mg/mL~0.1 mg/mL soybean extracts for 6 days, mixed in 1:1 ratio with osteoclast medium, and then added into RAW264.7 cells with receptor activator of nuclear factor kappa B ligand (RANKL), a differentiation inducer for 3 days. Of paracrine factors in the conditioned medium, the protein expression of osteoprotegerin (OPG) with soybean extract was specifically higher in a dose dependent manner than with $10^{-9}$ M~$10^{-6}$ M of estrogen, genistein or daidzein standards. In RAW264.7 cells, the conditioned medium with soybean inhibited RANKL induced osteoclastic differentiation as total number of multinucleated tartrateresistant alkaline phosphatase (TRAP)-positive osteoclasts and protein expression of MMP-9 were significantly decreased. Coupled with the low expression of estrogen receptor $\alpha$ and $\beta$ proteins in RANKL treated RAW264.7 cells, we demonstrate that the conditioned medium of soybean treated osteoblasts inhibits RANKL induced differentiation of osteoclasts with the selective expression of OPG in osteoblasts.

Impact of social relationships on self-related information processing and emotional experiences (사회적 관계가 개인의 정보처리와 정서경험에 미치는 효과)

  • Do social situations have an impact on an individual's information processing and emotional experiences? Two studies were conducted to investigate relationships between self-reference effects, emotional experiences and social information processing. Study 1 examined whether biases favoring self-related stimuli could occur automatically. Participants had to judge whether sequential geometric shape-label pairs matched or mismatched. The results showed that self-related stimuli are more rapidly processed than friends/others-related stimuli. In Study 2, the participants had to recall items which were presented with different instructions (either chosen by a friend or by the computer). Here we explored whether the self-reference effect is reduced in a social learning condition. When comparing the social learning condition (seated in pairs) with the nonsocial learning condition (seated alone), the participants recalled more self-related words in the nonsocial learning condition than in the social learning condition. Importantly, the automatic self-reference effect disappeared in the social learning condition. More friends-related words were recalled in the social condition than self-related words. In addition, while tasting chocolates, the participants judged them to be more likeable in the social condition than in the nonsocial condition. These results implicated that social processing can be useful for reducing the automatic self-reference effects and shared experiences are perceived more intensely than unshared experiences.

Constraints of Bio-augmentation in Improving Performances of Biological Treatment Process (미생물 활성 촉진에 의한 생물학적 처리효율 평가에 있어서의 제한성에 관한 연구)

  • 생물학적 처리공정내 미생물은 물리화학적, 생태학적으로 처리공정의 운전조건과 유입하·폐수 특성에 따라 존재량 및 반응조내 존재하는 우점종이 달라진다. 특히,폐수의 생물학적 처리시 유입수내 존재하는 독성 및 저해물질은 생물반응조내 존재하는 미생물의 양을 감소시키거나 미생물의 종류를 단순화시키거나 환경조건 및 폐수 특성변화에 능동적으로 대처할 수 있는 능력을 감소시킨다. 따라서 이러한 문제점을 극복하기 위한 방안으로 생물능력향상(bio-augmentation)이 필요하며, 여러 가지 방법으로 생물능력향상을 이룰 수 있다. 본 연구에서는 생물능력향상을 위하여 기존의 효소활성이 강한 미생물군을 투입함으로써 처리효율향상에 미치는 영향 및 제한성을 고찰하였다. 처리효율 향상을 위해서는 생존성, 폐수에 대한 순응기간 및 미생물의 체류가 매우 중요하였으며, 특히 누입된 미생물군이 처리효율을 향상시키기 위해서는 일정기간 이상의 순응기간이 필요하였다. 한편, 토착미생물군인 UB-1의 경우 기존 생물능력향상 미생물군과 거의 동일한 처리효율을 나타내고 있으며, 사전에 순양기간을 거친 UB-2의 경우도 기존 생물능력향상 미생물군과 비슷한 처리효율을 보였다. 따라서 본 결과로 볼 때 생물능력향상 미생물군 투입에 의한 처리효율의 향상은 크지 않으며, 하·폐수의 생물학적 처리공정에 적요하는데는 다소 제한성이 있은 것으로 판단되었다.

Analysis on Practicality of Seed Treatments for Medicinal Plants Published in Korean Scientific Journals (국내 학술지에 발표된 약용작물 종자처리의 실용성 분석)

  • Presowing seed treatments used to enhance the rates of germination and afterward seedling emergence have not occasionally shown the same rate in indoor and field. The treatments considering germination mechanism and factors affecting germination must be totally included in indoor experiments so that the results drawn can be reproduced in the field. Seed germination is controlled by Phytochrome-mediated action changed with composition rates of red and far-red lights. Sunlight can penetrate soil into $6{\sim}9\;mm$ depth, which in turn means that seeds having $2{\sim}3\;mm$ in their width may receive the light if soil was covered 3 times over them. The penetrating light, moreover, turns to more far-red light than red light reverse to the sunlight. For germination tests after the artificial presowing seed treatments, therefore, seeds of smaller than 2 mm (< 2 mm), $2{\sim}3\;mm$, and larger than 3 mm (> 3 mm) must be done with incandescent lamp (IL) having more far-red light, with IL or in darkness, and in darkness, respectively. The 96 papers published in 13 Korean scientific journals up to the end of 2003 were analysed on the basis of the above explanation. 91 species were used 147 times as experimental materials; 101 times for < 2 mm seeds, 24 times for $2{\sim}3\;mm$ seeds and 22 times for > 3 mm seeds. If they were analysed as the light sources used for germination tests, correct applications reached more and less than 60% in both $2{\sim}3\;mm$ and > 3 mm seeds but 23% in < 2 mm seeds, conclusionally meaning that when the experimental results in the scientific papers were applied into farming practices, care was taken of their application because most of medicinal plant seeds were very small.

Development of Isolation Process of Barley Starch Using $\beta$-glucanase ($\beta$-Glucanase를 이용한 보리전분 분리공정의 개발)

  • For the development of technique for isolation of naked barley starch from Youngsan variety, optimum conditions of the isolation process were investigated. The effect of blending was examined and the results showed that 29.7% starch yield was obtained by 6 times of blending. After the blending, the barley starch contained 3.2% protein, 0.7% fat, 0.4% fiber, 0.4% ash and 2.8% ${eta}$-glucan. The opitmum conditions of ${eta}$-glucanase treatment were studied and the results showed that the amount of ${eta}$-glucanase and barley flour-water ratio were 60,000 unit and 1/2, the optimum steeping temperature, pH were $45^{\circ}C$ and 6.5, respectively. The effect of alkali treatment which would be supposed to increase the yield and purity of the barley starch was also examined. 76.7% starch content was obtained by 2 hr of alkali treatment. After all the treatment of isolation process, the barley starch finally contained 0.2% protein and 0.1% ${eta}$-glucan.
