  • DNA 회복효소인 MPG는 DNA의 퓨린기에 결합되어 있는 메틸기 등 이물질을 염기와 함께 제거하는 작용을 한다. 본 연구에서는 노던 블롯팅 방법을 이용하여 Balb/c mice의 각 조직별로 발생단계별 mRNA 발현 정도를 조사하였다. 뇌와 콩팥조직에서는 출생직후에 발현이 가장 활발하였으며, 성체시기까지 비교적 높은 활성도가 유지되었다. 위장 조직에서는 출생직후에서 일주일 후까지는 명확히 관찰되었으나, 그 이후는 발현이 약화되었다. 간장과 폐조직에서는 그 발현 정도가 매우 약했으며, 특히, 간조직의 경우 출생 직후보다 성체에서 그 발현이 현저히 감소되었다. 이들 조직에서의 활성도는 출생후 24시간 이내에서 1주일후까지 상대적으로 높게 유지되다가 점차 감소되었다. 즉, 수유기(출생직후부터 1주일후)에는 그 활성도가 성체시기(4주에서 6개월)보다 높게 유지되었다. 이러한 결과들로 미루어 보아 늙은 생쥐가 젊고 어린 생쥐보다 alkylating mutagen들에 노출되었을대 암에 걸릴 위험성이 높다고 생각된다.

Effect of Shading and Supplemental Lighting for Greenhouse Cultivation of Cucumber in Summer Season (하절기 오이 온실재배 시 차광 및 보광 효과)

  • High solar radiation in summer season causes excessive respiration of crops and reduces photosynthesis. In addition, the rainy season, which mainly occurs in summer, causes a low light condition inside the greenhouse. A low light condition can reduce crop growth and yield. This study was conducted to evaluate the effect of shade and supplemental lighting on the growth and yield of cucumber during summer season. Cucumber grafted seedlings were transplanted in two plastic greenhouses on August 30, 2022. To reduce the light intensity inside the greenhouse, a 50% shading screen was installed in one greenhouse. Supplemental lighting was conducted from September 7, 2022 to October 20, 2022. HPS (high-pressure sodium lamp), W LED (white LED, red:green:blue = 5:3:2), and RB LED (combined red and blue LED, red:blue = 7:3) were used for supplemental lighting sources, and non-treated (nonsupplemental lighting) was as the control. The supplemental lighting was conducted before sunrise and after sunset for 2 hours with a photosynthetic photon flux density of 150 ± 20 µmol·m-2·s-1. The plant height, leaf length, leaf width, and SPAD value tended to increase in the shading group. RB LED increased stem diameter regardless of shading treatment. Fresh and dry weights of fruits were not significantly different in shading and supplemental lighting. Average fresh weight of fruits was not significantly different among supplemental lighting as the harvest date passed. In conclusion, in this study 50% shade treatment significantly improved the growth of cucumber during the summer season. In addition, the growth and fruit characteristics are better than the control without supplemental lighting. This study can be used as basic research data for applying supplemental lighting technology to cucumber cultivation.

Development of CFD Model for Estimation of Cooling Effect of Fog Cooling System in Greenhouse (온실 포그냉방시스템의 냉방효과 예측을 위한 CFD 모델의 개발)

  • This study was carried out not only to develop CFD model for numerically simulating fog cooling system but also to verify the validity of the developed model by data measured in fag cooling greenhouse. In addition the developed model was applied to investigate the effects of spraying water temperature, spraying water amount, spraying interval and evaporation percentage on the performance of the fog cooling system. According to the simulation results, the temperature differences between the measured and predicted temperatures at each measurement point were $0.1~1.4^{\circ}C$ in case of no shading and $0.2~2.3^{\circ}C$ in close of shading. The humidity differences were 0.3~6.0% and 0.7~10.6%, respectively in the cases of no shading and shading. Because the predicted data showed a good agreement with the measured ones, the developed model is supposed to be able to predict the cooling effect of the fog cooling system. The performance of fog cooling system was greatly influenced by spraying water amount, spraying interval and evaporation percentage, but it was not influenced by spraying water temperature.

Growth of Seedling and Germination Characteristics of Acanthopanax koreanum NAKAI (섬오갈피나무의 발아 및 유묘의 생육특성)

  • This experiment was conducted to investigate the propagation for mass production of Acanthopanax koreanum NAKAI utilized for health food and medicine material. Germination rate of A. koreanum seeds were 64% when seeds were stored at 15 for 60 days and then were treated with 50 ppm of kinetin and dormancy were broken at 5 for 60 days. Rooting rates of green-wood cuttings treated with IBA 100 ppm, NAA 50 ppm, and IAA 100 ppm were 61.7, 56.7, and 60.0%, respectively. Rooting rates of greenwood cuttings treated with Rooton in scoria + horticulture media and volcanic ash were 76.7 and 66.7%, respectively. Survival rate of seedlings planted on Aug. 10 was highest (96%) under 75% shading net while shoot growth was best under 55% shading net. Survival rate of seedlings planted on May 2 was highest (91%) 55% shading net, and shoot growth was also best 55% under shading net.

  • 정향풀[Amsonia ellipotica (Thumb.) Roem. & Schult.]은 5월에 백색의 취산꽃차례로 개화하여 관상가치가 높다고 판단되나, 전국에 자생지가 10곳 미만이며 한국적색목록에 위기종(EN)으로 평가되어 체계적인 번식 및 재배법 개발이 요구된다. 본 연구는 유전자원보전 및 신 관상소재로 개발 가능한 정향풀의 효과적인 육묘법을 확립하고자 수행되었다. 실험재료는 유용식물증식센터에서 분양받은 종자를 사용하였으며, 파종용기(72, 105 및 128구 트레이), 파종립수(셀 당 1, 2 및 4립), 토양종류(원예상토 단용 및 중화 피트모스와 펄라이트를 3:1 또는 4:1로 혼용), Hyponex[미분(6.5-6-19)] 추비농도(0, 250, 500 및 $1000mg{\cdot}L^{-1}$) 및 차광정도(0, 55 및 75%)를 달리하여 2019년 4월 20일부터 8주 동안 육묘하였다. 연구의 공통사항으로 종자를 $500mg{\cdot}L^{-1}$ $GA_3$에24시간 침지한 다음 원예상토를 충진한 105구 트레이에 셀 당 1립씩 파종하였으며, 이후 2, 4주차에 $500mg{\cdot}L^{-1}$ Hyponex를 엽면시비하였다. 연구의 결과, 파종용기는 셀 당 용적량이 많을수록 초장, 경직경, 엽수, 지상부 및 지하부 생체중이 증가하는 경향이었다. 파종립수에 따른 생육은 1립 파종 시 가장 우수하였으나, 2립 파종한 묘의 개체 당 생체중이 비교적 양호함에 따라 트레이 상 풍성한 볼륨감을 나타내었다. 추비처리의 농도가 증가할수록 유묘의 생육이 증가하는 경향이었으며, $1000mg{\cdot}L^{-1}$ 처리구에서 초장, 경직경, 엽수 및 생체중이 유의적으로 높았다. 한편 차광처리 시 차광정도에 관계없이 경직경, 엽수, 마디수 및 지상부 생체중이 유의적으로 감소하였다. 따라서 정향풀은 용적량이 큰 72구 트레이에 원예상토를 충진한 다음 셀 당 2립씩 파종하여 차광하지 않은 양지에서 Hyponex $1000mg{\cdot}L^{-1}$로 추비하는 것이 관상목적으로 육묘하기 위한 효과적인 재배법으로 판단된다.

옥천대 흑색 점판암의 납 동위원소 연대

  • 우리는 괴산 덕평리 지역의 소위 구룡산층과 대전 추부 지역의 창리층 흑색 점판암에 대한 납 동위원소 연대측정 결과를 보고한다. 덕평리 지역의 흑색 점판암은 270 Ma 내외의 Pb-Pb 연대를 보이고 U-Pb 연대는 정의되지 않는다. 그 Pb-Pb 연대는 같은 시료의 22개 uraninite 입자에 대한 CHIME 연대와 오차범위 내에서 일치한다. 이로 보아 uraninite는 형성 또는 변성작용에 의한 동위원소적 재평형 작용 이후 폐쇄계를 잘 유지하였지만 흑색 점판암이 지질학적으로 최근에 지표에 노출된 이후에는 전암 규모에서 개방계로 거동하였음을 알 수 있다. 박편 미조직 관찰에 의하면 흑색 점판암의 1차광물인 uraninite 외에 풍화기원 2차광물인 uranocircite, francevillite가 관찰된다. 덕평리 지역 흑색 점판암의 최고 변성온도 조건은 50$0^{\circ}C$ 내외이므로 (Kim et al., 2000) uraninite CHIME 연대의 폐쇄온도가 50$0^{\circ}C$ 이상이거나 uraninite의 형성시기와 변성시기 사이에 시간차가 거의 없었다고 판단된다. 덕평리 지역의 U 광화작용 시기는 이번 자료에 의해 고생대 말로 정의될 수 있으나 그 연대가 흑색 점판암의 모물질인 해저 흑색 유기질 퇴적물의 초기 속성작용과 관련 있는지 후기의 변성작용과 관련 있는지에 대해서는 광물학적인 연구가 더 진행되어야 한다. 옥천대 변성퇴적암의 일부가 고생대 말에 퇴적되었을 가능성은 황강리층 역의 xenotime 및 monazite에 대한 CHIME 연대측정 결과 (약 367 Ma; Adachi et al., 1996)에 의해서 지지된다. 추부 지역 흑색 점판암의 Pb-Pb 연대는 170 Ma 내외로서 인접한 쥬라기 화강암의 관입시기를 지시하는 것으로 생각된다. 이는 화강암체로부터의 거리로 볼 때 덕평리 지역과 추부 지역의 시료 채취 위치가 유사하지만 지하 천부에 관입한 백악기 속리산 화강암 (91$\pm$6 Ma, Cheong and Chang, 1997)에 의해서는 덕평리 지역 흑색 점판암의 납 동위원소계가 영향받지 않았다는 점과 대조적이다.

A Study on the Double-Wall Greenhouse Filled with Styrene Pellets (입자충전형 이중벽 온실에 관한 연구)

  • This study was conducted to develope the automatic insulation system which control inside temperature of the greenhouse. For this purpose, the double- wall greenhouse and system which could automatically supply and discharge styrene pellets were constructed and abrasion of the pellets, blower ability, insulating property, transmittance and shading effect were analyzed by the experiments. The results obtained from this study can be summarized as follows : 1. It took an hour and fifteen minutes to supply and discharge about 2㎥ pellets in the experimental greenhouse. However, it is possible to reduce the operation time by proper selection of the blower and exhaust port, and by proper control of the supply and return pipe. 2. It was founded that the indirect delivery way was more profitable than the direct one in the supply and return of pellets. 3. When the transmittance was measured between 10 a.m. and 3 p.m., the average light transmissivity rate was 67%. 4. In winter nighttime, the inside temperature of the double- wall greenhouse with out the pellets was higher than the outside temperature by 3.4$^{\circ}C$ on an average. However, the inside temperature of the double - wall greenhouse with insulated area 73% was higher than the outside by one 6.6$^{\circ}C$ on an average, and the inside temperature of the greenhouse with insulated area 100% was higher than outside one by 13.5$^{\circ}C$ on an average. Therefore, it was proved that the insulating ability of the double - wall greenhouse in nighttime was excellent. 5. When the outside temperature was 36.9$^{\circ}C$ on an average, the inside temperature of the double- wail greenhouse with insulated area 100% was 3$0^{\circ}C$ on an average. As the inside temperature was lower than the outside one by 7$^{\circ}C$ on an average, we could know that the shading effects of the double- wall greenhouse were excellent in summer daytime.

  • 광은 식물의 광합성 작용뿐만 아니라 조직이나 기관의 분화, 종자의 발달 등 식물의 형태형성에도 관여하는 중요한 요인 중 하나이다. 불량한 광환경조건 하에서는 작물의 생육부진 뿐만 아니라 다양한 생리장해 및 병발생 등을 유발하기 때문에 경제적으로 광효율을 증가시키기 위하여 적절한 시설구조를 채택해야 한다. 또한 작물의 생육과 품질을 향상시키기 위하여 보광 및 차광 등 광환경 관리기술 개발이 요구된다. 본 연구에서 도입한 적외선램프는 새로운 인공광원으로서 태양광과 80% 정도 유사하고 용도에 따라 근적외선, 중적외선 및 원적외선이 방사되도록 할 수 있으며, 적은 전력소모량과 발생되는 열원은 겨울철 온풍난방기를 보조하거나 대체할 수 있어 에너지 절감효과를 동시에 기대할 수 있다. 따라서 본 연구는 인공광원에 대한 기초연구로서 인공광원의 종류별 분광특성을 비교 분석하고, 일몰 후 적외선램프의 조사 시간이 배추와 상추 유묘의 생장에 미치는 영향을 구명하고자 실시하였다.

The Cooling Effect of Fog Cooling System as Affected by Air Exchange Rate in Natural Ventilation Greenhouse (자연환기 온실의 환기회수에 따른 포그냉방시스템의 냉방효과)

  • The cooling effect of a fog cooling system has a close relationship to air flow and relative humidity in the greenhouse. From the VETH chart for cooling design, a cooling efficiency can be improved by means of increasing the air exchange rate and the amount of sprayed water. In the no shading experimental greenhouse by time control, when average air exchange rate was 0.77 times.min$^{-1}$ and spray water amount was 2,009g, inside temperature of the greenhouse was 31$^{\circ}C$ that was almost close to outside temperature and cooling efficiency was 82%. When average air exchange rate was close to temperature of the greenhouse that was no cooling and 70% shading greenhouse environment. When average air exchange rate was 2.59times.min$^{-1}$ , spray water amount was 2,009g and shading rate was 70%, inside relative humidity of the greenhouse was increased was 2,009 g and shading rate was 70%, inside relative humidity of the greenhouse was increased, but temperature was not decreased. When average air exchange rate was 2.33 times.min$^{-1}$ and spray water amount was 2,009g, inside temperature was 31.4 and at that time maximum wind speed at the air inlet of greenhouse was 1.9m.s$^{-1}$ . Since time controller sprayed amount of constant water at a given interval, some of sprayed water remained not to be evaporated, which increased relative humidity and decreased cooling efficiency. Because the shading screen prevented air flow in the greenhouse, it also caused the evaporation efficiency to be decreased. In order to increase cooling efficiency, it was necessary to study on controling by relative humidity and air circulation in the greenhouse.

Extraction Efficiency and Stability of Anthocyanin Pigments in Black Soybean Seed Coat (검정콩 종피 안토시아닌의 적정 추출 효율 및 안정성)

  • This study was carried out to develop the methods for efficient extracting and stabilization of anthocyanin pigments in black colored soybean. The optimum solvent concentration for the highest pigment extraction from seed coat of black colored soybean was 60% MeOH with 0.1% HCl in laboratory test. The higher temperature of the solvent until $60^{\circ}C$ led to better extraction efficiency of anthocyanin. The anthocyanin extracts maintained almost its stability in strong acidity area whereas it destroyed markedly over pH 4. Also anthocyanin extracts was stable for a while under weak light condition, but it showed rapid degradation of pigment color and markedly decreased HPLC value for anthocyanin content after one month storage days. In case storage temperature and instrument, glass vessel on low temperature ($4^{\circ}C$) was good storage condition among some treatments. And C3G was more stable than other pigments.
