Effects of pig manure composts with different composting periods on feeding rate, biomass and cocoon production of earthworm(Eisenia fetida) (돈분 퇴비의 부숙기간이 줄지렁이의 섭식률, 생체량 및 산란율에 미치는 영향)

  • Effects of pig manure with various composting periods(0, 15, 30, 50, 80 days) on feeding rate, biomass, cocoon production of earthworm(Eisenia fetida) population and physicochemical property of vermicast produced from pig manure were investigated. The feeding rate of earthworm was increased with longer composting period of pig manure. But the biomass production of earthworm population was highest upon the pig manure composted for 30 days. Upon the pig manure composted for 80 days, the biomass was severely reduced. Cocoon production was decreased with longer composting period and especially lower on the pig manure composted for 80 days. Values of pH, EC, C/N ratio of vermicasts produced from pig manure composts were lower than those of pig manures. And the organic material contents of vermicasts were uniformly reduced irrespective of composting duration of pig manure, whose values were 35.9-39.8%. From these results, the optimum composting period of pig manure for vermicomposting could be 15-30 days. And the application of vermicomposting upon the composted pig manure could be an efficient way for the treatment of pig feces, which can stabilize and recycle the organic wastes more rapidly than the conventional composting method.

Influence of Artificial Rainfall on Wheat Grain Quality During Ripening by Using the Speed-breeding System (세대단축시스템을 이용한 국내 밀 품종의 등숙기 강우에 의한 품질변이 평가)

  • Wheat (Triticum aestivum L.) is an important crop in Korea, with a per capita consumption of 31.6 kg in 2019. In the southern region, wheat is grown after paddy rice, and it is harvested during the rainy season in mid-June. This timing, in combination with high humidity and untimely rainfall, activates the enzyme alpha-amylase, which breaks down starch in the wheat grains. As a result, sprouted grains have lower quality and value for flour. However, seeds that absorb water before sprouting are expected to maintain better quality. The aim of the study was to identify the critical period during wheat maturation when rainfall has the greatest impact on grain quality, to prevent price declines due to quality deterioration. Two wheat cultivars, Jokyoung and Hwanggeumal, were grown in a speed breeding room, and artificial rainfall was applied at different times after heading (30, 35, 40, 45, 50, and 55 days). The proportion of vitreous grains decreased from 40 to 55 days after heading (DAH). Both cultivars had chalky grain sections from 35 DAH, with Hwanggeumal having a higher proportion of vitreous grains. Starch degradation was observed using FE-SEM (Field Emission Scanning Electron Microscope) at 40 DAH for Jokyoung and 50 DAH for Hwanggeumal. Color measurements indicated increased L and E values from 40 DAH, with rain treatment at 55 DAH leading to a significant increase in L values for both cultivars. Ash content increased at 45 DAH, whereas SDSS decreased at 35 DAH. Overall, grain quality from 40 DAH until harvest was found to be affected to the greatest extent by direct exposure of the spikes to moisture. Red wheat showed better quality than white wheat. These findings have implications for the cultivation of high-quality wheat and can guide future research efforts in this area.

  • In this study, enzymatic saccharification of substrates from Alnus hirsuta Ruper (8-14 years). Quercus acutissima Carruthers, Betula platyphylla var. japonica Nera, Populus euramericana Guiner and Platanus orientalis L. were investigated using crude cellulase preparations of Trichoderma viride Pers. ex. Fr. SANK 16374, and conduced on the optimum treated conditions of the cellulase sacchrification and reactivation of residue of digested substrates. The Trichoderma viride cellulase was produced by the submerged culture process and produced in the culture fluid was salted out quantitatively by the use of ammonium sulfate. The method of dilignification from wood (5 species) was treated by the peracetic acid(PA) method. The reducing sugar was determined by the dinitrosalicylic acid (DNS) method. 1. The results of tests carried out for 96 hr. (Figure 1), show conclusively the initial substrates from 5 species ($S_3$) which has been rendered highly reactive form and the mean rate of reducing sugar was 28.3 %. 2. The results of tests carried out for 96 hr., the reactivation of residue of digested substrates (improvement in the quality of the substrate through preheating in air at $190^{\circ}C$. for 45 min. followed by milling was (60 mesh size) at the same substrate level, increased concentrations of cellulase at the same substrate level, and increased concentrations of cellulase increases the rate of hydrolysis considerably. 3. Figure 1. shows conclusively that the residue of digested substrates ($S_1$ dried at $60^{\circ}C$) which has been rendered extremly resistant to cellulase action can be reactivated into a highly reactive form ($S_2$), almost comparable to that of the initial substrates ($S_3$). And the reducing sugar formation did not show statistically significent differences at 5% levels by initial substrates and the residue of digested substrates (preheating in air at $190^{\circ}C$. for 45 min. fallowed by milling was (60 mesh size).

  • Panax ginseng C.A. Meyer (P. ginseng) is known to exert a wide range of pharmacological effects both in vitro and in vivo. Although studies on ginsenoside, antioxidant activity, and anticancer effect of the cultivated wild Panax ginseng (CWP) have been conducted, there is little research on the effect of CWP extract on bone metabolism. In this study, we investigated the potential anti-osteoporotic properties of CWP on the growth and differentiation of MC3T3-E1 cells. CWP significantly increased the viability and proliferation of MC3T3-E1 cells. CWP activated intracellular alkaline phosphatase (ALP) activity in MC3T3-E1 cells. In addition, CWP increased the mineralized nodules in MC3T3-E1 cells. Furthermore, CWP increased the expression of genes such as Runx2, ALP, OPN and OCN associated with osteoblast growth and differentiation in a dose-dependent manner.

The Bacterial Community Structure in Biofilms of the RABC Process for Swine Butchery Wastewater Treatment (돼지 도축폐수 처리를 위한 RABC 공정의 생물막 세균군집 구조)

  • Culture-independent microscopic observations and 16S rDNA analyses were applied to describe the bacterial community inherent to the biofilm structure of the RABC (Rotating Activated Bacillus Contactors) process for swine butchery wastewater treatment. The ratios of Gram-positive bacterial counts to total bacterial counts of the RABC process were significantly increased in the last aeration tank as well as returned sludge, while those of the existing A2O (Anaerobic-Anoxic-Oxic) process maintained constant from aeration tanks to returned sludge. Totally nine phyla were recovered by 16S rDNA analysis, two of which were major groups: the Proteobacteria (64.1%) and the Actinobacteria (18.4%). The third major group was the endospore-forming Firmicutes (5.4%). The remaining six minor groups are the Bacteroidetes (3.3%), the Chlorobi (2.2%), the Nitrospirae (1.1%), the Chlorofleix (1.1%), the Acidobacteria (1.1%), and the Fusobacteria (1.1%). The ratio of endospore-forming bacteria was 19.4%, which was composed of the members of the Firmicutes phylum (5.4%) and the Intrasporangiaceae family (14.0%) of the Actinobacteria phylum. Nitrifying and denitrifying related- and phosphorus accumulating related-sequences were composed of 6.5% and 5.4% of total community, respectively, these could mean the high capacity of the RABC process to remove odor compounds and reduce eutrophication by efficient removing inorganic nutrients.

Study on Sludge Reduction by Sludge Solubilization and Change of Operation Conditions of Sewage Treatment Process (하수슬러지 가용화와 하수처리 운전조건 개선을 통한 하수슬러지 발생저감 연구)

  • In order to find the way to solve the problem of sewage sludge discharge into the ocean, the sludge solubilization by ultrasonic and the improvement methods of wastewater treatment process were studied. In the membrane bioreactor the sludge retention time was stepwise increased from 5.1 day to 442 days where the biomass average concentration has been increased from $c_B$=3.4 $gTSSL^{-1}$ to $c_B$=14.5 $gTSSL^{-1}$ respectively. At the same time, the biomass yield coefficients were reduced from 0.5-0.7 at SRT=5.1 day to 0.005-0.007 at SRT=442 days which means the reduction of sludge production. Oxygen mass transfer coefficients and ${\alpha}$-factor were investigated with changing stirrer speed to find the relation between the high biomass concentration and aeration efficiency in the propeller loop reactor. As a result of sludge solubilization, the solubilization of sludge by ultrasound was increased with increasing energy input and it led to improved anaerobic digestion rate with more biogas production than that of nonsolubilized sewage sludge.

EFFECT ON THE OSTEOGENESIS OF DENTAL IMPLANTS AFTER HORIZONTAL DISTRACTION OSTEOGENESIS WITH NITRIFIED DISTRACTOR (질화 표면처리된 신장기를 이용한 수평적 치조골신장술 후 임플란트 식립시 골유착 효과에 관한 연구)

  • This study evaluated whether there is a difference between a conventional titanium distraction device and a TiN-coated device in terms of implant osseointegration after horizontal distraction osteogenesis in narrow alveolar bone. Four adult mongrel dogs, each weighing $9{\sim}10kg$, were used in this study. The lower premolars were extracted and horizontal distraction was performed with conventional titanium distraction devices (group 1) or TiN-coated devices (group 2). After an 8-week consolidation period, the implants were installed, and the dogs were sacrificed after another 8 weeks. The osseointegration around the implants was evaluated histomorphologically. There was one failure in experimental group 1 because of fracture of the device. Direct bone contact was achieved and there were no significant differences between the control group and experimental groups 1 and 2 in terms of osseointegration around the implants at 8 weeks. In summary, intraoral distraction osteogenesis is a good option for augmenting severely atrophied alveolar ridges, and the TiN-coated device may be useful because of its advantages, which include surface hardness, corrosion resistance, and reduced bacteria.

음극 아크 증착으로 코팅된 TiAlN 박막의 특성연구

  • 본 연구에서는 아크 소스를 이용하여 TiAlN을 코팅하였으며 공정 변수 중 질소 유량에 다른 TiAlN 박막의 물성 변화를 관찰하였다. TiAlN은 고경도 난삭재의 고능률 절삭 분야에 사용되어 공구의 수명을 향상하기 위한 표면처리 소재로 각광을 받고 있다. TiAlN 박막은 아크 소스에 장착된 TiAlN 타겟(Ti-50 at %Al)을 사용하여 스테인리스 강판 위에 코팅 하였으며 이 때 기판과 타겟 간의 거리는 약 30 cm이었다. 기판을 진공용기에 장착하고 ${\sim}10^{-6}$ torr까지 진공배기를 실시한 후 아르곤 가스를 진공용기 내로 공급하여 공정 압력인 $7{\times}10^{-4}$ torr로 제어한다. 공정 압력에서 아크 소스에 약 70 A의 전류를 인가하여 아크를 발생시키고 기판 홀더에 약 -400 V의 직류전압을 인가하여 약 5분간 청정을 실시하였다. 기판의 청정이 끝나면 기판에 인가된 전압을 차단하고 질소 가스를 진공용기에 공급하여 TiAlN을 코팅하였다. 질소 유량이 30 sccm일 경우 TiAlN 박막의 경도가 약 2510 Hv로 가장 높았으며, 질소의 유량이 40 sccm 이상으로 증가할 경우 TiAlN 박막의 경도는 1500 Hv로 주목할 만한 변화는 없었다. 질소 유량이 증가하면 TiAlN 박막의 색상은 회색에서 어두운 보라색으로 변화하였고 주사전자현미경 분석을 통해서 거대 입자(macro particle)가 감소하는 경향을 확인할 수 있었으며 이는 질소 유량이 증가할수록 TiAlN 박막의 표면조도 또한 증가하는 분석결과와 일치하였다. X-선 회절 분석을 통해 질소 유량이 30 sccm 이상에서 박막의 질화가 일어나고 2500 Hv 이상의 경도를 가지는 최적 조건임을 확인하였으며, 이는 절삭 공구 등과 같이 고경도 유지를 위한 코팅 분야에 적용이 가능할 것으로 판단된다. 본 연구에서 얻어진 결과를 바탕으로 질소 유량 외에 다른 공정 조건을 변화시켜 TiAlN 코팅을 실시한다면 다양한 색상 구현, 고경도, 내마모성 등 TiAlN 박막의 기능성을 향상할 수 있을 것으로 예상된다.

Effect of $CO_2$ Content on the Growth and Corrosion Characteristics of the Compound Layers in Gaseous Nitrocarburized Carbon Steels (가스 질화침탄처리한 탄소강의 화합물층 성장 및 부식특성에 미치는 $CO_2$함량의 영향)

  • This study has been performed to investigate the effect of $CO_2$ content on the growth characteristics of the compound layer, porous layer and corrosion characteristics of carbon steels after gaseous nitrocarburizing in $70%-NH_3-CO_2-N_2$ at $580^{\circ}C$ for 2.5 hrs. The results obtained from the experiment were the thickness of the compound and porous layers increased with increasing $CO_2$ contents. At the same fixed gas composition the thickness of the compound and porous layer increased with increasing carbon content of the specimens. X-ray diffraction analysis showed that compound layer was mainly consisted of ${\varepsilon}-Fe_{2-3}(N,C)$ and ${\gamma}^{\prime}-Fe_4N$ as the increased with $CO_2$ contents in atmosphere, compound layer was chiefly consisted of ${\varepsilon}-Fe_{2-3}(N,C)$ phase. With increasing $CO_2$ content and total flow rate in gaseous nitrocarburizing, the amount of ${\varepsilon}-Fe_{2-3}(N,C)$ phase in the compound layer was increased. The current density of passivity decreased with increasing $CO_2$ content due to the development of porous layer at the out most surface of ${\varepsilon}-Fe_{2-3}(N,C)$.

A Study on the High Temperature Gas Nitriding and Tempering in STD11 steel (STD11 고속도 공구강의 고온 가스질화 처리 및 템퍼링에 관한 연구)

  • The effects of the high temperature gas nitriding (HTGN), tempering and subzero treatment of STD11 steel have been investigated. HTGN treatment was carried out at $1050^{\circ}C$, $1100^{\circ}C$ and $1150^{\circ}C$ for 1 hr. in an atmosphere of $1\;kg/cm^2$ nitrogen gas. Tempering and double-tempering were performed at $550^{\circ}C$ for 1 hr. The surface layer of HTGN-treated steel appeared the precipitates of $M_2N$, $M_7C_3$ and $M_{23}C_6$ in the matrix of austenite. However, the interior region exhibited martensite with the precipitation of carbides. The nitrogen content of the surface layer appeared ~1.35 wt.%, ~0.83 wt.% and ~0.56 wt.% at the HTGN treatment temperature of $1050^{\circ}C$, $1100^{\circ}C$ and $1150^{\circ}C$, respectively. The surface hardness of double-tempered and subzero-treated steel measured the maximum value of 828 Hv, 960 Hv, 750 HV after HTGN treatment at the $1050^{\circ}C$, $1100^{\circ}C$ and $1150^{\circ}C$, respectively. These hardness value increased above 230~420 Hv compared with the HTGN-treated steel due to the decrease in retained austenite and existence of fine precipitates.
