The Effect of External Carbon Sources on Batch Denitrification Process. (회분식 탈질 공정에서 외부 탄소원에 따른 탈질효과)

  • Respiratory denitrification experiments were performed using different carbon sources (acetic acid, glucose, methanol, molasses). In the culture media with glucose and molasses, COD consumption and denitrification rates were higher than with acetic acid and methanol. However, up to 30-40% of reduced nitrate and nitrite were converted to ammonium in glucose and molasses media. In the culture media with acetic acid and methanol, ammonium was not accumulated. Some of the consumed COD seemed to be used by the acid formers for the acidification in glucose and molasses media. By initial pH control of with molasses media, higher denitrification rate (up to 99%) and faster response could be obtained.

  • An obligatory methanotroph Methylosinus trichosporium OB3b was cultivated for the production of poly-${\beta}$-hydroxybutyric acid(PHB) in shake-flask using liquefied natural gas(LNG) as the sole source of carbon and energy. The maximal specific growth rate decreased by 40% using LNG compared with that obtained with pure methane. This is attributed to the inhibition by ethane and propane presents in the LNG as impurities. For the production of PHB, two-stage culture separating the production stage from the growth stage was carried out. PHB accumulation was observed after switching nutrient-sufficient to nutrient-limited condition of non-carboneous component (NO3-, PO43-, K+, Na+, Fe2+, or Mg2+). The limitation of K+ or Mg2+ resulted in relatively high PHB content, but the highest content was obtained by nitrate limitation. The optimal pH and temperature for PHB accumulation was 7.0 and $30^{\circ}C$. Under the optimal condition the maximal PHB content was about 45% after 4-day cultivation.

  • Effects of nitrogen and carbon sources on the decolorization rate of poly R-478 dye by a white rot basidiomycete Coriorus versicolor IFO 30388 were examined. The fungus exhibited 87.2% of decolorization rate when it was cultured in the state of stationary in a nitrogen-limited medium (pH 4.5) which contained 2.0% glucose, 0.04% ammonium tartrate, 0.02% poly R-478 dye, 2% $KH_2PO_4$, 0.5% $MgSO_4{\cdot}7H_2O$, 0.1% $CaCl_2{\cdot}2H_2O$, 0.002% thiamine-HCl and 10 mM 2,2 dimethylsuccinate (sodium) at $28^{\circ}C$ for 10 days. Decolorization of the dye occurred in the presence of nitrogen source in the medium and decolorization rate increased rapidly after depletion of $NH_4^+$ from the medium.

  • The effects of temperature. pH, carbon and nitrogen sources on the growth of Rhizoctonia solani were studied by using five hyphal anastomosis groups(four cultural types, 7 isolates) of the fungus. The ranges of optimum temperature were $20^{\circ}C$ in the AG 2-1, AG 2-2 and AG 4, and $25^{\circ}C$ in the AG 1-IA, AG 1-IB, AG 3, AG 5. The optimum pH for the mycelial growth was 6-7 in the fungus. Glucose in the AG 1-lA, AG 1-IB, AG 2-2, AG 3 and AG 5, sucrose in the AG 2-1 and fructose in the AG 4 were the most effective for the mycelial growth, but glycerine, cellulose and lactose were not effectively utilized as nutrients. $Ca(NO_3)_2$ in the AG 1-IA, AG 1-IB and AG 4, asparagine in the AG 2-1, $KNO_3$ in the AG 2-2 and $NaNO_3$ in the AG 5 were the best nitrogen sources for the mycelial growth, but $NH_4NO_3$ was not easily utilized by the fungus. Nitrate and organic nitrogens for the fungal growth were utilized better than ammonium nitrogen.

Influence of Organic, Inorganic Nitrogen Sources and Amino Acids on the Biosynthesis of Coenzyme $Q_{10}$ by Agrobacterium tumefaciens Mutant (Agrobacterium tumefaciens 변이주에 의한 Coenzyme $Q_{10}$ 생합성시 유기, 무기질소원과 아미노산의 영향)

  • The effect of inorganic, organic nitrogen sources and amino acids on the coenzyme $Q_{10}$ production and coenzyme $Q_{10}$ component ratio was investigated. Among the nine organic nitrogen sources, CSP showed a remarkable enhancing effect on the production of coenzyme $Q_{10}$. But this enhancement was not observed in medium containing Bacto peptone, tryptone, casamino acid and soybean meal. These differences on the production of coenzyme $Q_{10}$ may be due to differences in kinds and amounts of component amino acids and peptides in the various organic nitrogen sources tested. In the addition of inorganic nitrogens, only $(NH_4)_2SO_4$ increase the coenzyme $Q_{10}$ production by 2.0 times compare to the control group. The addition of L-tyrosine to the medium containing Bacto tryptone, was also determined to be crucial for the coenzyme $Q_{10}$ production. But phenylalanin and tryptophan, other aromatic amino acids, had no stimulatory effect on the coenzyme $Q_{10}$ production. These results show that the production and components ratio of coenzyme $Q_{10}$ was greatly affected by the kinds and the concentration of inorganic, organic nitrogen sources as well as amino acids.

  • Trichoderma inhamatum KSJ1, a filamentous fungus, isolated from rotten wood showed high ability to hydrolysis of cellulosic materials. Enzyme productivity by strain KSJ1 was high in the cultivation using carbon sources such as cellulosic materials and lignocellulosic wastes as rice straw and paper waste. In previous study peptone was one of optimum organic nitrogen sources in producing cellulases for saccharification of food wastes. However, it was too expensive using peptone as organic nitrogen source, so, in this study, soybean and yeast were applicated to substitute peptone. Yeast showed producing high enzyme activity, so it was estimated that yeast is available in producing cellulase using Trichoderma inhamatum KSJ1 at industrial Production.

  • It was observed that the effect of Grand E.C, on the growth of Fusarium oxysporium f. cucumerinum mycellia, which was cultured in the media with different nitrogen and carton sources, varied with it's concentrations. (1) Mycellia of F. oxysporium f. cucumerinum were grown strongly in $(NH_4)_2HPO_4\;NH_4NO_3$ and fructose media. (2) In general, the diluted Grand E.C, $ (\times\;1,000)$ effected on check of fungal growth in any type of media with the different nitrogen and carbon sources. It was disturbed the growth of fungi in $NH_4NO_3$, lactose and fructose in the diluted fungicide $(\times\;5,000)$. In the diluted solution $(\times\;10,000)$ of fungicide, somewhat increased the viability of fungi, but it was disturbed the growth in $NH_4NO_3$ and fructose media. (3) In conclusion, it was thought to be that the diluted Grand emulsion effected in check of the growth of F. oxysporium f. cucumerinum which grown some nutritional conditions.

  • For the purpose of optimizing poly-l3-hydroxybutyrate (PHB) production from Alcaligenes eutrophus, two-stage fed-batch culture was adopted. In this system, specifk growth rate was maximized during the first stage whereas specific production rate was maximized during the second stage. The optimal concentrations of glucose and ammonium chloride were 16.6 and 0.54 g/I in the growth stage and 20.0 and 0.07 g/l in the production stage, respectively. Proportional feedback control considering time lag was suggested for PHB production process and a simulator was developed for real-time control purpose.

  • 충북 단양에 위치한 조일 광산에서 채취한 구리와 아연으로 오염된 광미 (광산 폐기물로서 금속 추출 후 남은 찌꺼기)를 효율적으로 처리하기 위하여 생물학적 용출기법(bioleaching)의 효율 증진에 관한 연구를 수행하였다. 기본 배지 조성(9K medium) 중 미생물의 성장 및 증식, 용출 효율에 영향을 미치는 인자인 에너지원, 인, 질소원의 농도를 변화시키며 배지 조성에 따른 중금속의 용출 효율을 관찰하였다. 그 결과 인 농도 변화는 인을 첨가하지 않았을 때, 아연과 구리의 용출 효율이 각각 98.8%와 47.5%로 나타났고, 질소원은 45mM 농도로 주입하였을 때 아연, 구리 각각 85%와 46.4%의 용출 효율을 보였다. 에너지원 변화에 서는 아연의 경우 에너지원을 첨가하지 않았을 때 93%의 용출 효율을 나타냈고, 구리의 경우는 160mM 첨가했을 때 46.4%로 가장 높은 용출 효율을 나타냈다.

Effect of Ammonium Ion on the Production of a Polysaccharide, Methylan from Methanol by Mentylobacterium organophilum (Methylobacterium organophilum에 의한 메탄올로부터 메틸란의 생산에 대한 암모니아 이온의 영향)

  • The effect of nitrogen source on production of a high viscosity exopolysaccharide, methylan, from methanol by Mentylobacterium organophilum was investigated in fed-batch culture. During the fermentation, cells continued to grow even after the nitrogen source added to the medium was depleted and methylan production was stimulated under the condition which ammonium ion was depleted. Cell growth increased proportionally to the initial concentration of ammonium ion in the medium, but methylan production was significantly inhibited at the high concentration of ammonium ion. As the initial concentration of ammonium ion increased, the specific growth rate, the specific product formation rate and the specific substrate consumption rate decreased due to the inhibitory effect of excess ammonium ions. In order to reduce the inhibitory effect by high concentration of ammonium ion. The control of ammonium ion concentration within the desired level(usually $0.45g/\ell$) was necessary. When ammonium ion concentration was maintained below $0.15g/\ell$ by exponential feeding, methylan production could be increased up to $12.5g/\ell$.
