Nitrogen fractionation of organic materials applied to Korean ginseng (고려인삼(高麗人蔘) 유기질비료의 질소성장(窒素性狀)에 대하여)

  • 1. Nitrogenous compounds of-organic materials as nitrogen sources for Korean ginseng were characterized according to their solubility and chemical forms. 2. The extractable fractionation was as follows : Yakto group : non-extractable N > acid hydrolyzable N > acid nonhydrolyzable N > water sol. N, Litter group : acid hydrolyzable N > non-extractable N > water sol. N > acid non-hydrolyzable N, Bone meal : acid hydrolyzable N > water sol. N > acid non-hydrolyzable N. 3. Nitrogenous compounds in the water sol. fraction were : Yakto group and Litter group : humus N > amino acid N > nitrate N (recognized only in Yakto group) > ammonia N > hexosamine N > amide N, Bone meal : amino acid N > humus N > ammonia N > amide N. And nitrogenous compounds in the acid hydrolyzable fraction were : amino acid N > humus N ${\simeq}$ ammonia N > hexosamine N. 4. Availability was discussed about the major nitrogenous compounds (amino acid, humus and inorganic N) and the solubility.

Comparisons of Decomposition Rates, Carbon and Nitrogen Dynamics of Branches in Pinus densiflora and Quercus variabilis Stands (소나무와 굴참나무 임분의 가지 분해율과 탄소 및 질소 동태 비교)

  • This study was carried out to understand how factors of decomposition such as quality of the substrate (tree species), forest types, and soil·meteorological conditions affect decomposition rates and dynamics of carbon (C) and nitrogen (N) of branches in adjacent Pinus densiflora and Quercus variabilis stands in Sancheong and J inju in Southern Korea. The branch bags (30 cm × 30 cm) with branch samples (a diameter of 2.0 - 4.0 cm and length of 10 cm) were placed on the forest floor at each stand in May 2018. The branch bags were collected in August and November 2018, February and May 2019, respectively. The decomposition rates of branches in P. densiflora stands were 4.49 % for Sancheong, and 5.75 % for Jinju. Whereas, the decomposition rates in Q. variabilis were 20.01 % for Sancheong, and 24.68 % for Jinju, respectively. The decomposition of branches was more rapid in Q. variabilis compared with P. densiflora in both regions. C and N in decomposed branches were more mineralized in Q. variabilis, whereas C and N were more accumulated in P. densiflora. These results indicated that the decomposition rates, C and N of decomposed branches may be affected by differences in substrate quality such as initial N concentration and C/N ratio rather than differences between both regions including different environmental factors.

Chemical Compositions of Sewage Sludges and Nitrogen Mineralization in Sewage Sludge Applied Soil (하수오니의 화학적 조성과 토양중 질소 무기화)

  • Swage sludge cakes produced from domestic wastewater treatment plants were collected from 21 different sites throughout Korea, and chemical properties of the sludge samples were determined. Inorganic nutrient contents did not indicate great differences among swage sludges from each sites, whereas the toxic heavy metal contents differed greatly. T-N, $NH_4{^+}-N$ and $NO_3{^-}-N$ contents from 21 sites sludges ranged 2.3-6.0, 291-4284, $1.4-58.8mg\;kg^{-1}$, respectively. Heavy metal (Cd, Cu, Pb and Zn) contents ranged 2.86-58.22, 144.0-5417.3, N.D.-943.5, and $N.D.-8,083mg\;kg^{-1}$, respectively. One of the sludges was treated to soils at rates of 12.5, 25, 50. and 100, $Mg\;ha^{-1}$ and incubated for 12 weeks to determine nitrogen materialization rate. Ammoniun nitrogen content decreased sharply at higher rates of sludge treatment up to 8 weeks after treatment and did not change much, while $NO_3{^-}$ increased at all treatment levels. The net amount of mineralized N of sludge treatment rates (12.5, 25, 50, and $100Mg\;ha^{-1}$) during 12weeks incubation were 189.0, 277.2, 303.8 and $376.6mg\;kg^{-1}$.

Quality Properties of Fermented Tofu Prepared with Different Molds and Coagulants (곰팡이와 응고제에 따른 발효두부의 품질특성)

  • Changes of quality properties of fermented tofu prepared with two molds like Actinomucor elegans (AE) and Rhizopus oligosporus (RO) and coagulants (CaCl$_2$ and citric acid) were investigated. Moisture and crude protein of fermented tofu were rapidly decreased during fermentation, the contents of crude lipid and crude ash were shown to be slightly increased, ad then total acidity was slowly decreased. The content of reducing sugar of fermented tofu was slowly increased for 7 day of fermentation, but rapidly increased after that time because of rapid hydrolysis of carbohydrate in fermented tofu. The contents of amino and ammonia type nitrogen were quickly increased during fermentation. The highest contents of amino type nitrogen of fermented tofu were found in sample of CaCl$_2$group as a coagulant and RO group as a mold. Contents of minerals in tofu fermented for 14 day were high in order of K>Ca>Mg>Na. Iin conclusion, AE was more effective than RO to enhance the contents of reducing sugar and amino type nitrogen as an indicator of fermentation within 7 day of fermentation, and then RO was more effective than AE after that time. Calcium chloride as a coagulant was more effective than citric acid in tofu fermented with the same strain for 14 day.

  • Fermentation waste from beer production was applied to a newly developed upland soil to evaluate the effect of lime added sludge on corn growth. The mineralization of lime added sludge was faster than that of raw sludge without lime treatment in the upland condition. It was accelerated by low C/N ratio and high lime content in the lime added sludge. The plant growth and yields of corn increased as the sludge application rate increased and so was nitrogen and organic matter contents in soil. Ammonium volatilization is considered to be high in the lime added sludge and thus the raw sludge is more promising as an organic sources.

  • Cultural characteristics of a strain of Streptomyces sp. K-14 (KFCC 35051) producing glucose isomerase were demonstrated. The glucose isomerase was produced when the strain was grown in the medium containing pure xylan or xylan.containing materials such as wheat bran or com cob. The optimum condition was attained in a culture medium composed of 3 % wheat bran or com cob, 2 % com steep liquor, 0.1% $MgSO_4$$7H_2O$ and 0.012 % $CoSO_4$$7H_2O$ for the production of the glucose isomera,e. The production of the enzyme reached to a maximum level when the strain was cultured for 40 hrs $30^{\circ}C$ and pH 7.0.

  • This study was intended to find out the effects of the inorganic nutrients in the soil and root on the available constituents contents in Angelica gigas Nakai. The correlation between the contents of decursin and decursinol in roots of the wild and cultivated and the inorganic nutrients contents in the soils and roots was investigated. The contents of decursin in the dried root of the wild was higher than that of the cultivated. The ammonium nitrogen contents in soil and the decursin contents in root had the significant negative correlation, whereas the significant positive correlation was obtained the available phosphorus contents. The significant negative correlation was obtained between the decursin contents and the nitrogen contents of root, and the significant positive correlation between the decursin contents and the contents of phosphorus or potassium in roots.

  • Field microplot(D 20cm, L 85cm) experiment filled with Bonryang sandy loam soil(Typic Udifluvents) was conducted to obtain quantitative information on the change of soil pH under different soil moisture regime and ladino clover cultivation, and the movement of applied nutrient under the rainfed bare soil condition. Slaked lime increased soil pH of the applied layer. $NH_4-N$ evolved from urea hydrolysis was more effective in raising the subsoil pH rather than the exchangeable Mg and Ca. Under the different soil miosture condition downward movement of Cl and $NO_3-N$ lowered subsoil pH during the middle of ladino clover growth. Leaching of CI and $NO_3-N$ down to the out of the soil microplot and increasing movement of Ca and Mg into the subsoil produced. a slight increase of subsoil pH during the late crop growth. The mean downward movement of applied nutrient 5.5 months after field installation of microplot in the bare soil under rainfed condition(1207mm) was in the order : Cl= more than 80.0>Inorganic nitrogen= 75.8>K= 67.3>Mg=62.7>Ca=18.1>P=2.3cm respectively.

Antagonistic Mechanisms and Culture Conditions of Isolated Microbes Applied for Controlling Large Patch Disease in Zoysiagrass (한국잔디 갈색퍼짐병 방제를 위한 선발 미생물의 길항기작 및 배양조건)

  • Our previous report demonstrated successful isolation of soil-borne bacteria that suppressed the potential of Rhizoctonia solani AG2-2 (IV) causing turfgrass large patch disease when applied to Korean lawngrass (Zoysia japonica). The current study aimed to uncover the mechanisms of this antagonism of Rhizoctonia solani and to define culture conditions for the isolated microbes. We found that two Bacillus isolates, I-009 and FRIN-001-1 strains, produced cellulase and siderophore, but not chitinase, while the Pseudomonas YPIN-022 strain was found to release only siderophore, implying that three antagonistic bacteria commonly interrupt Fe uptake by the large patch pathogen. The I-009 and FRIN-001-1 isolates grew best at 35 and $30^{\circ}C$ in growth medium of pH 5 to 8 for 32 and 28 h, respectively, while optimum growth for the YPIN-022 strain was found at $35^{\circ}C$ at pH 5 to 9 for 24 h. Good growth of I-009 and YPIN-022 over 24 h was obtained in M9 minimal medium supplemented with 1% sucrose, 0.5% yeast extract and 0.1% potassium chloride. FRIN-001-1 grew well in M9 medium with 1% mannitol, 0.5% yeast extract and 0.1% potassium phosphate dibasic.

  • A laboratory experiment was performed to investigate the effects on redox potential of submerged soil by application of CNP herbicide (2, 4, 6-Trichlorophenyl-4-Nitrophenyl ether) with or without rice straw. Two soils, sandy loam and clay loam textured, were incubated for sixty days at a constant temperature, $25^{\circ}C$. Sampling and analysis of pH, Eh and nitrogen mineralization were carried out during the incubation. The results were summarized as follows. 1. The CNP application decreased redox potential and increased soil pH. The higher the concentration of applicated CNP was, the effects on soil Eh and pH were higher. When rice straw was used with CNP, the Eh of soil remarkably decreased, but seperating the individual effects of rice straw and CNP was impossible in this study. 2. Ammonification increased as the concentration of applied CNP increased regardless of rice straw application or not. It was higher in sandy loam soil than clay loam soil. 3. Nitrification decreased as the concentration of applied CNP increased, especially in the sandy loam soil. Rice straw application reduced nitrification.
