Fungal Microbial Community Profiles of Meju, Solar Salt, and Doenjang Using Pyrosequencing (Pyrosequencing을 이용한 메주, 천일염, 된장의 곰팡이 군집 분석)

  • In order to evaluate the migration of fungi into doenjang from its materials, meju and solar salt, microbial communities were analyzed using pyrosequencing. Dominant fungi of meju were Botrytis spp. (57.94%) and Dothiorella samentorum (24.08%). Unidentified fungal species (37.53%), unassigned species (32.60%) and several fungal species of small portion were identified in solar salt. In doenjang, Candida versatilis were predominantly detected (92.62%). Non-halophilic mold were dominantly identified from meju (low-salt fermented soybean), while halophilic bacteria and archaea for solar salt and salt-tolerance fungi such as C. versatilis for doenjang (high-salt fermented soybean) were frequently detected. These results implied that most predominant fungal species might not be migrated from meju and/or solar salt into doenjang.

  • 하천 합류부에 지천이 유입되는 경우 복잡한 3차원적 흐름 구조를 발생시키고 이로 인해 유사혼합 및 지형 변화가 활발히 발생하게 된다. 특히, 하천 합류부에서 부유사 거동은 하천의 세굴과퇴적, 하천 지형 변화, 하천 생태계, 하천구조물 안정성 등에 직접적으로 영향을 미치기 때문에 이에 대한 정확한 분석이 하천 관리 및 재해 예방에 필수적인 요소이다. 기존의 하천 합류부 부유사 계측 자료들은 재래식 채취 방식으로 수행되어 시공간적 해상도가 매우 낮아서 실측 자료만으로 합류부에서 부유사 혼합을 분석하기에는 한계가 존재하기에 대하천의 부유사 혼합 거동 해석에 수치모형이 주로 활용되어 왔다. 본 연구에서는 하천 합류부에서 부유사 거동을 공간적으로 정밀하게 분석하기 위해 드론 기반초분광 영상을 활용하여 하천 합류부에 최적화된 부유사 계측 방법론을 제시하였다. 현장에서 계측한 초분광 자료와 부유사 농도간의 관계를 구축하기 위하여 기계학습모형인 랜덤포레스트(Random Forest) 회귀 모형과 합류부에서 분광 특성이 다른 두 하천의 특성을 정확하게 반영하기 위한 가우시안 혼합 모형 (Gaussian Mixture Model) 기반 초분광 군집화 기법을 결합하였다. 본 연구에서 구축한 방법론을 낙동강과 황강의 합류부에 적용한 결과, 초분광 군집을 통해 두하천 흐름의 경계층을 명확히 구별하였으며, 이를 바탕으로 지류와 본류에 대해 각각 분리된 회귀 모형을 구축하여 복잡한 합류부 근역 경계층에서의 부유사 거동을 보다 정확하게 재현하였다. 또한 나아가서 재현된 고해상도의 부유사 공간분포를 바탕으로 경계층에서 강한 두 흐름이 혼합되어 발생한 와류(Wake)가 부유사 혼합에 미치는 영향을 규명하였고, 하천 합류부에서 발생하는 전단층의 수평방향 대규모 와류가 부유사 혼합 양상에 지배적 영향을 미치는 것으로 확인하였다.

Phylogenetic Characteristics of viable but Nonculturable Bacterial Populations in a Pine Mushroom (Tricholoma matsutake) Forest Soil (송이 자생군락 토양 내 난배양성 세균군집의 계통학적 특성)

  • The CFDA (6-carboxyfluorescein diacetate) direct viable count method and plate count (PC) method using conventional nutrient broth (NB) medium and $10^{-2}$ diluted NB (DNB) medium were applied to samples collected from Mt. Yongdoo In Andong, in an effect to determine the number of living bacteria pine mushroom forest soil. The number of living bacteria determined via plate count in NB medium comprised $5{\sim}8%$ of the CFDA direct viable count, and the bacteria in the DNB medium comprised $40{\sim}47%$. This result indicated that viable but nonculturable (VBNC) bacteria existed in the pine mushroom forest soil at a high percentage. The phylogenetic characteristics of the VBNC bacterial populations in the samples of pine mushroom (Tricholoma matsutake) forest soil were analyzed via the direct extraction of DNA and 16S rDNA-ARDRA. The 115 clones from pine mushroom forest soil were clustered into 31 different RFLP phylotypes by ARDRA. Based on the 16S rDNA sequences, the 31 ARDRA clusters were classified into 6 phylogenetic groups: ${\alpha}-,\;{\beta}-,\;{\gamma}-Proteobacteria$, Acidobacteria, Actinobacteria and Firmicutes. Among these bacterial populations, approximately 85% were classified as members of phylum Acidobacteria. The Acidobacteria phylum was shown to exist abundantly in the pine mushroom forest soil.

Seasonal Variations in the Bacterial Community of Gwangyang Bay Seawater (광양만 해수의 세균 군집의 계절적 변화)

  • Seasonal variations in the bacterial community of Gwangyang Bay seawater were analyzed using both isolation and cultivation-independent methods. Amplified rDNA restriction analysis was applied to 200 bacterial isolates. Bacterial isolates were composed of four phyla: Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetes. Pyrosequencing was conducted, in addition to denaturing gradient gel electrophoresis (DGGE) of genomic DNA extracted directly from the water samples. The bacterial sequences obtained by pyrosequencing of 16S rRNA genes consisted of 24 phyla in the spring and summer, 39 in the fall, and 32 in the winter. The diversity index was high in the fall, whereas the dominancy index was high in the spring. In the spring, phylum Firmicutes was dominant, whereas phylum Proteobacteria dominated in the other three seasons. The second most dominant phyla were Proteobacteria in the spring, Firmicutes in the summer, and Bacteroidetes both in the fall and winter. Bacilliaceae was the most predominant family in the spring. Rhodobacteraceae and Bacilliaceae dominated in the summer, and Rhodobacteraceae dominated in the winter. Neither was dominant in the fall Twenty-seven bands purified from DGGE profiles were cloned and analyzed phylogenetically. In the spring, phylum Firmicutes dominated, followed by Proteobacteria. Proteobacteria dominated in all other seasons. Thus, two cultivation-independent methods for determination of seasonal variation patterns at the phylum level were in accordance with each other.

Comparative Analysis of Endophytic Bacterial Communities in the Roots of Rice Grown under Long-term Fertilization Practice using Pyrosequencing Method (파이로시퀀싱을 이용한 비료 장기 연용지의 벼 뿌리 내생세균의 군집 분석)

  • Bacterial endophytes may be important factors in plant growth and ecologically relevant functions in rice. Using pyrosequencing technology, we analyzed the composition of endophytic bacterial communities that colonized the roots of rice cultivated in long-term fertilized (APK) and non-fertilized (NF) paddy soils. A total of 1,900 reads were obtained from 2 samples. All sequences were classified into 177 OTUs (APK sample) or 72 OTUs (NF sample) at a 97% similarity cut-off. Twenty-two OTUs were shared between the 2 samples, and these were also the most dominant OTUs in both samples. Proteobacteria was the most dominant phylum with 90.2%, followed by Actinobacteria (7.1%) and Bacteroidetes (1.1%). Furthermore, Pseudomonas was the most abundant genus in both samples. We observed clear differences in the structure of the endophytic bacterial community structure between the 2 samples. Notably, the distributions of Alphaproteobacteria and Gammaproteobacteria were markedly different. The diversity index of the APK sample was higher than that of the NF sample. These findings showed that the endophytic bacterial community of rice roots was affected by the presence of fertilizers in the rice field soil.

An Analysis on Perception of Mothers about Career for Elementary Science-Gifted Children (초등과학영재 어머니들의 자녀 진로에 대한 인식 분석)

  • The purpose of this study is to try to structuralize the perception of the mothers of science-gifted elementary students using the concept mapping approach. The mothers who participated in this research had children who were 5th and 6th graders selected as science-gifted by a regional education office, a science high school and two national universities in a city. One of the authors interviewed 26 mothers, and extracted 50 general statements of their perceptions about the career path of their children. Ten mothers who participated in interviews sorted a shuffled pack of statement cards. The categorization of the statements into the dissimilarity matrix was carried out by SPSS multidimensional scaling analysis and hierarchical cluster analysis to generate a conceptual diagram. After that 140 mothers rated each statement using a Likert-type response scale from one to five. The result showed six clusters of parental views such as were 'Burden of private education, grades and going to the next grade,' 'Thinking about career guidance in gifted education and school,' 'Parental roles in child career education,' 'Difficulties in career guidance at home,' 'Demand for strengthening the parental capacity for career guidance,' and 'Demand for social support.' 'Demand for social support' obtained the highest sympathy from mothers of elementary science gifted.

Phylogenetic diversity of bacterial communities in a gray solar saltern and isolation of extremely halophilic bacteria using culturomics (토판염전 결정지 내 세균군집의 계통학적 다양성 및 Culturomics법을 이용한 고도 호염균의 분리)

  • In this study, we investigated the phylogenetic diversity of the bacterial community and isolation of extremely halophilic bacteria using culturomics in a gray solar saltern. The number of bacterial living cells, enumerated in a gray solar saltern by direct fluorescence microscopy was three to four orders of magnitude greater than those enumerated by plate counts, suggesting the distribution of 'viable but non-culturable bacteria'. The biodiversity of bacterial communities in a gray solar saltern was investigated by pyrosequencing, 1,778 OTUs of bacteria were comprised of 18 phyla 46 classes 85 orders 140 families 243 genera with 6.16 diversity index. Archaea communities were composed of 3 phyla 6 classes 7 orders 7 families 38 genera with 4.95 diversity index from 643 OTUs. Totally 137 isolates were isolated by 59 different cultural methods based on culturomics considering culture media and conditions suitable for the growth of extremely halophilic bacteria. Phylogenetic analyses of extremely halophilic isolates based on 16S rRNA gene sequences, extremely halophilic isolates were composed of 4 phyla and 11 genera. Haloterrigena and Haloferax can be successfully isolated from culturomics. These culturomics were effective methods for collection of diversity of extremely halophilic bacteria.

Temporal and Spatial Distribution of Microbial Community and Odor Compounds in the Bukhan River System (북한강 수계 미소생물 군집 및 이취미 물질의 시공간적 분포 특성)

  • Odor compounds (geosmin, 2-MIB) have been causing problems in the Bukhan River system, but the causative organisms have not been clearly identified. To evaluate the relationship between dynamics of microbial community and odor compounds, two times monthly monitoring of water quality and microbial community from the three serial lakes (Lake Uiam, Lake Cheongpyeong and Lake Paldang) in the Bukhan River system were conducted from April to October 2017. The odor compounds were analyzed by HS-SPME analysis method using GC/MS. Bacteria communities were identified at the class level by NGS analysis. Actinobacteria and Betaproteobacteria were dominant taxon in bacteria community of three serial lakes. In the case of phytoplankton communities showed that seasonal changes by Bacillariophyceae and Cryptophyceae in spring, Cyanobacteria in summer, and Bacillariophyceae and Cryptophyceae in autumn. Dominant species was Dolichospermum (=Anabaena), Microcystis and Pseudanabaena in Bukhan River system in all study period. At the same time the odors geosmin and 2-MIB were also detected at high concentration. There is a significant positive correlation between proportion of Actinobaceria and 2-MIB concentration (r=0.491, p<0.01). In addition, proportion of cyanobacteria showed a significant correlation of geosmin (r=0.381, p<0.05) and 2-MIB (r=0.386, p<0.05) concentration. In this study, odor compounds in the Bukhan River system are considered to be a direct relationship between with Actinobacteria and cyanobacteria.

Impacts of Sand Mining on the Macrobenthic Community in Gyeonggi Bay, Korea (경기만에서 해사채취가 대형저서동물 군집구조에 미치는 영향)

  • Small-scale effects of sand mining on macrobenthic communities have been reported previously. However, little information is available as to how dredging affects the macrobenthic community structure. The objective of this study was to determine the impacts of large-scale exhibition dredging on the macrobenthic community of Gyeonggi Bay, Korea, where sand mining has continued for 20 years. Prior to dredging, the macrobenthic species composition was similar to that of areas near the dredging site, with several common dominant species found before dredging. After dredging, the number of species, density, and diversity (H') in the experimental area (sites 0 and 1) decreased significantly, but no difference was observed at other sites. Multivariate analysis (multidimensional scaling) revealed significant differences in community structure before and after dredging. The amphipod Urothoe grimaldii japonica, which was the most dominant species at sites 0,1, and 2, decreased rapidly at sites 0 and 1 after dredging, but no difference was observed at site 2. In addition, the index of multivariate dispersion (IMD) and the relative IMD (r. IMD) were large at sites 0 and 1, suggesting that the effects of dredging were direct at site 0 and 1, but indirect at site 2. The macrobenthic communities at sites 3 and 4 were not affected by dredging, but they were affected by physical conditions and biological interactions. We suggest that benthic biotope indices such as the IMD and r.IMD may constitute a valid tool for assessing the effects of dredging on the macrobenthic community; long-term monitoring is required to verify this.

SWAT Direct Runoff and Baseflow Evaluation using Web-based Flow Clustering EI Estimation System (웹기반의 유량 군집화 EI 평가시스템을 이용한 SWAT 직접유출과 기저유출 평가)

  • In order to assess hydrologic and nonpoint source pollutant behaviors in a watershed with Soil and Water Assessment Tool (SWAT) model, the accuracy evaluation of SWAT model should be conducted prior to the application of it to a watershed. When calibrating and validating hydrological components of SWAT model, the Nash-Sutcliffe efficiency coefficient (EI) has been widely used. However, the EI value has been known as it is affected sensitively by big numbers among the range of numbers. In this study, a Web-based flow clustering EI estimation system using K-means clustering algorithm was developed and used for SWAT hydrology evaluation. Even though the EI of total streamflow was high, the EI values of hydrologic components (i.e., direct runoff and baseflow) were not high. Also when the EI values of flow group I and II (i.e., low and high value group) clustered from direct runoff and baseflow were computed, respectively, the EI values of them were much lower with negative EI values for some flow group comparison. The SWAT auto-calibration tool estimated values also showed negative EI values for most flow group I and II of direct runoff and baseflow although EI value of total streamflow was high. The result obtained in this study indicates that the SWAT hydrology component should be calibrated until all four positive EI values for each flow group of direct runoff and baseflow are obtained for better accuracy both in direct runoff and baseflow.
