A Study on the Effects of Retardation with Pb in the Biofilter (생물여과층에 의한 납의 이동억제효과에 관한 연구)

  • The purpose of this study was to show the application of the Biofilter for treatment of the soil contaminated by lead and to investigates the effect of the biofilter on the retardation of lead with pilotplants that were simulated with different media and the number of bed. and to testify the inoculation by seeding microbes. The ratio of the degradation of soil contaminant was verified as CODcr/TOC in order to find a variation of the stabilization index in soil. The Biofiltration was one of biological processing methods for treatment. The contaminants were transported through the biofilter that was filled with the media. The surface of media formed biofilm which was surrounded by microbes and through its boundary, some materials were exchanged and migrated into the cell of microbes in an orderly manner. To investigate the effect of the Biofiltration, contaminated soil with lead nitrate of 1000mg/kg in dry was made artificially. The tests were simulated such as compost, Bioceramic and compost with bioceramic by 7:3 in weight. The bed consisted of three layers in order to find effect of the number of bed. Aspergillus niger was used as a biosorbent could probe the effect on the retardation of lead.

Behavior of Refractory Organic Matter in Leachate from Landfill Contaminated by Foot-and-mouth Disease (구제역 매몰지역 침출수에서 발생하는 난분해성 유기물질 거동)

  • The leachate from landfill (Andong city) contaminated by foot-and-mouth disease (FMD) contains 44%-50% hydrophilic organic matter, compared with 22%-27% in natural water bodies such as ground water, lake water, and river water. In such natural water, the organic matter content is reduced by the metabolism of microbes in the water. However, in the case of leachate-1 and -2, the concentrations of RTOC (refractory total organic carbon) and RDOC (refractory dissolved organic carbon) were higher than the initial TOC and DOC after burial. According to time elapsed after burial, the concentrations of RTOC and RDOC were decreased below the initial TOC and DOC. In the case of leachate-6 (386 days after burial), RDOC made up 91% of RTOC. This result shows that organic matter in the leachate was composed dominantly of RDOM, most of which was not removed by the metabolism of microbes. Hence, the presence and characteristics of RDOM provide a valuable indication of the effect of leachate on the quality of surface water and ground water. Such information is useful in understanding leachate environments.

  • Cambodia is a typical country that a very famous tourist destination in South Asian regions, but water has some serious problems. As a public hygiene point of view, water problems in Cambodia can be roughly classified into two categories. A numbers of the report revealed that thermotolerant coliform(TTC) bacteria and arsenic(As) pollution representative species as a representative pollutant for surface water and ground water problems, respectively. In recent years, home water purifier systems for various filter systems including BSF(Bio-Sand Filter), MF and RO have been spread and adopted in rural areas. However, BSF and composite membrane processes are recognized as an undesirable and representative method due to unreliable efficiency, high cost and maintenance issues. In order to develop a small-scale ceramic filter as a home water purifier, the applicability of two types of gravity ceramic filter using clay and red soil in the part of the Northwest was made and compared in this research.

Optimization of L-leucine Fermentation (L-leucine 발효의 최적화)

  • The effects of pH, temperature and oxygen transfer rate(OTR) on L-Ieucine fermentation were investigated employing Corynebacterium glutamicum CH 1516 in 71 fermentor. The optimum pH, temperature and OTR were determined to be 7.0, $30^{\circ}C$ and 0.21 kmole $O_2$/$m^3{\cdot}hr$, respectively. For the values of OTR lower than 0.19 kmole $O_2$/$m^3{\cdot}hr$ a significant amount of lactic acid was accumulated, while the packed cell volume(PCV) was rapidly increased at higher OTR values above 0.23 kmole $O_2$/$m^3{\cdot}hr$ and glutamic acid was produced to some extent. Scale-up studies for L-Ieucine fermentation which was carried out in 12001 pilot scale fermentor reaffirmed the results of 71 fermentation. The optimum redox potential value for L-Ieucine production was found to be -150 to -170 mY.

Toxic effects of Aroclor 1016 and bisphenol A on marine green algae Tetraselmis suecica, diatom Ditylum brightwellii and dinoflagellate Prorocentrum minimum (해양 녹조류 Tetraselmis suecica, 규조류 Ditylum brightwellii, 와편모조류 Prorocentrum minimum에 대한 Aroclor 1016과 비스페놀 A의 독성 효과)

  • Microalgae are the potential bioindicators of environmental changes, for the environmental risk assessment as well as to set limits for toxic chemical release in the aquatic environment. Here, we evaluated the effects of two endocrine disrupting chemicals (EDCs), namely bisphenol A (BPA) and Aroclor 1016, on the green algae Tetraselmis suecica, diatom Ditylum brightwellii, and dinoflagellate Prorocentrum minimum. Each species showed wide different sensitivity ranges when exposed to these two EDCs; the 72 h effective concentration ($EC_{50}$) for these test species showed that Aroclor 1016 was more toxic than BPA. $EC_{50}$ values for the diatom D. birghtwellii were calculated at 0.037 mg/L for BPA and 0.002 mg/L for Aroclor 1016, representing it was the most sensitive when compared to the other species. In addition, these results suggest that these EDC discharge beyond these concentrations into the aquatic environments may cause harmful effect to these marine species.

Transfer RP4::Mu cts and RP4::mini-Mu from E. coli to Pseudomonas sp. (RP4::Mu cts 및 RP4::mini-Mu Pseudomonas sp.로의 전달)

  • Chromosomal gene transferable hybrid plasmids, RP4::Mu cts and RP4::mini-Mu, were transferred by conjugation from E. coli to Pseudomonas strains. In order to use for recipient cells of RP4::Mu cts and RP4:: mini-Mu, plasmid-free Pseudomonas strains were characterized for their antobiotic resistance, aromatic hydrocarbon utility and degradation patterns of chlorinated herbicide. Transfer frequencies of RP4::mini-Mu exhibited about $10^{-2}$ to $10^{-4}$, while those of RP4::Mu cts exhibited very low value of $10^{-7}$ in recipients tested except Pseudomonas aeruginosa KU557. Existance of hybrid plasmids in Pseudomonas transconjugants were identified by their antibiotic resistance and agarose gel electrophoresis. In case of RP4::Mu cts transconjugants it was also confirmed by demonstrating that they were capable of releasing phage and forming plaques at $43^{\circ}C$. Plaque forming unit of the transconjugants was about $10^{5}$. It was shown by the stability test that RP4::Mu cts and RP4::mini-Mu in Pseudomonas were relatively stable.

Properties of biparental clones formed by spheroplast fusion of pseudomonas putida (원형질체 융합에 의한 pseudomonas putida의 biparental clones의 형성과 성질)

  • Biparental clones and recombinant clones were obtained by spheroplast fusion of Pseudomonas putida KU218R-3 and P.putida KU428. Formation of the fusion product was the most effective when the Pseudomonas spheroplast mixture were treated with 40% plyethyleneglycol(PEG) 6000 for 10min at room temperature, The fusants which selected by indirect method were obtained at an average frequency of 10.8%. Most of the fusants were biparental clones (10.4%) and the recombinant clones were produced in low yield (0.42%). Fusants, at the frequency of 4% were obtained without PEG 6000, which shows that fusion is not strictly dependant on PEG. The stability of fusants were examined. Most of the biparental clones were segregated to parental form amd late recombinants were formed on further propagation of biparental clone but the recombinant clones were nery stable.

  • Kimchi cabbage is one of the four major vegetable crops in Korea. The total annual production of kimchi cabbage, the main material of kimchi, was 20,559 tons in 2015. Kimchi cabbage is one of the majer crops produced by farmers which accounts for about 80% of the total leaf vegetable production in Korea. As the consumption of environmental-friendly agricultural products increases, food safety is one of the major public health concerns. We analyzed the biological hazards of kimchi cabbage produced by two types of cultivation methos such as organic farming and conventional farming using various culture media and microscopy. A total of 432 samples were analysed for presence of sanitary indicator microorganisms (aerobic plate count, coliform count, yeast & mold) and food-borne pathogens (Staphylococcus aureus, environmental Listeria, Bacillus cereus). The population of sanitary indicating microorganisms and food borne pathogens was under 5 Log CFU/g in all tested samples. The results of total microorganism numbers of leaf surface showed a positive correlation to those of soil samples. Additionally, we examined chemical factors such as pesticide residues and heavy metals in soil samples. All tested samples did not shown contamination levels higher than the standard limit.

Effects of Electrical Conductivity on the Soil Microbial Community in a Controled Horticultural Land for Strawberry Cultivation (시설딸기재배지 토양에서 염류농도가 미생물 생태에 미치는 영향)

  • Total soil microbial activities have great impact to soil management for organic farming. This study was evaluated in the soil microbial community by fatty acid methyl ester (FAME) in a controlled horticultural field for strawberry organic farm. Experimental plots were prepared with a high level of soil electrical conductivity (EC) and a optimum level of soil EC. Soil microbial biomasses and communities of total bacteria, Gram-negative bacteria, Gram-positive bacteria, actinomycetes, fungi, and arbuscular mycorrhizal fungi in the high level of soil EC were significantly larger than those in the optimum level of soil EC. Lower ratios of cy17:0 to 16:$1{\omega}7c$ and cy19:0 to 18:$1{\omega}7c$ were found in the optimum level of soil EC than those in the high level of soil EC, indicating that microbial stress decreased.

Investigation of Microbiological Hazard from Korean Leeks and Cultivation Area to Establish the GAP Model (Good Agricultural Practices (GAP)모델 개발을 위한 부추 및 생산환경에서의 위해요소 조사)

  • This study is to investigate microbiological hazards which can be used as fundamental data to adequately control leeks hazards and develop leeks GAP model for those who want to get GAP system. The microbiological investigations on cultivation environments (soil and water), crops (leeks), personal hygiene (workers' hands, clothes and gloves) and working tools (boxes) have been conducted for one year, so the period was classified under non-cultivation, cultivation, and post harvest. Total bacteria was detected from soil (4.0~6.7 log CFU/g), leeks (4.6~5.1 log CFU/g), hands (ND~3.3 log CFU/hand) and gloves ($ND{\sim}5.4\;log\;CFU/cm^2$) while nothing was detected from the other samples. The coliform contamination of leeks (4.8~5.0 log CFU/g) was more high than that of soil (3.9~4.2 log CFU/g). In case of foodborne pathogens, only B. cereus was detected at the level of 0.5~4.6 log CFU/g (or hand, $100cm^2$). Fungi was observed at the level of 2.1~3.8 log CFU/g (or hand, $100cm^2$) excepting water and some working tools. These results demonstrate that the contamination of leeks is comparatively higher than that of soil sample. The reason may be the cross-contamination by biological hazards presenting on soil. Therefore, it is necessary to properly control soil and fertilizer for safety against biological hazards.
