Antioxidant effect of Aspalatone on KA-induced neurodegeneration in rat. (카인산 유발 신경세포 변성 모델에 대한 Aspalatone의 항산화 작용.)

  • 뇌신경 변성 / 퇴행과 관련된 중요한 병인론 중의 하나는 변성 과정에서 형성된 유리기(free radical)로 인한 항산화계의 평형 소실로 알려져 있다. Aspalatone (APT)의 예상되는 항산화 효능을 검정하기 위하여 본 실험에서는 Kainic acid (KA) 유발 뇌변성 모델을 적용하였다. KA 모델은 변연계의 간질성 경련과 신경세포 변성에 대하여 재현성 있는 병변 모델을 제공해 주며, 이와 같은 신경세포의 병독 기전에 산소 유리기가 관여함이 강력히 시사되고 있기 때문이다. KA 투여로 인하여 지속적이고도 전형적인 간질성 경련이 관찰되고 1일 이내에 높은 치사율을 보였으나 APT으로 인하여 그 간질성 경련 행위와 비율이 억제되고 KA 유발 치사율도 억제되었다. 최종 KA 투여 3일 후에 얻어진 흰쥐 해마 및 대뇌 피질에서 항산화 효소인 Superoxide dismutase (SOD), Catalase (Cat.), Glutathione peroxidase (GSH-PX) 및 과산화지질의 지표인 Malondialdehyde (MDA)를 검정하였다. 대조군에 비하여 KA는 뇌조직의 SOD-1을 유도하였으나, Cat.와 GSH-PX의 활성은 현저히 유도되지 않았고, 반면에 MDA 치는 현저히 증가하였다. 즉, Cat., GSH-PX와 같은 $H_2O$$_2$중화제가 동반 유도하지 않는 SOD의 유도는 세포내 축적되는 $H_2O$$_2$로 인하여 Fenton/Haber-Weiss 반응을 가속화하여 과산화지질화를 촉진함을 시사한다. APT 병용 투여로 SOD는 현저히 유도되지 않았으나 특히 Cat.가 현저히 유도되어지고 MDA는 억제되었다. 이와 같은 생화학적인 결과는 다음의 형태학적인 소견과 일치한다. Fos 관련 항원 (FRA)와 SOD-1을 면역세포화학 (Immunocytochemistry)적 방법으로 이중 표식 (double-labelling) 하였다. FRA는 KA로 인한 신경세포의 자극에 대한 지표로 응용하였고, SOD-1은 퇴행성 뇌질환에서 산화적 손상의 지표로 사용하였다. KA 투여로 해마의 dentate gyrus (DG) 내에 강한 면역환성 (immunoreactivity)이 나타났고 pyramidal cell layer (PCL)와 glia에 SOD-1이 강하게 염색되었다. APT 병용 투여로 상당수의 경련이 일어나지 않은 흰쥐는 해마의 DG에 FRA가 경미하게 염색되었고, PCL에 SOD-1도 경미하게 나타났으나, 경련이 나타난 쥐에서는 KA만을 투여한 흰쥐와 구별되지 않았다. 이상의 APT의 항산화 효과는 KA로 인한 뇌세포 변성 개선에 중요한 인자로 작용할 것으로 사료되나, 보다 명확한 APT의 기전을 검색하고 직접 임상에 응응하기 위하여는 보다 다양한 실험 조건이 보완되어야 찰 것으로 생각된다.

사상체질(四象體質)과 비만(肥滿)의 상관성에 관한 임상적 연구

  • 연구 목적 : 복지부 발표 '1993년도 대한민국 국민들의 영양상태'보고서를 보면 1인당 섭취열량은 1969보다 257Kcal가 줄어든 것으로 나타났다. 그런데도 불구하고 일부의 사람들은 과도한 열량을 섭취하고 있기 때문에 비만인 사람이 점점 늘어나고 있다. 그렇다보니 신문이나 잡지에서는 거의 매일 살빼는 약이나 신종 다이어트방법에 대한 광고와 기사를 앞다투어 내보내고 있는 실정이다. 비만증은 근래에 발병률이 현저히 증가하면서 중풍, 당뇨병, 고혈압, 암, 간경화증, 당석증 등의 만성질환의 이환율을 증가시키고, 관절에 큰 부담을 주어 골관절염을 보다 빠르게 진전시키며, 사람들의 수명을 단축시키는 등 심각한 건강상의 문제를 일으키고 있다. 또한 비만증은 용모의 손상을 일으켜 정신적인 스트레스의 주요 인자로 등장하고 있기도 하다. 비만인이 증가하는 원인으로는 서구화된 음식습관으로 동물성 지방질과 단백질의 섭취증가, TV 자동차 세탁기 오디오 리모콘 등 생활이기의 사용으로 인한 운동부족, 체질과 질병 등이 재기되고 있다. 이제까지 비만을 치료하기 위한 많은 시도가 있었으나 주된 이론은 섭취음식의 절재와 운동량을 증가시키는 것이었으며, 체질적인 요소를 중심으로 비만증을 해결하려는 시도는 없었다. 이에 저자는 1992년 8월 20일부터 1995년 8월 19일까지 만 3년동안 상지대학교 부속한방병원 체질의학과에서 치료받는 461명의 환자를 대상으로 사상체질과 비만과의 상관성을 임상적으로 연구하고 그 효과를 보고하는 바이다. 연구 결론 : 상지대학교 부속한방병원에 내원치료를 받고 있는 비만증환자들을 대상으로하여 체질과 비만과의 관계를 연구한 결과는 다음과 같다. 1. 비만증 환자의 70.2%가 태음인이었다. 2. 치료결과 4주동안에는 $1.48{\pm}1.64kg$, 5주부터 8주동안에는 $2.05{\pm}2.10kg$, 9주부터 12주동안에는 $2.18{\pm}2.27kg$, 13주부터 16주동안에는 $2.08{\pm}2.88kg$이 감량되었다. 3. 체중감량에 있어 태음인이 가장 많은 감소를 보였다. 연령별로는 10대에서 가장 많은 감소를 보였다. 4. 비만환자의 혈액 가운데 총 콜레스테롤, 저밀도 지방단백이 높온 사람보다 유리지방산과 중성지방이 높은 경우가 대부분을 차지했다. 5. 비만인의 혈청지질에서는 FFA, TG가 높았다. 그 이유는 육식보다는 당질과 지방질의 섭취로 나타난 것이었다.

  • Starch solid model system was employed to investigate the effect of lipoxygenase, ${\beta}-carotene$, ${\alpha}-tocopherol$ and water activity on the oxidation of linoleic acid. The rate of oxidation of linoleic acid by lipoxygenase was increased with the increase in water activity, Addition of ${\beta}-carotene$ and ${\alpha}-tocopherol$ to this system has been shown to inhibit the oxidation of linoleic acid and ${\alpha}-tocopherol$ was more effective antioxidant than ${\beta}-carotene$. However, an increase in the concentration of ${\beta}-carotene$ was found to have a strong antioxidant effect in the solid model system. And also the antioxidative action of ${\beta}-carotene$ was increased with increasing water activity in this system.

  • Gymnema sylvestre, a traditional India medicine called a 'Gagaimogouip' in Korea, has been used to regulate sugar metabolism in patients with diabetes mellitus. First, we induced callus from Gymnema sylvestre leaf and mode extract from cultured callus. In this study, we investigated the anti-obesity effect of tissue-cultured Gymnema sylvestre leaf extract in 3T3-L1 cells. The tissue-cultured Gymnema sylvestre extract inhibited the cytoplasmic lipid accumulation as well as adipogenic differentiation of preadipocytes. The tissue-cultured Gymnema sylvestre extract inhibited insulin-induced predipocyte differentiation through the inhibition of expression of the early adipogenic transcription factor C/EBP-${\alpha}$ that regulate adipogenesis. These results indicate that tissue-cultured Gymnema sylvestre extract may be potential natural ingredient for slimming cosmetic products.

Association of Osteoarthritis with prevalence of Metabolic syndrome: based on 2009-2010 Korean National Health & Nutrition Survey (골관절염과 대사증후군과의 상관관계: 2009-2010년 국민건강영양조사 이용)

  • This study was attempted to investigate the possible association of osteoarthritis with the metabolic syndrome (MetS) and its components in Korean adults based on the Korean National Health and Nutrition Examination Survey (KNHANES). The prevalence of MetS was significantly increased in OA group in women. To investigate the association between the components for MetS and osteoarthritis, we analyzed the mean values of 5 components of MetS, waist circumference (WC), triglyceride (TG), HDL-cholesterol, systolic blood pressure, and fasting plasma glucose in normal and OA group. In women, WC, TG, HDL, and systolic blood pressure were associated with the development of OA. Moreover, accumulation of components of MetS was significantly related to the prevalence of OA in Korean men and women. These data suggest that MetS might be a novel risk factor for OA and MetS should be taken into account in the prevention and treatment of OA for Korean adults as well.

A Clinical Study on Blood Lipids as a Risk Factor of Cerebral Infarction (뇌경색 위험인자로서의 혈중 지질에 대한 임상적 연구)

  • Objective The purpose of this case-control study was done to examine the relationship among the acute brain infarction, silent brain infarction and blood lipids. Methods We compared the components of blood lipids among acute brain infarction patients group (n=99), silent brain infarction patients group(n=101) and controls group (n=153). The analysis of the data was done by Frequency Analysis, one-way ANOVA, Post Hoc Test(Duncan), Pearson's Correlation. Results The results were as follows. High density lipoprotein cholesterol(HDL-Chol) was significantly lower in patients of acute brain infarctions group. The blood levels of total cholesterol(T-Chol) was significantly higher in patients of silent brain infarctions. A study on the relationship among the significant variables ; In T-Chol and HDL-Chol, DM(No=0, Yes=1) and Age, they had positive correlation each other. Between Sex(Female=0, Male=1) and HDL-Chol, Age and HDL-Chol, DM and HDL-Chol, they had negative correlation each other. Conclusions These results suggest that low HDL-Chol may be risk factor of acute brain infarction.

  • The purpose of this study was to evaluate the effect of vegetation on the cutting slope applied by vegetation system in the whole land. The engineering characteristics of slopes were investigated using the face mapping and physical tests. From the statistical methods, a relationship between a geologic stratum and degree of covering was independent. Therefore, the afforestation of slope was affected more by condition of vegetation as direction and opening of joints, and a topsoil state than by conventional classification of layer. It was concluded that the adjustable vegetation on the slope considering condition of geometric shape of slope, joint data and spontaneous type could make useful early afforestation.

  • Although it is important to reflect the accurate information of the ground condition in the tunnel design, the analysis and design are conducted by limited information because it is very difficult to consider various geographies and geotechnical conditions. When the tunnel is under construction, examination of accurate safety and prediction of behavior are overcome the limits of predicting behavior by Artificial Neural Network in this study. First, construct the suitable structure after the data of field was made sure by the multi-layer back propagation, then apply with algorithm. Employ the result of measured data from database, and consider the influence factor of tunnel, like supporting pattern, RMR, Q, the types of rock, excavation length, excavation shape, excavation over, to carry out the reliable analysis through field applicability of Artificial Neural Network. After studying, using the ANN model to predict the shearing displacement, convergence displacement, underground displacement, Rock bolt output follow the excavation over of tunnel construction field, then determine the field applicability with ANN through field measured value and comparison analysis when tunnel is being constructed.

Numerical Analysis on the Effect of Fractured Zone on the Displacement Behavior of Tunnel (파쇄대가 터널 주변 암반의 변형 거동에 미치는 영향에 대한 수치해석적 연구)

  • Anisotropic/heterogeneous rock mass shows various deformation behavior types due to tunnelling because deformation behavior is largely controlled by the spacial characteristics of geological factors such as faults, joints and fractured zone in rock mass. In this paper 2-dimensional numerical analysis on the several influencing factors is performed considering fractured zone located near tunnel. This numerical analysis shows that deformation behavior of tunnel are very different according to the width and the location of fractured zone and supper method. However, 3-dimensional analysis is necessary to consider 3-dimensional geometrical characteristics sufficiently since discontinuity and fractured zone have 3-dimensional geometry. Also flexible design/construction guidelines for tunnelling are required to cope with uncertain ground condition and circumstance for technically safe and economic tunnel construction.

Effect of Potato Polyuphenolics on the Hyperlipidemia in Rats (식이성 고지혈증에 미치는 감자 폴리페놀의 영향)

  • Effect of potato polyphenolic compound on the concentrations of serum and liver lipids, serum glucose, and urine protein was investigated in Sprague Dawley rats by feeding a diet containing 0.5% of cholesterol for 2 weeks. Polyphenolic compound extracted from potato (Solanum tuberosum variety Dejima) was supplemented at a 0.5% level in the basal diet or the cholesterol diet. The supplementation of potato polyphenolic compounds decreased slightly the concentrations of total cholesterol and VLDL+LDL-cholesterol in serum, and atherosclerotic index in rats fed the cholesterol diet, while those measurements were not altered by the supplementation of potato polyphenolic compounds in rats fed the basal diet. In the rats fed the basal diet or the cholesterol diet containing potato polyphenolic compounds, urine protein increased by 12% and 27% at 1st week, respectively, but this change was not seen at 2nd week. The concentration of serum glucose, however, was not significantly different in the dietary groups.
