Nutritional Components and Physicochemical Properties of Lipids Extracted from Forest Resources (산림자원에서 추출한 유지자원의 영양성분 및 이화학적 특성 검토)

  • Nutritional constituents and physicochemical properties of lipids of forest resources were studied in order to examine their practical utilization in the lipid industry. In this study, Garae, Dongback, Mougwi, and Muwhanja were chosen as sources of fat-soluble components. Fatty acid profiles of forest resources showed more than 80% polyunsaturated fatty acids in total fatty acids. For total tocopherol contents, Garae showed higher content than others; moreover, Dongback was a good source of ${\alpha}$-tocopherol. Phytosterols of forest resources ranged from $55.96{\pm}2.23$ to $194.94{\pm}21.42mg$/100 g, and Muwhanja showed the highest phytosterol contents. Chemical properties such as acid value, peroxide value, and p-anisidine value showed good oxidative stability of lipids of forest resources. For physical properties, browning intensity and color parameters were studied. Induction times, as an indicator of oxidative stability, were measured and ranged from $0.70{\pm}0.01$ to $18.40{\pm}1.02h$ in four forest resources. Taken together, contents of lipid constituents and physicochemical properties can be used as an important preliminary database for utilization of lipids of forest resources.

Relationships between Fatty Acids and Tocopherols of Conventional and Genetically Modified Peanut Cultivars Grown in the United States (미국산 전통품종과 유전자 재조합 땅콩품종의 지방산과 토코페롤의 상관관계)

  • Relationships between fatty acids and tocopherols in conventional and genetically modified peanut cultivars were studied by gas chromatography with flame ion detector and high performance liquid chromatography with fluorescence detection. Eight fatty acids and four tocopherol isomers in the sample set were identified and quantified. Oleic acid and linoleic acid are major fatty acids and the ratio of oleic and linoleic acids ranged from 1.11 to 16.26. Tocopherols contents were 6.76 to 12.24 for ${\alpha}$-tocopherol (T), 0.08 to 0.39 for ${\beta}$-T, 5.28 to 15.02 for ${\gamma}$-T, and 0.17 to 1.17 mg/100 g for ${\delta}$-T. Correlation coefficient (r) for fatty acids and tocopherols indicated a strong inverse relationship between oleic & linoleic acids (r=-0.97, P<0.05) and positive relationships between palmitic & linoleic acids (r=0.95, P<0.05) and ${\gamma}$-T & ${\delta}$-T (r=0.83, P<0.05). Principal component analysis (PCA) of fatty acids and tocopherols gave four significant principal components (PCs, with eigenvalues>1), which together account for 85.49% of the total variance in the data set with PC1 and PC2 contributing 45.27% and 21.33% of the total variability, respectively. Eigen analysis of the correlation matrix loadings of the four significant PCs revealed that PC1 was mainly contributed by palmitic, oleic, linoleic, and gondoic acids, while PC2 was by behenic acid, ${\beta}$-T, and ${\gamma}$-T. The score plot generated by PC1-PC2 identified sample clusters in the two spatial planes based on the oleic and linoleic acids. The score plot PC3-PC4 didn't separate sample groups.

Effects of Polyacetylenes in Ginseng on Activity of Enzymes Related to Post-translational Modification of Ras Protein and Effects of Petroleum Ether Extract of Ginseng on Progression of Cell Cycle (인삼의 polyacetylene 계 성분이 Ras 단백질 변형 효소의 활성에 미치는 영향과 인삼의 석유에텔 추출물이 세포주기에 미치는 영향)

  • We investigated the effects of polyacetylenes of ginseng on farnesyl protein transferase (FPTase) and carboxyl methyl transferase (CMTase) activities related to post-translational modification of Ras protein. We also investigated the effect of petroleum ether extract (PEE) of ginseng on progression of cell cycle. FPTase activity was respectively inhibited 16.2% by 10mM panaxynol and 21.3% by 10mM panaxydol, whereas CMTase activity was not inhibited by panaxynol or panaxydol. Treatment of PEE significantly reduced the numbers and size of human colon cancer cell (HT-29) and human liver cancer cell(HepG2) cultured, respectively. To investigate the mechanism of growth inhibition by PEE of ginseng, we analyzed the cell cycle progressions of PT-29 and HepG2 cells, respectively. We found that PEE significantly inhibited progression of cell cycle from G1 to S phase. These results suggest that anticancer effects of PEE were derived from the arrest of G1 phase in cell cycle progression.

동결농축유의 미량성분 분석 및 관능적 특성에 관한 연구

  • 본 연구는 동결농축유의 미량성분을 분석하고 관능적 특성을 알아보기 위하여 실시하였다. 농축유의 수용성비타민은 동결농축유가 진공농축유보다 비타민의 감소율이 적었고, 같은 동결농축유의 경우에 17% 고형분인 농축유가 27% 고형분인 농축유보다 비타민의 감소율이 적었다. 지용성비타민도 동결농축유가 진공농축유보다 비타민 감소율이 적었으며, 수용성비타민 보다는 그 감소율이 크지 않았다. 유리아미노산은 진공농축유보다 동결농축유에서 많이 측정되었고 단쇄유리지방산은 진공농축유가 동결농축유보다 생성량이 많았으며 고형분이 많을수록 많이 측정되었다. 지방산화도는 단쇄유리지방산의 결과와 같이 동결농축유보다 진공농축유가 더 높게 측정되었다. 관능검사는 cooked flavor, oxidized flavor, cheesy flavor, fat-taste, oxidized taste, off-taste에서 동결농축유가 진공농축유보다 양호하게 평가되었다. 결과적으로 동결농축유가 진공농축유보다 미량성분의 손실이 적고 관능적으로 양호하게 평가되었기 때문에 영양학적, 관능적으로 우수하다고 사료된다.

Isolation and Identification of Red Color Pigments from the Korean Lithospermum erythrorhizon (한국산 자초 적색소의 분리 및 확인)

  • Conditions of isolation for the red color pigments from the Korean Lithospermum erythrorhizon were investigated and identification of the red pigment was analysed. Non-polar solvents were more effective than water. Especially, 95% ethanol was observed as optimum solvent for the pigments extraction. About 20 minutes at 40$^{\circ}C$ with 95% ethanol was enough for the extraction of the red pigments. The major pigment was analysed as acetylshikonin by TLC, IR, NMR and GC/MS.

비타민(IV) 수용성비타민

  • 수용성 비타민은 지용성비타민과는 달리 체내에 축적이 많이 되지 않기 때문에 자주 계속적으로 공급해 주어야 한다. 수용성비타민은 주로 탄수화물, 지방, 단백질의 대사작용에 작용하며 조효소의 구성성분이다. 수용성 비타민에는 비타민 B군(群)과 비타민 C가 있다. 비타민 B군에 속하는 비타민에는 비타민$B_1$(thiamin), 비타민 $B{_2}$(riboflavin), 나이아신(niacin), 비타민$B{_6}$(pyridoxine), 판토텐산(pantothenicaicd), 바이오틴(biotin), 콜린(choline), 엽산(folacin), 비타민$B_{12}$ 등이 있다. 이들 B군에 속하는 비타민들은 대체로 자연에 함께 존재한다. 즉, 어느 한가지가 풍부한 곳에는 B군에 속하는 다른 비타민들도 풍부하게 존재하고 있다. 따라서 인간이나 동물이 순수한 어느 한가지 비타민 B의 결핍증에 걸리기는 매우 어렵다.

In Vitro Antioxidant and Anticancer Potential of n-Hexane Extract from Ginseng Marc (인삼박 n-Hexane 추출물의 in vitro 항산화 및 항암 활성)

  • A lipid-soluble extract in ginseng marc was prepared by n-hexane extraction to evaluate its antioxidant and anticancer potential. A hexane extract of ginseng marc (HEGM) possessed a 2,2-diphenyl-1-picryl-hydrazyl free radical scavenging activity which was related to the amount of total phenolics. Also, HEGM showed a potent inhibitory activity on human non-small cell lung cancer (A549, $GI_{50}=34.0{\mu}g/mL$) and colon cancer (SNU-C4, $GI_{50}=45.2{\mu}g/mL$) cells proliferation in vitro in a concentration-dependent manner as did the hexane extract of ginseng with $GI_{50}$ values of $20.0{\mu}g/mL$ in A549 and $37.0{\mu}g/mL$ in SNU-C4. These results imply that HEGM can be utilized as an antioxidant and anticancer substance.

Effects of Petroleum Ether Extract of Ginseng Root on Some Enzyme Activity in Human Colon Cancer Cells (고려인삼중 지용성 성분이 인체암 세포의 수종 효소활성에 미치는 영향.)

  • This study was devised to observed the growth inhibition and change of disaccharidase activities of human colon cancer cells cultured in medium containing the ginseng extract. Three species of human colon cancer cell lines, HRT-18, HCT-48 and HT-29, were used for the experiment. The activities of sucrease, lactase, maltase and trehalase in the cancer cells were determined. The results obtained are summarized as follows; 1. The doubling times of the HRT-18, HT-29 and HCT-48 were about 20,22 and 24 hours, respectively. 2. The growth rates of the HRT-18 and HCT-48 in culture medium containing the ginseng extract were inhibited gradually according to increase of the concentration of ginseng extract and extension of the incubation time. 3. The activities of disaccharidase in HRT-18 and HCT-48 cultured in the medium containing the ginseng extract were increased compared with control group as follows;

  • Manuka oil named New Zealand’s tea tree oil is oil-soluble and comes firom nature. Manuka oil and its extract $\alpha$-pinene, Oxy’less clear, R-limonene which is one of the component of Citron extracted from Grapefruit seed and Citrex were used to estimate the antimicrobial activity and to improve the capability of antiseptic. Disk diffusion method was used to measure the antimicrobial activity. Escherichia coli which is gram-negative bacteria and Staphylococcus aureus which is gram-positive bacteria were used as strain. The antimicrobial activity of Manuka oil and $\alpha$-pinene for Escherichia coli, Staphylococcus aureus was similar when the concentration of Manuka oil and $\alpha$-pinene are 10u/paper disk. However, antimi-crobial activity of Manuka oil fDr Escherichia coli, Staphylococcus aurem was better than that of $\alpha$-pinene when the concentration of Manuka oil and $\alpha$-pinene was low. Antimicrobial activity of Oxy’less clear is better than that of propyl para hydroxybenzoate(PPHB), R-limonene at all the concentration and is similar to that of $\alpha$-pinene. A Study on the Cytotoxic Activity of Garlic(Allium Sativum) Extract Against Cancer Cells (마늘중 지용성 성분의 암세포증식 억제효과 연구)

  • This study was devised to observe the cytotoxic activities of garlic extracts against various cancer cells, that is, murine leukemic lymphocyte(L1210 and P388) and human rectal(HRT-18) and colon cancer cells(HCT-48 and HT-29) in vitro, and murine ascitic tumor cell(S-180) in vivo. Each cell-line except S-180 was cultured in medium containing serial concentration of the garlic extract in vitro. Inhibitory effect n the growth rate of the cancer cells was stronger in extracts of petroleum ether than that of ethanol. A lipid soluble compound in the extracts of garlic was cytocidal to murine leukemic cells, human rectal and colon cancer cells in vitro. The growth rates of the cancer cells in medium containing garlic extracts were inhibited gradually to a significant degree in proportion to the increase of the extract concentration. The cytotoxic activity of garlic active fraction from TLC was about 2.3 times more potent than that of crude garlic extract, one unit of cytotoxic activity against L1210 cells being equivalent to 4.2$\mu\textrm{g}$ and 1.8$\mu\textrm{g}$ from the crude garlic and active fraction, respectively. The Rf value of the active fraction on silica-gel TLC was 0.18 in condition that petroleum ether/ethyl ether/acetic acid mixture(90:10:1, v/v/v) was used as a developing solvent. The survival times of mice inoculated with S-180 cells were extended about 1.5 to 2 times in the groups treated with garlic extract(through i.p. and oral administration) compared with their control group(no garlic extract treatment). Observations were carried out on S-180. Ethanol extracts of garlic injured markedly tumor cells within 3 hours after injection.
