Effect of N-3, N-6 Fatty Acid and d-Limonene Treatment on Membrane Lipid Composition and Protein Kinase C Activity in Experimental Rat Hepatocarcinogenesis (쥐의 간 발암과정에서 N-3, N-6 지방산 섭취 및 d-Limonene 투여가 생체막 지질조성 및 Protein Kinase C 활성도에 미치는 영향)

  • This study was done to investigate the effects of n-3, n-6 fatty acid and d-limonene on the hepatic membrane lipid composition, protein kinase C (PKC) and glutathione S-transferase (GST) activities in experimental rat hepatocarcinogenesis. Sprague-Dawley female rats were fed with two different types of dietary oil for 20 weeks. Corn oil (CO) and sardine oil (SO) were used at 15% by weight as a source of n-6 and n-3 fatty acid, respectively. One week after feeding, rats were intraperitoneally injected twice with a dose of diethylnitrosamine (DEN, 50 mg/kg body weight) and after 1 week 0.05% phenobarbital (PB) was provided with drinking water. Membrane fractional lipid composition showed that the content of cholesterol was higher in 50 group than CO group and also significantly decreased by d-limonene. The content of phospholipid was increased by carcinogen treatment but not affected by dietary oils or d-limonene. Membrane C/PL molar ratio was significantly decreased by d-limonene or carcinogen treatment in 50 groups but not in CO groups. Fatty acid composition was changed by dietary oils but not by carcinogen treatment or d-limonene. Cytosolic PKC activity was not significantly different by dietary oils, d-limonene or carcinogen treatment. However, membrane PKC activity was significantly increased by carcinogen treatment and decreased by d-limonene. Cytosolic GST activity was affected by d-limonene or carcinogen treatment in all dietary groups. These data indicate that dietary oils, d-limonene and carcinogen treatment can not change much membrane phospholipid composition. But membrane C/PL molar ratio was changed by carcinogen treatment and d -limonene although the effect was different between dietary oils. Therefore, it is suggested that different dietary oils and d-limonene can somewhat modulate the changes of membrane fluidity and activities of membrane bound enzymes like membrane associated PKC during carcinogenesis.

  • 호우는 우리나라에 매년 약 60여명의 인명과 6,000억원에 달하는 재산 손실을 가져오는 가장 파괴적인 자연 재해이다. 그러므로, 강수 행태의 변화, 특히 호우 빈도와 규모의 변화를 이해하는 것은 악기상과 관련된 재해를 최소화하는데 필요한 환경영향평가에 필수적이다. 최근 기후변화의 징후로써 뿐만 아니라 사회에 미치는 영향 때문에 강수 극값에 대한 관심이 크게 증가하고 있다. 기후변화가 사회에 영향을 미치게 될 규모는 기후변동성의 변화, 특히 기후극값의 강도와 빈도에 의해서 결정 될 것이다. 본 연구의 목적은 한반도 남부지방의 장기간의 강수강도와 극값의 변화 경향을 파악하는 것이다. 장기간의 일 강수 자료(1920-1999)를 보유하고 있는 대구, 전주, 부산, 목포의 자료를 산술 평균하여 남부지방의 지역 일 강수 계열을 구축하였다. 남부지방의 연강수일수는 뚜렷한 감소현상이 나타나는 반면 연강수량은 약한 증가현상을 보인다. 이로 인해서 강수일수당 강수량을 보여주는 강수강도는 뚜렷하게 증가하고 있다. 이 모든 경향은 통계적으로 유의한 수준의 변화이다. 계절별로는 여름의 강수량 증가와 가을의 강수일 수 감소가 가장 뚜렷하다. 또한 강수극값의 규모를 나타내는 90번째, 95번째, 99번째 백분위수의 값도 증가하는 경향을 보여주고 있다. 호우 사상의 발생빈도와 그에 의한 강수량은 증가하는 반면, 비호우 사상의 발생빈도는 감소하고, 그에 의한 강수량에는 뚜렷한 경향이 나타나지 않았다. 남부지방에 나타난 강수일수의 감소는 비호우 사상의 감소에 의한 것이었고, 강수량의 증가는 호우 사상의 발생빈도와 그에 의한 강수량 증가에 의한 것 임을 알 수 있다. 위의 결과는 한반도에서도 기후변화에 의한 수문순환의 강화를 나타나고 있음을 보여주는 것으로, 기후변화에 대응하기 위한 적절한 지역영향평가를 위해서는 강수 극값에 대한 보다 상세한 분석이 필요함을 제안하고 있다.

Effects of Eriobotrya japonica Lindl. (Loquat) Leaf Ethanol Extract on Cholesterol and Antioxidative Activity in Rats Fed a High-Fat/High-Cholesterol Diet (비파잎 에탄올 추출물이 고지방-고콜레스테롤 식이를 급여한 흰쥐의 콜레스테롤 저하 및 항산화 활성에 미치는 영향)

  • We investigated the effects of an ethanol extract of Eriobotrya japonica Lindl. (loquat) leaves (EJ) on the lipid metabolism of serum, liver, and adipose tissue, and antioxidative activity in rats fed a fat/cholesterol diet for four weeks. Male Sprague-Dawley rats weighing 207 g were divided into 4 groups: a normal diet group (N), a high-fat/high-cholesterol diet group (HFC), a high-fat/high-cholesterol diet group administered 200 mg/kg day EJ (HFC-EJL), and a high-fat/high-cholesterol diet group administered 400 mg/kg/day EJ (HFC-EJH). The serum ALT and AST activities of the EJ groups were lower than those of HFC group, but there was no significant change in serum ALP or LDH activities. The serum total and LDL-cholesterol, atherogenic index, and cardiac risk factor tended to be decreased in the EJ groups compared to the HFC group, while the serum HDL-cholesterol decreased in the HFC group and increased only minimally in the EJ groups. The total cholesterol in liver and mesenteric adipose tissues was lower in the EJ groups than in the HFC group. Triglycerides in the mesenteric and epididymal adipose tissues were lower in the EJ groups than in the HFC group. The liver GSH levels of the EJ groups were significantly lower than the HFC group. The liver TBARS content was significantly lower in the EJ groups than in the HFC group. These results suggest that EJ ethanol extract may improve the lipid metabolism of serum, liver, and adipose tissue and prevent oxidative stress by stimulating antioxidative systems in rats fed a high-fat/high-cholesterol diet.

The Effect of Nutrition Education on Visceral Fat Reduction and Diet Quality in Postmenopausal Women (폐경 여성의 내장지방 및 식사의 질에 미치는 영양 교육의 효과)

  • This study investigated the effects of the nutrition education on body weight, visceral fat and diet quality in the postmenopausal women. The subjects (n = 101) were randomly divided into two groups: Nutrition education + Exercise (NEE) group (n = 51) and Exercise only (EO) group (n = 50). Nutrition education was consisted of counseling in portion control, food selection for low carbohydrate, high fiber food items and for the improvement in micronutrient intakes and diet quality. After 6 months, the reduction in the body weight and visceral fat area was significantly greater in the NEE than in the EO group. The NEE subjects were further divided into two groups according to the amount of visceral fat area reduction; high visceral fat area loss (HVL) group with a visceral fat area reduction 2.35% or greater and low visceral fat area loss (LVL) group with a reduction less than 2.35%. In the HVL group, the reduction in body weight, BMI, percent body fat, waist to hip ratio and visceral fat area was significantly greater than that in the LVL group. We observed a significant increase in the serum HDL-cholesterol level and a decrease in systolic blood pressure, fasting blood sucrose, total and LDL-cholesterol levels in the HVL group compared to the LVL group. The energyadjusted protein, fiber, calcium, vitamin $B_6$, vitamin C, vitamin E intakes were significantly increased in the HVL compared to LVL group. The index of nutritional quality (INQ) and mean adequacy ratio (MAR) were also increased in the HVL group compared to the LVL group. These results show that our nutrition education program was an effective intervention measure for the reduction of body weight and visceral fat, blood pressure, glucose and lipid levels in the blood and also for the improvement of nutrient intake and diet quality in postmenopausal women who are overweight.

Effect of Addition of Fractionated Milk Fats on Fat Composition and Melting Behavior of Cocoa Butter (유지방 획분 첨가가 코코아 버터의 지방조성과 용융거동에 미치는 영향)

  • The effect of addition of fractionated milk fats on the composition and melting behavior of cocoa butter was investigated. High melting fraction (HMF) of milk fat fractions had the highest contents of long chain fatty acid $(C16{\sim}C18)$ and saturated fatty acid followed by medium melting fraction 1 (MMF1), medium melting fraction 2 (MMF2), anhydrous milk fat (AMF), and low melting fraction (LMF) in a decreasing order. MMF2 had the highest contents of the short chain fatty acid $(C4{\sim}C10)$ and medium chain fatty acid $(C12{\sim}C14)$ followed by AMF, HMF, MMF1, and LMF in a decreasing order. When the fractionated milk fats were added to cocoa butter, the long chain fatty acid contents increased with increasing the ratio of fractionated milk fats. The saturated fatty acid contents decreased only when the LMF was added. The higher content of long chain triglyceride and the lower contents of short chain triglyceride and medium chain triglyceride were obtained from the fractionated milk fat of higher melting point. When the fractionated milk fats were added to cocoa butter, long chain triglyceride contents decreased with increasing the ratio of the fractionated milk fats. The melting points of cocoa butter, AMF, HMF, MMF1, MMF2, LMF were $33.3^{\circ}C,\;31.2^{\circ}C,\;40.6^{\circ}C,\;37.4^{\circ}C,\;33.5^{\circ}C$, and $6.5^{\circ}C$, respectively. Cocoa butter had the highest content of solid fat followed by HMF, MMF1, MMF2, AMF, and LMF in a decreasing order. When the fractionated milk fat was added to cocoa butter at various temperatures, the solid fat content in the mixture of fractionated milk fat and cocoa butter decreased with increasing the ratio of fractionated milk fat. This results suggested that anhydrous milk fat and fractionated milk fats had a good compatibility with cocoa butter.

Effect of Brassica rapa Sprouts on Lipid Metabolism in Rats Fed High Fat Diet (순무 싹이 고지방식이를 급여한 흰쥐의 지질대사에 미치는 영향)

  • This study was performed to investigate the effect of Brassica rapa (BR) sprouts on weight reduction and cholesterol-lowering action in rats fed high fat diet for 4 weeks. Weight-matched male Sprague-Dawley rats were assigned to four groups according to dietary fat levels (10% or 20% of diet wt.). Experimental groups were normal diet group (N), high fat diet group (HF), high fat diet with 5% BR sprouts powder group (HF-BRL), and high fat diet with 10% BR sprouts powder group (HF-BRH). The body weight gain was increased in HF group, but gradually decreased to the corresponding level of the N group fed BR sprouts powder. The concentrations of serum LDL-cholesterol, atherogenic index and cardiac risk factor tended to decrease in the BR sprouts powder fed groups compared with the HF group. However, HDL-cholesterol concentration in serum decreased in the HF group and markedly increased in the BR sprouts powder fed groups. Concentrations of triglyceride and total cholesterol in liver were also markedly decreased in the BR sprouts powder fed groups. Triglyceride concentrations of epididymal and mesenteric adipose tissues in the BR sprouts powder fed groups were also decreased compared with the HF group. These results indicate that BR sprouts powder may reduce fat accumulation and body weight, and have cholesterol-lowering effect.

Changes in Amino Acids and Fatty Acids Composition during Germination of Rapeseed (유채(油菜) 발아중(發芽中) 아미노산과 지방산 조성(組成)의 변화(變化))

  • Rapeseeds (Brassica napus L.)were germinated at $25^{\circ}C$ for 60 hours under dark condition in order to investigate the compositional changes in amino acids and fatty acids. The weight loss was most significant after 30 hours of germination and root growth was observed from that time. Analysis showed that Glu, Lys and Asp were the major amino acids and Ile was the limiting one. Germination caused an increase in Lys, Thr, Val and Leu while Met and Phe decreased among essential amino acids, resulting Met to be the limiting amino acid. The major fatty acids were oleic (49.3%), linoleic (22.0%) and eicosenoic (10.5%) acids. The decrease in oleic and stearic acids was measured while linoleic, linolenic, eicosenoic and erucic acids showed initial decrease followed by gradual increase.

Inhibitory Effects of Allium sacculiferum Max. Methanol Extracts on ROS Production and Lipid Accumulation during Differentiation of 3T3-L1 Cells (참산부추(Allium sacculiferum Max.) 메탄올 추출물의 지방세포 내 ROS 생성 및 지질 축적 억제 효능)

  • Allium sacculiferum Max. (ASM) is a perennial plant of the Liliaceae family and grows over the entire regions of Korea. Obesity is a serious health problem worldwide and has currently become a prevalent chronic disease. Adipocytes produced by preadipocyte differentiation during adipogenesis and adipocytes combined with abnormal accumulation cause obesity. Recently, intracellular reactive oxygen species (ROS) were shown to accelerate lipid accumulation in 3T3-L1 cells. In this study, we investigated the effects of ASM methanol extracts on ROS production and lipid accumulation in 3T3-L1 adipocytes. Our results indicate that the 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity of ASM methanol extracts increased in a dose-dependent manner. ASM methanol extracts suppressed ROS production and lipid accumulation during adipogenesis. In addition, ASM methanol extracts inhibited the mRNA expression of both pro-oxidant enzymes such as glucose-6-phosphate dehydrogenase as well as the transcription factors, including sterol regulatory element-binding proteins 1c, peroxisome proliferator-activated receptor ${\gamma}$, and CCAAT/enhancer-binding protein ${\alpha}$. Our results suggest that ASM methanol extracts inhibit ROS production and lipid accumulation by controlling ROS regulatory genes and adipogenic transcription factors. Thus, ASM has potent natural antioxidant, anti-adipogenic properties and have potential in the development of a potent anti-obesity agent.

Effect of Acacia catechu Extract on 3T3-L1 Preadipocyte Differentiation (지방세포의 분화에 미치는 Acacia catechu 추출물의 항비만 효과)

  • The purpose of this study was to investigate the effects of catechu water extract on adipogenesis in 3T3-L1 adipocytes. 3T3-L1 preadipocytes were differentiated with adipogenic regents by incubation for 9 days in the absence or presence of catechu extract ranging from $1{\sim}200{\mu}g/mL$. The effect of catechu extracts on cell proliferation of 3T3-L1 preadipocytes was investigated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. The effect of catechu extracts on 3T3-L1 differentiation was examined by measuring intracellular lipid droplet and triglyceride contents. These results were obtained from preadipocyte proliferation and adipocyte differentiation of 3T3-L1. Catechu extracts inhibited lipid accumulation and remarkably decreased triglyceride contents in 3T3-L1 preadipocytes at a concentration showing no cytotoxicity. The anti-adipogenic effects of catechu appeared to be mediated by significant down-regulation of expression of peroxisome proliferator-activated receptor ${\gamma}$, CCAAT/enhancer-binding protein ${\alpha}$, and sterol regulatory element-binding protein 1c proteins apart from expression of hormone-sensitive lipase. We suggest that catechu extracts significantly inhibit adipogenesis and can be used for regulation of obesity.

Development of Polyclonal Antibodies to Abdominal and Subcutaneous Adipocyte for Reducing Body Fat of Rat (흰쥐 체지방 감소를 위한 지방부위별 특이 다클론 항체 개발 연구)

  • Polyclonal antisera against regional (abdominal and subcutaneous) fats were developed to reduce body fat in rats. Isolation and culture of abdominal and subcutaneous adipocytes of rats were performed for analyzing lactate dehydrogenase (LDH) concentration. At the level of 1:1,000 dilution, little antibody reactivity appeared in non-immunized serum whereas both of antisera against abdominal (AAb) and subcutaneous adipocyte plasma membrane proteins (SAb) had relatively strong reactivity till the level of 1:128,000 dilutions. Compared with regional fats, extremely low reactivities of AAb and SAb were detected with PMP of the organs (p<0.001). Both AAb and SAb were most strongly reacted with each adipocyte plasma memebrane proteins and showed statistically (p<0.01) higher cross-reactivities compared with non-immunized serum based on LDH analysis. In conclusion, these results may indicate that the present polyclonal antibodies against regional inedible adipocyte plasma membrane proteins are well developed and have safety in cross-reactivities with body organs.
