스테로이드 치료 전 DHEA 단독투여와 DHEA 투여와 운동의 동시적용이 스테로이드에 의해 유발되는 쥐 뒷다리근의 위축 예방에 미치는 효과 (Effect of DHEA Administration Alone or Exercise combined with DHEA before Steroid Treatment on Rat Hindlimb Muscles)

  • Purpose: The purpose of this study was to determine the effect of Dehydroepiandrosterone(DHEA) administration alone or exercise combined with DHEA before steroid treatment on rat hindlimb muscles. Methods: Male Sprague-Dawley rats were assigned to one of three groups: a steroid group(S, n=10) that had no treatment for 7 days before steroid treatment; a DHEA-steroid group(DS, n=8) that had 0.34 mmol/kg/day DHEA injection once a day for 7 days before steroid treatment and an exercise+DHEA-steroid group(EDS, n=9) that ran on the treadmill combined with 0.34 mmol/kg/day DHEA injection for 7 days before steroid treatment. At 15 days all rats were anesthetized and soleus, plantaris and gastrocnemius muscles were dissected. Body weight, food intake, muscle weight, myofibillar protein content and cross-sectional area of the dissected muscles were determined. Results: The DS group showed significant increases(p<.05) as compared to the steroid group in body weight, and muscle weight of gastrocnemius muscles. The EDS group showed significant increases(p<.05) as compared to the S group in body weight, muscle weight, myofibrillar protein content, and Type II fiber cross-sectional area of soleus, plantaris and gastrocnemius muscles. Conclusion: Exercise combined with DHEA administration before steroid treatment prevents steroid induced muscle atrophy, with exercise combined with DHEA administration being more effective than DHEA administration alone in preventing muscle atrophy.

스테로이드 치료 후 회복기의 규칙적인 운동이 쥐의 스테로이드 유발성 위축 뒷다리근에 미치는 영향 (Effect of regular exercise during recovery period following steroid treatment on the atrophied hindlimb muscles induced by steroid in rats)

  • This study was conducted to determine whether low intensity regular exercise following steroid treatment could attenuate steroid-induced muscle atrophy. Thirty-eight Sprague-Dawley rats weighing $165{\sim}175g$ were divided into six groups ; control group(C), dexamethasone administration group(D), sedentary normal saline administration group(C+Se), exercise after normal saline administration group(C+Ex), sedentary group after dexamethasone administration(D+Se), exercise group after dexamethasone administration(D+Ex). Either dexamethasone(5mg/kg) or normal saline was injected for 7days accordingly. Exercise was started at 10m/min on the $10^{\circ}$ grade treadmill and gradually increased up to 15m/min by the 7th day for 60minutes/day($20min{\times}3$). The data were analyzed by Kruskal-Wallis test and Mann-Whitney U test using the SPSS WIN 9.0 program. Body weight, muscle weight and myofibrillar protein content of both plantaris and gastrocnemius, Type I, II muscle fiber cross-sectional area of plantaris, and Type II muscle fiber cross-sectional area of gastrocnemius in D group were significantly lower than those of C group(p<0.05) respectively. Hindlimb muscle weight, myofibrillar protein content of both plantaris and gastrocnemius. Type I muscle fiber cross-sectional area of soleus and Type I, II muscle fiber cross-sectional area of plantaris in D+Ex group tended to increase compared to those of D+Se group. Myofibrillar protein content of both plantaris and gastrocnemius, Type I muscle fiber cross-sectional area of plantaris in D+Ex group tended to increase compared to those of C+Se group. Based on these results, it is suggested that regular low-intensity exercise during recovery period after steroid treatment might facilitate the recovery from steroid-induced muscle atrophy.

시스플라틴에 의해 유발된 식욕부진이 쥐의 뒷다리근에 미치는 영향: 저용량 연속투여요법과 고용량 투여요법 간의 비교 (A Comparison between Effects of Anorexia Induced by Consecutive Low-Dose Cisplatin and High-Dose Cisplatin on Hindlimb Muscles of Rats)

  • Purpose: The purpose of this study was to compare the effects of anorexia induced by consecutive low-dose and high-dose of cisplatin (CDDP) on the hindlimb muscles of rats. Methods: Male Sprague-Dawley rats were assigned to three groups: Control group (C) received a saline (the same dose and duration as the low CDDP group), the high-dose cisplatin (High CDDP) group received a single 5 mg/kg dose of cisplatin, the consecutive low-dose cisplatin (Low CDDP) group had 1 mg/kg of cisplatin administered for five consecutive days. On the 8th day the soleus and gastrocnemius muscles were dissected. Body weight, food intake, activity, muscle weight, Type I, II fiber cross-sectional area (CSA) of the dissected muscles were measured. Results: Body weight, food intake, muscle weight and Type I, II fiber CSA of the High CDDP and Low CDDP groups were significantly less than the C group. The High CDDP group showed significant decreases, compared to the Low CDDP group, in body weight, food intake, activity score, muscle weight and Type I, II fiber CSA. Conclusion: Hindlimb muscle atrophy occurs due to anorexia induced by both consecutive low-dose and high-dose cisplatin. The muscle atrophy induced by consecutive low-dose cisplatin is less apparent than high-dose cisplatin.

단기간의 영양 섭취 저하가 쥐 뒷다리근에 미치는 영향 (Effect of Short-term Undernutrition on Hindlimb Muscles in Rats)

  • Purpose: The purpose of this study was to examine the effect of short-term undernutrition on muscle weight and Type I and II fiber cross-sectional area of hindlimb muscles in undernourished rats. Methods: Adult male Sprague-Dawley rats were randomly assigned to one of two groups: The undernourished (UN) group (n=9) and the control (C) group (n=9). A control group was allowed to have water and pellet ad libitum for 5 days. Undernutrition was induced by providing 32% of total intake of the control group for 5 days. Body weight of two groups and food intake of the control group were measured every day. At 6 days all rats were anesthetized and soleus, plantaris and gastrocnemius muscles, and liver were dissected. Body weight, food intake, muscle weight, liver weight and cross-sectional area were determined. Results: The UN group at 6 days after undernutrition showed significant decreases, as compared to the control group in body weight, liver weight, muscle weight of soleus, plantaris, and gastrocnemius, and Type I fiber cross-sectional area of soleus and gastrocnemius muscles and Type II fiber cross-sectional area of plantaris and gastrocnemius muscles. Conclusion: Hindlimb muscle atrophy occurs from the short-term undernutrition.

말초신경 손상에 의한 신경병증성 통증으로 유발된 쥐 뒷다리근 위축 (Hindlimb Muscle Atrophy of Rat Induced by Neuropathic Pain)

  • Purpose: The purpose of this study was to examine the effect of neuropathic pain by peripheral nerve injury on mass and Type I and II fiber cross-sectional areas on hindlimb muscles of the neuropathic pain model rat. Method: Adult male Sprague-Dawley rats (body weight 200-220 g) were assigned to one of two groups: a neuropathic pain group (n=7) that had a ligation of the left L5 spinal nerve, a control group (n=5), a naive rat without any procedures. Withdrawal threshold, activity, body weight and food intake were measured daily. At 8 days after neuropathic pain, all rats were anesthetized and the soleus and plantaris muscles were dissected from the both hindlimbs. Body weight, food intake, muscle weight and Type I and II fiber cross-sectional area of the dissected muscles were determined. Result: The neuropathic pain group showed a significant decreases (p<.05) as compared with the control rats, in diet intake, body weight, muscle weight and Type II fiber cross-sectional area of the left (affected side) soleus and plantaris muscles, and the right (unaffected side) muscle weight of plantaris and Type II fiber cross-sectional area of the soleus muscle. Conclusion: The hindlimb muscle atrophy occurs in both affected and unaffected side due to neuropathic pain by the peripheral nerve injury. The hindlimb muscle atrophy of the affected side is more pronounced than that of the unaffected side.

저강도 격일 운동과 매일 운동이 쥐 뒷다리근에 미치는 근 비대 효과 비교 (Comparison of Hypertrophic Effects of Low-Intensity Exercise on Rat Hindlimb Muscles between Every Other Day Exercise and Everyday Exercise)

  • Purpose: The purpose of this study was to compare the hypertrophic effects of low-intensity exercise on weight, myofibrillar protein content and Type I, II fiber cross-sectional area of hindlimb muscles of rats between every other day exercise and every day exercise. Methods: Adult male Sprague-Dawley rats were assigned to 1 of 3 groups: control group (C, n=6), experimental group 1 (E1, n=7) and experimental group 2 (E2, n=7). Rats in E1 group had 7 sessions (every other day) and those in E2 group had 14 sessions (every day) of exercise in which they ran on a treadmill for 30 min/day at 10 m/min. Results: Muscle weight, cross-sectional area of type I fiber and myofibrillar protein content of soleus and myofibrillar protein content of plantaris in E1 group, and myofibrillar protein content of soleus and cross-sectional area of type I fiber of plantaris in E2 group were greater than those in C group. Cross-sectional area of type I fiber of soleus of E1 group was higher than E2 group while cross-sectional area of type I fiber of plantaris of E2 group was higher than E1 group. Conclusion: Hypertrophy of hindlimb muscles occurs from every other day exercise similar to every day exercise.

쥐에서 이중에너지 방사선 흡수법을 이용한 골밀도의 측정: 다양한 골부위에서 재위치 여부에 따른 생체내 측정의 정밀도 (Bone Mineral Density Measurement of Rats Using Dual-energy X-ray Absorptiometry: Precision of In Vivo Measurements for Various Skeletal Sites with or without Repositioning)

  • 목적: 살아있는 쥐를 실험대상으로 DXA를 이용하여 검사사이마다 자세변화가 있는 경우와 자세변화 없는 경우에 다양한 부위의 골밀도를 반복적으로 측정하여 각 개체의 자세 변화와 측정부위에 따른 정밀도에 차이가 있는지를 평가하고자 하였다. 대상 및 방법: 실험은 총 12마리의 수컷 쥐를 대상으로 하였다. 아이소후루란(isoflurane)을 이용한 흡입 마취하에 DXA 기기와 기기에 내장되어 있는 소동 물 프로그램(software)를 이용하여 쥐들을 스캔하였다. 스캔 후 얻어진 영상에서 오른쪽 뒷다리(hind limb), 요추(lumbar spine), 두개골(skull) 그리고 골반골(pelvic bones)에 각각 관심영역(ROI)을 그려서 골밀도($g/cm^2$) 값을 구하였다. 실험에 포함된 12마리 쥐 모두에 대해서는 처음 스캔 테이블에 올린 자세를 유지한 상태 그대로 반복해서 4번씩의 스캔을 시행하였다(그룹 1a). 그리고 일주일 후에는 12마리 중 6마리에 대해서만 검사를 시행하였고 한번의 스캔 후 쥐를 테이블에서 완전히 내렸다가 다시 스캔 테이블로 올려서 자세를 잡고 스캔을 반복하는 방식으로 스캔 사이사이마다 새롭게 자세를 잡고 검사를 시행하였다(그룹 2). 자세변화유무와 상관없이 각각의 그룹 내에서의 부위별 정밀도의 차이는 Kruskal-Wallis one-way analysis of variance(ANOVA) test로 비교하였으며 자세변화에 따른 그룹간에 부위별 정밀도의 차이는 그룹 1의 12마리 중 그룹 2에 해당하는 6마리(그룹 1b)와 그룹 2를 Wilcoxon Signed Rank Sum Test로 비교하였으며 얼마나 일관성이 있는지 Bland Altman 도표분석을 하였다. 결과: 각각의 그룹 내에서 측정 부위에 따라 정밀도의 차이는 그룹 1a는 유의한 차이가 없었으나(p=0.657) 그룹 2의 경우 정밀도에 유의한 차이가 있었다(p=0.003). 그룹간에 부위별 정밀도는 차이는 오른쪽 뒷다리가 p=0.173, 제1-4 요추가 p=0.600 두개골이 p=0.046 그리고 골반골이 p=0.075로 두개골을 제외한 모든 부위에서 검사간 자세변화 따른 정밀도의 차이가 통계적으로 유의하지 않았다. 또한 Bland Altman도표에서 자세변화에 따른 두 그룹의 부위별 측정값의 차이에 치우침은 두개골을 제외한 모든 부위에서 없었으며 차이의 2표준편차는 오른쪽 뒷다리가 2.3%, 제1-4요추가 5.9%, 두개골이 4.4% 그리고 골반골이 3.0%이었다. 결론: 본 연구에서 재위치를 시행한 그룹 내에서 부위에 따른 정밀도의 차이는 나타났지만 재위치 유무에 따른 각 부위의 정밀도는 두개골을 제외한 나머지 부위들에서는 유의한 차이를 없었다. 결론적으로 본 연구의 결과는 DXA는 쥐와 같은 살아있는 소동물에서 다양한 부위에 대해 반복적으로 골밀도 값을 측정하고 추적관찰 할 때 자세의 재위치에 상관없이 충분한 정밀도를 가지고 있으며 골밀도를 평가하는데 있어서 매우 믿을만한 검사라는 것을 시사한다.

지구력 운동이 급성기 뇌졸중 쥐의 뒷다리근 질량에 미치는 영향 (Effect of endurance exercise during acute stage on hindlimb muscles of stroke induced rat)

  • The purpose of this study was to identify hindlimb muscle atrophy in stroke induced rat and determine the effect of endurance exercise on body weight, weight of hindlimb muscle during 7 days after stroke induction. Thirty four male Sprague-Dawley rats with 200-270g body weight were divided into four groups : control, control+exercise(Con+Ex), stroke, and exercise after stroke(St+Ex) group. The control group and Con+Ex group received sham operation and the stroke group and St+Ex group received right MCA occlusion operation by using silicon-coated probe. The Con+Ex and St+Ex groups ran on a treadmill for 20min/day at 10m/min and $10^{\circ}grade$. Daily body weight and diet intake were measured every morning for 7 days. Cerebral infarction of stroke and St+Ex groups were identified by staining with TCC for 30minutes. The data were analyzed by Kruskal-Wallis test and Mann-Whitney U test using the SPSSWIN 9.0 program. Body weight of the control group at the 7th day increased by 18.3% significantly from the first day of experiment, that of the stroke group at the 7th day decreased by 6.7% significantly compared to the day of receiving right MCA occlusion operation. Body weight of the Con+Ex group at the 7th day increased by 10.3% significantly form the first day of experiment, that of St+Ex group at the 7th day also increased by 13.4% significantly compared to the day of receiving right MCA occlusion operation. The total amount of diet in stroke group decreased significantly compared to that of St+Ex and that of control group. In stroke group the wet weight of both sides of soleus, plantaris, and gastrocnemius muscles decreased significantly compared to that of control group. The relative weight of affected(left) plantaris and gastrocnemius muscles decreased significantly compared to that of the control group. The difference between the weight of affected and unaffected soleus, plantaris, and gastrocnemius muscles were not significant in stroke group. The wet weight of right gastrocnemius muscles in Con+Ex group increased compared to that of control group. The relative weight of right gastrocnemius muscle increased significantly compared to that of the control group. The wet weight of St+Ex group increased significantly compared to that of the stroke group in both sides of soleus, plantaris, and gastrocnemius muscles. The relative weight of affected plantaris muscle increased significantly compared to that of the stroke group. The difference between the weight of affected and unaffected soleus, plantaris, and gastrocnemius muscles were not significant in St+Ex group. Body weight and wet weight of soleus, plantaris, and gastrocnemius muscles in the St+Ex group did not recover to the values of control group. Based on these results, it can be suggested that endurance exercise during acute stage of stroke can reduce muscle atrophy related to denervation, inactivity and undernutrition.

  • The purpose of this study was to determine the effect of endurance training prior to occurrence of muscle atrophy on the mass, myofibrillar protein content and fiber crossectional area of atrophied hindlimb muscles of rats. Adult female Wistar rats were trained prior to occurrence of muscle atrophy induced by hindlimb suspension. Training began on the 1st day for 10min /day at 15m /min on a 0% grade, training exercise increased daily in time and intensity so that by the 4th week rats were running 60min /day, at 34m /min on a i3.5% grade. Wet weight and relative weight of soleus, plantaris and gastrocnemius muscle decreased significantly after seven days of hindlimb suspension. Wet weight and relative weight of soleus tended to increase and that of plantaris and gastrocnemius tended to decrease in the exercise group as compared to the control group. Myofibrillar protein content of soleus and gastrocnemius tended to increase and that of plantaris tended to decrease in the endurance trained group as compared to the control group. Fiber crossectional area of Type I, II fiber in soleus and plantaris muscle tended to increase in the exercise group as compared to the control group. Wet weight and relative weight of soleus. plantaris and gastrocnemius decreased significantly, myofibrillar protein content of soleus, plantaris and gastrocnemius increased in hindlimb suspended rats following endurance training as compared to the control group. There was no change in fiber type percentage and crossectional area of type I and II fiber in soleus muscle and that of type I and IIfiber in plantaris muscle decreased in the hindlimb suspended rats following endurance training as compared to the control group. Wet weight and relative weight of soleus and plantaris tended to increase, that of gastrocnemius increased significantly, myofibrillar protein content of soleus and plantaris muscle increased significantly and that of gastrocnemius tended to increase in the hindlimb suspended rats following endurance training as compared to sedentary rats following endurance training. Crossectional area of type I fiber of soleus muscle tended to increase. that of type I fiber of plantaris muscle increased significantly and that of type II fiber tended to increase in hindlimb suspended rats following endurance training as compared to sedentary rats following endurance training. The results suggest that endurance training prior to occurrence of muscle atrophy can attenuate the decrease of mass, myofibrillar protein content and fiber crossectional area induced by hindlimb suspension.

  • Inpatients are mostly occupied in bed with restricted activity, nearly all patient populations are at risk for the occurrence of skeletal muscle atrophy due to decreased level of activity. Restriction of mobility is far greater in pediatric patients compared with adult patients since almost all the activities of daily living is performed by parents or caregivers. It could be assumed that pediatric patients are more vulnerable to skeletal muscle atrophy than adult patients, however, there have been no attempts to reduce the atrophy of developing muscle. Therefore it is important to determine the effect of exercise in developing muscle during decreased activity. The purpose of this study was to determine the effect of periodic weight support during hindlimb suspension on the mass and cross-sectional area of Type I and II fibers in developing soleus(Type I ) muscle. To examine the effectiveness of periodic weight support activity in maintaining mass and fiber size. the hindlimb of young female Wistar rats was suspended(HS) and half of these rats walked on a treadmill for 45min / day(15min every 4h) at 5m / min at a 15 grade(HS-WS). After 7days of hindlimb suspension, soleus wet weight was 28. 57% smaller and relative soleus weight was 28. 21% smaller in comparison with con-trol rats (p〈0.05) Soleus wet weight and relative soleus weight increased by 67.72% and 71.43% each with periodic weight support activity during hindlimb suspension (p〈0.01, p〈0.005), moreover soleus wet weight and relative soleus weight of the HS -WS rats were greater than those of the control group. No change was observed in fiber type percentage of the developing soleus muscle after 1 week of hindlimb suspension plus weight support activity. Type I and II fiber cross-sectional areas of the developing soleus muscle were 50.45% and 43.39% lower in the HS group than in the control group (p〈0.0001), type I and II fiber cross-sectional areas of the developing soleus were 24.49% and 29.93% greater in the HS - WS group than in the HS rats (p〈0.0001), whereas Type I and II fiber cross-sectional areas of HS - WS group were less than those of the control group, The results suggest that periodic weight support activity can ameliorate developing soleus muscle atrophy induced by hindlimb suspension, even in type II fibers that would not have been expected to be recruited by this type of neuromuscular demand. Clinical experimental study is needed to deter-mine the effect of periodic weight bearing exercise on developing atrophied leg muscle based on these results.
