Optimum Stocking Density of 3-year-old Pacific Abalone, Haliotis discus hannai Reared in Net Cage Culture (3년산 북방전복, Haliotis discus hannai의 해상가두리 양성 시 적정 수용밀도)

  • The effects of different stocking densities on the growth and survival rate of the 3-year-old pacific abalone, Haliotis dicus hannai were investigated in marine net cage for a year. Stocking densities in net cage ($2.4{\times}1.2m$) was set 15, 30, 45 and 60 percentage (= per)/sq m (square meter, $m^2$) with share to cross-sectional area per shelter. The water temperature during the testing period was $8.2^{\circ}C-22.1^{\circ}C$, and salinity is $33.5{\pm}0.6psu$, and dissolved oxygen is $7.87{\pm}0.86mg/L$. In the shell length (initial size : $71.50{\pm}2.28mm$) growth and shell breadth (initial size : $46.43{\pm}2.28mm$) of the test abalones, the absolute growth rate (ARG), daily growth rate (DGR) and specific growth rates (SGR) of the 15 per/sq m and 30 per/sq m were higher than those of 45 per/sq m and 60 per/sq m density group (P < 0.05). Also in the weight (initial weight : $35.7{\pm}8.1g$), it showed the same results. In survival rates, it were that 15 per/sq m and 30 per/sq m is significantly higher than 45 per/sq m and 60 per/sq m. Therefore, it was that the 15 per/sq m is optimized stocking density in marine net cages about the 3-year-old pacific abalone over 70 mm size. The result shown that total cross-sectional area under the shelter is based on 15 per/sq m ($2.4{\times}2.4m$, 354 number in a net cage) is suitable for fast growth and survival. But if the economy consider, optimized stocking density would be appropriate to accept 30 per/sq m ($2.4{\times}2.4m$, 710 number in a net cage).

The effect of environmental condition to the mycelial browning of Lentinula edodes (Berkeley) Sing. during sawdust bag cultivation (환경조건이 표고톱밥배지의 갈변에 미치는 영향)

  • Recently sawdust cultivation of Shiitake mushroom (Lentinula edodes ) is getting increased because log cultivation is getting difficult to get oak logs. It is important to make mycelia browning on the substrate surface in sawdust cultivation. This browned surface plays an important role like as artificial bark of the oak log, which protects the other pests and suppresses water evaporation in the substrate. The period for mycelia browning is so long that the sawdust cultivation of Shiitake mushroom can not spread well into the mushroom farms. In this article we would like to discuss about the effect of environmental condition to the mycelial browning during sawdust bag cultivation for the To reduce the period required for browning of substrates, sawdust substrates was illuminated light with difference intensity. One hundred Lux light illumination was needed for producing normal yield of fruit body but fruit body yield was low and abnormally shaped fruit body was produced when cultured under the dark condition of incubation. Illumination over 200lux is necessary for the successful browning of substrates during incubation. Optimum incubation temperature for browning of substrates and fruiting was $25^{\circ}C$. The treatment of cotton plug with different size to identify the effect of aeration on the browning of substrates and fruiting showed rapid mycelial growth and reduced the periods for browning as the size of cotton plug was bigger. However, yield of fruit body was the highest at 16mm diameter cotton plug as compared to 20mm of that. $CO_2$ content in vessel of substrates was low as the size of cotton plug was bigger during incubation. $CO_2$ content during incubation of substrate was highest in periods between 8 week and 14 week after inoculation of shiitake when substrate was changed color into brown. $C_2H_4$ content in vessel with substrates was highest at 8mm diameter cotton plug and it was increased by order of 12, 16, 20, 0, 4 mm diameter cotton plug during substrate incubation. Sawdust substrate was soaked in cold water for different time to identify soaking effect of sawdust substrate on fruit body yield and activities of enzymes in these substrates were investigated. The fruit body yield was increased up to 40% by soaking substrates in comparison with unsoaked substrates. The soaked substrates showed 165, 175g/1,000ml at treatment of 4 and 15 hours, respectively. Cellulose activities in soaked substrates were not changed with soaking time, but activities of laccase, lignin degradation enzyme, were drastically increased up to 4 times in comparison with unsoaked substrates.

Effect of Low Storage Temperature on Quality of Fresh Ginseng (저온저장 온도가 수삼의 품질에 미치는 영향)

  • To investigate optimum temperature for storage of fresh ginseng (Panax ginseng C. A. Meyer), the quality of the ginseng was compared during its storage at $-3^{\circ}C$, $-1.5^{\circ}C$ and $0^{\circ}C$. The deterioration rate of fresh ginseng stored at $-3^{\circ}C$ was the lowest for 8 weeks after storage. The rate was rapidly increased after that time and the rate at $-3^{\circ}C$ was higher than that of fresh ginseng stored at $-1.5^{\circ}C$ or $0^{\circ}C$ after the 12th week of storage. The deterioration severity of the fresh ginseng stored at $0^{\circ}C$ was much higher than that of the ginseng stored at $-1.5^{\circ}C$ and $-3^{\circ}C$. The weight loss of fresh ginseng ranged from 0.7---- to 1.6---- after 16th week; it was the lowest in the ginseng stored at $-1.5^{\circ}C$ and similar in fresh ginseng stored at $0^{\circ}C$ and $-3^{\circ}C$. The number of viable cells and molds in the fresh ginseng stored at $-3^{\circ}C$ was smaller than the fresh ginseng that was stored at other temperatures for 12 weeks, but did not differ with different storage temperatures after the 14th week of storage. The surface color of the fresh ginseng at $0^{\circ}C$ or $-1.5^{\circ}C$ was changed little while the discoloration of fresh ginseng at $-3^{\circ}C$ was relatively great. The electrolytic leakage from the rhizome of the fresh ginseng stored at $-3^{\circ}C$ was higher than that of the rhizome stored at $-1.5^{\circ}C$ and $0^{\circ}C$. The overall sensory quality of the fresh ginseng dropped below 3.0 in the S-point scale after the 10th week of storage at $-3^{\circ}C$ and after the 14th week of storage at $-1.5^{\circ}C$ and $0^{\circ}C$ (p<0.05).

Seed Germination, Plant Growth and Antioxidant Capacity of Limonium tetragonum under Different Salt Concentrations (염농도에 따른 갯질경(Limonium tetragonum) 종자의 발아와 식물체의 생장 및 항산화 활성)

  • Limonium tetragonum is a halophyte grown naturally in the coastal region in South Korea. This study was conducted to investigate the effects of salt concentrations on seed germination, seedling growth, and antioxidant capacity of L. tetragonum. Seeds were collected from naturally grown plants of L. tetragonum and those at full maturity were used in this experiment. All experiments were performed at 0%, 0.5%, 1.0%, or 2.0% of salt concentrations. Seed germination rate was highest as 86% at $20^{\circ}C$ and followed as higher in order of $25^{\circ}C$, $30^{\circ}C$ and $15^{\circ}C$. The germination rate was about 80% at 0% or 0.5% of salt concentration, but it was very low at the salt concentrations higher than 1%. Growth of L. tetragonum seedlings showed no difference in Hoagland solution containing NaCl in the range of 0% to 1.0% and seedlings survived at 2.0% of NaCl concentration. As the salt concentration increased, the content of $Na^+$ in the shoot increased, but that of $K^+$, $Ca^{{+}{+}}$, or $Mg^{{+}{+}}$ decreased. The antioxidant activity and the content of total polyphenol and total flavonoid in the shoot were similar at 0% and 0.5% of NaCl and were highest at 2.0% of NaCl concentration. In conclusion, performance of seed germination and plant growth of L. tetragonum was highest at 0% and 0.5% of NaCl concentration, and showed no difference in antioxidant activity, total polyphenol contents, and total flavonoid contents at the same salt concentrations.

Studies on the Storage of a New Rice Variety Tongil (수도신품종(水稻新品種) "통일(統一)"의 저장성연구(貯藏性硏究))

  • To investigate the preservability of Tongil, three types(rough, brown and milled rice) of Tongil and the common variety (being composed of typical varieties) in Korea were stored at a ambient and a low temperature $(15^{\circ}C)$ warehouse from April to October of each year for three years (1972-1974). The results obtained are described as follows; 1) Temperatures of atmosphere and the natural temperature warehouse changed in the same way. 2) Similar changes of relative humidity were observed in atmosphere and both warehouses. 3) Equilibrium moisture content of Tongil at $25^{\circ}C$ was 0.55 % lower in rough and brown rice. 0.33% lower in milled rice than each type of the common variety. 4) Weight loss rate in the natural temperature storage and in the low temperature storage was 1.35% and 0.74% after 6 month storage. 5) The change of fat acidity in browe rice was greater than any others (rough and milled rice). 6) Hardness of Tongil was 1.64kg higher than that of the common variety. 7) Germination rate was about the same during the low temperature storage, but that of brown of the natural temperature storage was abruptly decreased to $23%{\sim}37%$ after August. As a result, it is possible Tongil is applicable to the long-term storage of rice due to Tongil's superior preservability.

Elemental analysis of the fluoride varnish effects on root caries initiation (불소 바니쉬 도포 후 초기 치근 우식 발현에 대한 정량원소분석)

  • Objectives: The usage of fluoride varnish for a moderate to low caries-risk group has not been well validated. This study aimed to evaluate the preventive and therapeutic efficacies of fluoride varnish on the initiated root caries. Materials and Methods: Ten premolars were sectioned into quarters, further divided into two windows, one of which was painted with Fluor Protector (1,000 ppm fluoride, Ivoclar Vivadent). An initial lesion with a well-preserved surface layer was produced by pH cycling. Scanned line analysis using energy dispersive spectrometry determined the weight percentages of Ca and P in the demineralized layer. Scanning Electron microscopy and confocal laser scanning microscopy (CLSM) evaluated the varnish-applied root surfaces. Results: The mean lesion depth (SD) was 12.3 (2.6) ${\mu}m$ (single cycling) and 19.6 (3.8) ${\mu}m$ (double cycling). Double cycling extended the lesion depth, but induced no more mineral loss than single cycling (p < 0.05). The mean weight percentages of Ca and P between groups with and without varnish were not significantly different (p < 0.05). A CLSM showed varnish remained within 15 ${\mu}m$ of the surface layer. Conclusions: When a mild acid challenge initiated root tissue demineralization, the application of low-concentration fluoride varnish did not influence the lesion depth or the mineral composition of the subsurface lesion.

Dietary Effects of Post-fermented Green Tea by Monascus pilosus on the Body Weight, Serum Lipid Profiles and the Activities of Hepatic Antioxidative Enzymes in Mouse Fed a High Fat Diet (Monascus pilosus로 발효시킨 후발효 녹차가 고지방 식이 마우스의 체중과 혈청 지방함량 및 간 조직 항산화계 효소활성에 미치는 영향)

  • The anti-obese, hypolipidemic and hepatoprotective effects of post-fermented green tea by Monascus pilosus was tested with mice fed with high-fat diet for 7 weeks. The body weight gain and feed efficiency ratio (FER) in normal control group (NC), CHA (2% non-fermented green tea powder supplemented high-fat diet group) and mCHA (2% green tea powder post-fermented by M. pilosus supplemented high fat diet group) groups were significantly lower than those of high fat diet control group (HC). Epididymal fat weight in mCHA and NC were significantly lower than HC. The hepatic lipid peroxide was dramatically higher in HC than that of NC and was significantly lower in CHA and mCHA. In addition, dehydrogenase type activity of xanthine oxidoreductase in HC was lower than that of NC, but significantly higher than CHA and mCHA. In histopathological findings, hepatic fat accumulation in HC was higher than that of NC, CHA and mCHA. Antiobese, hypolipidemic and antifatty liver effect of green tea powder post-fermented by M. pilosus was slightly higher than that of non-fermented green tea. In conclusion, the constituents of green tea fermented by M. pilosus has been proven to not only inhibit obesity and hyperlipidemia but also decrease the hepatic fat accumulation in high fat diet-induced obese mice.

Mineralogical and Physico-chemical Properties of Sludge Produced During Artificial Sand Processing (국내 화강암류를 이용한 일부 인공쇄석사 제조과정에서 발생되는 슬러지의 광물.물리화학적 특성)

  • The consumption of artificially crushed sands exceeds more than 30 percent of the domestic sand supply in South Korea, and its rate is still increasing. For the manufacture of crushed sand granites and granitic gneisses are preferred, fine fractions (i.e. sludge, particles finer than 63 microns) are removed by use of flocculation agents, and its amount occupy about 15 wt%. The sludges consist of quartz, feldspars, micas, chlorite/vermiculite, kaolinites, smectites and occasionally calcite. Among the clay minerals micas are usually predominant, and $14{\AA}$ minerals, kaolinites and smectites are rather scarce. Jurassic granites usually contain more kaolinites and smectites than those of Cretaceous to Tertiary granites, probably due to longer geologic ages. On the other hand, sludge from Precambrian gneiss does not contain kaolinites and smectites. Chemical analyses for the granites and their sludges show rather clear differences in most of major chemical components. Except for $SiO_2,\;Na_2O\;and\;K_2O$, all other components represent rather clear increase. Decrease of $SiO_2$ content is attributed to the relative decrease of quartz in the sludges. And the $Na_2O decrease is caused by a relatively stronger weathering property of albite compared to Ca plagioclase. The $K_2O$ content shows rather small differences throughout the whole samples. The increases of $Al_2O_3$ and other major components resulted from weathering processes and most of colored components are also concentrated in the sludges. Particle size analyses reveal that the sludges are categorized as sandy loams in a sand-silt-clay triangular diagram. The sludge is now classified as industrial waste because of its impermeability, and this result was also confirmed by rather higher hydraulic conductivities. For the environmental problems, and accomplishing effective sand manufacture, more fresh rocks with little weathering products must be chosen.

Effects of Medicinal Enzyme Powder on Intestinal Mobility, Lipid Level, and Blood Parameters of Loperamide-Induced Constipation in Rats (약선효소 분말식이가 Loperamide의 투여로 유발된 변비 쥐의 장 운동과 지질 함량 및 혈액학적 성분 변화에 미치는 영향)

  • This study was aimed at investigating whether dietary therapy using medicinal enzyme powder is effective in reducing constipation caused by loperamide in rats. Nine-week-old male Sprague Dawley were subdivided into 4 groups: normal diet group (C), loperamide treatment and normal diet (CL), medicinal enzyme powder diet (E), and loperamide treatment and medicinal enzyme powder diet (EL). Constipation was induced by subcutaneous injection of loperamide (1.5 mg/kg) 3 days prior to sacrifice. The treatment with loperamide led to an increase in weight gain, a decrease in the number and wet weight of fecal pellets, and a decrease in intestinal motility. The administration of the medicinal enzyme powder significantly reduced weight gain but increased intestinal mobility compared with the loperamide-treated group. The treatment with loperamide in the normal diet group reduced the activities of both suggesting that constipation may be involved in the low level of glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase (GPT). Additionally, the loperamide treatment in the medicinal enzyme powder diet group increased the level of GOT, but reduced the level of GPT. Loperamide treatment also reduced cholesterol and increased the atherogenic index (AI) and cardiac risk factors (CRFs). Interestingly, the treatment with the medicinal enzyme powder effectively attenuated both the increase in AI and the reduction in high density lipopretein (HDL)-cholesterol, caused by the treatment with loperamide. Although there were no significant differences in the blood protein level, including hemoglobin and hematocrit, between the normal diet group and the loperamide-treated group, the administration of the medicinal enzyme powder to the loperamide-treated group effectively increased the levels of both hemoglobin and hematocrit. Collectively, the results demonstrate that the medicinal enzyme powder can help to combat the negative events caused by constipation.

Studies on the CA Storage of Sweet Persimmon in Polyethylene Film Pack (Polyethylene film포장(包裝)에 의(依)한 단감의 CA저장(貯藏)에 관(關)한 연구(硏究))

  • For development of long-term storage method of sweet-persimmons using polyethylene film bags, basic experiment was conducted with 30 boxes of sweet-persimmons in 1973 and the same experiment was extended for industrial application with 2,500 boxes of the persimmons in the cold storage of Jinyoung Sweet-persimmon Association in 1974. Investigation was made on change of the quality by storage period. At the same time, persimmons put in the cold storage test were shipped to market at different time in order to monitor consumer response and commercial feasibility. The followings are conclusion obtained from the result of this experiment. 1. Storage of sweet-persimmon, Buyu, produced in Jinyoung, Kimhae was possible for 1 month at $2^{\circ}C{\pm}1^{\circ}C$ cold storage. This storage period was extended to 4 months until the end of February in case that the fruits were hermetically sealed in P.E. film bags of 0.08 to 0.1 mm thickness. 2. During the storage period of sweet-persimmons packed in the film bags, the loss of weight due to evaporation was effectively prevented with use of the film of bag thicker than 0.04 mm. 3. The storage ability of 3-5 persimmons per small bag was somewhat superior to that of many persimmons packed in the large box of 15kg capacity. 4. The thicker the film of bags, the more $CO_2$ gas was accumulated inside, however, from 1 month after beginning of the storage the rate of $CO_2$ accumulation became very low maintaining the stabilized level of 5-6% at the plot of 0.06-0.08mm thick bags. 5. While the persimmons were in storage, decreased was the content of total sugars, total acids, and vitamin C, of which the phenomenon was remarkable especially with the fruits of non-packed plot. 6. The sweet-persimmons in the film bags subjected to cold storage when shipped to market in their intact condition were more beneficial than when they were shipped out in unpacked condition. The intact fruits packed in the P.E. film bags were able to keep their commercial value for 10days in the outdoor situation. 7. The sweet-persimmons that were packed in the film bags and put in the cold storage had maintained promissing marketability and the economic feasibility was acknowledged when the experimental practice was applied to industrial scale.
