The damage of plant growth and alteration of cellular tissues of barley infected by Barley yellow mosaic virus (BaYMV) was explored. The infected plots significantly damaged in all of measured factors by the disease. In severely diseased plant, the viral infection affected on plant growth like as shorten culm length about 25cm, 36% constrained ratio, comparing to healthy. The yield decreased over 70% in diseased plots by fewer numbers of spike and kernel per square meter and spike, respectively. BaYMV constructed typical inclusion body like a pinwheel type inside barley leaves, and the infection affected on cellular elongation or growth not cell division in examined three parts as stem, neck of panicle and node, related to dwarfness of infected barley. The stem tissues were most severely affected on cell growth as restrained epidermis cell length in diameter and vascular bundle size. In neck of panicle tissues, distribution and size of tissues of fiber and cortex parts, respectively, showed differences between healthy and infected plants. In node part, healthy plant showed bigger tissue size as 1.5 times than infected plant. Theses results suggest that BaYMV infection could affect on the cell growth not cell division, and which resulted shorten culm length in plant growth and decreased yield, finally.
To establish the system of in vitro plant regeneration, the different explants (stem with axillary bud and stem without axillary bud) of Hylotelephium ussuriense were cultured on the Murashige and Skoog's medium containing 6-benzylaminopurine (BA) and indolebutyric acid (IBA). The adventitious shoot induction was more effective in the stem with axillary bud explants than the stem without axillary bud explants, and was the best on MS medium containing 3.0 mg/L BA and 0.01 mg/L IBA. Frequency of plantlet growth was not significantly treated on MS and sucrose. Total chlorophyll contents under ventilation treatment were higher than those in control (non-ventilation). This in vitro propagation protocol will be useful for conservation and mass propagation of this endangered plant.
This study was conducted to determine the effect of a light-emitting diode (LED) on in vitro shoot growth and rooting in teak (Tectona grandis L.). In the experiments with apical bud explants, the greatest shoot elongation (3.2 cm) occurred when they were cultured on DKW medium under 50% blue and 50% red LED mixture (BR), whereas no differences in growth were observed in different light sources (florescent light [F] or BR) or media (MS or DKW). The highest number of shoot multiplication (2.4/explant) or elongation (4.94 cm) was achieved with 0.5 or 1.0 mg/L 6-Benzyladenine (BA) treatment under BR. In addition, the best rooting rate (93.8%) or root length (1.3 cm) was recorded with 0.5 mg/L indole-3-butyric acid (IBA) treatment under BR, and the highest root induction (3.1/explant) was observed in 0.2 mg/L IBA under BR. The in vitro rooted plantlets were hardened and survived well on soil.
Mature nodal segments of 2-year-old greenhouse stock plant were cultured on Murashige and Skoog basal medium supplemented with the different kinds and various concentrations of cytokinins to produce multiple shoots in in vitro condition. The most adventitious shoots were produced from excised ends of nodal segments. The highest average number $(24.6\;{\pm}\;4.6)$ of shoots was produced with the combination of BA 1.0mg/L and TDZ 0.1mg/L in MS medium. In addition, several shoots were formed from lenticels of bark cambium with the same treatment. These concentrations promoted high shooting capability upto $94.6\%$ and NAA was the best cytokinin among five different PGR sources.
Field experiment was carried out to investigate the effect of plant growth regulator (PGR); IAA, GA, Kinetin to regrowth of sorghum and pearlmillet according to variety and plant growth stage. Kinetin application after cut increased tiller number and decreased dry weight of regrowth, but its application on sorghum stubble in water stress increased tiller number and leaf elongation rate, consequently increased regrowth dry weight. GA application reduced tiller production in both species, but tiller formation in pearlmillet was decreased more than in sorghum by promoting leaf elongation of old tiller. Nonstructural carbohydrate (NSC) of stubble during regrowth was consumed less at anthesis than at stem elongation stage because of senescence of tiller primordia. GA treatment reduced NSC content more than other PGR in both plant species, by consuming reserve NSC and stimulating rapid elongation of old tiller after cut. Dry matter increase during regrowth had high correlation with tiller number and tiller elongation a week after cut, while it did not have any correlation with NSC at cutting stage or with consumption of NSC during regrowth. Therefore, regrowth in sorghum and pearmillet must depend upon activity of tiller primordia more than upon amount of reserved NSC.
An experiment was carried out to investigate the effects of plant growth regulators on the growth and fiber yield of ramie. Gibberellin (GA) and Atonic acid were applied at the various levels of application dates, application frequencies and concentration. Stem growth of ramie increased greatly by applying GA on the meristem of shoot apex. Among three treatments of application frequencies of GA, that is, one time application at the stage of stem length with 50cm, twice applications at the stem length of 50cm and 100cm, and three times applications at stem length of 50cm, 100cm and 150cm, stem growth increased more as application frequency increased. GA application is more effective on stem growth at the later stage of growth than the earlier growth stage. GA treatment of 100 to 300 ppm is more effective on stem elongation than 50 ppm, which was due on the elongation of internodes without increase in number of nodes. One time application of GA enhanced leaf growth more or less regardless of concentration, but GA was applied more frequently, leaf growth was inhibited more at higher concentration. Fiber yield was the highest at the treatment of three times application of GA with 100 ppm. This treatment also showed the highest percentage of fiber with 5.3%, which is much higher value compared with that of control treatment with 4.6%. Atonic acid was less effective on stem elongation than GA, but it also seemed to be effective on the enhancement of fiber development.
This work was conducted to investigate the field growth and yield of the sweetpotato (Ipomoea batatas) slips grown under different light emitting diodes (LEDs). Sweet potato cuttings of 3 cultivars ('Matnami', 'Shinhwangmi', and 'Yeonhwangmi') were cultivated under fluorescent lamp (FL) and several LEDs (PPF $150{\pm}5{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ at 20cm distance) in deep flow culture system for 20 days. The plants were acclimatized under sunlight for 10 days, and then cuttings (30cm length) were planted with $75{\times}25cm$ planting density on June 10th, covered with black vinyl film during growth period. Length and diameter of vine, number of root were excellent in the red plus blue (7:3) LED than the other treatments. At 30 days after planting, the survival rate in red plus blue (7:3) LED was significantly higher than that in FL and red LED, and it was not different among cultivars. Vine length, vine diameter, and number of node were not significant among LED light qualities and cultivars. After 120 days in the field cultivation, vine length, vine diameter, number of node, number of branch, and fresh weight of shoot were not significant among LED light qualities, but those except the number of branch showed significant differences among cultivars. Yield characteristics among LED light colors were not significant, but weight of storage root per plant, mean weight of storage root, and yield showed significant differences among cultivars. The yield per 10a in 'Matnami', and 'Yeonhwangmi' was significantly higher than that in 'Shinhwangmi'.
Stem cell-based therapy is under intensive investigation to treat acute renal failure (ARF). The purpose of this study was to evaluate available ARF models, and suggest a model appropriate to therapeutic evaluation of the stem cells in preclinical approach by determining the optimum concentration of nephrotoxic agents and duration of ischemia induction. Three different types of available acute kidney injury (AKI) animal models were analyzed using rats: Cisplatin (saline, 5 and 7.5 mg/kg, IP) or glycerol (saline, 8 and 10 ml/kg, IM)-induced nephrotoxicity as toxic models and ischemia-induced (sham, 35 and 45 minutes) nephropathy as an ischemic model. The relevance and applicability to investigate especially the regenerative ability of stem cells were evaluated regarding morphology, renal function and survival at this time point. In the point of renal function, 10 ml glycerol/kg and 7.5 mg cisplatin/kg model in toxic models and 45 min model in ischemia models showed significant decrease for the longer observation time compared to 8 ml glycerol/kg, 5 mg cisplatin/kg and the 35 min ischemia models, respectively. All groups were observed no mortality except 45 min-ischemia model with 50% survival. Histological significant alterations including cast formation in the tubular lumen, tubular necrosis and apoptosis were revealed on the second day in either ischemiaor glycerol-induced models, and on day 5 in cisplatin-induced models. The results indicate that ischemia 35 min-, cisplatin 7.5 mg/kg- and glycerol 10 ml/kg-induced AKI would be ideal animal models to monitor a outcome parameter related to the therapeutic effects on renal function with noninvasive techniques in the same animal at multiple time points. Our findings also suggest that the best time points for the functional or histological interpretation of renal will be on day 2 in both glycerol- and ischemia-induced AKI models and on day 5 in cisplatin-induced AKI.
The effect of plant growth regulators on proliferation of shoot from stem node culture of Japanese yew (Taxus cuspidata Sieb. et Zucc.) was studied using Quoirin and Lepoivre (1977) medium. Among the cytokinin tested, BAP, kinetin, and thidiazuron at various concentrations had no effect on shoot multiplication However when zeatin at 5$\times$10$^{-5}$ M was added to the medium, an average of 6 shoots were regenerated per explant after 8 weeks of culture. The ratio of rooting ex vitro was remarkably increased up to 34% by dipping the basal end in 0.5 to 1.0% IBA on talc compared with 3% in vitro rooting. Rooted plantlets were acclimated in greenhouse conditions for one month and successfully transplanted to the field.
The control of stem length is most important external quality aspect in cut chrysanthemum. The present work in conducted in growth chamber and aims at investigating the effect of light intensity on the photosynthesis capacity and stem elongation in cut chrysanthemum. To evaluate the effect, different level of assimilation lighting (LEDs) was given to canopy level 60, 100, 140, and $180mol{\cdot}m^{-2}{\cdot}s^{-1}$, individually, under short-day conditions, and the light treatments were initiated from 7days after planting. There was a positive linear-regression relationship between the light intensity and the net assimilation rate. On the other hand, there was not significant difference in flower buds induction. The growth of stem length, leaf area, and dry weight was increased by increasing the light intensity, whereas the plants grown under 140 and $180mol{\cdot}m^{-2}{\cdot}s^{-1}$ was not differences in those of growth. It indicates that the increased net assimilation rate is not continually coincided with the maximized growth in cut chrysanthemum. It might be considered that the optimal light intensity for stem elongation of cut chrysanthemum is $140mol{\cdot}m^{-2}{\cdot}s^{-1}$ under short-day condition.
