Studies on the Fertilization of Pulsatilla koreana (할미꽃(Pulsatilla koreana)의 수정현상(受精現象)에 관(關)한 연구(硏究))

  • This experiment was carried out to investigate the fertilization and the size of mature female and male gametophytic parts of Pulsatilla koreana after artificial pollination. The size of pollen is $26.5{\mu}m$ at the time of anther dehiscence, that is, about $3{\sim}4$ days after pollination. Synergid nucleus, egg nucleus, and polar nucleus are 10.0, 15.0, and $32.5{\mu}m$ respectively at the time of completing egg apparatus formation, that is, about 2 days after pollination. Poller tubes germinate on stigma about 10 hours, passing lower part of style about 30 hours, penetrating into micropyle about 35 hours after pollination. Sperm nucleus penetrates into polar nucleus about 40 hours and egg cell about 48 hours after pollination. But, there seems to be different among the individuals. Multinuclei and multinucleoli are formed in egg cell, synergid, and polar nucleus about the time of fertilization. Proembryo is formed about 4 days, being changed to large globular form about $6{\sim}8$ days after pollination. Endosperm nuclei divide into free nuclei after fertilization and change to cotyledon in gymnosperm. There seems to be same phenomena in Pulsatilla koreana.

Study on Species Identification for Pungnammun Gate (Treasure 308) in Jeonju, Korea (보물 제 308호 전주 풍남문 주요 부재의 수종 연구)

  • This study is for species identification for each structure member such as Pillar, Bo, Changbang, Dori, Jangyeo, Judu, Donjaju, Chunyeo, Guitle, and Jongdae, of Pungnammun Gate (Treasure 308). Jeonju is the birthplace of Joseon Dynasty and Pungnammun Gate was the southern gate of old Jeonjueupseong which was walled town. Provincial Governor of Koryeo Dynasty, Yu Gyeong Choi built Jeonjubuseong and four gates at all cardinal points in 1388. And the gate was burnt down by Jeongyujaeran (war with Japan in 1597). It was rebuilt by King Yeongjo (Joseon Dynasty) in 1734 and renamed 'Pungnammun' after 34 years. It was designated for Treasure 308 for its unique style of architecture and historic values in 1963. In this study, all of wooden structure members were Pinus spp.. This result was matched for the result of major species for wooden building of late Joseon Dynasty. It can be used to complete database for architecture of Castle's Gate and help for restoration of cultural heritage in the future.

Characterization of Amino Acid Contents in Grain of Core Collections of Korean Native Rice (벼 재래종 핵심 유전자원의 현미 아미노산 함량 평가)

  • Amino acid composition in grain of Korean native rice core collections was evaluated to identify the useful genotypes for quality enhancing rice breeding and value-added food development. Large variations were found in the contents of 16 amino acids among 388 genotypes tested with 14.9% of the minimum coefficient of variation (CV), and the contents of methionine and histidine showed the highest CV and the second high, respectively. The average values of total amino acid, total essential amino acid, lysine and threonine contents of Korean native rice collections were almost similar with those of cultivating japonica rice, Saechucheong. Among 388 native core collections Baekkokna, Daesona and Saducho were identified as high amino acid germplasm in total amino acid, total essential amino acid, lysine and threonine contents. And Seoksanjo and Sodujo were identified as low amino acid collections. Rice collections showing purple apiculus color tended to distribute in lower serine, proline, glutamic acid, glycine, alanine, valine, leucine, phenylalanine, arginine, and total amino acid contents compared with those of yellowish apiculus color collections. The glutinous rice collections showed the tendency of higher contents of serine, glutamic acid, alanine, valine and leucine compared with those of non-glutinous collections.

'Glory Pink' a FA Intersectional Lily with Unspotted Pink Petals (연한 분홍색 FAA 아속간 교잡나리 'Glory Pink' 육성)

  • 'Glory Pink' an interspecific lily cultivar was released in 2007 at National Horticultural Research Institute (NHRI), Rural Development Administration (RDA), Suwon, Korea. The cross and ovule culture was made between FA interspecific hybrid 'FA97-30' (L. formolongi 'Raizan' ${\times}$ Asiatic lily 'A61'), a red colored cultivar, and Asiatic lily 'Sanzio', light red colored cultivar in 2001. Multiplication and bulbing, and characteristic tests were conducted from 2005 to 2006. Flowering time is around the first of July and the plant height is 131.7 cm. Flowers are upward-facing and red-pink (RHS 63C), with size of 11.3 cm. Petal length and width is 9.1 cm and 4.1 cm, respectively. Leaves are 13.8 cm long, 1.4 cm wide. The throat is dark green, and the stigma is light yellow and pollen is light brown. The weight and size of bulb is 51.1 g and 13.2 cm, respectively. For long-term storage, bulbs can be stored under -1 to -2 for year-round forcing.

Fragrance Analysis Using GC-MS and Electronic Nose in Phalaenopsis (GC-MS와 전자코를 이용한 팔레놉시스 향기 분석)

  • Phalaenopsis (P.) has various species, and some of them have strong fragrance. There are fragrant species such as P. bellina, P. violacea, P. schilleriana and used in breeding program for fragrant Phalaenopsis. This study was performed for establishment of fragrance analysis system using GC-MS and electronic nose in eight P. resources. We analyzed fragrant compound using the tissue of sepal, petal, column, and lip of P. '3010'. The percentage of the major compound was high in the petal and lip tissues. The main compound emitted from P. bellina was linalool (21.21%). It was possible that fragrance pattern could be analyzed among the resources using the electronic nose. Discriminant function analysis (DFA) was more useful than the principal component analysis (PCA) in statistics program. We utilized GC-MS method for the major compounds of flower from our breeding materials. This study would be useful to the fragrant analysis system for the fragrant orchid breeding in the future.

Hybridity Verification of Progenies Obtained from Ovule Culture by Using RAPD Markers in Reciprocal Crosses of Alstroemeria (알스트로메리아 배주배양을 통하여 획득한 정역교배 자손의 혼종성 분석)

  • In this study, we performed ovule culture after reciprocal crosses of two Alstroemeria accessions and investigated genetic contribution of parents by using RAPD markers. The best method was half-ovule culture on MS medium supplemented with $60g{\cdot}L^{-1}$ sucrose and $2.2g{\cdot}L^{-1}$ gelrite at 14 days after pollination. Embryos began to germinate after 6 weeks of culture. The complete plantlets were formed after 4 months of culture. In eight progenies and two parental cultivars, 59 polymorphic bands were obtained out of 89 total bands by RAPD analysis using 7 primers. Eight $F_1$ progenies from the crosses between two accessions using reciprocal crosses showed 1:1 contribution of maternal and paternal parents. It is confirmed that $F_1$ progenies were obtained from parental accessions by using RAPD markers. We conclude this cross combination showed pre-fertilization barriers with incompatibility between stigma or style, and pollen because progeny number was different in each cross combination. Thereby, it warrants overcoming pre-fertilization barrier together with post-fertilization barrier in order to broaden the heterozygosity within progeny populations in Alstroemeria breeding program.

Correlation between Body Fat Percent Estimated by Bioelectrical Impedance Analysis and Other Variable Methods (생체전기 저항법에 의한 체지방율과 다른 계측치간의 상관성 연구)

  • Purpose : Obesity is a significant health problem with medical and psychological consequences for children and adolescents. The purpose of this study was to assess the correlation between body fat percent using bioelectrical impedance(BI) and other variable methods. Methods : We measured height, weight, body mass index(BMI) and body fat percent by skinfold thickness(ST) and BI in 1,035(496 male; 539 female) children from seven to 18 years of age. The correlation coefficients between BI and each of the other different methods were obtained. The sensitivity and specificity to predict obesity by these several methods were studied. Results : Fat percent estimated by BI analysis and BMI showed a strong correlation(r=0.749). Fat percent estimated by BI analysis and ST showed a very strong correlation(r=0.835). At the 95th percentile cut-off point for BI, ST showed a sensitivity of 57.7%, and a specificity of 97.6% for estimating body fat. At the 95th percentile cut-off point for BI, BMI showed a sensitivity of 84.9%, and a specificity of 99.3% for estimating body fat. Conclusion : The fat percent estimated by BI analysis correlated strongly with ST or BMI. BI analysis is an objective and accurate method for estimating body fat in childhood obesity.

Taxonomic study on viola albida var. albida and its related taxa (태백제비꽃과 근연분류군의 분류학적 연구)

  • Taxonomic studies were conducted to evaluate the interspecific relationships in Viola albida var. albida and its related taxa using seven populations for morphology, palynology and anatomy. Molecular phylogenetic studies were also examined in 28 populations including 19 Korean, four Chinese, two Japanese, one American population and two outgroups using nrITS, and 27 populations except V pinnata for trnL-F region of chloroplast DNA. Morphological differences was observed among seven populations of three species in leaf shape, but characters such as serrate number of leaf margins, petal size, pistil shape were showed overlap between populations. Pollen shape of seven populations was monad and grain shape on the polar axis was semi-angular. Morphology of aperture was tri-colporate, and the surface sculpturing was scabrate in rugulate. The grain shape of equatorial view of five populations was prolate whereas V albida var. taknhashii type 1 and V albida var. chaerophylloides type 3 were subprolate. The anatomical characters of rnidvein of leaf, petiole, peduncle, root were also described for the species. The stomatal apparatus of the leaves was observed only in abaxial surface, and the number of stoma per unit ($mm^2$) were abundant in incised than lobate or cleft margin of the leaves. The nrITS analysis shows that V. pinnata and V. dissecta was monophyletic and occupied a basal position in the V. albida var. albida and its related taxa. The other clade including infraspecific populations of V. albida, and V. eizanensis was paraphyletic. The trnL-F noncoding region analysis was similar to the ITS tree. According to the above observations in morphology, palynology, anatomy, and molecular phylogenetic analysis, the significant differences were not found except for leaf shape in Viola albida var. albida and its related taxa, therefore V. albida var. takanhashii and V. albida var. chaerophylloides were considered to be an infraspecific taxa of V. albida var. albida rather than an independent species, subvariety or variety of V. pinnata and V. dissecta.

Tensile Performance of Machine-Cut Dovetail Joint with Larch Glulam (낙엽송집성재를 이용한 기계프리커트 주먹장접합부의 인장성능)

  • Members used for the Korean traditional joints have been processed by handicraft, especially with domestic red pine species. Dovetail joint is most commonly used in woodworking joinery and traditional horizontal and vertical connections. It is able to be processed much easier to cut by handicraft and machines. However, although it is processed straight forwards, it requires a high degree of accuracy to ensure a snug fit. Also, tenons and mortises must fit together with no gap between them so that the joint interlocks tightly. A few scientific studies on the dovetail joints have been conducted so far. For the effective applications of traditional joints and domestic plantation wood species, dovetail joints were assembled by larch glulam members processed by machine pre-cut. To identify the tensile properties of through dovetail joints, larch glulam with 150 150mm in cross section were prepared. Furthermore, various geometric parameters of dovetai joints such as width, length, and tenon angle, were surveyed. The ends in the mortise was cracked mainly at a low strength level in the control specimens without reinforcements. The maximum tensile strengths of reinforced specimens considering real connections such as capital joint and headpiece on a column, increasedby handicraft, especially with domestic red pine species. Studies on Heterostylism, Fertility, and Embryological Characteristics in Buckwheat. Fagopyrum esculentum (메밀의 이형예현상과 수정력 발생학적 특징에 관한 연구)

  • Fifty-five local collections of buck wheat, Fagopyrum esculentum, were investigated their ratios of long-styled (LS) and short-styled (SS) flowers, fertility, meiosis of megaspore and microspore mother cell, female and male gametogenesis, and egg apparatus in accordance with the sowing seasons (spring, summer), altitudes (20m, 50-100m, 300m), and parent style types (L, S). Also they were embryologically investigated the fertility, fertilizing phenomenon and proembryogenesis by the legitimate and illegitimate pollination. There were no differences in the ratios of long-styled and short-5tyled flowers along with altitudes, but more irregularness was observed in plain area than that in the mountaineous or coastal area. LS versus SS ratios by sowing seasons were significantly separated into 1 : 1 in the summer sowing (P 0.1), but they were irregularly separated in the spring sowing. The segregating ratios by parent style types showed more number of short-styled flower in the spring sowing, and were statistically seperated into 1 : 1 in the summer sowing (P 0.25), regardless to parent style types. In the artificial legitimate union, the seed setting rates of the summer sowing (59-61%) were much higher than those of the spring sowing (about 30%), but in the artificial illegitimate union the seed setting rates were only fructified about 0.8-1.8% in the spring sowing. The seed setting rates in accordance with flowering stages were larger in turn early, middle, late, in the summer sowing. The grain number and grain weight per plant of short-styled flower were more than those of long-styled one regardless to style types. The 1,000 grain weight of long-styled flower was heavier than that of short-styled one in large grain, but it was lighter than that of short-styled flower in small or medium grain. The percentage of normal female and male gametogenesis in the summer sowing were higher than those in the spring sowing. The ovule was atropous and two polar nuclei were a synkarion before flowering. The pollens germinated at 30 minuts after pollination and the pollen tube grew continually and penetrated into micropyle at 1.5-2 hours and the two male nuclei fertilized with egg nucleus at 3 -5 hours after pollination. Flertilizing times in summer were shorter than in autumn. The fertilized egg was divided in a small apical cell toward the interior of the embryo sac and a large basal cell toward the micropyle cell at 15-24 hours after pollination, and division times in summer were shorter than in autumn. The proembryo began the embryogenesis at 7-8 days and formed itself into the perfect embryo at 15 days after pollination.
