Isolation and Identification of Fatty Acid and Volatile Compounds from Tuna Fish Oil with Supercritical Carbon Dioxide (초임계 이산화탄소를 이용한 참치안구유로부터 지방산 및 휘발성 성분의 분리 동정)

  • Isolation and Identification of fatty acid and volatile compounds in tuna fish oil were successfully carried out using supercritical carbon dioxide. Samples of the oil were extracted in a 56 ml semi-batch stainless steel vessel under conditions which ranged from 80 to 200 bar and 40 to $60^{\circ}C$ with carbon dioxide flows from 10 ml/min. Volatiles in the oil extracted from the samples with supercritical carbon dioxide were analyzed by gas chromatography, mass detector with canister system. The extracts were contained with various fatty acids, 57.0% of unsaturated fatty acids such as docosahexaenoic acid(DHA) and eicosapentaenoic acid(EPA), and 43.0% of saturated fatty acids. The aroma compounds in the oil showed over 129 peaks, of which 100 compounds were identified. Volatile components included 2,4-hepatadienal(fishy), dimethyldisulfide (unpleasant), dimethyltrisulfide (unpleasant) and 2-nonenal(fatty). The isolation efficiency of the volatile compounds from the samples was 99.4% at $50^{\circ}C$ and 200 bar.

Isolation and Diversity of Wild Yeasts from the Waters and Bank Soils of Daejeoncheon, Gapcheon, and Yudeungcheon in Daejeon Metropolitan City, Korea (대전광역시 대전천과 갑천 및 유등천의 물과 주변 토양들로부터 야생효모들의 분리 및 분포 특성)

  • In order to investigate the diversity of yeasts from major rivers (the Daejeoncheon, Gapcheon and Yudeungcheon) located in Daejeon city, we isolated wild yeasts by plating diluents of samples collected during the summer and winter of 2016 onto yeast extract-peptone-dextrose (YPD) medium, then identified them using Basic Local Alignment Search Tool (BLAST) analysis to compare the nucleotide sequences of the PCR amplicons for the D1/D2 domain of 26S rDNA. In total, we isolated 191 yeast strains belonging to 104 species from 148 soil or water samples from the rivers and their junctions. Candida spp. (45 strains) including Candida tropicalis (22 strains) were the most abundantly isolated strains from the Daejeoncheon. Candida spp. (16 strains) including Candida vartiovaarae (8 strains) and Candida spp. (18 strains) such as Candida sake (4 strains) were also the dominant isolates from the Gapcheon and Yudeungcheon, respectively. In conclusion, Candida spp. and Cryptococcus spp. were the most dominant strains, corresponding to 42% and 7% of the 191 yeast strains isolated in this study, respectively.

Species Identification of Nontoxic Alexandrium tamarense (Dinophyceae) from Chinhae Bay, Korea, Using Molecular Probes (진해만에서 분리한 무독성 Alexandrium tamarense (Dinophyceae) 동정을 위한 molecular probe 이용)

  • The morphology of the apical pore complex, the first apical plate and the posterior sulcal plates in a new isolate of Alexandrium tamarense (Lebour) Balech from the Bay of Chinhae was compared with other that of toxic strains of A. tamarense previously isolated from Korean waters. Although this isolate was morphologically identical to these toxic strains, high performance liquid chromatography and mouse bioassay showed no evidence of toxin production. The nontoxic A. tamarense strain showed a strong positive binding activity with PNA lectin, indicating a high density of lactose and galactose residues on the cell surface, and in SDS-PAGE and Western blot analysis a unique protein of about 21-kDa molecular sizes was observed. These findings demonstrate that the use of PNA and immunobioassay could be used to discriminate between toxic and nontoxic strains of A. tamarense.

Diversity of Endophytic Fungi Isolated from Pinus densiflora in Chuncheon city, Gangwon province, Korea (강원도 춘천시의 소나무(Pinus densiflora)에서 분리한 내생균류의 다양성)

  • Red pine (Pinus densiflora) is a dominant tree species in Korean forests. The diversity of endophytic fungi was investigated by isolating fungi from different parts of a red pine tree during the fall and spring seasons. One hundred and fifty-two strains of endophytic fungi were isolated and classified into 42 species belonging to 29 genera. Among these, 74 (49%), 47 (31%) and 31 strains (20%) were isolated from branches, leaves, and stems, respectively. The most frequently isolated species were Daldinia childiae and Biscogniauxia maritima (28.9% and 9.8%, respectively). D. childiae was the dominant endophytic fungus on branches (20 strains) and leaves (19 strains). B. maritima was most frequently isolated from branches. The isolation frequencies on stems of B. maritima (6 strains) and D. childiae (5 strains) were also high. Fifty-seven isolates belonging to 19 species and 17 genera were isolated in the spring, and 95 strains belonging to 35 species and 24 genera were isolated in the fall. D. childiae was the most frequently isolated fungus in both spring and fall (21 and 23 strains, respectively). B. maritima was also frequently isolated in both spring (7 strains) and fall (8 strains).

Distribution and Species Diversity of Wild Yeasts Isolated from Flowers in Korea (국내에 서식하는 꽃에서 분리한 야생 효모 분포 및 종 다양성)

  • Various indigenous yeasts were isolated and obtained from flowers in the Republic of Korea, and their distribution and species diversity were studied. Seventy-seven flowers were collected from 25 areas in Korea, and 502 yeast strains were isolated from these flowers. A total of 50 species were identified by comparing large subunit rDNA gene sequence homology with the type strains of yeasts. The analysis of yeast distribution showed that the dominant yeast species were Aureobasidium pullulans, A. leucospermi, and Filobasidium magnum in each region and flower samples. Except for the above three yeast species, no species of yeasts showed any meaningful distribution among the habitat regions and sources. In conclusion, 50 species of indigenous yeasts were obtained from flowers that can be used as industrial resources, and the data could be used for further research on yeast diversity and interactions between yeast and its host.

Isolation and Identification of Protease Producing Bacteria in Kimchi (김치에서 단백질분해효소활성 균주분리 및 동정)

  • Strains producing the protease, which is essential for growth of lactic acid bacteria and fermented kimchi, were screened and identified. Among five types of selected pulmuone kimchis (Jeonlado kimchi, ripened kimchi, yeolmu kimchi, kakdugi, and baechu kimchi), nine strains of bacteria were screened and identified by whole cell protein pattern and API test. The nine strains consisted of one of Lactobacillus sp., one of Leuconostoc sp., six of Bacillus sp., and one of Brevibacillus sp. The protease activities of these strains were compared with known strains (Bac. subtilis KCCM 12248 and Bac. licheniformis KCCM 11851) producing protease. Among tested strains, K-2 (Brevibacillus sp.) showed the highest value (0.11 unit/mg) in protease activity.

Analysis of Volatiles in Sesame Oil Collected by Simultaneous Distillation/Extraction(SDE) and Dynamic Headspace Sampling (DHS) (연속수증기증류법(SDE)과 동적헤드스페이스법(DHS)에 의한 참기름 중의 향기성분의 분석)

  • The flavor compounds of the oil from roasted sesame seeds were collected by simultaneous steam distillation/ extraction(SDE) and dynamic headspace sampling(DHS) and were analyzed using a gas chromatograph equipped with a flame ionization detector and connected into a mass spectrometer. Among the flavor compounds collected by the SDE method, 46 compounds were identified. They consisted of 6 alcohols, 6 aldehydes, 5 ketones or acids, 4 furans or phenols, 12 pyrazines, 4 pyridines or thiazoles, and others. Thirty six compounds were identified by DHS and many of them were the same as those identified by the SDE method. However, some compounds such as 1-hexanol, pentanal, and dimethylsulfide were identified only by the DHS method.

Identification of Orchid Mycorrhizal Fungi Isolated from Five Species of Terrestrial Orchids in Korea (한국에 자생하는 5종의 지생란에서 분리한 난균근균의 동정)

  • This study was performed to isolate and identify the orchid mycorrhizal fungi (OMF) from roots of five species of terrestrial orchids in Korea; Cymbidium goeringii, Spiranthes sinensis, Calanthe discolor, Bletilla striata, Pogonia minor. DNA was extracted from isolated OMF and ITS region was amplified using primers, ITS1-OF and ITS4-OF. Four species of OMF belong to Tulasnellaceae and Sebacinaceae were identified; Tulasnella calospora, Tulasnella irregularis, Tulasnella sp., Sebacina vermifera.

Two Unreported Glomeromycota Fungi Isolated from Rhizospheres of Smilax china (청미래덩굴의 근권에서 분리된 2종의 Glomeromycota 미기록종)

  • We isolated fungal spores belonging to the phylum Glomeromycota from the rhizospheres of Smilax china, cultured in a greenhouse. We identified the isolated spores using sequence analysis of 18S partial rDNA region, internal transcribed spacer and 28S rDNA regions. We confirmed 2 unreported spores of Glomeromycota fungal species, Diversispora eburnea and Paraglomus laccatum. Here, we described the morphological characteristics and results of phylogenetic analysis of the confirmed species.

Three Novel Endophytic Fungal Species Isolated from Roots of Medicinal Crops in Korea (약용작물의 뿌리에서 분리된 3종의 국내 미기록 내생균)

  • We isolated endophytic fungal strains from the roots of three medicinal crops, Ligusticum chuanxiong, Angelica gigas, and Cnidium officinale, cultivated at Yeongju, Korea. The fungal strains were identified based on their morphological characteristics and molecular analyses of their internal transcribed spacer, large subunit rDNA, and beta-tubulin regions. Thereby, we identified three previously unrecorded endophytic fungal species, Dactylonectria pauciseptata, Rhizopycnis vagum, and Sistotrema sernanderi, in Korea. In this report, we describe the morphological characteristics and phylogenetic analyses of these three fungal species.
