  • This study used loess from the source region of Asian dust (a desert area in China) to analyze the effects of Asian dust on fruiting and fruit quality in Korean fruit trees. Asian dust and loess from the source region were highly alkaline compared to average Korean field soil. Organic material contents of Asian dust were about three times as high as that of the average field soil. Loess was dusted onto the stigma of apple, pear, and peach trees. fruiting, seed number, fruit quality and pollen tube elongation were investigated. Pollination and fertilization were not influenced while pollen tube elongation was slightly influenced. Consequently, fruiting, seed number and fruit quality were not affected by the dusting treatment.

  • In order to study the rate control step in the protein metabolic course of the chlorophyll formation, the transaminase activities which are obtained freely in the extracts of cotyledons of germinating peas at the light and the dark places, are measured in Beckman mopel D.U, spectrophoto meter at 490 mu. In this case, of two enzymatic reaction products; oxalacetic acid and pyruvic acid, the former is converted to pyruvic acid by aniline citrate and after each pyruvate phenyl hydrazones are extracted by toluenes: when this is treated with strong alcoholic alkali, a colored hydrazone is formed and it is measured by above apparatus. The estimated G.O.T. and G.P.T. in the germinated cotyledons at dark and light places considerably differ in their activities; G.O.T. and G.P.T. activities which are formed at the light are more increased than at the dark and also they differ in their rates through germination, though G.O.T. activity increment is smoothly but that of G.P.T. is more sharply, and they are considered to be directly affected to the chlorophyll formation and indirectly to the growth. G.O.T. and G.P.T. in each fractions of cell in the cotyledons should be formed by dissociation of zymogens in the microsomal fractions and it seems to promoted by light. In the formation of the chlorophyll, the protein metabolism occurred mainly in the microsomal fractions and the rate determining step is found at the point where the zymogene that is able to produce G.P.T. is activated, and this activation is promoted by light as noted above.

  • Purpose: Nondestructive evaluation of seed viability is a highly demanded technique in the seed industry. In this study, hyperspectral imaging system was used for discrimination of viable and non-viable radish seeds. Method: The spectral data with the range from 400 to 1000 nm measured by hyperspectral reflectance imaging system were used. A calibration and a test models were developed by partial least square discrimination analysis (PLS-DA) for classification of viable and non-viable radish seeds. Either each data set of visible (400~750 nm) and NIR (750~1000 nm) spectra and the spectra of the combined spectral ranges were used for developing models. Results: The discrimination accuracy of calibration was 84% for visible range and 76.3% for NIR range. The discrimination accuracy of test was 84.2% for visible range and 75.8% for NIR range. The discrimination accuracies of calibration and test with full range were 92.2% and 92.5%, respectively. The resultant images based on the optimal PLS-DA model showed high performance for the discrimination of the nonviable seeds from the viable seeds with the accuracy of 95%. Conclusions: The results showed that hyperspectral reflectance imaging has good potential for discriminating nonviable radish seeds from massive amounts of viable seeds.

  • Purpose: The objective of this research was to select high quality cucumber (cucumis sativus) seed by classifying into viable or non-viable one using Raman spectroscopy. Method: Both transmission and back-scattering Raman spectra of viable and non-viable seeds in the range from $150cm^{-1}$ to $1890cm^{-1}$ were collected with a laser illumination. Results: The Raman spectra of cucumber seed showed Raman peaks with features of polyunsaturated fatty acids. The partial least squares-discriminant analysis (PLS-DA) to predict viable seeds was developed with measured transmission and backscattering spectra with Raman spectroscopy and germination test results. Various types of spectra pretreatment were investigated to develop the classification models. The results of developed PLS-DA models using the transmission spectra with mean normalization or range normalization, and back-scattering spectra with mean normalization treatment or baseline correction showed 100% discrimination accuracy. Conclusions: These results showed that Raman spectroscopy technologies can be used to select the high quality cucumber seeds.

  • It failed occasionally to take a reasonable emergence rate since tobacco (Nicotiana tabacum L.) seeds were planted on late Jan. showing lowest temperature. This experiment was done to measure the effect of GA3 (concentration ; period) , light quality (red ; white ; dark) during or after its treatment, daily irradiation hours( 0 ; 8 ; 12 ;16) and germination termperature (20 or 10 $^{\circ}C$ ocnstant ; 20/1$0^{\circ}C$ alternating) on the germination rate. Red and white light given during grmination showed no differences between the other daily irradiation hours except that 8 hours red light delayed germination although their 12 hours irradiation had the gratest rate. The rate was increased with increased concentration to GA3 0.01 mM or increased imbibition period to 3 days although the rate of cv. NC 82 was less than that of cv. Burley 21 in the case of dark imbibition of GA3 but daily 12 hours irradiation during germination. Light quality forced during GA3 imbibition eliminated such effect of GA3 shown in the darkness so that only light quality pretreatment and termperature during germinition were affected on the rate. The germination rate of thecultivars was decreased in the order of red, white light, darkness meaning that it was highly influenced by the light quality during GA3 treatment. Regardless of GA3 or light quality treatment,on the other hand, the rate was greater in 20 $^{\circ}C$ constat than 1$0^{\circ}C$ constant and 20/1$0^{\circ}C$ alternating germination temperature having similar germinative patterns.

  • Several presowing seed treatments have been used to facilitate the seed germination and seedling emergence. The study was performed to design a model of presowing seed treatments in Taraxacum platycarpum. Germination and seedling growth responses to temperature, drying, chilling, $GA_3$ and priming were investigated to know the optimum germination conditions and presowing treatment. The optimum temperature condition for germination and seedling growth were $15{\sim}20^{\circ}C$ at dark condition. The seeds exposed to infra red light, however, did not germinate. The moisture content of seeds dried at $35^{\circ}C$ for 3.5 hours was returned to the level of unimbibed seeds. The germination of seeds dried under dark condition was better than those dried under infra red light. Prechilling treatment imposed for 6 weeks showed higher germination rate than $GA_3$ treatment or priming. In field test, the highest field emergence was observed in the seeds dried after prechilled for 6 weeks and followed by drying followed by $GA_3$ > low temperature > $KNO_3$ priming. Imbibition with 0.1 mM $GA_3$ solution for 2 hours then drying at 35 for 3.5 hour under dark condition was effective to the practical presowing treatment for Taraxacum seeds.

  • Background: Many Taraxacum spp. plants are widely used in medicine, but some of them have propagation problems, such as strong dormancy and poor germination rates. This study investigated the effects of temperature, gibberellic acid ($GA_3$), and potassium nitrate ($KNO_3$) on seed germination in Taraxacum ohwianum. Methods and Results: The seeds (NIBRGR0000135524) were exalbuminous, and their length and width were $4.54{\pm}0.032mm$ and $0.97{\pm}0.029mm$, respectively. Various germination temperatures were tested (15, 20, 25, and $30^{\circ}C$), the optimum temperature for germination was found to be $20^{\circ}C$ (31.3%). High temperature ($30^{\circ}C$) resulted in non-typical seedlings (thickened and crumpled cotyledons, with a restricted root system). To increase the germination capacity, $GA_3$ (200, 500, and $1,000mg/{\ell}$) or $KNO_3$ (20, 50, 100, 200, and $500mg/{\ell}$) solutions were used as pre-soaking solutions instead of water. The $GA_3$ treatments increased the germination percentage and rate, but germination percentage was higher with the $KNO_3$ treatments. Under the $50-200mg/{\ell}\;KNO_3$ treatments, the germination percentage exceeded 80% after 12 days, and $50mg/{\ell}\;KNO_3$ was notably effective (91.2% after 15 days). Conclusions: T. ohwianum seeds showed improved germination at low temperatures. The best method for germination was presoaking in $50mg/{\ell}\;KNO_3$ for 24 h, in the dark at $4^{\circ}C$, and then incubating the germinated seed at $20^{\circ}C$ for 15 days.

  • Characteristic of seed for Sageretia thea fruits collected from the habitat of harvest season in Jeju and their germination ratio in different temperatures were considered. The average weight was 0.2 g, average diameter was 7.2 mm, average length was 6.5 mm, and size distribution range was 5.1 ~ 10.0 mm. The number of seeds per fruit was 1.8, and 1000 grain weight was 7.77 g with diameter of 3.7 mm and thickness of 1.7 mm in size. The fruit maturation was investigated to be from April 27 to June 1, when the best maturation period was about a week, May 11 to May 18. Also, the fruits grown on May 4 to May 11 were seen to have the bigger in transverse size and weight with a tendency of the earlier maturation date has the greater the number of seeds. S. thea seeds pretreated for 24 hours at $50^{\circ}C$ were not germinated, while most of those pretreated for 24 hours at $4^{\circ}C$ were successfully germinated at any degree of temperature (except at a temperature of $10^{\circ}C$). Particularly, the highest germination rate of 55% was made at $15^{\circ}C$, and plumule and radicle were best grown within the temperature range of $25^{\circ}C$.

  • This study is part of research to develop the technology for managing major medicinal crops after harvest. We studied the optimal germination conditions of $Cynanchum$ $wilfordii$ (Maxim.) Hemsl. The mean germination time (MGT) of $C.$ $wilfordii$ seeds was higher after soaking for 4 days after storage at $5^{\circ}C$ for 8 weeks, than with germination at $25^{\circ}C$ (3 days). However, the germinative energy (GE) decreased as the number of days soaking increased. The greatest germination rate ($83.3{\pm}8.8%$) was at $25^{\circ}C$ with no soaking of seeds stored at $-20^{\circ}C$ for 8 weeks. Based on these results, we characterized the germination conditions of a major medicinal crop.
