Chlorella cultivation for mass culture of rotifer, Brachionus Plicatilis I. Selection of suitable Chlorella species (Rotifer (Brachionus Plicatilis) 사육을 위한 Chlorella의 배양 1. 적종 Chlorella의 선택)

  • Rotifers became an important live food for fish larvae, especially for marine fishes, and Chlorella has been used as a very useful food for the mass culture of rotifer. However, not many tests for suitable Chorella species for the mass culture of rotifer were done and many of Chlorella sp. have been used without consideration of species for this purpose. There-fore, two species of marine Chlorella and four species of freshwater Chlorella were tested to select suitable species for the mass culture of a rotifer, Brachionus plicatilis. These Chlorella species were cultured in five different culture media; f/2, Erdschreiber, Complesal for marine species and S$\cdot$K, Wai and Complesal for freshwater species. Proximate analyses were done to see the protein, lipid and ash contents of a marine species, C. ellipsoidea and a freshwater species, C. vulgaris. Amino acids content of these species were also tested. C. ellipsoidea and C. vulgaris showed better growth than the other species. For marine Chlorella sp., f/2 media was better than Erdschreiber and Complesal. But for the freshwater species, Complesal showed the best result in growth. By the proximate analyses, C. ellipsoidea has higher lipid contents whereas C. vulgaris has higher protein and ash. In the analysis of amino acid of Chlorella, it was remarkable that freshwater Chlorella, C. vulgaris, has high content of $NH_3$ comparing with marine Chlorella, C, ellipsoidea. According to the above results, C. vulgaris seems have higher possibilities for mass culture of rotifer but further studies are needed.

Prediction of weight loss of low temperature storage tomato (Tiwai 250) by non-destructive firmness measurement (비파괴적인 경도 측정을 통한 저온저장 토마토(티와이250)의 감모율 예측)

  • This study was conducted to investigate the weight loss, firmness, external color and vitamin C (VC) content of tomatoes (Lycopersicon esculentum) using non-destructive method to measure identical tomato samples during 15 days storage at low temperature and high humidity. Tomatoes were harvested at the light red stage, sorted, box packed and then stored in thermo-hygrostat ($10{\pm}1^{\circ}C$, $90{\pm}10%RH$). The quality changes in weight loss, firmness and external color were measured every 3 day interval. Weight loss was increased by $1.13{\pm}0.15%$, but it may not be considered to affect quality. Surface color of fruit was changed, especially in lightness and hue angle value. The color values were analyzed by analysis of variance (ANOVA), and the results were significant (p<0.001). Firmness of fruit declined during storage, but it did not decrease in direct proportion. On the storage of day 15, firmness was decreased to 40% of initial state. At last, all the experiment data are summarized and the relationship between firmness and weight loss is analyzed to construct a linear regression mathematical model that can predict the weight loss with the firmness value measured by non-destructive method. This research result could be useful in helping tomato exporters and suppliers to get real-time quality factor by using proposed method and regression model.

The Classification of Forest Vegetation Types and Species Composition in the Sector between Danmoknyeong and Guryongnyeong of Baekdudaegan (백두대간 단목령-구룡령 구간의 산림식생유형 분류 및 종조성)

  • This study was intended to provide basic data for more ecological and systematic preservation and management planning of forest vegetations in the Baekdudaegan sector between Danmoknyeong and Guryongnyeong where biodiversity is considered to be very important. A vegetation survey using the Zurich-Montpellier (Z.-M.) method of phytosociology was carried out in a total 142 sites to examine the environmental characteristics. The survey showed the forest vegetation types of one community group, two communities, four groups, and four subgroups. Quercus mongolica and Acer pseudosieboldianum, which fall under Constancy Class V, were dominant species and deemed to be the key species of the forest ecosystem management plan for the investigated areas. Regarding species diversity, it was found that the dominant species of the VT3 type were evenly distributed in the area while the species of the VT4 type showed not only a diverse composition but also the most abundant number of species. Moreover, there were 24 of rare plants found in the Danmoknyeong-Guryongnyeong section: the critically endangered species include Dracocephalum rupestre; the endangered species include Saxifraga octopetala; the vulnerable species include Paeonia japonica, Euphorbia fauriei, Hanabusaya asiatica, and Parasenecio pseudotaimingasa; the least concern species include Aristolochia manshuriensis, Rodgersia podophylla, Salvia chanryoenica, Scopolia japonica, and Parasenecio firmus.

Three Crystal Structures of Dehydrated $Ag_{12-x}Na_x-A$ (x: 4, 6, and 8) Treated With Cesium Vapor (탈수한 $Ag_{12-x}Na_x-A$(x :4, 6, 및 8)를 세슘 증기로 처리한 세가지 결정구조)

  • Ag+ 이온이 부분적으로 치환된 3가지 제올라이트 A(Ag4Na8-A, Ag6Na6-A 및 Ag8Na4-A)를 완전히 탈수한 후 280℃에서 24시간동안 약 0.1 Torr의 Cs중기로 처리하 였다. 이들의 결정구조는 22(1)℃ 노에서 입방공간군 Pm3m (단위세포상수 a가 각각 12.321(3) A, 12.295(1) A 및 12.380(7) A임)을 사용하여 단결정 X-선회절법으로 해석 하였다. 이들 세가지 구조에서 Cs+이온은 각각 서로 다른 4개의 결정학적 위치에서 발견되었다. 단위세포당 3개의 Cs+이온은 8-링 중심에 위치하고, 약 6.9-7.3개의 Cs+이온 은 큰 동공의 6-링과 마주보는 위치에 있는 3회 회전축상에서 발견되었다. 그리고 약 2.17-2.74개의 Cs+이온은 소다 라이트 동공내에서 발견되며 약 0.5-1.0개의 Cs+'이온은 4링과 마주보는 곳에 위치한다. 또한 이들 구조에서 단위세포당 각각 1.88(5),2.30(3) 및 5.28(10)개의 Ag종이 존재하며 이들은 큰동공의 중심에서 헥사실버 클러스트를 형성한다. 8-링위치가 Cs+이온으로 모두 차있어서 Ag0가 골조밖으로 이동하는 것을 막을 수 있다. 각각의 헥사실버 클러스터는 서로 다른 좌표에 위치하는 14개의 Cs+이온에 의해 안정화된다. 이들 구조에서 발견되는 약 12.35-13.49개의 Cs+이온들은 Cso의 흡착이 일어나 제올라이트 기본 골 격에 있는 음이온 전하와 균형을 맞출 수 있는 12개의 Cs+ 이온 이상의 이온 또는 원자로 존재하고 있다. Cs+의 배열 은 다음과 같은 두 가지 배열로 쉽게 설명할 수 있다. 일부는 2개의 Cs+이온이 소다이트 동공내에 있는 6-링과 마주보는 곳에 위치하고 큰동공내에는 6개의 Cs+이온이 6링 근처에 위치하며 1개는 4-링 근처에 위치한다. 그 나머지는 소다라이트 동공내에 위치하는 3개의 Cs+이온과 한변의 길이를 3.52 A로 갖는 삼각형을 형성한 후 6-링을 통 하여 큰 동공 내에 위치한 3개의 Cs원자와 결합하게 되므로 3m (C3v)의 대칭구조를 갖는 (CS6)4+클러스터를 형성한다. 그 밖의 5개 Cs+이온은 비어있는 큰동공의 6-링에 위치한다.

Fermentation Characteristics of Whole Soybean Meju Model System Inoculated with 4 Bacillus Strains (Baciilus속 세균 4종을 이용한 콩알메주 Model System의 발효특성)

  • Whole soybean meju was fermented with four Bacillus strains for 45 hr in its model system. The pH range of the product was $7.98{\sim}8.68$, the contents of amino nitrogen and ammoniacal nitrogen were $286{\sim}439\;mg%,\;0.11{\sim}0.23%$, respectively and that of reducing sugar ranged $0.65{\sim}2.24%$. During fermentation, the enzyme activities increased up to $30{\sim}40\;hr$ of fermentation and slightly decreased after 45 hr. Stachyose was special sugar components for B. licheniformis and raffinose was for B. natto. The components of the organic acid showed distinctive patterns among four products and the patterns of amino acids and fatty acids were almost similar to those of other reports. The main and common odor concentrates of meju were pyrazine components, 3-methyl-1-butanol, acetic acid and ethanol. Chunggukjang, mixed with B. natto and B. licheniformis showed more acceptabilities than other combinations. Soybean paste, mixed with B. megaterium and B. subtilis, soysauce, mixed with B. megaterium and A. oryzae showed excellent acceptability, respectively.

Volatile Flavor Components in Mash of Takju prepared by using Aspergillus kawachii Nuruks (Aspergillus kawachii 누룩으로 담금한 탁주 술덧의 발효 과정 중 휘발성 향기성분)

  • Volatile flavor components of Takjus mash prepared using Aspergillus kawachii nuruk were identified by GC and GC/MS. Twenty-two esters, 20 alcohols, 10 acids, 8 aldehydes, and 3 others were found in Takju mash. Thirty two components including 13 esters and 13 alcohols were detected at beginning of fermentation. Thirteen more components were detected after second day of fermentation, and 63 additional components after 12 days of fermentation. Twenty nine flavor components including 12 alcohols such as ethanol, 3-methyl-1-butanol, 2-methyl-1-propanol, and benzeneethanol, 12 esters such as ethyl acetate, ethyl caprylate, and ethyl butyrate 3 aldehydes, and 2 acids were detected during fermentation. Major volatile components detected during fermentation included 3-methyl-1-butanol, ethyl caprylate, and benzeneethanol. Peak areas of 2-methyl-1-propanol, 1-hexanol, 2, 3-butanediol (D.L), 1-dodecanol, 2-phenylethyl acetate, ethyl acetate, and monoethyl butanoate were higher than those of other components depending upon fermentation period.

Antibacterial Activity of Xanthium strumarium L. Extract Against Bacteria Causing Eye Disease (안질환세균에 대한 도꼬마리 추출물의 항균 활성)

  • Purpose: The purpose of this study is to investigate the antibacterial activity Xanthium strumarium L. extract against bacteria causing eye disease and to examine the possibility as a natural preservative on behalf of synthetic preservatives which are added to the contact lens solution. Methods: Antibacterial activity of Xanthium strumarium L. extract against the bacteria causing eye disease was assessed using agar diffusion method, and determined by whether clear zone was formed around paper disc and in terms of the size(mm) of clear zone.Staphylococcus aureus and Pseudomonas aeruginosa were used in the study. Results: Xanthium strumarium L. extract provided activity against Staphylococcus aureus KCCM 40050 in a concentration of $1000{\mu}g/20{\mu}l$with an clear zone of 11.5 mm. Moreover, Xanthium strumarium L. extract showed an activity against Pseudomonas aeruginosa KCCM 11803 in a concentration of $1000{\mu}g/20{\mu}l$with an clear zone of 9.0 mm, and against Pseudomonas fluorescens KCCM 40223 with an clear zone of 7.0 mm. Conclusions: Antibacterial activity of Xanthium strumarium L. extract against the bacteria causing eye disease was proved. This result suggests that Xanthium strumarium L. extract can be a potential natural preservative, which is added to contact lens solution.

Antibacterial Activity of Sea-mustard, Laminaria japonica Extracts on the Cariogenic Bacteria, Streptococcus mutans (충치균 (Streptococcus mutans)에 대한 다시마 추출물의 항균활성)

  • In previous reports the authors have screened the inhibition effects of marine algae extracts on halitosis, and demonstrated that a brown algae, Eisenia bicyclis (' Daehwang') possess not only strong deodorant effect bug also considerable anticariogenic activities. In this study, we screened antibacterial effects of various marine algae, and measured minimum inhibitory concentration (MIC) value of them against mutans streptococci in vitro. Among the 27 species of marine algae, $80\%$ ethanol extract of dried sea-mustard, Laminaria japonica ('Dasima') showed the strongest inhibition activity against Streptococcus mutans KCTC 3300. The extracts of Ulva lactuca ('Galparae'), Codium fragile ('Cheonggak'), Ecklonia cava ('Gamtae'), E. stolonifera ('Gompi') and Undalia Pinnatifida ('Miyeok') showed slightly weaker inhibitory potency than L. japonica. Differences of MIC values in $80\%$ ethanol extract of some species of marine algae were observed depending on test bacterial species, i.e., S. mutans KCTC 3300 or S. sobrinus KCTC 3307. Eighty percent ethanol extract of dried L japonica was fractionated with diethyl ether, chloroform, ethyl acetate, n-buthanol and water successively, The ether-soluble fraction had inhibitory effect on S. mutans KCTC 3300, however the inhibitory effects were not found in the other fractions. The MIC values of $80\%$ ethanol extract and ether fraction were 180 and 105 $\mu$g/mL respectively, while no significant inhibition activity of water-soluble fraction was found even when the fraction was added up to 5,500 $\mu$g/mL.

Enhancement of Species-specific Attraction by Addition of a Minor Component of Sex Pheromone Gland of Grapholita dimorpha (복숭아순나방붙이 성페로몬샘의 미량성분 첨가에 따른 종특이적 유인력 증가 효과)

  • Two congener species of Grapholita molesta and G. dimorpha have similar host ranges and adult occurrence patterns. In addition, the two species commonly use cis-8-dodecenyl acetate (Z8-12:Ac) and trans-8-dodecenyl acetate (E8-12:Ac) as their major sex pheromone components. The commercial lures made of the two components, therefore, suffer a mixed attraction of these two species. This study was conducted to assess any effect of their known minor sex pheromone components to reduce the mixed attraction. When cis-8-tetradecenyl acetate (Z8-14:Ac) was added to the commercial sex pheromone consisting of Z8-12:Ac/E8-12:Ac = 85/15, the attraction to G. dimorpha was not affected, but its attraction to G. molesta was significantly reduced. However, the addition of either dodecanyl acetate (12:Ac) or tetradedecanyl acetate (14:Ac) did not influence on the attraction of both species to the commercial lure. The addition of E8-14:Ac to 3-component sex pheromone, Z8-12:Ac/E8-12:Ac/Z8-14:Ac = 85/15/10, the attraction to G. dimorpha was rather significantly suppressed. From these results, the 3-component sex pheromone, Z8-12:Ac/E8-12:Ac/Z8-14:Ac = 85/15/10 was suggested as an improved composition of sex pheromone lure for G. dimorpha.

Analysis of Explanations and Examples of the Brønsted-Lowry Model Presented in Chemistry Textbooks Developed by 2009 Revised Curriculum (2009 개정교육과정의 화학교과서에 제시된 Brønsted-Lowry 모델에 관한 설명과 예시의 문제점 분석)

  • In this study, we analyzed the explanations and examples of Brønsted-Lowry model in Chemistry I and Chemistry II textbooks of the 2009 revised curriculum. In particular, the definition of the Brønsted-Lowry model, the examples, and the content of experiments were analyzed by the process perspective of chemical equilibrium, emergent process. The analyzed textbooks were 4 kinds of Chemistry I textbooks and 4 kinds of Chemistry II textbooks in 2009 revision curriculum. As a result, Chemical I textbooks did not adequately show the chemical equilibrium viewpoint when explaining the Brønsted-Lowry model. In the Chemistry II textbooks, the examples of Brønsted-Lowry model were not present emergent process viewpoint, and those were described as sequential viewpoint of Arrhenius model. In addition, examples of experiments to demonstrate the Brønsted-Lowry model of Chemistry II textbooks were insufficient. The experimental examples related to the definition of acid bases were at the level of classification by the color change of indicators. The experimental examples for explaining the strength of acid and base were to compare current intensity or amount of hydrogen gas generated from the reaction with metal. In addition, all textbooks presented the state of aqueous solution when describing the Brønsted-Lowry model, causing problems with differentiation from the Arrhenius model. Therefore, it is necessary to develop examples of experiments to help students understand Brønsted-Lowry model by presenting acid and base reaction in the non-aqueous solution state.
