  • A pot (1/200a) experiment was conducted to find out the changes of soil and water temperature on the several paddy soil weeds (Duck weed, Algae and Floatin fern) growing condition. The results obtained were shown as follows; 1. Diurnal maximum soil and water temperature were lowered by 2 to $3^{\circ}C$ under duck weed growing condition while minimum soil and water temperature were higher in $0.5^{\circ}C$ and/or similar tendency than that of bare paddy soil condition. 2. The changes of soil and water temperature were lowered by 0.1 to $0.3^{\circ}C$ at sunny day under Duck weed growing condition but was higher in 0.1 ta $0.4^{\circ}C$ at rainy, cloudy and after rainy day than that of bare paddy soil. 3. Soil and water temperature were higher about 0.3 to $1.0^{\circ}C$ under algae growing condition than that of bare paddy condition. 4. The effects of keeping warm efficiency were high in order of Duck weed > Duck weed with algae > algae > bare paddy soil condition.

  • High frequency plant proliferation system via direct frond regeneration of endemic duckweed plants Lemna paucicostata and Spirodela polyrhiza was established. Fronds of L. paucicostata and S. polyrhiza were able to multiply half-strength MS basal medium without plant growth regulators. However, addition of BA at a range of 1 to 3 mg/L was more effective than high concentration of BA treatments for fronds proliferation. Also half-strength MS salts was suitable for the fronds proliferation. Increase of salts concentration had inhibitory effect on fronds proliferation. Also the frequency of callus formation from fronds of L. paucicostata was 3.3%, when they cultured onto 1/2 MS medium supplemented with 1 mg/L of BA. Similarly the frequency of callus formation from S. polyrhiza was very low. After subculture of white globular structures derived from fronds of L. paucicostata, numerous globular somatic embryos and calluses were developed onto the surface of fronds. However these somatic embryos did not fully develop into normal plants when transferred to 1/2 MS basal medium. Therefore direct frond regeneration system was more efficient for mass proliferation of L. paucicostata and S. polyrhiza. The plant regeneration system of L. paucicostata and S. polyrhiza established in this study, might be applied to mass proliferation and genetic transformation for molecular breeding.

  • The golden apple snail-Pomacea canaliculata was an alien species and introduced into Korea as table use in 1983. This species is possible pest in direct seeded rice paddy in Korea. We investigated the effects of this exotic snail on the macrophytes and the water quality in water canal. Also, the basic life cycle of this species and the overwintering ecology in Chungnam and Jeonbuk province were elucidated. A survey of natural water canal in Korea showed that high densities of the snail were associated with absences of macrophytes-Hydrilla verticillata, Ceratophyllum demersum, Spirodela polyrhiza and Lemna perpusilla. Experiments in water tank also demonstrated that the golden apple snail can cause the loss of macrophytes, and increase the EC(Electric conductivity), COD(Chemical oxygen demand), T-N(Total nitrogen) and T-P(Total phosphorus) in water.

  • A phytotoxin was purified by repeated chromatography from liquid cultures of Fusarium oxysporum BG isolated from barnyardgrass. Its chemical structure was determined to be dehydrofusaric acid by mass and NMR spectral analyses. The substance showed a potent phytotoxic activity against growth of duckweed with a $EC_{50}$ value of $1.5{\mu}g/ml$. It also inhibited the root growth of barnyard millet, cress, barnyard grass, and rice cultivar 'Dongjin'. However, it had no inhibitory activity against seed germination of barnyard millet and cress, and the shoot growth of the four plant species.

  • Lemna gibba was newly cultured and provided for toxicity tests. In this study, the chromate toxicity tests for Lemna gibba were performed according to the OECD Lemna growth inhibition test guideline. The test species was Lemna gibba, and the tests were repeated 5 times. To evaluate the toxicity test results, the average specific growth rate, EC50, 95% confidential limit, and variances were calculated. The test performance was analyzed by the doubling time and test statistics. The average values of EC50 data determined by logistic and linear interpolation curves were 25.9 ppm and 35.4 ppm respectively (by chromate concentration). The doubling time of all controls were below 2.5 day, so all tests passed the criteria for the test performance. This study introduced a new test method, Lemna growth inhibition test, which is provided for the hazard assessment of aquatic environment.

  • In order to select potent bioactive isolates, 70 Fusarium isolates obtained from soil and 21 plant species were screened by antifungal, insecticidal, herbicidal, and duckweed bioassays after culturing in potato dextrose broth and rice solid media. Eight (11.4%) of the 70 liquid broth cultures showed disease-controlling activities more than 80% against at least one of the 6 plant diseases tested. Fusarium sp. FO-68 isolate exhibited the most potent antifungal activity; it controlled rice blast, wheat leaf rust, and barley powdery mildew with control values more than 95%. Out of 70 solid cultures, 21 (30.0%) controlled at least one plant disease more than 80% and F. equiseti FO-68 isolate showed disease-controlling activities more than 95% against 3 plant diseases such as rice blast, tomato late blight, and wheat leaf rust. As for tile insecticidal activities, 2 liquid and 1 solid cultures showed potent insecticidal activities against pest insects more than 80%, Liquid cultures of F. oxysporum FO-61 and Fusarium sp. FO-80 isolates exhibited insecticidal activities more than 80% against green peach aphid and diamondback moth, respectively. The solid culture of Fusarium sp. FO-510 isolate had 80% insecticidal activity against green peach aphid. However, none of liquid and solid cultures of the 70 Fusarium isolates showed potent herbicidal activities against 10 upland weeds. As the results of duckweed assay, 3 liquid cultures showed 70% growth inhibitory activity at concentrations less than 1.25% of culture supernatants and 9 solid cultures had a potent inhibitory activity against duckweed growth. On the other hand, there was a significant correlation between antifungal activities and herbicidal activities against duckweed of both liquid and solid cultures of tile 70 Fusarium isolates.

  • The climate change affects the growth and development of weeds as well as the outbreak of weeds. Especially, the occurrences of problematic paddy weeds due to climate change might cause the difficulties in weed control. This study therefore, investigated the current dominance and distribution of paddy weeds. As a result of the study on paddy weeds in northern Gyeonggi-do, there were total of 65 taxonomy groups including 23 family, 41 genus, 57 species, 7 subspecies and 1 variety. Among all the plants, 46 species were annual plants and 16 were perennial plants. Echinochloa crus-galli was the highest in importance analysis and the followings were in order of Ludwigia prostrate and Lemna paucicostata. The similarity of different paddy weeds in different regions observed through TWINSPAN analysis was distinguished by Fimbristylis miliacea, Rotala indica and Cyperus flaccidus. Regional differences shown in CCA analysis using weed species and soil environment revealed that Gimpo-si and Namyangju-si has difference soil and weeds, which are features that distinguished them from other regions. In northern Gyeonggi-do the result of paddy weed research showed the interregional difference not in dominant weeds but in distribution species.

  • The membrane potential in the subepidermal cells of Lemna gibba G3 fronds was measured in the dark with glass capillary microelectrodes. At pH 7, the membrane potential, approximately-215 mV, could be depolarized to -82∼-88 mV by 0.1 mM dicyclohexylcarbodiimide (DCCD) or by KCN at 0.3 mM or higher concentrations. When the pH of the medium was altered the potential showed reversible changes, while it revealed no response to the external pH changes when energy transduction across the membrane was being blocked by 0.1 mM DCCD. The results support an assumption that the active component of the membrane potential of Lemna subepidermal cells is generated by electrogenic H+ -pump. By the addition of 0.10∼5.00 mM salicylic acid(SA) to the bathing medium the membrane potential was depolarized to a great extent, and the removal of SA from the medium repolarized the potential showing almost complete recovery, 92.3∼97.6% to the initial levels. Although the potential was greatly depolarized by 5.0% or higher concentrations of dimethylsulfoxide (DMSO), the recovery rate by DMSO removal was decreased as the pretreatment concentration had increased. Twenty percent DMSO pretreatment limited the recovery at only 47.1%. The presence of SA in the bathing medium could reversibly increase the permeability of the plasmalemma. DMSO at its concentration of 5.0% or higher increased the permeability of the membrane by irrevesibly impairing the membrane component involved in the membrane permeability.
