Effects of Shading Rate, Rooting Media and Plant Growth Regulators on Rooting of Veronoca L. by Cuttings (삽목 번식 시 차광정도, 삽목용토 및 생장조절제 처리가 꼬리풀(Veronica L.)의 발근에 미치는 영향)

  • This study was conducted to develop a mass production for a commercial use by cutting of 4 kinds of V. glabrifolia Kitag., V. pusanensis Y.N.Lee, V. glabrifolia Kitag. × V. Spicata 'Alba' and V. spicata 'Ulster Blue Dwarf' × V. longifolia. Veronica L. Effects of shade, media and concentrations of plant growth regulators on the rooting of Veronica L. were examined. The rooting rate of V. glabrifolia Kitag. was higher as a 60% than the V. pusanensis Y.N.Lee as a 20% in the control of commercial media. In shading treatment, the rooting rate was highest at 30% or 60% shading, and the 30% shading was the best in number of root and root length, but the 90% shading was lower than no shading treatment. For cutting media, the rock wool and 100% perlite increased the rooting rate by more than 10% compared to commercial media, and increased the number of roots by 2 or 3 times. However the Cocopeat of media was lower of rooting rate, root number, and root length compared to another treatment. In the plant growth regulator treatment, the rate of rooting increased in all treatment compared to control, and was highest at IBA 1,000mg·L-1 as a 82.2% and NAA 200 or 400mg·L-1 as a 82.2% or 84.4% respectively, in the V. glabrifolia Kitag. × V. Spicata 'Alba'. However, the root number and root length was decreased as the concentration of growth regulators increased.

Plug Seedling Production Using Tissue Cultue Regenerants and Their Seeds in Bupleurum falcatum L. (자호(紫胡)의 조직배양묘(組織培養苗)와 배양식물(培養植物)의 종자(種子)를 이용(利用)한 프러그묘(苗) 생산(生産))

  • This study was conducted to establish technologies for plug seedling production using somatic embryos-derived regenerants and their seeds in Bupleurum falcatum L. Among distilled water, GA (0.1mg/l) and putrescine (0.lmg/l) treated to regenerants for acclimatization, GA was most effective to develop shoots and roots, 1/2X MS medium and NAA 0.1mg/l + BA 0. 5mg/l enhanced the growth rates of the regenerants and increased dry weight. Activated charcoal effected to grow markedly leaves and roots of the regenerants at the level of 0.4 %. Regenerants increased their plant height, root length and dry weight at $30^{\circ}C$. Plug seedlings originated from seeds of the tissue culture regenerants showed the maxium growth on the mixture of peatmoss soil (2) and mountain sand (1) .Root length, leaf area and dry weight of plug seedlings increased significantly when No.1, 2 and 3 of Wondergrow solution were mixed in the ratio of 1.3 - 0.9 - 0.1. Light supplement (4%) and high tem­perature $(30^{\circ}C)$ promoted the growth of plug seedlings as well as dry weight. Ninety days seedlings were more vigorous and adaptable for transplanting than other seedlings.

Uptake of Fe and Mn in Red Pepper and Tomato Plants under Different Soil Conditions (토양조건에 따른 고추와 토마토의 철 및 망간 흡수특성)

  • This experiment was performed to understand the relationship between uptake of Fe and Mn by plants, red pepper and tomato, and soil physico-chemical properties under different soil conditions at an environmentally controlled chamber in NAAS(National Academy of Agricultural Science) in 2008. After the dipping for 3 days, four treatments, dipping, dipping+aeration, drainage, drainage+aeration, were set up to investigate the changes in soil redox potential and moisture content. Drainage+aeration changed soil to the oxidation condition from 72 hrs of treatment, and soil moisture content was immediately reduced after treatment. Uptake of Fe and Mn of red pepper was investigated with two treatments, soil only and the mixed[soil(50%) : bed soil(35%) : bark(15%)]. Red pepper leaves taken at 30 days after treatment absorbed excessively Mn from the treatment of soil only and the mixed, and thus uptake of iron was strongly reduced. Also, uptake pattern of Fe and Mn of tomato was examined with four treatment, soil only, soil(50%) + rice straw(50%), soil(50%) + compost(50%) and soil + aeration. Contents of Fe and Mn in tomato leaves was measured at 60 days after treatment. Fe content was the greatest in soil(50%) + compost(50%) whereas Mn content was the highest in soil only. As a result of this experiment, plant growth was stronger influenced by soil moisture content than redox potential or porosity, and the oxidation status of soil was likely to promote that plant predominantly absorbed Mn from soil and thus resulted in Fe deficiency.

Effects of Temperature, Light and Plant Growth Regulators on the Seed Germination of Lavandula angustifolia Mill (잉글리쉬 라벤다의 종자발아(種子發芽)에 대한 온도(溫度), 광(光) 및 생육조절물질(生長調節物質)의 영향(影響))

  • This study was conducted to establish mass propagation system from seeds of Lavandula angustiolia Mill. Only 4% of dry seeds were germinated when they were incubated in $4^{\circ}C$. Germination rate of stratified seeds for 9 weeks was 15% higher in light than darkness. Soaking with 1000mg/l of $GA_3$ was helpful to overcome the effect of darkness. The optimal temperature for germination was $25^{\circ}C$ for the seeds that were treated with$GA_3$, solution. For the improvement of germination rates, pretreatment of $GA_3$ at 500-2000mg/l showed about 75% of germination, and in the combination treatment of $GA_3$, and BA, germination rate increased by about 10% in the treatment of 1000mg/l $GA_3+10mg/l$ BA compared with the 1000mg/l $GA_3$ treatment. Cold stratification treatment was very effective for seed germination, and over 70% of seeds were germinated when they were incubated in $4^{\circ}C$ for 9-12 weeks. In addition, there was synergic effect on the seed germination subject to stratification and 1000mg/l $GA_3$, treatment for 6 months. In the mixture of vermiculite: peatmoss (1 : 1, v/v), emergence rate was 6.7% in control and 65% in 1000mg/l $GA_3$ treatment, respectively.

Propagation Efficiencies at Different LED Light Qualities for Leaf Cutting of Six Echeveria Cultivars in a Plant Factory System (에케베리아 6품종의 엽삽 시 식물공장시스템 내 LED 파장에 따른 번식 효율)

  • The succulent plants of Echeveria genus are in increasing demand worldwide, but it is difficult to supply good quality young plants throughout the year because propagation efficiencies are depend on cultivar and environmental factors. This study was carried out to investigate the propagation efficiencies of leaf cutting in Echeveria cultivars at different LED light qualities in a closed-type plant factory system. Leaf cuttings cut from stock plants of six difficult-to-propagated cultivars 'Afterglow (AG)', 'Berkeley Light (BL)', 'Mason (MS)', 'Subsessilis Light (SL)', 'Cream Tea (CT)', and 'Ben Badis (BB)' were put into cutting media in the plant factory system maintained at a temperature of $24{\pm}2^{\circ}C$ and relative humidity of $60{\pm}10%$, and watered with over-head irrigation twice a week. Cuttings were irradiated with sole or mixed red (R, 660 nm), blue (B, 450 nm), green (G, 530 nm), and far-red (FR, 730 nm) LEDs as follows: R10, R8+B2, R5+B5, R7+B2+FR1, and R7+B2+G1. PPFD just above the cuttings was $200{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and photoperiod was 16/8 (light/dark) hours. As a result, propagation efficiencies were dependent on cultivar. Rooting and shooting were relatively easy in 'SL' but shoot formation in 'AG' was very difficult. Light qualities from LEDs also affected plant regeneration. Light conditions with a higher ratio of B, R5+B5, R7+B2+FR1, and R7+B2+G1, promoted shoot formation and growth but inhibited rooting and root growth. R10 and R8+B2 with a higher ratio of R promoted rooting and root growth and inhibited shoot formation and growth of cuttings. In addition, the treatment with FR increased leaf size and biomass of the all plants. Therefore, further studies are needed to investigate the optimum compositions of LED light quality for the improvement of leaf cutting efficiency in difficultto-propagated Echeveria cultivars.
