Perforation Adjustment of Unit Package for 'Fuji' Apples during Short-term Cold Storage and Export Simulation ('후지' 사과의 단기 저온저장 및 모의수출 과정에서 소포장의 천공도 조절 효과)

  • Various types of unit packaging methods were applied for 'Fuji' apples during short-term cold storage and export simulation. Gas tightness of the package was controlled stepwise in the successive two-year experiments using different perforation treatments (none, punch hole, or pinhole) and sealing methods (tie v s. heat seal). Risk of tight packaging and effectiveness of macroperforation on weight loss and quality maintenance were analyzed as related to changes in gas concentration inside the packages. Immediately after harvest, each 5 apple units were packaged in $40{\mu}m$ polypropylene (PP) film bags, stored 4 weeks at $0^{\circ}C$, and then put on the shelf for one week at ambient temperature in the preliminary experiment, In the main experiment, export process was imposed after storage simulating 2 week refrigerated container shipment at $0^{\circ}C$ plus one week local marketing at ambient temperature. Non-perforated film packaging with relatively high gas tightness induced flesh browning caused by carbon dioxide accumulation regardless of the sealing methods. Among perforated film packaging, in contrast, atmospheric modification was partly established only in the pinhole treatment and flesh browning symptom was not observed in all the treatments. Even the punch hole perforated film packaging without gas tightness effectively reduced the weight loss, whereas had slight benefits for quality maintenance. Reduced perforation using pinhole treatment seemed to improve sensory texture, while effects on physicochemical quality were insignificant. Overall results suggest the need of more minute perforation treatments on the packaging film to ensure modified atmosphere effects on quality maintenance.

Effect of starvation on tissues in Far Eastern catfish(Silurus asotus) (메기(Silurus asotus) 기아 시 조직학적 변화)

  • A 210-day experiment was conducted to examine the effects of starvation on survival, the gonadosomatic index (GSI), hepatosomatic index (HSI), and the intestinosomatic index (ISI), and histological changes in the renal tubule epithelium, midgut epithelium, and hepatocytes in Far Eastern catfish (Silurus asotus). The survival rate decreased to 92.2±0.47% in the fed group and 74.4±2.59% in the starved group during the 210-day experimental period. GSI, HSI, and ISI were lowest in the starved group (p<0.05). The hepatocyte nuclear area, hepatocyte cell area, the nuclear height of the midgut epithelium, and the nuclear height of the kidney were highest in the fed group (p<0.05). The hepatocyte nuclear area, nuclear height of the midgut epithelium, and nuclear height of the kidney were lowest in the starved group(p<0.05). The numbers of melano-macrophages (MMs) found in the kidney cells increased during starvation in this species. This suggests that thinner body cavity regions, the contraction of hepatocyte nuclear sites, and the spreading of kidney cell MMs in this species could be used as alternative indicators for identifying starvation conditions. Therefore, the results from our study provide accurate indications of the nutritional status of Far Eastern catfish.

Quality characteristics of outer leaves of Kimchi cabbage according to various blanching treatment conditions (국내산 배추 우거지의 blanching 처리에 따른 품질 특성)

  • The aim of this study was to establish an optimum blanching condition for outer cabbage leaves to increase utilization of the byproduct. Outer leaves of Kimchi cabbages were cut to size ($4{\times}4cm$) and blanched at 80, 90, $100^{\circ}C$ for 1, 3, and 5 min. Subsequently, weight loss, pH, total bacteria, coliform and texture properties were investigated. Sensory evaluation of a ready-to-cook soup prepared from blanched outer leaves was investigated to determine whether the quality of the blanched outer leaves was acceptable. Both weight loss and pH of outer leaves of Kimchi cabbage were increased by blanching. Compared to raw samples, total bacterial counts decreased and coliform bacteria were not detected. The hardness of raw leaf samples was $1.5kg{\cdot}force$ (N). Sample hardness was 18.7~21.7% lower after blanching for 3 min and 34.8~36.7% after blanching for 5 min. Sensory evaluation of outer leaves blanched at $100^{\circ}C$ for 5 min revealed significant differences from the other treatments (p<0.05). In addition, sensory evaluation scores for texture and appearance of outer leaves blanched at $100^{\circ}C$ for 3 min were higher than those for commercially available products. The results of this study indicate that blanching can be effective for developing a food product from the outer leaves of Kimchi cabbage.

Effect of Monascus purpureus-Fermented Korean Red Ginseng Powder on the Serum Lipid Levels and Antioxidative Activity in Rats (홍국발효홍삼 분말 급여에 의한 흰쥐의 혈청 지질 농도 및 항산화 활성에 미치는 영향)

  • Monascus-fermented Korean red ginseng (MFRG), fermented by Monascus purpureus KCCM 12002, may be an ideal candidate for the hepatoprotectic, hypolipidemic and antioxidative activities. Effect of MFRG powder on these parameters in rats was investigated. Body weight gain, food intake, and water consumption were not significantly different among the groups. Total and relative weights of liver were significantly higher in MFRG group than that in other groups. The activities of AST and $\gamma$-GTP were highly lowered in MFRG group compared to control group. Contents of serum total lipid and triglyceride were significantly lowered in silymarin group and were significantly increased in MFRG group compared to control group, but tended to be lowered in RG group. Serum content of total cholesterol tended to be lowered in silymarin, RG, and MFRG groups compared to control group. HDL-cholesterol contents was only significantly increased in MFRG group compared to control group. At the same time, atherogenic index (AI) was also significantly lowered in silymarin, RG and MFRG groups compared to control group, and this effect was more pronounced in MFRG group. Content of thiobarbituric acid reactive substances (TBARS) in the liver was significantly lowered in MFRG group and tended to lowered in silymarin and RG groups compared to control group. The hepatic glutathione concentration was significantly higher in silymarin and MFRG groups. Hepatic morphology in all experimental groups revealed clear-cut hepatic lobules with the uniform pattern of the polyhedral hepatocytes radiating towards the periphery from the central vein. These results suggested that MFRG may have anti-atherogenic index (AI) and antioxidative activity in normal dietary feeding rats.

  • In this study, we investigated the preventive and therapeutic effects of antler-extract for osteoporosis. Rats were ovariectomized bilaterally and were fed up with Ca- and P-free diet in order to induce osteoporosis. Body weight, organ weight, the weight of femur and bone ash quantity were examined for 5 weeks. We also performed histological and electronical microscopic examinations. 1. After adminstration of female and male antler extract to osteoporosis-induced rats at the doses of 625 and 1250 mg/kg, respectively, the body weights were significantly increased compared with those of normal control group's which was 230.2:t2.3-281.0:t2.5g (p<0.05). 2. The weights of both right and left femur of osteoporosis-induced rats, administered with female or male antler-extract, little decreased compared with those of normal control group. 3. The bone ash quanties of femur of osteoporosis-induced rats, administered with female or male antler-extract, little decreased compared with those of normal control group. 4. The weights of liver, spleen, and kidney of osteoporosis-induced rats, administered with female or male antler-extract, decreased compared with those of normal control group. 5. Histological and electronic microscopical findings were (1) that in normal control rats the connectional of lacunae appeared well and were without loss of bone mineral, (2) that in ovariectomized rats the connections of lacunae were mostly broken and were with loss of bone mineral compared with those of normal control rats, (3) that in osteoporosis-induced rats, administrated with female or male antler-extract, the shape of lacunae and the connections of them were similar to those of normal control rats. These findings suggest a possible protective and therapeutic effects of female or male antler extract against bone loss in ovariectomized rats.

Effect of Supplement of Various Antibiotics Alternatives on Performance, Organ Weight, Cecal Microflora, and Blood Characteristics in Broilers (다양한 항생제 대체제의 첨가 급여가 육계의 생산성, 조직 중량, 장내 미생물 균총 및 혈액 특성에 미치는 영향)

  • This study was conducted to investigate the effect of dietary supplementation with Lactobacillus, medicinal plant extracts, and plant extracts on growth performance, cecal microflora, relative organ weight, and lymphocyte profile in broiler chickens. One hundred broilers (5 wk old) were used and divided into five (treatment) ${\times}$ two (challenged Salmonella) groups containing 10 broilers. The treatments were as follows: no antibiotics group (NC), antibiotics group (PC), 0.1% lactic acid bacteria group (LB), 100 ppm medicinal plant extract group (MPE), and 100 ppm herb extract group (HE). A basal diet was formulated as 3,100 kcal/kg ME and 20% CP based on corn and soybean meal. Broilers were fed with the experimental diets with no challenge for 1 wk and with challenge of SG for 2 wk. SG were added in water as $1.0{\times}10^6cfu/L$. Final body weight and weight gain were significantly decreased following challenge with SG (P<0.05). However, with the addition of antibiotic alternatives, growth performance was improved and reduction of performance following challenge with SG was lowed compared with that of the NC treatment (P<0.05). Coliform bacteria and Salmonella but not lactic acid bacteria increased with the addition of antibiotic alternatives (P<0.05). Lactobacillus increased significantly with the addition of lactic acid bacteria compared with the NC and PC treatments (P<0.05). The weight of liver, spleen, and bursa of Fabricius increased with addition of antibiotic alternatives (P<0.05). WBC was highly reduced at 4 days after challenge with SG, but was normally maintained thereafter. There was no significant difference in coliform bacteria, Salmonella, organ weights, or WBC with the addition of antibiotic alternatives. Finally, Lactobacillus, medicinal plant extract, and herb extract can be used as antibiotic alternatives; but one alternative completely can't alternate as antibiotics. Therefore, further studies are needed to investigate the synergistic effects of two or more antibiotic alternatives.

  • 식품동결에 의한 품질손상에 있어 크게 영향을 미치는 인자 중의 하나가 체적변화에 의한 조직 파괴이므로 식육 및 과채류의 냉동시 내부압력에 따른 조직파괴를 방지하기 위한 기초연구로 다양한 동결방법에 따른 내부압력 변화에 대하여 조사한 결과, 급속 및 완만동결시 우육의 내부압력 변화 경향은 동결 직전에 급격히 상승한 후 동결 초기부터 최대빙결정생성대를 통과하는 동안은 급격히 감소하다가 다시 온도가 내려감에 따라 압력변화는 증가와 감소를 반복하여 상승하는 경향을 보여 주었고, 동결시의 내부압력 크기는 약 8~10 psig 수준이며, 내부압력의 변화 경향은 정지공기식보다 침지식에서 약 1 psig 정도 크게 나타났다. 냉동냉장시에 일어나는 내부압력의 상하변화는 돈육의 품온차가 ${\pm}1^{\circ}C$일 때, 시료중량에 따라 1.84~2.32 psig 정도의 내부압력 차가 반복적으로 발생됨을 알 수 있었다. 또한, 돈육의 해동시 내부압력은 해동 개시 후 급격히 상승하여 5분 이내에 최고압력에 도달한 이후에는 서서히 하강하였으며, 해동시에 발생한 내부압력의 값은 동결시의 내부압력 값보다도 대부분 크게 나타났다. 또한, 균온처리한 우육의 동결시 내부압력 값은 약 1~4 psig 수준으로 균온처리하지 않은 우육에 비하여 매우 적게 나타났으며, 동결소요시간도 약 10~20% 정도 짧게 나타났다. 그리고 균온처리 및 다양한 동결방법에 따른 동결식육의 조직과 냉동냉장 중의 품질변화를 비교 검토한 결과, 동결방법에 따른 드립손실율은 우육의 경우 정지공기식으로 처리한 시료는 타 처리구에 비해 계속 높게 나타났으며, 돈육은 우육에 비해 상대적으로 드립손실량이 적게 나타났으나 송풍식으로 처리한 시료에서 저장 40일째, 7.39%로 가장 높게 나타났다. 동결 우육 및 돈육의 pH변화는 뚜렷한 차이를 나타내지 않았으며, VBN 및 TBA값에 있어서는 균온 처리한 시료가 완만 및 급속동결 처리한 타 시료에 비해 저장 200일까지 가장 변화가 없었다. 동결저장온도의 상하변동에 있어서는 온도변동 횟수가 증가할수록 pH 및 수분함량은 뚜렷한 변화가 없었으나 드립손실율, VBN 및 TBA값은 온도 변동횟수가 증가할수록 서서히 증가하는 경향을 보여 주었다.

Effect of Salicornia herbacea L. Supplementation on Tissue Triglyceride Concentrations and PGC-1α & PPAR-γ Expression of Skeletal Muscle of Rats Fed a High-fat Diet (함초의 보충식이가 고지방식이 흰쥐의 혈청 및 조직의 중성지방 농도와 골격근 내 PGC-1α 및 PPAR-γ 단백질 발현에 미치는 영향)

  • This study examined whether the supplementation of Salicornia herbacea L. (SH), a member of the Chenopodiaceae subfamily, affects tissue specific triglyceride (TG) accumulation and the peroxisome proliferator-activated $receptor-{\gamma}$ $coactivator-1{\alpha}$ ($PGC-1{\alpha}$) and peroxisome proliferator-activated $receptor-{\gamma}$ ($PPAR-{\gamma}$) protein expressions of skeletal muscle in rats with a high-fat diet. Sprague-Dawley male rats were randomly divided into three groups: control normal diet group (CD), high-fat diet group (HD), and 5.0% SH supplemented high-fat diet group (SD). The weights of fat tissue of the SD group were reduced by approximately 25%(p<0.01), while the skeletal muscle weight of the SD group increased approximately 5% compared to those in the HD group (p<0.01). The serum and hepatic TG of the SD group decreased approximately 20% compared to those of the HD group (p<0.05). In the protein expression levels in the skeletal muscle, the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ expressions of the SD group were 1.5-folds higher than those of the HD group (p<0.01). From these results, SH supplementation contributes to the improvement of the serum and hepatic TG concentrations, and the $PGC-1{\alpha}$ and $PPAR-{\gamma}$ protein expression levels in the skeletal muscle of fed a high-fat diet. Thus, SH supplementation was effective in reducing fat mass and increasing muscle mass.

Making Fish Paste with Yam(Dioscorea japonica Thumb) Powder and Its Characteristics (마 분말을 첨가한 어묵의 제조 및 특성)

  • This study was conducted the physicochemical and sensory characteristics of fish paste with yam powder(0~5%). The pH of the samples ranged from 6.89 to 7.02, and moisture content ranged from 79.53 to 80.44%. Increasing the amount of yam powder in the fish paste tended to decrease the lightness(L) in Hunter color value while increasing the redness(a) and yellowness(b). For the textural characteristics, the addition of yam powder increased strength, gumminess, cohesiveness and springiness. Fish paste with 3~5% yam powder had good flexibility and wasn't broken even after 4 times folds. In sensory evaluation, the addition of 2% yam powder had the best score in color, taste and overall preference. Therefore, this results suggest that 2% yam powder can be applied to fish paste for the purpose of high quality, preference and functionality.

Anti-obesity Effect of Sargassum confusum Ethanol Extract in Obese Rats (알쏭이모자반 주정추출물의 항비만 효과)

  • Effect of Sargassum confusum extract on the reduction of body weight gain and lipid contents in obese rats were evaluated to find natural materials with anti-obesity benefits. After inducing obesity by feeding 42.5% high-fat diet for 5 weeks, each 10 Sprague-Dawley rats were randomly assigned to high-fat diet control (HFD) group and high-fat diet group containing 3% Sargassum confusum extract (HFDSC). Weight gain of HFD group ($2.96{\pm}0.31g/day$) was significantly (p<0.05) higher as compared to that of normal diet (ND) group ($2.19{\pm}0.17g/day$). Weights of adipose tissues of HFD group were higher than those of ND group. Body weight gain of HFDSC group, however, was $2.36{\pm}0.24g/day$, which was significantly (p<0.05) lower by 21% than that of HFD group. In addition, weights of epididymal and perirenal adipose tissues were lower by 15% and 16%, respectively, as compared to those of HFD group. Biochemical analyses showed that concentration of triglyceride, total cholesterol, and fatty acids were significantly (p<0.05) lower in HFDSC group. These results suggest that Sargassum confusum extract has a high potential as an anti-obesity material by reducing weight gain and obesity-related factors in serum.
