Peeling Operations of Root Vegetables: Potato, Sweet Potato and Carrot (근채류(감자, 고구마, 당근)의 탈피조작)

  • The effect of peeling methods, spherecity and weight of potatoes and carrots on the peeling efficiency were investigated. The changes in the surface texture by peeling were estimated by Rheometer and were related to the changes in the microstructure. The optimum mechanical peeling conditions using abrasion type rotary peeler were 90 sec. at 300 rpm for potatoes, 70 sec. at 300 rpm for sweet potatoes and 60 sec. at 300 rpm for carrots. The peeling loss was influenced by the sphericity and weight of the sample. The optimum conditions for alkali peeling were 90 sec. immersion in boiling 10% NaOH solution for potatoes, 300 sec. in boiling 10% NaOH solution for sweet potatoes and 60 sec. in boiling 6% NaOH solution for carrots. Severe damage of surface structure was noticed by alkali peeling, demonstrated by denaturation of starch granules in the cell. The structural damage observed by microscope was related to the reduction of cutting force after peeling.

고진공 수산물 조미통조림의 향기성분 분석

  • 일반적으로 시중에 유통되고 있는 일반통조림 제품은 조미액, 시럽, 물 등의 액즙을 함유하고 있어 이로 인해 제품의 중량의 증가, 가용성 성분의 희석, 액즙의 흡수와 팽윤으로 인한 조직감의 연화, 원료의 고유의 향기성분 손실 등과 조미액은 대부분 버리게 되므로 제품의 경제적 손실도 크다. 따라서 본 연구에는 종래의 통조림보다 가볍고, 조직감이 있으며 원료 고유의 향미를 살린 제품을 만들기 위한 기초적인 연구로 packing medium이 없는 고진공 통조림제품을 제조하여 일반통조림 제품과의 향기성분을 비교분석하였다. Effects of Water-Soluble Extract from Leaves of Morus alba and Cudrania tricuspidata on the Lipid Peroxidation in Tissues of Rats (뽕나무(Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎의 수용성 추출물이 흰쥐 각 조직중의 지질 과산화물 함량에 미치는 영향)

  • 뽕나무 (Morus alba) 및 꾸지뽕나무(Cudrania tricuspidata) 잎으로부터 추출한 수용성 물질을 0.5% 콜레스테롤 투여 식이에 1%(w/w) 수준으로 첨가하여 각 조직에서의 지질 과산화물 생성정도를 나타내는 TBARS 측정법으로 항산화 활성을 검토하였다. 흰쥐의 체중 및 식이 섭취량은 실험군 간의 유의적인 차이는 없었다. 간장 및 뇌 조직 중량은 뽕잎 추출물군에서 증가하였다. 신장의 지질 과산화물 함량은 대조군에 비해서 뽕잎 추출물군에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화에서 유의적으로 감소하였다. 뇌 및 심장의 지질 과산화물 함량은 대조군에 비교해서 꾸지뽕잎 추출물군에서 유의적으로 증가하였다. 간장 및 비장의 경우에는 각 실험군 간에 유의적인 차이가 없었다. 각 조직의 microsomes 분획으 지질 과산화물은 심장과 비장에서 뽕잎 및 꾸지뽕잎 추출물군에서 감소하였으나, 다른 조직에서는 현저한 차이가 없었다. 한편, 각 조직의 microsomes 분획을 얻어서 Fe2+/ascorbate로 지질 과산화물을 유도하기 위하여 37$^{\circ}C$에서 한시간 반응을 시킨 결과, 뽕잎 및 꾸지뽕잎 추출물군에서 약 절반 정도 감소하였으며, 상대적 증가량은 뇌조직 microsomes에서 현저하였다. 본 실험의 결과 신장조직 및 간장 microsome막 지질 과산화물 함량은 뽕잎 및 꾸지뽕잎 수용성 추출물에 의해 감소되었으며, 뇌 및 심장조직의 지질 과산화물 함량은 꾸지뽕잎 수용성 추출물에 의해 증가됨으로써 봉잎 추출물간의 차이를 나타내었다.

Quality Characteristics of Sulgidduk Containing Chlorella Powder (클로렐라를 첨가한 설기떡의 품질특성)

  • The quality characteristics of sulgidduk, a steamed rice cake, with chlorellaa sulgidduk) were evaluated. Chlorella sulgidduk was prepared in the different ratio of chlorella powder (0, 0.2, 0.5 or 1%, w/w) and water content (15 or 20%, w/w). In mechanical texture characteristics, hardness, gumminess and brittleness were higher in 15% water content chlorella sulgidduk and lower in 20% water content chlorella sulgidduk than in the absence of chlorella (the control sulgidduk), wherease 20% water content chlorella sulgidduk and a significantly higher adhesiveness and cohesiveness values (p<0.05) compared with control 24 hours storage at $25^{\circ}C$, the good texture characteristics were maintained in 20% water content chlorella sulgidduk. In sensory evaluation, the most favorite quality characteristics were shown in the sulgidduk with 0.2 and 0.5% chlorella powder.

The Effect of Various Culture Media on Histological Anatomy and Saikosaponin Content in Hairy Root Culture of Buplurum falcatum (시호 모상근 배양에서 배지성분에 따른 사이코사포닌 함량과 조직학적 형태의 차이)

  • Growth rate and saikosaponin content of Buplurum falcatum hairy root on MS and RCM basal media were measured. And using hairy root cultured in those media, it was investigated whether some correlation between differences in growth rate and saikosaponin content and histiological difference was or not. Results obtained showed that among media tested, MS and 3RCM medium which showed the faster growth in fresh weight and dry weight, respectively. And saikosaponin content of hairy root cultured in 3RCM was $2.2{\sim}5.5$ times higher than that of 1 year cultivated roots, whereas that in MS was extremely low. On the other hand, histological differences of root tips and about 1 cm region from root tip cultured in both MS and 3RCM were not prominent, however, in mature region, a lot of cells only in 3RCM contained densely stained vacuoles or organelles which probably contain tannin, suggesting that development of those has a correlation with biosynthesis of secondary metabolites containing saikosaponin.

  • Adult mice were injected with a single sublethal dose of cyclophosphamide. Effects of the drug on the body weight, spleen weight, and morphology of the peripheral lymphoid system have been analysed. The body weights of the mice given cyclophosphamide(300mg/kg body weight) decreased slightly and returned to normal quickly. Spleen weights, however, changed greatly by keeping the process of decrease, recovery, overshoot, and gradual return to normal only by 20 days. Histologic examinations of spleen and popliteal lymph node showed that follicles disappeared 1 to 2 days before periarteriolar lymphatic sheath or paracortex. At the peak of splenomegaly, the architectures of spleen and lymph node were replaced with the interstitial tissue composed of dense and uniform layer of lymphoid cells. With the return of spleen weight to normal range, the architecturles returned to normal. Our results clearly indicated that cyclophosphamide affected not only B cells but also T cells. These results seemed to suggest that augmentation of delayed-type hypersensitivity by cyclophosphamide may be due to the eliminateion of the suppressor T cells.

한우체세포 복제란이식우 및 정상우의 분만전후 태반특성 및 단백질분리양상

  • 본 연구실에서는 체세포복제 수정란 이식우의 분만전후 태아와 모체 및 태반과의 내분비적인 관계를 검토하기 위해 복제수정란 이식우의 분만 전후에 있어서, 혈중progesterone 과 TGF-$\beta$$_1$ 수준을 분석한 결과 정상우의 분만직전과 달리 혈중progesterone 및 TGF-$\beta$$_1$ 농도는 감소되지 않고 높은 수준을 그대로 유지되어 복제수정란이식우의 경우 정상적인 분만 signal이 나타나지 않는 것으로 나타났다. 본 연구는 체세포 복제소 수정란 이식우에 있어서 태반조직의 특성을 정상우의 동일한 시기의 태반과 비교 검토하였으며 분만시기에 있어서 태반의 역할을 검토하였다. 태반은 복제란 이식우 및 정상우 임신우를 술을 실시하여 태반을 채취하여 실험에 공시하였다. 정상우의 임신기간중 혈중 progesterone 농도는 임신초기와 중기에 다소 증가되어 유지되다가 임신말기에는 급격한 progesterone의 감소현상과 더불어 분만이 유기되는 것으로 나타났으나 복제우의 경우 분만예정일 직전까지 progesterone의 급격한 감소는 일어나지 않고 높은 농도로 유지되는 현상을 보였다. 이때 제왕절제 수술중 제대근방에 있는 태반궁부를 적출하여 중량을 측정한 결과 정상우의 궁부에 비해 유의적으로 무거웠으며, 이것은 태아의 중량과 정의 상관관계가 있음이 확인되었으며 태아 및 궁부의 중량이 무거울수록 분만 전후의 태아사망율이 높은 것으로 나타났다 이때의 궁부를 조직학적으로 검토한 결과, 복제우의 태반궁부는 정상우의 궁부에 비해 cytotrophoblast 및 syncytiotrophoblast가 적었으며 치밀하지 못한 형태를 확인할 수 있었다. 이때의 태반조직를 이용하여 세포질을 추출하여 단백질양상을 검토한 결과, 정상태반에 비해 복제태반은분자량 90kd, 65kd 및 18kd의 특이단백질이 존재하고 있음을 확인할 수 있었으며 이들 단백질을 HPLC system에 의해 분리한 결과 fraction 2와 3에서 특이한 단백질을 분리하였다. 이와 같이, 복제우의 태반에는 정상적인 임신과 분만을 비롯하여 복제송아지의 생존에 중요한 인자가 관여하고 있을 확인할 수 있었다.

Physicochemical and Sensory Characteristics of Persimmon Jelly Added with Different Levels of Daebong Persimmon Puree (대봉감 퓨레의 첨가량에 따른 감 젤리의 이화학적 및 관능적 특성)

  • Physicochemical and sensory characteristics of persimmon jelly products with different levels (3, 5, and 7% w/w) of Daebong ripened persimmon puree was investigated. The moisture content of all jelly products used in this experiments was maintained within $8{\pm}2%$ of the products. Levels of crude chemical composition (moisture, crude protein, crude lipid, and crude ashes), texture, soluble materials, vitamin C, dietary fiber and ${\beta}$-carotene were increased as amount of puree addition increased in the products. Color values, $L^*$, $a^*$, and $b^*$ of the persimmon jelly products were 33.08-42.04, 0.93-1.31 and 8.85-11.21, respectively. There was no significant difference in pH (5.51-5.61) as the levels of Daebong ripened persimmon puree increased. Sensory evaluation in terms of appearance, color, taste, flavor, chewiness, and overall acceptance resulted in the highest score in the persimmon jelly product with 5% level of Daebong ripened persimmon puree. In conclusion, level of Daebong ripened persimmon puree affected color value, texture and vitamin C, dietary fiber, and ${\beta}$-carotene content of the persimmon jelly products. In addition, optimum level of Daebong ripened persimmon puree would be 5% for manufacturing the persimmon jelly product.

Effect of Dietary Lactobacillus, Medicinal Plants Extract, Herb Extract on the Laying Performance, Cecal Microflora, Relative Organs Weight, Leucocyte Profiles in Laying Hens against Salmonella gallinarum (Salmonella gallinarum에 대한 Lactobacillus, Medicinal Plants Extract, Essential Oil 급여가 산란계의 생산성, 맹장 내 미생물 균총, 조직중량 및 백혈구 조성에 미치는 영향)

  • The study was conducted to investigate the effect of dietary supplementation of Lactobacillus, medicinal plants extract, and essential oil on laying performance, cecal microflora, relative organic weight, leucocyte profiles in laying hens against Salmonella gallinarum. A total of 100 Hy-Line Brown laying hens of 65 weeks of age were assigned into 5 dietary treatments supplemented with no antibiotic (negative control), antibiotic (positive control), Lactobacillus 0.1% (LB), medicinal plants extract 100 ppm (MPE), herb extract 100 ppm (EO). After 7 days of treatment, the 20 hens in each treatment group were divided into challenge group (n=10) and non-challenge group (n=10) for oral administration of Salmonella gallinarums, and were maintained in the same dietary treatments for two additional weeks. Laying hens were selected 20 by each treatment, splitting up into oral Salmonella group and nontreated group. The feeding trial lasted for one weeks and Salmonella gallinarum was orally administered for two weeks. Egg production, egg weight, egg mass, and feed conversion ratio were calculated and cecal microflora, relative organ, and blood were collected at the end of experiment. Compared with the control, laying hens fed Lactobacillus, medical plant extract, and essential oil were not observed to change of laying performance. Salmonella count increased significantly when Salmonella gallinarum was orally administered (P<0.05) but coliform bacteria and Lactobacillus counts were not affected. The relative organs weight was not different in hens with oral administration of Salmonella gallinarum and also in hens fed Lactobacillus, medical plant extract, and essential oil. H/L ratio was not significantly different, but total leucocyte level was somewhat increased in the treatment with oral administration of Salmonella gallinarum and more leucocyte increased in laying hens fed Lactobacillus, medical plant extract, and essential oil than control.

Quality properties of texturized vegetable protein made from defatted soybean flour with different soybean seed coat contents (대두껍질 함량에 따른 탈지대두분말 식물조직단백의 품질 특성)

  • The texturization characteristics of textured vegetable protein (TVP) were investigated based on the extent of soybean decoating during the pretreatment of defatted soybean flour used for TVP. The raw materials for TVP consisted of 50% defatted soybean flour, 30% gluten, and 20% corn starch. The weight ratios of soybean seed coat to soybean flour were 9%, 6%, 3%, and zero. Extrusion was performed using an extruder equipped with a cooling die, maintaining a barrel temperature of 190℃ and screw speed of 250 rpm, Water was injected at a rate of 9 rpm using a metering pump. Regarding the textures of the extruded TVPs produced from defatted soybean flour, an increase in the soybean seed coat content led to a decrease in the apparent fibrous structural layer and an increase in hardness. However, there were no significant changes in elasticity and cohesion. Moreover, as the soybean seed coat content increased, the pH of TVPs decreased. A higher soybean seed coat content also tended to lower the moisture content, increasing water absorption, solids elution, and turbidity. These results suggest that an increased seed coat content reduces the proportion of protein, and the fibers present in the seed coats prevent texturization.
