Saline Water Treatment by Underwater Plasma

  • 수중방전은 다양한 라디칼을 직접 물 속에서 발생시키기 때문에 수처리 공정에 다양한 응용이 가능하다. 특히, 최근에 선박평형수 등의 살균이 국제적인 이슈가 되고 있고, 2017년까지는 모든 선박에 살균을 위한 수처리 설비가 의무화된다. 본 연구에서는 염분이 있는 수체에서의 방전공정을 연구하고 이를 수처리공정에 적용할 수 있는 방법에 대해 연구하였다. 해수의 경우 전도도가 53mS로 자유로운 전하의 이동이 가능하기 때문에 일반적인 민물방전의 전원과 전극 등으로는 방전을 할 수 없다. 이에 세라믹과 금속의 이중구조로 되어 있는 모세관전극을 개발하여 전도성이 있는 수체에서의 방전을 이루었다. 전원장치로는 60 Hz, 380 V를 1차측에 인가하여 2차측에서 약 3 kV, 10 kW의 파워가 발생하는 12위상차 교류전원장치를 개발하여 사용하였다. 모세관 내부에 전압이 인가되면 전류가 발생하여 joule heating에 의하여 모세관 내부에 기포가 형성된다. 이 때, 전류의 단락이 이루어지면서 고전압쪽에 전하가 축적되며 기포내부의 E-field가 상승한다. 이후 기포 내에서 방전이 개시되며 각종 라디칼을 생성한다. 방전에 의해 생성되는 산화제로는 오존, OH라디칼, 과산화수소 등이 있으며, 해수에서는 Cl-의 결합에 의하여 Cl2 가스가 발생한다. 약 30,000 J/L의 체적에너지에 대하여 생성되는 총염소의 농도는 2.5 mg/L이다. 수중방전의 적용대상으로 선박평형수, 멤브레인과의 결합, 용존기포부상법을 선정하여 적용가능성을 연구하였다. 먼저 선박평형수 살균처리를 위해 해수의 처리유량을 20 lpm으로 유지하고 대장균, 바실러스, 조류(테트라셀미스) 등을 투입하여 전극 12개가 삽입된 12위상차 플라즈마 반응기를 통과시켰더니, 약 30,000 J/L의 체적에너지에 대하여 1일 후의 살균력이 각각 99.99, 99.99, 99.9%의 살균력을 나타내었다. 이는 국제해사기구에서 권장하는 살균수준인 99.9%를 초과하는 수치이다. 플라즈마를 이용한 해수살균공정의 안정적 운전을 위해 후단에 UF멤브레인을 추가하여 잔류생존 미생물을 제거할 수 있다. 이를 위해 플라즈마가 후단의 멤브레인 운전에 미치는 영향을 평가하였다. 카올린과 탄산칼슘을 오염원으로 각각 투입하여 멤브레인으로 처리를 하였을 때, 방전 직후 멤브레인에 걸리는 막간압력차가 약 30% 감소하였는데, 이는 막에 형성된 파울링이 방전에 의해 제거된 것으로 평가할 수 있다. 수중방전은 다양한 산화제를 생성함과 동시에 미세기포를 발생시키는데 이는 수중유기물의 부상분리에 적용될 수 있다. 방전모세관전극의 내부직경을 1 mm로 유지하고, 60 Hz, 교류전원으로 방전한 결과 평균입경 44 um의 기포를 발생시켰고, 이는 일반적으로 용존공기부상법에 사용되는 기포의 크기와 일치한다.

Water Quality of Streams in Some Agricultural Areas of Different Agricultural Practices along Nakdong River Basin (낙동강 유역 주요 농업지대 소하천 수질의 영농형태별 비교)

  • A survey on four tributary streams in agricultural areas along Nakdong River was carried out to evaluate the effect of agricultural practices on the quality of streamwater. Typicalpaddy and upland farmings were major agriculturalpractice in two survey areas. Apple orchards were located along Imgo -Cheon. Intensive farming in plastic film house was conducted along the Habin cheon. Electriclal condutivity and nutrient contents were measured. Comparing to the reference water sample collected from very upper part of Yangsang -Cheon in Moonkyong, water in the streams studied were quite polluted and such pollution could be due to the farmings conducted along the streams. Phosphorus content were higher than the minimum level for eutrophication (0.01-0.05 mg/L). Nitrogeon content were also significantly high in many sites to cause harmful effects on crops when normalfertilizer level was applicated. Among the four stream, water quality in paddy area were relatively less polluted. High nitrogen level in Imgo-Cheon and high level of EC in Habin-Cheon were problematic. As farming is the major sources of pollution in the streams studied, this are traceable to the agricultural nonpoint sources. To maintain water quality of the stream, the agricultural nonpoint source along thributary streams should be properly controlled.

Seasonal Variation of Primary Productivity and Pigment of Phytoplankton Community Structure in the Seomjin Estuary (섬진강 하구역의 계절별 일차생산력 및 식물플랑크톤 색소 조성 변화)

  • Four in situ incubation experiments were conducted in May, August and November 2009, as well as February 2010 to determine the seasonal primary productivity and the community structure of phytoplankton at the Seomjin estuary. The primary production of phytoplankton ranged from 9 to 3560 mgC $m^{-2}\;d^{-1}$. Primary productivity was the highest in the summer season (August), which was influenced by improved optical and temperature conditions of the water, as well as the supply of nutrients derived from its surrounding watershed. Particularly, the upper station (SJ-1, SJ-2) of Seom-jin estuary showed a higher productivity, as a result of inflow of input nutrients originated from the terrestrial source. The fucoxanthin, as an index pigment of diatoms showed the highest concentration (0.74~9.51 ${\mu}g\;L^{-1}$) at all stations, occupying 30~80% to total Chl a concentrations. The phytoplankton species composition determination, using a microscope showed similar results to the pigment analysis, which indicated diatom Skeletonema costatum, as the dominant species. The primary productivity in Seom-jin estuary indicates temporally and spatially large variation, according to different environmental conditions. Also, Skeletonema costatum has euryhaline features with relatively higher contribution.

The Effect of Floating Wetland on Water Quality Improvement in a Eutrophic Lake (부유습지를 이용한 부영양수계 현장 수질개선 효과)

  • At weekly intervals, we monitored continuous changes in water quality by constructed floating wetland equipped with the four different filter media (sponge, volcanic stone, activated carbon and magnesium hydroxide) in a eutrophic lake from March 2011 to May 2012. We also investigated phyto- and zooplankton communities both in the influent and the effluent water through the floating wetland. Over a 10-month time period, average turbidity (66%), suspended solids (79%) and chlorophyll-a (80%) concentrations were remarkably reduced in the effluent water compared to the influent (P<0.001). The average removal rates of $NO_2-N$ and $NH_3-N$ were 24% and 20%, respectively (P<0.05). The average removal rates of $NO_3-N$ and TN were less than 10% (P>0.05). On the other hand, the average removal rates of $PO_4-P$ and TP were more than 65% (P<0.01). Interestingly, the abundance of phytoplankton in the effluent was decreased about 2.6 times compared to that of the influent, whereas the abundance of zooplankton in the effluent was increased about 3.5 times compared to that of the influent. Overall, particulate matters (SS, Chl-a and TP) and dissolved nutrients ($NO_2-N$, $NH_3-N$ and $PO_4-P$) were particularly reduced at high rates. Therefore, application of our constructed floating wetland in a eutrophic lake improved the water quality and demonstrated a potential for algal bloom mitigation.

Evaluation of In-vitro Anticoagulation Activity of 35 Different Seaweed Extracts (35종 해조류 추출물의 in-vitro 항혈전 활성 평가)

  • Seaweeds have been recognized as a health food, having anti-obesity, anti-constipation and anticoagulation activities, and the use of seaweeds in the food, medicine, and cosmetic industries have recently significantly increased. In this study, methanol extracts were prepared from 35 different seaweeds (17 phaeophyta, 11 rhodophyta and 7 chlorophyta), and thrombin time (TT), prothrombin time (PT) and activated partial thromboplastin time (aPTT) were determined in order to develop safe and novel anticoagulation agents from natural products. In TT experiments, Ecklonia cava, Ecklonia stolonifera, Eisenia bicyclis (Kjellman) Setchell, Ishige foliacea, I. okamurai, Sargassum confusum and S. yamade showed strong thrombin inhibition activity among the 35 different seaweeds. In PT experiments, the inhibitions of prothrombin were identified in the selected seaweeds from TT experiment, with the exception of S. yamade. In aPTT experiments, the seaweeds with blood coagulation inhibition factors were E. cava, E. stolonifera, E. bicyclis (Kjellman) Setchell, I. foliacea, I. okamurai, S. confusum and Hixikia fusiforme Okamura. Further anticoagulation assay with the selected 8 seaweeds suggested that S. confusum is most effective in antithrombosis, and E. stolonifera, E. bicyclis (Kjellman) Setchell, and I. foliacea have high potential as antithrombosis agents. Based on components-activity correlation analysis, flavonoids are considered as active anticoagulation components of seaweeds These results suggest that edible seaweeds, especially S. confusum, have potential as safe and novel anticoagulants, and S. yamade and H. fusiforme Okamura could be used as a thrombin-specific and coagulation factor-specific inhibitors.

Characteristics of Acid-hydrolysis and Ethanol Fermentation of Laminaria japonica (다시마의 산 가수분해와 에탄올 발효 특성)

  • In order to study the utilization of brown seaweed Laminaria japonica as an alternative renewable feedstock for bioethanol production, the properties of acid hydrolysis and ethanol fermentation were investigated. The acid hydrolysis enhanced the final yield of fermentable sugars, which led great increase of ethanol productivity. The maximum yield of reducing sugars reached 135 mg/g-dry Laminaria japonica after 1.0N sulfuric acid-hydrolysis at $130^{\circ}C$ for 6 h. The Saccharomyces cerevisiae (ATCC 24858) could ferment $C_6$-sugars like glucose, galactose and mannose into ethanol, but not $C_5$-sugars like arabinose and xylose. Optimal fermentation time varied with sugars; 48 h for glucose, 72 h for galactose, and 96 h for mannose. Nevertheless, the ethanol yield from the hydrolysate reached 242 mg/g-dry Laminaria japonica after fermentation by the S. cerevisiae at $35^{\circ}C$ for 96 h, which corresponds to approximately 4 times more than the theoretical yield from total reducing sugars in the hydrolysates. It indicates that the non-reducing sugars or oligosaccharides dissolved in the hydrolysate played an important role in producing bioethanol. The ethanol concentration linearly increased from 2.4 to 9.2 g/L, while the ethanol yield per dry weight of biomass decreased from 242 to 185 mg/g, with increasing the ratio of biomass to acid solution from 1 to 5% (w/v). The bioethanol yield estimated was approximately 7,400~9,600 kg/ha/year, and indicated that Laminaria japonica is a promissing feedstock for bioethanol production.

Relations of Nutrient Concentrations on the Seasonality of Algal Community in the Nakdong River, Korea (낙동강 조류군집의 계절적 변화와 영양염 농도와의 관계)

  • The construction of the eight large weirs in the Nakdong River, the second largest river in Korea, caused big changes in the physical environment of the water system. Algal communities and their correlations with environmental factors, mainly nutrients were studied at five weir areas in the Nakdong River from 2010 to 2013. Water quality, hydrodynamics and algal composition were investigated. Results showed that flow velocities were reduced compared with that before weir construction near the areas where are located in the mid and upstreams of the Nakdong River. A seasonal algal community succession was observed and it was mainly correlated with temperature and phosphorus. Diatoms were dominated from winter to spring months and massive diatomic blooms of Stephanodiscus sp. occurred early in March during survey period. Cyanobacterial blooms of Microcystis sp. occurred from July to September 2013 and was preceded by the lower total phosphorus concentration of $0.05mg\;L^{-1}$. The correlations between total phosphorus concentrations and algal abundances were not significant during the survey periods. However, significant correlation with cyanobacteria was found in the period of weir construction after only at the GG survey site and blooms periods of 7 times in the survey sites, and its correlation coefficients were 0.53 (p<0.001) and 0.42 (p<0.01) respectively. When algal bloom was observed, partially low nutrient concentration was observed in the Nakdong River. In conclusion, partially low nutrient concentration which may result from algal bloom was observed, and we presume it caused the reduction of algal abundunces.

Anti-inflammatory and Anti-allergic Effects of Phlorofucofuroeckol A and Dieckol Isolated from Ecklonia cava (감태에서 분리된 플로로탄닌의 염증 및 알러지 억제 효과)

  • The anti-inflammatory effect of brown alga Ecklonia cava is well known, and several phlorotannins have also been reported. In this study, major active components for anti-allergy and anti-inflammation were identified by NMR and MS analysis, and the levels of effectiveness were compared. Six major phlorotannins-phloroglucinol, eckol, eckstolonol, triphlorethol-A, phlorofucofuroeckol A, and dieckol-were isolated from the ethyl acetate fraction of E. cava. In order to analyze the major active substances in E. cava, antioxidant, anti-inflammatory, and anti-allergic effects were evaluated for the six separate substances. Antioxidant capacities of each phlorotannin were evaluated using the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals, where phlorofucofuroeckol A and triphlorethol-A had the highest radical scavenging capacity in respective radical scavenging assays. Phlorofucofuroeckol A exhibited the highest inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells among phlorotannins tested. Dieckol inhibited the release of ${\beta}-hexosaminidase$, a marker for the release of histamine in mast cells, in a dose-dependent manner in antigen-stimulated RBL-2H3 mast cells. Additionally, no cytotoxicities were observed at 1 and $2{\mu}g/ml$ in both phlorofucofuroeckol A and dieckol. These results suggest that phlorofucofuroeckol A and dieckol may play a key role in allergic inflammatory reactions.

Conditions for Ideal Draw Solutes and Current Research Trends in the Draw Solutes for Forward Osmosis Process (정삼투 공정 적용에 적합한 유도 용질의 조건과 최근 동향)

  • Water is an essential resource for humans, but fresh water becomes scarce due to population growth and contamination of limited resources. Membrane technology has been widely used for water treatment, and forward osmosis is a process which does not need high hydraulic pressure for the operation. However, there are needs for (1) development of novel draw solutes causing low internal concentration polarization and reverse salt flux for high water flux, and (2) development of economic recovery method of the draw solutes in the diluted draw solution. Previous researches on the draw solute include $NaHCO_3$ which can be regenerated by about $60^{\circ}C$ heating, sucrose which can make potable water without additional process, and magnetic nanoparticles which can be regenerated by external magnetic field. Using the principles of forward osmosis process, sea water desalination, wastewater treatment, refinement of proteins, energy generation using pressure retarded osmosis process, preparation of diluted fertilizer, and growing algae for biofuel can be conducted. This paper summarizes characteristics of ideal draw solutes, recovery method of the draw solutes, and various application examples.

Antibacterial Activity of Sea-mustard, Laminaria japonica Extracts on the Cariogenic Bacteria, Streptococcus mutans (충치균 (Streptococcus mutans)에 대한 다시마 추출물의 항균활성)

  • In previous reports the authors have screened the inhibition effects of marine algae extracts on halitosis, and demonstrated that a brown algae, Eisenia bicyclis (' Daehwang') possess not only strong deodorant effect bug also considerable anticariogenic activities. In this study, we screened antibacterial effects of various marine algae, and measured minimum inhibitory concentration (MIC) value of them against mutans streptococci in vitro. Among the 27 species of marine algae, $80\%$ ethanol extract of dried sea-mustard, Laminaria japonica ('Dasima') showed the strongest inhibition activity against Streptococcus mutans KCTC 3300. The extracts of Ulva lactuca ('Galparae'), Codium fragile ('Cheonggak'), Ecklonia cava ('Gamtae'), E. stolonifera ('Gompi') and Undalia Pinnatifida ('Miyeok') showed slightly weaker inhibitory potency than L. japonica. Differences of MIC values in $80\%$ ethanol extract of some species of marine algae were observed depending on test bacterial species, i.e., S. mutans KCTC 3300 or S. sobrinus KCTC 3307. Eighty percent ethanol extract of dried L japonica was fractionated with diethyl ether, chloroform, ethyl acetate, n-buthanol and water successively, The ether-soluble fraction had inhibitory effect on S. mutans KCTC 3300, however the inhibitory effects were not found in the other fractions. The MIC values of $80\%$ ethanol extract and ether fraction were 180 and 105 $\mu$g/mL respectively, while no significant inhibition activity of water-soluble fraction was found even when the fraction was added up to 5,500 $\mu$g/mL.
