Background : Salmeterol, a new ${\beta}_2$-adrenergic receptor agonist, is a long-acting bronchodilator and benefits patients with asthma who have nocturnal symptoms. We wished to assess the efficacy of inhaled salmeterol ($50{\mu}g$ bid) compared to inhaled salbutamol ($200{\mu}g$ qid) for the treatment of bronchial asthma, particularly nocturnal asthma. Method : We randomly assigned 35 patients (25 female and 10 male patients, 15 to 50 years old) to one of two treatment groups : one group received $50{\mu}g$ of salmeterol twice daily and another did $200{\mu}g$ salbutamol four times per day. And this study was performed as an open-label and the 6 weeks inhalation period. Results : Analysis of symptam score ; Day and night time symptom score showed significant difference between salmeterol and salbutamol Group (p<0.05). Number of days for additional bronchodilator requirements; The number of days and puffs for additional bronchodilator were lower in the salmeterol group in either day and night time (p<0.05). Pulmonary function test ; $FEV_1$ showed significant increase in salmeterol group compared to salbutamol group after 2 and 4 weeks inhalation period. Adverse effects ; We found no evidence of tolerance to the bronchodilating effects of salmeterol, and adverse reactions to all the treatments were infrequent and mild. Conclusion : For the management of bronchial asthma, salmeterol given twice daily is superior to salbutamol given four times daily.
Background : Mucus hypersecretion in the patients with airway diseases represents poor prognosis as well as discomfort. However, there is no known therapy for its effective control. One important component of mucus is mucin, a glycosylated protein, which endows mucus with viscosity. We studied whether a proteinase has a role in mucin secretion and if so, which. Methods : (1) Inhibition of mucin secretion Group-specific proteinase inhibitors were tested to evaluate whether a proteinase belonging to a group of proteinases plays a role in mucin secretion. Phenylmethylsulfonyl fluoride(PMSF, a serine proteinase inhibitor), E-64(a cysteine proteinase inhibitor), Pepstatin(an aspartic proteinase inhibitor) and 1, 10-Phenanthroline(a metalloproteinase inhibitor) were treated into the Calu-3 cell line for 24 hours. The enzyme linked immunoabsorbant assay(ELISA) for MUC5AC was performed to evaluate the amount of mucin secretion and to compare with a control. (2) Stimulation of mucin secretion Matrix metalloproteinase-9(MMP-9), MMP-12 and TACE(TNF-alpha converting enzyme), which are known to be related with airway diseases, were used to be treated into Calu-3 for 24 hours. ELISA for MUC5AC was performed to evaluate the amount of mucin secretion and to compare with the controls. Results : (1) PMSF($10^{-4}M$), E-64($10^{-4}M$), Pepstatin($10^{-6}M$) and 1, 10-Phenanthroline($10^{-4}M$) reduced the MUC5AC secretion by $1{\pm}4.9%$(mean${\pm}$standard deviation; P=1.0 compared with the control), $-6{\pm}3.9%$(P=0.34), $-13{\pm}9.7%$(P=0.34) and $41{\pm}8.2%$(P=0.03), respectively. (2) The amounts of MUC5AC secretion stimulated by MMP-9(250ng/ml), MMP-12(100ng/ml) and TACE(200ng/ml) were $103{\pm}6%$(P=0.39), $102{\pm}8%$(P=1.0) and $107{\pm}13%$(P=0.39), respectively, compared with the controls. Conclusion : Metalloproteinase(s) is (are) suggested to play a role in mucin secretion. It appears that metalloproteinases, other than MMP-9, MMP-12 or TACE, affect the mucin secretion in this in vitro model.
Objective: This study was performed to evaluate the effect of oxytocin antagonist on the outcome of IVF/ICSI cycles in infertile patients with repeated failure of IVF/ICSI treatment. Method: Forty patients who had experienced two or more failures of IVF/ICSI treatment without low ovarian reserve, were recruited for this prospective randomized study. All patients received controlled ovarian stimulation (COS) using GnRH antagonist multidose protocol (MDP). For the intervention group, intravenous administration of atosiban (mixed vasopressin $V_{1A}$/oxytocin antagonist) started with a bolus dose 6.75 mg one hour before embryo transfer (ET) and continued at an infusion rate of 18 mg/hour. After ET, administered atosiban was reduced to 6 mg/hour and continued for 2 hours. The main efficacy endpoints were clinical pregnancy rate and implantation rate. Results: Patients' characteristics were comparable in the intervention and control groups. COS parameters and IVF results were also similar. The number of uterine contractions for 3 minutes measured just before ET was significantly lower in the intervention group than control group ($3.5{\pm}1.4$ vs $8.7{\pm}2.2$, p<0.001). While there was no statistically significant difference in the clinical pregnancy rate between control group and intervention group (20.0% and 40.0%, p=0.168), the implantation rate was significantly higher in the intervention group, with 16.9% (11/65) compared with 6.0% (4/67) in the control group (p=0.047). There were no differences in ectopic pregnancy rate and miscarriage rate between the two groups. Conclusion: This study demonstrates that administration of oxytocin antagonist during ET can improve the implantation rate probably by decreasing the frequency of uterine contractions in infertile patients undergoing IVF/ICSI treatment.
Herbal medicines are widely used to treat a variety of human diseases. However, therapeutic effect of herbal medicine on the canine facial nerve paralysis (FNP) has not been investigated. This study examined therapeutic effects of Oyaksungisan on the canine FNP. Ten dogs with the induced canine FNP were equally divided into the control and an herbal medicine-treated groups (5 dogs, Oyaksungisan-treated group), respectively. No treatment was given to the control group. In the Oyaksungisan-treated group, Oyaksungisan was administered orally, twice per day for 2 weeks (50 mg/kg) after inducing FNP. Significant differences in the clinical scores were detected between the Oyaksungisan-treated and control groups on day 14 (p < 0.05). The serum creatine kinase (CK) activities in the Oyaksungisan-treated group showed a tendency to decrease but there was no significant difference compared with those of the control group. In conclusion, Oyaksungisan was effective for the induced canine FNP.

The behavior of glycogen and histological changes of hepatic tissues in the liver of rats, aged 6 to 7 weeks, fed 18% casein diet under control, gramoxone and gramoxone + vitamin C(Vt. C) diets has been investigated in a comined histropathological and histochemical studies. Cloudy swelling and fat changes of hepatic cells were observed in the gramoxone group with the duration of feeding time. Fat changes of hepatic cells were observed more obviously than cloudy welling, especially in the hepatic cells of periportal area. The number of Kupffer's cells increased signifciantly in the gramoxone group fed for 4weeks. The cloudy swelling and fat changes decreased obviously in the gramxone + Vt. C group. Glycogen content of heaptic cells tended to increase slightly in the gramoxone group as compared with the control group . moreover, glycogen depositons were higher in the hepatic cells where fat changes were obvious. It seems to be that Vt.C alleviating effects on the gramoxone toxicity in the patterns of glycogen distribution and histological structure of heaptic tissues.

An electron microscopic study has been made of the effects of change of cell structure of Escherichia coli treated with several disinfectants. The alterations observed as follows: 1. Nucleoid, cytoplasm which contain ribosomes and cell wall appeared to be composed of a parallel triple. layered membrane can observed in control Escherichia coli. 2. The nuclear material was no longer demonstrable in its normal sites. The cytoplasm lost its granularity, became homogeneous and disruption of cell wall were observed by the treatment with 70% ethyl alcohol and 3% $H_2O_2$. 3. Aggregation of ribosomes and condensation of nuclear material were also observed by the treatment of 5% lysol solution and 1% dodecyl-diamino-ethyl-glycin-hydrochloride. 4. In autoclaved group, the each layer of cell wall was separated and destroyed in some sites where cytoplasm was extracted.

We have developed a sodium fluoride containing gelatin and methyl cellulose gel. Cariostatic abilities of those gel were investigated and compared with APF gel and fluoride varnish($Cavityshield^{TM}$). We prepared the bovine tooth samples and divided into two surface, control side and experimental side in same specimen for exclusion of difference between specimens. The experiment was consisted of 4 groups : (I) APF gel : (II) $Cavityshield^{TM}$ : (III) Gelatin F gel : (IV) Methyl cellulose F gel Decalcification were produced by placing each specimen into artificial acidic solution(pH 4.0) for 72 hours. Surface microhardness were measured and depth of demineralization lesion were measured by polarizing light microscope. The results were as follows: 1. The difference of VHN between control and experimental side is smallest in group I (p<0.05). 2. The largest difference was shown in group II (p<0.05). 3. There were no significant difference between group III and IV in microhardness test (p>0.05). 4. The difference of lesion depth is smallest in group I (p<0.05). 5. There were no significant difference between group II, III and IV in lesion depth (p>0.05). The result of the present study indicate that the fluoride containing gelatin and methyl cellulose gel is more effective than APF gel and is similar to fluoride varnish application for prevention of demineralization.

Purpose : To study whether a low protein diet increase the efficacy of antihypertensive therapy on the progression of renal failure, we conducted an experimental study using 5/6 nephrectomized rats(n=63). Methods : At 7 days after surgery, rats were randomly assigned to three groups according to receiving antihypertensive drug: no antihypertensive drug (U), enalapril (E), and nicardipine (N), respectively and fed a low protein diet (6$\%$ protein). Proteinuria, mesangial matrix expansion score and glomerular volume were assessed at 4, 12 and 16 weeks after renal ablation. Results : Group U rats on a low protein diet developed progressive hypertension ($140{\pm}8,\;162{\pm}5,\;171{\pm}5\;and\;184{\pm}11\;mmHg$ at 4, 8, 12 and 16 weeks) which were controlled by E and N. Group U rats on a low protein diet developed proteinuria ($74{\pm}15\;mg/day$ at 16 weeks) which were decreased by E ($42{\pm}12 mg/day$) or N ($48{\pm}8 mg/day$) (p<0.05). Mesangial matrix expansion score and glomerular volume were not different between groups U, E and N on a low protein diet regardless of the antihypertensive drugs administered. Conclusion : A low protein diet did not affect blood pressure. Enalapril and nicardipine-treated rats on a low protein diet did not have different mesangial matrix expansion and glomerular volumes from rats on a low protein diet at 12 weeks and 16 weeks, in spite of the better controlling of systemic hypertension and lessening of proteinuria. Thus, combined treatment with a low protein diet and antihypertensive drugs didn't appear to show any addition,11 effects to attenuate glomerular injury.
Fischer 344 rats aged six weeks were diYided into four groups and group 1, 2, and 3 of rats were given an intraperitoneal injection of diethylnitrosamine at 200 mg/kg body weight and group 4 was given saline alone. Two weeks after beginning of the experiment, group 1 and 2 of rats were begun to feed on diets containing 0.02% 2-acetylaminofluorene as a promoter for four weeks. Three weeks after beginning of the experiment, all groups were performed partial hepatectomy. During the last two weeks, group 1 and 3 of rats were received subcutaneously 3 consecutive weekly doses of iron dextran at 0.125 ml/100 g body weight. Subcutaneous injection of iron dextran resulted in hepatic siderosis in group 1 and 3 of rats. Pre neoplastic nodules were identified histopathologically by two markers, resistance to exogenous iron accumulation and glutathione S-transeferase placental form (GST-P) activity, while early carcinogen induced foci were hardly resistant to iron accumulation and though a few lesions were identified, it could hardly be distincted from normal hepatocytes of surroundings. However, GST-P positive nodules as well as foci were clearly distincted from normal hepatic cells of surroundings. In the quantitative analysis of carcinogen-induced nodules and foci, more lesions were detected by immunohistochemical method for GST-P than by prussian blue staining for resistant to iron accumulation. It is concluded that immunohistochemical marker for GST-P is more sensitive and reliable than iron-resistance marker, and that iron-resistance is not useful marker for early detection of carcinogen-induced hepatic lesions.
The effectiveness of Palmarosa and Neroli essential oil on dry skin of rat induced by kitchen detergent are investigated. The experimental groups were divided the control group (C), group treated with surfactant (A1), group treated with Palmarosa (A2), group treated with Neroli (A3), group treated with Palmarosa and Neroli (A4). The protein analysis of all experimental groups was performed with SDS polyacrylamide gel electrophoresis and observation of epidermis and the alteration of mast cell were performed with photomicroscope. According to the protein analysis, the A3 group treated with Neroli essential oil was appeared the most similar with the control group. And then the A4 group treated with Palmarosa + Neroli essential oil was appeared the most similar with the control group. According to the results of morphologic view with keratin layer, the keratin layer's breakaway resulting from Palmarosa essential oil, the keratin layer's restoration resulting from Neroli essential oil was appeared. And then the structure of the epidermal layer was preserved by hyperkeratosis reaction. In photomicrosopic obersevation of mast cell to examine the inflammatory reactions, the increase in size and number of mast cell were showed in A1 group treated with surfactant compared to the control group (C). The number of mast cells definitely decreased in groups (A3, A4) which were treated with Neroli essential oil.
