반응표면분석에 의한 쇠고기 야채 쌀죽의 이화학적 및 관능적 특성 변화

  • 현대 사회는 서구적인 식생활의 변화로 인해 조리가 간편하고 조리 시간이 짧은 즉석식품과 영양 기호식품을 동시에 충족시켜주는 음식에 대한 소비가 늘고 있는 실정이다. 또한 최근 미곡의 공급량에 비해서 소비량이 해마다 감소하여 재고미의 증가를 볼 때, 쌀의 새로운 이용방법 모색이 절실히 요망된다. 따라서 쌀의 소비촉진과 현대사회의 소비형태를 접목시켜서 쇠고기와 야채를 이용한 즉석쌀죽을 개발하고자 하였다. 쇠고기, 야채 및 쌀가루를 이용한 soup mix의 최적 배합비를 설정하기 위하여 제조조건에 따라 다르게 제조한 쇠고기 야채 쌀죽의 이화학적 및 관능적 특성에 미치는 변화를 조사하였다. 이때 야채의 배합비에 따른 이화학적 및 관능적 특성을 모니터링 하고자 반응표면분석법 (response surface methodology, RSM)을 이용하였다. 요인변수(Xn)를 쌀의 양에 대한 버섯의 비율 (X$_1$), 당근의 비율 (X$_2$), 대파의 비율 (X$_3$)로 하여 중심합성계획에 따라 17실험구로 구분하여 조리실험을 실시하였고, 반응변수(Yn)는 soup mix를 이용하여 제조한 쇠고기 야채 쌀죽의 이화학적 특성인 색도의 L*값 (Y$_1$), a*값 (Y$_2$), b*값 (Y$_3$), 점도(Y$_4$), 퍼짐성 (Y$_{5}$), 고형분 함량(Y$_{6}$), PH (Y$_{7}$)으로 하였으며 관능적 특성인 색 (Y$_{8}$), 향 (Y$_{9}$), 점성 (Y$_{10}$), 맛 (Y$_{11}$), 전체적인 기호도 (Y$_{12}$)를 종속변수로 하여 회귀분석에 이용하였다. 회귀분석에 의한 모델식의 예측에는 SAS (statistical analysis system)program을 사용하였으며, 3차원 반응표면 분석법으로 해석하였다. 야채의 배합비에 따라 제조한 쇠고기 야채 쌀죽의 물리적 특성인 색도의 L*, a*, b* 값에 대한 반응표면 회귀식의 $R^2$은 각각 0.6098(p> 0.05), 0.8803 (p <0.05), 0.6781(p> 0.05)로서 b값에 있어서 그 유의성이 5%수준에서 인정되어 b값에 미치는 영향이 크다는 것을 알 수 있었다. L*값은 63-68사이로, a*값은 0.13에서 -0.89사이를 b*값은 2-5값 사이에서 변화하여 제조한 죽의 색이 옅은 황색임을 알 수 있었다. 고형분 함량, 퍼짐성과 pH에 대한 $R^2$은 각각 0.4280, 0.5433과 0.2406임을 볼 때 버섯, 당근, 대파의 비율에 따라 제조한 쇠고기 야채 쌀죽의 고형분 함량, 퍼짐성과 pH는 설정된 범위내에서 그 유의성이 인정되지 않아 큰 영향을 미치지 않음을 알 수 있었다. 관능검사 결과, 색과 향에 대한 반응표면 회귀식의 $R^2$은 각각0.6000과 0.7825이고 P-value는 각각 0.4290과 0.0942로서 5% 수준에서 유의한 상관성이 없음을 확인할 수 있었다. 맛과 점성에 대한 $R^2$은 0.8717과 0.8068이고 P-value는 각각 0.0195 (p <0.05)와 0.0612로서 야채의 배합비에 따라 맛에 있어서 유의확률 5%수준에서 그 유의성이 인정되었으며, 전체적인 기호도에 대한 유의성은 $R^2$이 0.8463이고 P-value는 0.0344 (p <0.05)임을 볼 때, 설정된 범위내에서 야채의 배합비에 따라 제조한 쇠고기 야채 쌀죽의 맛과 기호도에 큰 영향을 미치는 것을 알 수 있었다. 그리고 최대 임계점이 버섯의 첨가량은 0.99%, 당근의 첨가량은 0.97%, 대파의 첨가량은 0.59%에서 최적 반응표면을 나타내었다. 이상의 결과로 볼 때, 야채의 배합비에 따른 맛과 전체적인 기호도에 있어서 그 유의성이 5%수준에서 모두 유의한 상관관계를 보였으며, soup mix 제조시 쌀가루 양에 대한 야채의 최적 배합비는 버섯, 당근, 대파에 있어서 각각 0.99, 0.97과 0.59%임을 알 수 있었다.

Quality Characteristics of Jocheong Containing Various Level of Letinus edodes Extracts (표고버섯 추출액을 이용한 조청의 품질특성)

  • Jocheong was prepared by adding Lentinus edodes extract to improve its quality and to give some functional properties. Contents of crude protein, ash and crude lipid were similar to those of control, while carbohydrate content was decreased. Total mineral content were $1,916.03\~2,674.24mg/kg$ that was no difference between test samples. From HPLC determination of free sugars, Jocheong was found to contain maltose as the highest sugar, followed by glucose and fructose. In amino acid analysis, seventeen amino acid were identified and quantified. Glutamic acid in Jocheong was major amino acid. The major fatty acids in Jocheong $(0\%,\;control)$ were linoleic acid, palmitic acid, oleic acid, myristic acid and caproic acid. There was no significant differences in fatty acid composition, pH and reducing sugar content among the Jocheong samples. The viscosity and solid contents tended to decrease with the addition of Lentinus edodes extract. Increasing the ratio of mushroom extract in Jocheong tended to decrease the lightness, yellowness and redness in Hunter's color value. Although sensory value decreased with increasing Lentinus edodes extracts, use of mushroom extracts (7: 3; saccharification liquids: Lentinus edodes extracts) is recommended for making Jocheong.

Phosphate Concentration Dependent Degradation of Biofilm in S. aureus Triggered by Physical Properties (인산염 농도에 따른 물성 변화로 발생하는 황색포도상구균 바이오필름 제거 현상)

  • The objective of this study was to establish technology for removing bacteria with human- and eco-friendly material. Staphylococcus aureus as an important component for balanced equilibrium among microbiomes, was cultured under various concentrations of phosphate. Experimental observation relating to physical properties was performed in an addition of phosphate buffer. Statistically minimum value of size and hardness using atomic force microscope was observed on the matured biofilm at 5 mM concentration of phosphate. As a result of absorbance for the biofilm tagged with dye, concentration of biofilm was reduced with phophate, too. To identify whether this reduction by phosphate at the 5 mM is caused by counter ion or not, sodium chloride was treated to the biofilm under the same condition. To elucidate components of the biofilm counting analysis of the biofilm using time-of-flight secondary ion mass spectrometry was employed. The secondary ions from the biofilm revealed that alteration of physical properties is consistent to the change of extracellular polymeric substrate (EPS) for the biofilm. Viscoelastic characterization of the biofilm using a controlled shear stress rheometer, where internal change of physical properties could be detected, exhibited a static viscosity and a reduction of elastic modulus at the 5 mM concentration of phosphate. Accordingly, bacteria at the 5 mM concentration of phosphate are attributed to removing the EPS through a reduction of elastic modulus for bacteria. We suggest that the reduction of concentration of biofilm induces dispersion which assists to easily spread its dormitory. In conclusion, it is elucidated that an addition of phosphate causes removal of EPS, and that causes a function of antibiotic.

Change in the Fish Fauna and Fish Community Characteristics in the Upper Reaches of the Seomgang (River), Korea (섬강 상류의 어류상 변화 및 어류군집 특성)

  • The survey conducted from 2018 to 2020 aimed to investigate the changes in fish fauna and community characteristics in the upper reaches of the Seomgang River, Korea. During the survey period, 35 sites were selected, resulting in the collection of 7,817 fish belonging to 12 families and 40 species. The dominant species was Zacco koreanus, with a relative abundance of 34.5%, followed by Z. platypus at 28.7%. Other significant species included Rhynchocypris oxycephalus (10.2%), Pungtungia herzi (5.3%), and Squalidus gracilis majimae (4.3%). Notably, four protected species - Acheilognathus signifer, Gobiobotia brevibarba, and Cottus koreanus, designated as class II endangered wildlife by the Ministry of Environment- were identified. These species predominantly inhabit the middle and lower reaches, except for Gobiobotia brevibarba, which is found in the upper reaches. Nineteen species, accounting for a 47.5% endemism rate, were endemic to Korea. The study also noted the presence of one climate-sensitive species, Cottus koreanus, and two exotic species, Carassius cuvieri and Micropterus salmoides. Community analysis indicated a trend of decreasing dominance and increasing diversity and richness from upstream to downstream, with a distinct division into uppermost reaches, upper reaches, middle and lower reaches, and lakes. The construction of the Hwaseong Dam has had a significant direct and indirect impact on the fish community. The habitat and abundance of endangered species such as R. pseudosericeus, A. signifer, and G. brevibarba decreased dramatically immediately after the dam's construction, transforming the submerged area from lotic to lentic environments. Approximately 20 years later, the habitats have stabilized, leading to an increase in the fish population and a recovery of the previously diminished endangered species. The river health (FAI) was also evaluated, with 27 sites rated as very good (A), seven as good (B), and one as fair (C). However, endangered species such as A. signifer continue to face threats from dam and river construction, while C. Koreanus has experienced a severe population decline due to river works. Additionally, the presence of the ecosystem-disrupting species M. salmoides in Hwaseong Lake raises concerns. To ensure a stable habitat for fish in the upper reaches of the Seomgang River, it is crucial to avoid indiscriminate river construction, urgently implement restoration policies for endangered species such as A. signifer, and develop management strategies to control the spread of invasive species such as bass.

A Dynamic Simulation on the Squeezing Flow of ER Fluids (전기유변 유체의 압착유동에 대한 동적 수치모사)

  • Electrorheological(ER) fluid is a material that shows the dramatic change of rheological properties under an electric field and responds reversibly in a few milliseconds. ER fluid's response to an electric field along with its fast switching capability allows ER devices to be precisely controlled. The real application with ER fluid, however, has many limitations to be overcome; temperature fluctuation, moisture, dust, aggregation, precipitation, and low yield stress, for example. The magnitude and the characteristics of yield stress of ER fluid plays an important role in practical applications. In this research, a dynamic simulation on the squeezing flow of the ER fluid was carried out. Numerical simulation on isolated chains was performed to find out the effect of hydrodynamic and electrostatic force depending on the chain location, the squeezing rate, and the chain structure. Suspension model that is composed of a large number of particles was also investigated. The increase of normal stresses as well as the existence of a yield stress at an earlier stage could be observed, and the effective control of the normal stresses could be achieved at an optimal condition of the hydrodynamic force and the electrostatic force.

Influences of Nutrient Deficiency on the Phytoplankton Community in Pal'tang Reservoir (몇가지 영양염 결핍이 팔당댐의 식물플랑크톤군집에 미치는 영향)

  • To understand the effect of nutrient deficiency on the plankton community in three stations with different water qualities in Pal'tang Reservoir, Korea, phytoplanktons(> 10 ${\mu}m$) were cultured in nutrient enrichment Allen's media (AM) and nutrient-deficient Allen's media. A distinct shift in the species composition and biomass of phytoplankton (as chlorophyll- a) showed in all treatments. In particular, it was very interesting that the new development of cyanobacterium Microcystis aeruginosa occurred by the Fe-deficient AM. Except for Si, a community growth (as chlorophyll- a) was inhibited in all nutrient deficient treatments. Species diversity after nutrient deficiency was changed to below 2.0; slightly increased in N and P-deficiency, while decreased in Si and Fe, respectively. As suggested, dominance was entirely opposite to diversity. Therefore, the nutrient deficient effectively induced the succession of species and biomass, phytoplankton community, suggesting a possibility as a reliable tool to control the algal bloom in eutrophic lakes and reservoirs.

Drug Interactions between Cardiovascular Agents and Psychotropic Drugs (심혈관질환약물과 향정신성약물의 약물상호작용)

  • There are numerous drug interactions related to many psychotropic and cardiovascular medications. Firstly, the principles in predicting drug interactions are discussed. Cytochrome P (CYP) 450 plays a significant role in the metabolism of these drugs that are substrates, inhibitors, or inducers of CYP450 enzymes. The two most significant enzymes are CYP2D6 and CYP3A4. The ability of psychotropic drugs to act as inhibitors for the enzymes may lead to altered efficacy or toxicity of co-administered cardiovascular agents as a substrate for the enzymes. The following is also a review of the known interactions between many commonly prescribed cardiovascular agents and psychotropic drugs. Most beta blockers are metabolized by CYP2D6, which may lead to drug toxicity when they use in combination with potent CYP2D6 inhibitors including bupropion, chlorpromazine, haloperidol, selective serotonin reuptake inhibitors, and quinidine. Concomitant administration of lithium with angiotensin converting enzyme inhibitors, angiotensin receptor blockers, and diuretics may increase serum lithium concentrations and toxicity. Calcium channel blockers and cholesterol lowering agents are subject to interactions with potent inhibitors of CYP3A4, such as amiodarone, diltiazem, fluvoxamine, nefazodone, and verapamil. Prescribing antiarrhythmic drugs in conjunction with medications are known to prolong QT interval and/or inhibitors on a relevant CYP450 enzyme is generally not recommended, or needs watchful monitoring. Digoxin and warfarin also have warrant careful monitoring if co-administered with psychotropic drugs.

Microbiological, Physicochemical, and Antioxidant Properties of Plain Yogurt and Soy Yogurt (플레인 요구르트와 소이 요구르트의 미생물학적, 물리화학적 및 항산화 활성)

  • This study evaluated the physicochemical and microbiological characteristics and antioxidant properties of yogurt samples fermented with lactic acid bacteria (LAB) obtained from pickled cabbage. API 50 CHL systems and 16S rRNA nucleotide sequence analyses revealed that the isolates were Lactobacillus casei PC05 and L. acidophilus PC16. Cell counts, titratable acidity, and viscosity of the yogurt samples fermented with L. acidophilus PC16 were significantly higher than those of the samples fermented with L. casei PC05 (P<0.05). The detected cell counts and physicochemical properties were significantly lower in plain yogurt than in soy yogurt (P<0.05). Yogurt samples fermented with L. acidophilus PC16 exhibited higher antioxidant activity, measured as ability to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals and chelate ferrous ions, than those fermented with L. casei PC05. However, the ability to scavenge superoxide anions and superoxide dismutase (SOD) activity were significantly (P<0.05) higher in yogurt samples fermented with L. casei PC05 compared to those in samples fermented with L. acidophilus PC16. The antioxidant activity of soy yogurt was significantly (P<0.05) higher than that of plain yogurt. The antioxidant activity of the tested strains resulted in lipid peroxidation inhibition (in vitro), which may be related to the elimination of free radicals, chelating ability, and reducing power. There were no significant differences in the physicochemical properties and antioxidant activities of the yogurt samples during cold storage.

Characterization of interfacial chemistry on the coal bottom ash (저회의 계면 화학적 특성 규명)

  • Landfill is the main treatment method for bottom-ash because it has not only an irregular particle size and ingredients but also not proper recycling treatment. The aim of this study is to raise recycling rate of bottom-ash(nonplasticity pulverulent) and for the purpose of alternatives of clay to investigate the properties of Bottom-ash (B/A)-Hard Clay (H/C) bodies with controlled interfacial chemistry properties. After investigating the sedimentation height of suspensions with controlled pH, it was discovered that there was no hetero-polar aggregation for mixed slips because hard clay and bottom-ash had similar interfacial chemistry properties. Also, bulk density, water absorption, and microstructure properties of each pellet was observed that made by silp casting method and manufactured at $50^{\circ}C$ intervals between $1000{\sim}1250^{\circ}C$. As a result, dispersed slip of clay and bottom ash are possible for slip casting and plastic forming process because they exhibit Bingham plastic behavior. Products that made by slip with dispersed clay and bottom ash are not only suitable for KS L 4201 and KS L 1001 at $1250^{\circ}C$ but it is also possible to apply for ceramic and sanitary ware because specific gravity was about 15 % lighter than general ceramic materials.

  • Grading of the urushi lacquer is quite difficult because of large variations depending on origin, growing conditions, time acquisition, storage conditions. It is urgently required to establish a grading system of urushi lacquer by combining traditional method with scientific methods. Traditional grading of urushi lacquer was done by visual inspection of 10 experts who are working on urushi lacquer industry. Common aspects of the experts were color, odor, viscosity, spread properties on Hanji, color and drying properties on glass based the traditional grading system. In addition, rubbing on Hannj and heating with alcohol lamp was also used to grade urushi lacquer. The grading results of 10 experts showed that chinese urushi lacquer (E) of 7.03, japanese urushi lacquer (C) of 6.84, domestic urushi lacquer of (A) of 6.41 and another chinese urushi lacquer (D) of 5.27, and domestic urushi lacquer (B) of 2.50 in descending order. The degree of spread on Hanji was not consistent among 10 experts. These results indicated that the traditional grading system was pretty much personal and required of developing more objective method based on scientific background.
