Morphological Characteristics of Bemisia tabaci(Gennadius) (Homoptera: Aleyrodidae) and Discrimination of Their Biotypes in Korea by DNA Makers (담배가루이 Bemisia tabaci(Gennadius)(Homoptera: Aleyrodidae)의 형태적 특징과 DNA 표식자에 의한 biotype 판별)

  • The sweetpotato whiteflies, Bemisia tabaci(Gennadius), were found recently in Korea on Glycine max, Euphorbia pulcherrima, and Rosa hybrida. The biotype identity of Bemisia tabaci in Korea was determined by several DNA markers including the random amplified polymorphic DNAs, and restriction fragments length polymorphism of mitochondrial 12S and 16S rRNA genes. The electromorph profiles of DNA fragments from the rose(Jincheon) and poinsettia(Seoul) populations in Korea are both identical to those of B biotypes distributed in Australia, Israel, and Japan. The populations of B. tabaci collected on Glycine max, Ipomea batatas, and Perilla frutescens in different localities retained the same DNA markes with the population from Lonicera japonica and shikoku of Japan. These populations are non-B biotype and considered as an indigenous type in the Far Eastern Asia Region including Korea and Japan, Morphological Characteristics of B. Tabaci were also observed by the scanning electron microscope and described with the comparison to the other important whitefly pest, Trialeurodes vaporariorum (Westwood).

Factors Associated with Smoking Differences of Korean Adolescents with Smoking Experience : 11th(2015) Korean Youth Risk Behavior Web-based Survey (우리나라 흡연경험이 있는 청소년의 흡연유형 관련요인 : 제11차 청소년건강행태온라인조사)

  • This study was conducted to investigate the factors related to smoking and smoking differences among adolescents who have experienced smoking using data from the 11th (2015) Korean Youth Risk Behavior Web-based Survey. The data used in this study were from 5,347 middle and high school students who had cigarette smoking and electronic cigarette smoking experience among 68,043 respondents to the 11th (2015) Korean Youth Risk Behavior Web-based Survey. The data were evaluated by frequency analysis, the ${\chi}^2$ test and logistic regression analysis using weighted composite sample analysis. Overall 20% of the total smoking adolescents were dual users and 85.6% of the users of electronic cigarettes used nicotine. The results from multinomial regressions show that: male students (OR 1.63; 95% CI 1.15-2.31), living in the metropolitan area (OR 1.76; 95% CI 1.32-2.33), an average consumption of fast food of once or twice a day (OR 2.48; 95% CI 1.33-4.64), first started smoking during high school (OR 0.76; 95% CI 0.58-0.98), having experience with passive smoking (OR 1.24; 95% CI 1.07-1.44), experience drinking alcohol (OR 2.03; 95% CI 1.65-2.48) and those with higher weekly allowances were more likely to be at risk than dual users. This study provides basic data regarding the dual use of electronic cigarettes and cigarettes by adolescents. The results presented herein will facilitate development of a smoking cessation program and policy.

Electronic cigarettes recognition and influence factors of electronic cigarettes of among smoking university (흡연 대학생의 전자담배에 대한 인식과 전자담배 사용 영향 요인)

  • Objectives: This study is purpose to recognition the of smoking behavior and the electronic cigarette of university students have a current smoking. Methods: The subjects were university living in Busan, the survey was conducted from March 23 to April 12, 2015, 314 except for 24 copies of non-response and error response among a total of 340 questionnaires were analyzed. Statistical analysis methods used in this study are $x^2$-test, Linear Regression Analysis and other basic statistics such frequency, percentage using SPSS version 22.0. Results: Electronic cigarette has been analyzed to be recognized non smoking, smoking reduces, good health than conventional cigarettes, convenient to use, there is no smell, can smoke in any place and as safe. Gender, grade, non smoking experience, non smoking and accept factors were analyzed to influence the use of electronic cigarettes. Conclusions: We suggest an established institutional arrangements and regulations, take advantage of various health programs development, and ongoing health education.

Comparison of the Factors for Attempts to Quit Smoking by Adolescent using Tobacco Cigarettes only and those Adolescents using Tobacco Cigarettes Together with Electronic Cigarettes (궐련 흡연 청소년과 전자담배를 중복 사용하는 흡연 청소년의 금연시도 영향요인 비교)

  • Purpose: This study was conducted to compare and analyze the smoking-related social-ecological factors affecting attempts to quit smoking by adolescents using tobacco cigarettes only and also those adolescents using tobacco cigarettes along with electronic cigarettes. Methods: This study, as secondary analysis research, used the raw data from the 14th Korea youth risk behavior survey 2018. The data was analyzed by frequency analysis, the Rao-scott χ2-test and logistic regression analysis when considering the complex sample's analysis. Results: On logistic regression analysis, during their first smoking period, intense physical activities and having friends who smoked were associated with significantly more attempts to quit smoking by cigarettes smokers, and their first smoking period and experiences of undergoing smoking cessation education were associated with significantly more attempts to quit smoking for dual smokers of both tobacco cigarettes and electronic ones. Conclusion: As a result of this study, we identified the need to differentiate different types of smoking cessation counseling and education according to the types of smoking in adolescents.

The Relationship between Attitude and Coping Skills toward Secondhand E-cigarette Smoking among Nonsmoking College Students (비흡연 대학생의 전자담배 간접흡연에 대한 흡연태도와 흡연대처행동의 관계)

  • Purpose: The purpose of the study was to examine the relationship between smoking attitude and coping skills toward secondhand e-cigarette smoking among nonsmoking college students. Methods: This study was conducted with 148 students from a University in Seoul, Korea. Participants completed questionnaires and the data were analyzed using a T-test, ANOVA, and Pearson's correlation coefficients. Results: The mean score of smoking attitude is $51.35{\pm}4.73$. The mean score of coping skills is $13.89{\pm}2.53$ in verbal skills, $10.16{\pm}2.37$ in behavioral skills, and $8.32{\pm}2.30$ in assertive skills. Coping skills toward secondhand smoking are noted as being different by current cohabitation smoker. A significant relationship between smoking attitude and behavioral coping skills(r = .23, p= .004) was found, which means that a better smoking attitude was correlated with more active behavioral coping skills. Conclusion: The results showed that nonsmoking college students with better smoking attitude react to the E-cigarette smoking with more active behaviors. Therefore, interventions are needed to be developed to encourage better smoking attitude and coping skills toward secondhand E-cigarette smoking.

  • Spinaches showing dark green mosaic symptoms were used for identification of broad bean wilt virus. In host reaction test, that virus caused local lesions on the inoculated leaves and mosaic symptoms on upper leaves of Chenopodium amaranticolor, Chenopodium quinoa and Vicia faba, and developed mosaic symptoms on Physalis floridana, Spinacia oleracea, Nicotiana tabacum, (White burley, Bright yellow) Nicotiana glutinusa. In agar gel-diffusion test, the virus showed positive reaction with broad bean wilt virus antiserum. Spherical virus particles with size of 25nm in diameter were observed in electron microscope.

Characterization of New Bacillus thuringiensis Isolated with Bioactivities to Tobacco Cutworm, Spodoptera litura (Lepidoptera: Noctuidae) (담배거세미나방에 살충효과를 나타내는 새로운 Bacillus thuringiensis 균주의 특성)

  • Bacillus thuringiensis with selected high toxicities against tobacco cutworm, Spodoptera litura were isolated from domestic soils. When being observed under a phase-contrast microscope, the insecticidal crystal proteins were showed a bipyramidal crystal types. New CAB 109 isolate was identified to B. thuringiensis subsp. aizawai in the H serotype. As a results of insecticidal activities between CAB 109 isolate and 3 existing ready-made products against 3rd larva of S. litura, CAB 109 isolate showed 100% mortality with spore concentration $(1.3{\times}10^7cfu/ml)$. It was a very high insecticidal activity compared with a existing ready-made B. t. products. $LD_{50}$ values of CAB 109 isolate was $9.78{\times}10^5,\;6.87{\times}10^6\;and\;1.83{\times}10^7cfu/ml$ spore concentration against 2nd, 3rd and 4th larva of S. litura, respectively. Unlike Plutella xylostella, S. litura was slowly died after application up to 7 days. The weight of S. litura larva applied with CAB 109 isolate were 6-7 times less than controlled group. Even though it didn't die, it did not grow into next larva. The result observed with scanning electron microscope was that CAB 109 isolate of B. t. aizawai formed a typical bipyramidal crystal protein type. Otherwise, when CAB 109 isolate was examined with SDS-PAGE and with trypsin, there was no difference between CAB 109 strain and ready-made products of B. thuringiensis.

Factors Associated with Smoking Behaviors according to the Types of Cigarettes, Conventional and Electronic, among Adolescents (청소년의 일반담배 및 전자담배 사용 유형에 따른 흡연행위 관련 요인)

  • Purpose: The purpose of the study was to verify the degree of smoking behaviors including conventional cigarettes, electronic cigarettes, and dual use of conventional and electronic cigarettes among adolescents, and to explore associated intrapersonal and interpersonal factors of the three smoking behavior groups. Methods: The secondary data analysis was performed using the 17th 2021 Korea Youth Risk Behavior Survey. The types of smoking behaviors were grouped into conventional cigarettes, electronic cigarettes, and dual use for data analysis. Descriptive statistics and a χ2 test were performed. Multinomial logistic regression analysis was conducted to identify factors associated with of the three smoking behavior groups. Results: Among the 54,848 adolescents, 3.6%, 1.2% and 6.2% were involved in conventional cigarettes, electronic cigarettes, and the dual use, respectively. The factors associated with the three types of smoking behaviors were depression, suicidal ideation, having a smoking mother, having smoking brothers or sisters, and having smoking friends. Recovery from fatigue by sleep was associated with the dual use of conventional and electronic cigarettes. Anxiety was associated with conventional cigarettes. Having a smoking father was associated with conventional cigarettes and dual use. Conclusion: A significant number of adolescents were involved in not only the use of conventional or electronic cigarettes, but also dual use of them. Adolescents who had depression, suicidal ideation, having a smoking friend, and having a smoking family member were more likely to be involved in the three types of smoking behaviors. Customized interventions to decrease smoking should be developed considering intrapersonal factors as well as interpersonal factors.

Expression and Inheritance Patterns of Gus Gene Driven by an Endosperm-Specific Promoter in Transgenic Tobacco (배유 특이 프로모터에 의해 유도된 GUS 유전자의 형질전환 담배 내에서의 발현 및 유전 양상)

  • This study was carried out to investigate the tissue-specific expression of ${\beta}$-glucuronidase (gus) gene driven by endosperm-specific promoter (Z4 promoter) in the transgenic tobacco and to find out inheritance pattern of transgene to the next generation. Tobacco (Nicotiana tabaccum cv. Havana SR1) was transformed with Agrobacterium tumerfaciens LBA4404 harboring BV3 construct containing gus gene driven by Z4 promoter and a kanamycin resistant gene. Seven hundred bp PCR products, indicating the presence of npt II gene, were found in the all eight transformants by PCR analysis using nptII primers. To study the expression pattern of the two different kind of promoters, leaf disks of the Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. As a result, leaf disks of Z4pro-gus-transformed plants showed very weak and partial positive gus activity. In contrast, leaf disks of 35Spro-gus-transformed plants showed relatively strong positive gus activity. To investigate the expressed position of Z4 promoter, seeds from Z4pro-gus-transformed plants and 35Spro-gus-transformed plants were analyzed histochemically for gus activity. Z4pro-gus-transformed seeds showed positive gus activity restricted to the endosperm. However, the blue-colored product in 35Spro-gus-transformed seeds was observed in all the area including endosperm. Kanamycin resistance assay showed that transgenes were stably inherited to next generation in all lines.
