Identification of a Newly Isolated Protease-producing Bacterium, Bacillus subtilis FBL-1, from Soil (토양으로부터 새로이 분리된 단백질 분해효소 생산 미생물 Bacillus subtilis FBL-1의 동정)

  • A novel proteolytic bacterium was isolated from soil at Yeungnam University, South Korea. The strain, named FBL-1, was rod-shaped with a smooth surface. Biolog and API 50CHB test results revealed that strain FBL-1 was a Bacillus species. Based on 16S rDNA sequencing and chemotaxonomic characterization, the strain was identified as Bacillus subtilis because it had the highest homology with Bacillus subtilis subsp. subtilis NCIB 3610 (99.5%). In liquid culture at 37℃ with shaking at 200 rpm, fructose and yeast extract were found to be the best carbon and nitrogen sources, respectively, for cell growth and protease production. The highest protease activity (451.640 U/ml) was obtained when the strain was cultured in medium containing 20 g/l of fructose and 5 g/l of yeast extract. Although further studies are needed to characterize the protease and enhance its activity, the newly isolated protein-degrading B. subtilis FBL-1 can be applicable for the production of peptides and for the degradation of proteins in various industries.

  • This study discussed the meaning of a drone, and especially drone journalism and legal and ethical issues around that, at an introductory dimension, which is used in various social bases, but is still just an academic discussion at the beginning stage. As a methodology, content analysis was used. It seems that drone journalism has high diffusibility as a technology with high 'relative advantage', 'compatibility', 'trial ability' and 'observability' and low 'complexity' in terms of the diffusion of innovation theory. However, it will be very likely that controversies will be raised, such as safety issue due to collision and crash, a dispute over violation of privacy that may seriously infringe privacy like individual portrait rights and a controversy about the accuracy and source of information as drone filming low price and ease of use. Suggest solutions to legal and ethical issues based on existing research. Technical stability is required. Also, it is necessary to change the awareness of journalists about the drones coverage and to educate ethics, and it is necessary to establish social public opinion on issues such as privacy violation and establish system and legal measures through it. Future research is expected to carry out empirical research including journalists and public awareness surveys.

Effect of LED Light Strength for Enhancing Rutin Content in Tatary Buckwheat Sprouts and Antioxidant Activity (타타리메밀싹의 루틴 함량 향상을 위한 LED 광량 효과와 항산화 활성)

  • This study aimed to enhance rutin contents by controlling germination condition for manufacturing buckwheat sprouts. Two kinds of buckwheat, a common buckwheat (Fagopyrum esculentum Moench) and a tartary buckwheat (Fagopyrum tataricum Gaertner) were used. By comparing the rutin content of two buckwheats, tartary buckwheat was 487 ppm, about 36 times higher than common buckwheat. Both common buckwheat and tartary buckwheat which germinated and grew under the light had higher rutin content relatively. In case of tartary buckwheat, rutin content of over 10 cm sprout was 4,579 ppm (without the light), and 5,160 ppm (with the light). Furthermore, tartary buckwheat was germinated and grew under different light strengths from 2,000 to 22,000 Lux. The rutin contents of tartary buckwheat sprout that was grown under the 22,000 Lux light was the highest. The rutin content was increased dramatically at 14,000 Lux of light. From 14,000 to 22,000 Lux, there was a little change on rutin content. Therefore, the condition of 14,000 Lux light was determined optimal for manufacturing tartary buckwheat sprouts. Also, rutin contents of extracts treated with 60, 70, 80 and $90^{\circ}C$ during different time had no significant difference. Therefore, rutin of tartary buckwheat sprout extract had thermostability up to $90^{\circ}C$.

  • 연구목적: 생산자와 소비자의 물리적 사회적 거리가 가까워지고, 1일 유통체계 준수에 의한 높은 신선도 유지, 먹거리에 대한 생산자와 생산정보 공유에 의한 농산물 안전성 향상 등 여러 장점 때문에 늘어나고 있는 로컬 푸드의 지속발전을 모색하고자 로컬 푸드 참여농가의 농작업 어려움 조사 및 기계화 방안 제시 조사방법: 전북 완주군 소재 용진농협에서 운영 중인 로컬 푸드 직매장에 농산물을 공급하는 농업인 21인을 대상으로 2016년 11월에 로컬 푸드의 어려운 점, 주요 생산품목, 가장 힘든 농작업, 기계화가 시급한 농작업, 농기계 도입에 어려운 점, 의견 및 건의사항을 설문 조사 분석하였음 결과 및 고찰: 로컬 푸드 참여 농가들은 4~5개 품목을 소량 생산하고, 농가에서 수확 소포장 라벨 부착 후 매장에 직접 진열하고 있었음. 농협은 정선 선별기, 소형 도정기, 포장기, 바코드 기계 등을 설치하여 농가 공동이용, 안전성 신선도 관리 등 교육, 스마트폰 앱을 매장 재고 현황을 농업인에게 실시간으로 제공 등 지원하고 있음. 조사에 응한 농업인 21명 중 60대(13명) > 50대(5명) > 70대(3명)로 모두 50대 이상이었음. 어려운 점에 대한 물음에 대해서는 '판매가 어렵다'(45%) > '인력이 부족하다'(40%) > '생산비가 많이 든다'(8.5%) > 기타(6.5%) 순이었음. '인력부족'은 파종과 수확 시기에 노동력이 집중적으로 필요한 것에 기인하고, 경지 규모가 다소 큰 농가에서는 농사일을 전문으로 하는 외국인 노동자를 고용함으로 노동력 부족 문제를 해소하지만, 소규모 농가는 품삯을 주고 고용할 경우 오히려 적자이기 때문에 고되더라도 자체 노동력으로 해결하고 있는 것으로 조사되었음. 가장 힘든 작업은 수확(81%) > 파종 정식(19%)으로 수확이 절대적으로 힘든 작업이라고 응답하였고, 기계화가 시급한 작업은 수확(71%) > 파종 정식(29%)으로 응답하였음. 힘든 이유로는 적기에 수확을 끝내야 상품성을 인정받을 수 있기 때문에 단기간에 많은 노동력이 요구된다고 응답하였음. 농기계 도입에 어려운 원인으로는, '적합한 수확기계가 없다'(48%)와 '재배면적이 적어 필요성을 못 느낀다'(29%)가 대부분이었고, 이밖에 '가격이 비싸다'(10%), '기계 정확도가 떨어진다'(10%), '기계 조작이 어렵다'(5%)로 나타났음. 결론: 로컬 푸드 참여 농가를 대상으로 농작업 기계화 현황에 대하여 조사하였음. 로컬 푸드의 지속 발전을 위한 방안을 제시하면; 첫째, 중 소농을 위한 수확기 개발이 가장 시급한 것으로 조사되었고, 둘째, 농기계가 아니더라도 우선 힘든 일을 해소하면서 편하게 자세를 유지할 수 있는 작업 보조기구 또는 편이장비의 개발 보급이 필요함. 셋째, 농가의 영세성과 지역의 특성을 고려하여 기술센터 농기계임대사업이나 시군 보조사업을 통해서 소형 농기계, 농기구, 보조 및 편이장비 보급이 확대되어야 함.

  • 생력화를 위한 구기자의 수확 기계화는 열악한 수확작업환경을 쾌적한 작업환경으로 개선하고 노동력 감소, 생산비 절감을 할 수 있다. 관행 손 수확과 진동 고리형 수확기 방법보다 높은 작업 능률 향상으로 영농규모의 확대 촉진 및 안정적인 영농 구조를 구축하여 재배농가의 생산비를 절감하여 경쟁력을 높일 수 있으며, 기존 인력에 의존하였던 수확작업을 기계화함으로서 전업농 및 대단위 경작이 가능하게 함으로서 국내에서 생산한 양질의 구기자를 국민에게 안정적으로 제공할 수 있다. 따라서, 본 연구는 구기자 수확작업의 생력화를 위하여 개발 보급된 수목형의 재배법 특성을 분석하고 이를 토대로 타격장치를 적용한 보행형 구기자 수확기를 개발하는데 목적이 있다. 수목형 구기자나무의 분지에 착과되어 있는 숙과를 주행하면서 탈과 할 수 있는 탈과 장치를 제작하기 위하여 타격형 탈과 장치를 3D 모델링 작업(Inventor V.11, Autodesk, USA) 후 시작기를 제작, 구기자 수확 시작기는 주행부, 타격장치, 집과부, 분지유인부로 구성하였다. 구기자 수확 시작기의 최대 높이는 형태학적 특성을 토대로 타격봉의 높이를 900 mm 이하로 제한하였으며, 조향장치의 높이는 800 mm로 하였다. 주행부는 구기자 재식 조사결과를 이용하여 고랑 폭 1,500 mm 이하에서 자유롭게 전 후진 이동이 가능하고 경사로 등을 주행 시에도 안전성을 높이기 위해 자동브레이크 기능이 있으며 타격장치의 타격 봉은 알루미늄 재질로 지름 100 mm, 길이 400 mm로 설계 제작하였으며, 구기자 분지 타격 시 분지와 타격 봉이 수직 상태로 타격이 가능하도록 제작, 집과장치는 포장의 두둑, 고랑은 일괄 표준화가 되어 있지않아 청양구기자시험장에서 측정한 재배법을 바탕으로 설계된 수집부 프레임의 적용범위는 폭 450 mm, 길이 720 mm, 높이 1,500 mm를 집과 범위로 하여 설계 제작하였다. 타격 방식을 적용한 구기자 수확기 성능평가 결과 조숙기에 30초 이상의 탈과 시 87.5 % 이상 탈과는 어려울 것으로 판단되었으며, 성숙기에는 타격시간에 관계없이 92 %의 매우 우수한 탈과율이 나타났다. 성숙기의 주행속도 48 m/h 일 때 탈과율과 집과율은 89 %, 92 %로 나타났다.단위작업시간당 최대 수확 능력은 관행작업 2.9 kg/hr, 진동고리형 수확기 5.2 kg/hr, 타격방식을 적용한 구기자 수확기는 최소 7.6 kg/hr, 최대 24.1 kg/hr로 관행작업과 비교하여 주행속도와 시기별 최소 2.6배, 최대 8.3배의 작업 성능 차이가 나타났다. 재배양식에서는 기계화 수목형이 적합한 것으로 나타났고, 타격방식을 적용한 보행형 구기자 수확기를 이용하여 수형별 시간대별 수확성능을 시험한 결과 우수한 결과가 나타났다. 이에 따라 구기자 재배 농가에 기계화수목형 재배법을 보급하고 타격방식을 적용한 구기자 수확기를 이용하면 작업환경 개선과 노동력, 인건비 절감을 통한 영농규모의 확대 촉진 및 안정적인 영농 구조로 구기자 경쟁력 제고를 할 수 있을 것으로 판단되어진다.

Behavior Case Study of Temporary Structures during Underground Extension Work by Field Measurement (현장계측을 통한 지하증축공사 중 가설구조물의 거동 사례연구)

  • During the construction of underground space expansion of old facilities, it is necessary to secure temporary residence space for existing residents as well as noise and vibration issues during construction, and in the case of commercial, industrial, and social use, damage is expected from suspension of the use of facilities. There is a need for a technology that minimizes noise and vibration during underground expansion, enabling the use of existing facilities even during construction. In this study, a practical underground extension model is proposed by analyzing the behavior of the temporary structure and the surrounding ground as a result of measurement at each construction stage for a actual construction site. In order to solve the problems that occurred during construction, the basement slabs were placed in advance after the initial excavation. The measurement results (building inclinometer, crack measurement system, structure inclinometer and surface settlement meter) at the site were reviewed to analyze the behavior of the temporary structure and surrounding ground. As a result, it was confirmed that the inclinometer of the building and the structural inclinometer showed a tendency that the displacement after the slab line was placed was reduced or converged. The placement of basement slabs during underground extension not only relived the noise and vibration problems during construction, but also secured the stability of structures.

Evaluation of Optimal Time Between Overhaul Period of the First Driving Devices for High-Speed Railway Vehicle (고속철도차량 1차 구동장치에 대한 완전분해정비의 최적 주기 평가)

  • The first driving device of the power bogies for the Korean high-speed railway vehicle consists of the traction motor (TM) and the motor reduction gears unit (MRU). Although TM and MRU are the mechanically integrated structures, their time between overhauls (TBO) have two separate intervals due to different technical requirements(i.e. TBO of MRU: $1.8{\times}10^6km$, TBO of TM: $2.5{\times}10^6km$). Therefore, to reduce the unnecessary number of preventive maintenances, it is important to evaluate the optimal TBO with a viewpoint of reliability-center maintenance towards cost-effective solution. In this study, derived from the field data in maintenance, fault tree analysis and failure rate of the subsystem considering criticality of the components are evaluated respectively. To minimize the conventional total maintenance cost, the same optimal TBO of the components is derived from genetic algorithm considering target reliability and improvement factor. In this algorithm, a chromosome which comprised of each individual is the minimum preventive maintenance interval. The fitness function of the individual in generation is acquired through the formulation using an inverse number of the total maintenance cost. Whereas the lowest common multiple method produces only a four percent reduction compared to what the existing method did, the optimal TBO of them using genetic algorithm is $2.25{\times}10^6$km, which is reduced to about 14% comparing the conventional method.

Study on a Propulsion Control of the Roller Coasters Train based on Air Cored Linear Synchronous Motor (공심형 선형동기전동기 기반의 궤도열차 추진제어에 관한 연구)

  • To accelerate a heavy roller coaster train with over 1G force, a lot of thrust is required and linear synchronous motor(LSM) as propulsion method is suitable for this kind of system. To increase the propulsion efficiency of LSM, precise and real-time position information of vehicle is required for accurate phase control. However, the discontinuous position information with relatively long time interval is usually transmitted from the hall-sensors on the track every magnet length. In this paper, the basic motor model based on traditional dq-axis equations is described and the motor dynamic model is produced by considering the cogging force and friction loss. To improve the position accuracy, the position estimator is also proposed for LSM control system. Simulations were performed to check the characteristics of the torque control system which includes the position estimator based on the motor model. Simulation results based on the linearized model show that this control system has an enough bandwidth and phase margin and the executed algorithm achieves an ideal effect to follow the real-time position signal. Therefore, the feasibility of position estimator is also confirmed.

Hardware-Based High Performance XML Parsing Technique Using an FPGA (FPGA를 이용한 하드웨어 기반 고성능 XML 파싱 기법)

  • A structured XML has been widely used to present services on various Web-services. The XML is also used for digital documents and digital signatures and for the representation of multimedia files in email systems. The XML document should be firstly parsed to access elements in the XML. The parsing is the most compute-instensive task in the use of XML documents. Most of the previous work has focused on hardware based XML parsers in order to improve parsing performance, while a little work has studied parsing techniques. We present the high performance parsing technique which can be used all of XML parsers and design hardware based XML parser using an FPGA. The proposed parsing technique uses element analyzers instead of the state machine and performs multibyte-based element matching. As a result, our parsing technique can reduce the number of clock cycles per byte(CPB) and does not need to require any preprocessing, such as loading XML data into memory. Compared to other parsers, our parser acheives 1.33~1.82 times improvement in the system performance. Therefore, the proposed parsing technique can process XML documents in real time and is suitable for applying to all of XML parsers.

Feasibility Study of a Series Hybrid-Electric Propulsion System for a Fixed Wing VTOL Unmanned Aerial Vehicle (고정익 수직이착륙 무인항공기를 위한 하이브리드-전기 추진시스템의 타당성 연구)

  • General VTOL aircraft uses gas turbine engine which has high power to weight ratio. However, in the VTOL UAV in small sector, the gas turbine as a prime mover is not adequate because of the limitation of the high fuel consumption ratio of the gas turbine. In this research, The Series Hybrid-Electric Propulsion System(SHEPS) has been proposed and technology survey & comparison analysis has conducted to constitute propulsion system for engine, electric motor and battery. To achieve this object a 65kg-class P-UAV from "Company I" was used. And to estimate the validity of power control algorithm and developed power management control, Matlab/simulink$^{(R)}$ has been used for the simulation. As a result, the developed algorithm worked comparatively well and the research has predicted that SHEPS was satisfied enough for 7 hour of endurance for mission profile.
