Clinical Significance of Maspin Expression in Osteosarcoma (골육종에서 Maspin 발현의 임상적 의의)

  • Purpose: This study was performed to investigate the maspin gene expression from osteosarcoma and to determine whether its expression correlates with clinical course of the cancer. Materials and Methods: Between 2001 and 2006, 39 patients who were diagnosed and treated surgically for osteosarcoma were included in the present study. We estimated the maspin gene expression from osteosarcoma tissue samples using RT-PCR. And we examined the correlations between the maspin expression and clinical data (post-chemotherapeutic response, local relapse or metastases). Results: Maspin was over expressed in 21 cases of 39 osteosarcoma tissues. There were significant correlations between maspin expression and the response to neoadjuvant chemotherapy, distant metastases & metastasis-free survival. In multivariate analysis, maspin low-expression was significant risk factor for distant metastases. Also, there was significant difference in metastasis-free survivals between maspin hi- expression group ($69.0{\pm}10.5%$) and low-expression group ($25.4{\pm}13.0%$). Conclusion: The degree of maspin expression in osteosarcoma was significant risk factor for distant metastases and predictive factor for metastasis-free of overall survivals. Maspin may be a useful biologic marker in evaluating the prognosis in patients with osteosarcoma and could be used as a therapeutic target clinically.

$SO_2$가 흰쥐 기관의 Glycoconjugates에 미치는 영향에 관한 연구

  • $SO_2$ 폭로군의 기관 상피층의 섬모에서 DBA 및 SBA는 50ppm 이하 $SO_2$ 폭로군에서는 대조군에 비해 증가하였으며 sWGA 및 UEA-1은 대부분의 $SO_2$ 폭로군에서 대조군에 비해 증가하였다. 섬모세포에서 LCA 및 Con A는 100ppm 및 200ppm $SO_2$ 폭로군에서 대조군에 비해 약간 증가하였다. 배상세포에서는 대부분의 렉틴의 반응성이 50ppm 이하의 $SO_2$ 폭로군에서는 증가하였다가 100ppm 및 200 ppm $SO_2$ 폭로군에서는 현저히 감소하였다. 장액선포에서는 LCA 및 Con A는 모든 $SO_2$ 폭로군에서 대조군보다 증가하는 경향을 보였으며 점액선포에서는 대조군에서 관찰되지 않았던 RCA-1은 50ppm 및 100ppm $SO_2$ 폭로군의 일부 선포에서 증가하였다. 위의 결과로 보아 $SO_2$는 기관에 심대한 병리조직학적 변화뿐만 아니라 glycoconjugates대사에 심대한 영향을 미쳐 심한 병변을 야기시킴을 알 수 있었다. 전반적으로 $SO_2$의 호흡기 영향은 고농도로 갈수록 심한 영향을 나타내었다. 저농도에서도 폭로 시간이 길어짐에 따라 나타나는 $SO_2$의 영향이 고농도와 유사한 경향을 나타내며 또한 모든 $SO_2$의 농도를 비교해보면 폭로 시간이 길어질수록 그 영향이 더 심하게 나타났다.

울진 2호기 5주기 저출력 노물리 시험에서의 감마 Background 의 영향 분석

  • 울진 2호기 5주기 노물리 시험 결과 기준 제어군의 제어능과 기준 제어군 삽입 전후의 붕소 농도의 차이의 비로 정의되는 평균 붕소능의 측정 결과가 제한치를 위배하였다. 이것은 노물리 시험시 사용하는 노외 계측기가 비보상형 전리함이기 때문에 중성자속 준위가 낮을 때 감마 background 에 의한 영향이 커지며, 이런 경우에 발생될 수 있는 현상으로 판명되었다. 따라서 본 연구에서는 이러한 감마 background로 인한 반응도 증가 현상을 분석하기 위해 점 동특성 방정식을 수치 해석적으로 풀고 이로부터 감마 background가 없을 때의 기준 제어군 제어능 측정 시험을 모사 계산하였다. 이러한 방법으로 계산한 결과, 기준 제어군의 예상 측정제어능이 실제 측정치보다 약 6%증가하였으며 이 값을 사용하여 평균 붕소능을 계산한 바에 의하면 그 제한치를 만족하는 것으로 평가되었다.

Relationship Between Mitochondrial DNA Copy Number, Metabolic Abnormalities and Hepatic Steatosis (지방간 및 대사 인자들과 말초혈액 백혈구의 사립체 DNA copy 수와의 연관성)

  • Insulin resistance plays a central role in fatty liver, a part of the metabolic syndrome. This study examined the relationship between fatty liver, metabolic abnormalities and mitochondrial DNA [mtDNA] copy number in peripheral blood that is correlated with diabetes or metabolic markers. Fatty liver was assessed by questionnaire on alcohol consumption and abdominal ultrasonography. MtDNA copy number in peripheral leukocytes was measured by a real-time quantitative polymerase chain reaction [PCR]. Among 445 subjects, 148 subjects had hepatic steatosis and 297 were controls. mtDNA copy number was significantly lower in fatty liver group in comparison with that of normal finding group. This result is similar in both groups, alcoholic or non-alcoholic fatty liver group. MtDNA copy number was inversely correlated with alanine aminotransferase [ALT], aspartate aminotransferase [AST], gamma-glutamyltransferase [$\gamma$-GTP], body mass index [BMI], waist circumference, diastolic blood pressure, and free fatty acid. MtDNA copy number in peripheral leukocytes was associated with fatty liver and insulin resistance related factors.

  • 육군은 국민들이 육군의 정책을 흥미 있으면서도 쉽게 이해할 수 있도록 유머 기법을 적용한 사진영상 홍보콘텐츠를 온라인 홍보에 적극 활용하고 있다. 본 연구에서는 유머가 육군에 관한 메시지 전달 기법으로 유용한지를 정교화 가능성 모델을 바탕으로 고 저유머 홍보 동영상을 고 저관여 집단에게 보여준 후 인지 정서적 요소(주의-기분-신뢰)와 태도에 어떻게 영향을 미치는지 살펴보았다. 그 결과 유머는 육군 홍보에 긍정적 요소로 작용하며, 주의와 기분 요소를 증가시키는데 기여하고, 이러한 주의와 기분 요소는 태도 형성에 상관관계가 있고 영향력이 높은 것으로 나타났다.

The Clinical Significance of Serum Vascular Endothelial Growth Factor Levels Measured at Ovulation Triggering Day In Intrauterine Insemination Cycles (자궁강 내 인공수정을 위한 과배란유도 시 hCG 투여 일에 측정한 혈중 Vascular Endothelial Growth Factor의 임상적 의의)

  • Objective: The objective of this study was to investigate whether serum levels of vascular endothelial growth factor (VEGF) measured at ovulation triggering day reflect ovarian response in intrauterine insemination (IUI) cycles. Methods: Forty-nine fertile women who undergoing superovulation and IUI were included. Superovulation was performed using clomiphene citrate (100 mg/d on day 3$\sim$7) in combination with human menopausal gonadotropin (150 IU every other day starting on day 5). Serum samples were obtained on the day of hCG administration and the levels of VEGF-A and estradiol were measured. The numbers of mature follicle $\geq$17 mm in diameter were also counted. Results: Serum VEGF-A levels did not correlate with the numbers of mature follicle count nor serum estradiol levels. Serum estradiol level was positively associated with mature follicle count. Serum VEGF-A levels tended to be lower in women with mature follicle count less than three or women with more than five. Conclusion: Our results indicate that serum VEGF-A levels do not have an association with superovulation outcome in IUI cycles. However, a tendency of lower VEGF-A level in poor and high responder suggests that those with extreme response to superovulation may be related with abnormal angiogenesis. Further studies should be warranted in larger populations.

Neuroprotective effects of resveratrol via anti-apoptosis on hypoxic-ischemic brain injury in neonatal rats (신생 백서의 저 산소 허혈 뇌손상에서 항세포사멸사를 통한 resveratrol의 신경보호 효과)

  • Purpose : Resveratrol, extracted from red wine and grapes, has an anti-cancer effect, an antiinflammatory effect, and an antioxidative effect mainly in heart disease and also has neuroprotective effects in the adult animal model. No studies for neuroprotective effects during the neonatal periods have been reported. Therefore, we studied the neuroprotective effect of resveratrol on hypoxic-ischemic brain damage in neonatal rats via anti-apoptosis. Methods : Embryonic cortical neuronal cell culture of rat brain was performed using pregnant Sprague-Dawley (SD) rats at 18 days of gestation (E18) for the in vitro approach. We injured the cells with hypoxia and administered resveratrol (1, 10, and $30{\mu}g/mL$) to the cells at 30 minutes before hypoxic insults. In addition, unilateral carotid artery ligation with hypoxia was induced in 7-day-old neonatal rats for the in vivo approach. We injected resveratrol (30 mg/kg) intraperitoneally into animal models. Real-time PCR and Western blotting were performed to identify the neuroprotective effects of resveratrol through anti-apoptosis. Results : In the in vitro approach of hypoxia, the expression of Bax, caspase-3, and the ratio of Bax/Bcl-2, indicators of the level of apoptosis, were significantly increased in the hypoxia group compared to the normoxia group. In the case of the resveratrol-treated group, expression was significantly decreased compared to the hypoxia group. And the results in the in vivo approach were the same as in the in vitro approach. Conclusion : The present study demonstrates that resveratrol plays neuroprotective role in hypoxic-ischemic brain damage during neonatal periods through the mechanism of anti-apoptosis.

Dose Response Relationship in Local Radiotherapy for Hepatocellular Carcinoma (원발성 간암의 국소 방사선치료 시 선량반응 관계)

  • Purpose : In this study, it was investigated whether dose response relation existed or not in local radiotherapy for primary hepatocellular carcinoma. Materials and Methods : From January 1992 to March 2000, 158 patients were included in present study. Exclusion criteria included the presence of extrahepatic metastasis, liver cirrhosis of Child's class C, tumors occupying more than two thirds of the entire liver, and performance status on the ECOG scale of more than 3. Radiotherapy was given to the field including tumor with generous margin using 6, 10-MV X-ray. Mean tumor dose was $48.2{\pm}7.9\;Gy$ in daily 1.8 Gy fractions. Tumor response was based on diagnostic radiologic examinations such as CT scan, MR imaging, hepatic artery angiography at $4\~8$ weeks following completion of treatment. Statistical analysis was done to investigate the existence of dose response relationship of local radiotherapy when it was applied to the treatment of primary hepatocellular carcinoma. Results : An objective response was observed in 106 of 158 patients, giving a response rate of $67.1\%$. Statistical analysis revealed that total dose was the most significant factor in relation to tumor response when local radiotherapy was applied to the treatment of primary hepatocellular carcinoma. Only $29.2\%$ showed objective response in patients treated with dose less than 40 Gy, while $68.6\%\;and\;77.1\%$ showed major response in patients with $40\~50\;Gy$ and more than 50 Gy, respectively. Child-Pugh classification was significant factor in the development of ascites, overt radiation induced liver disease and gastroenteritis. Radiation dose was an important factor for development of radiation induced gastroduodenal ulcer. Conclusion : Present study showed the existence of dose response relationship in local radiotherapy for primary hepatocellular carcinoma. Only radiotherapy dose was a significant factor to predict the objective response. Further study is required to predict the maximal tolerance dose in consideration of liver function and non-irradiated liver volume.

  • Nonspecific suppressions of the immune responses have been reported to occur in the cancer-bearing patients. The mechanisms responsible for these immunosuppressions remain unclear but some investigators suggested that the immunosuppressions may result from immunosuppressive factors exudated from tumor masses. This study was undertaken to evaluate the in vivo effects of ascitic and pleural fluids from patients with cancers metastatic to peritoneum or pleura on the immune responses of mice. Non-cancerous ascitic and pleural effusions were used as controls. The ascitic fluids from hepatomap atients and pleural fluids from pulmonary carcinoma patients decreased both the delayed-type hypersensitivity and the antibody formation only when these were injected before antigenic stimulation. Control effusions exerted no effect on the immune responses to mice. These results suggested that cancerous fluids suppressed the immune responses by affecting the afferent arc of the immune system.

염분변화에 따른 황복의 산소소비와 암모니아 질소배설

  • 황복(Takifugus obscurus)은 주로 황해에 분포하며 봄철에 서해안으로 흐르는 금강, 한강 및 임진강으로 소상하여 강의 중상류에서 산란하는 소하성어류이다. 특히 황복은 바다와 하천을 오고 가는 관계로 해수뿐만 아니라 민물에서도 양식이 가능한 어종으로 다른 소하성어류와 마찬가지로 염분변화에 따른 삼투조절능력이 매우 우수할 것으로 생각된다. 본 연구는 황복이 급격히 염분을 달리하였을 때 나타나는 생리적 반응을 산소소비량과 암모니아 질소배설량의 변화로 염분변화에 따른 생리적 적응력을 살펴보고자 하였다. 본 연구에 사용한 황복은 전장 10cm(체중 15~25g) 전후의 치어로서 염분 32, 22, 12, 및 2 ppt로 조절되어 있는 FRP수조(500 L)에서 2개월간 사육 후, 각기 염분을 달리한(32, 22, 12, 2 ppt) 호흡측정 장치에 황복을 수용하여 산소소비량과 암모니아 질소배설량을 측정하였다. 한편, 사육 시스템내에서의 산소소비와 암모니아 질소의 배설경향을 보기 위하여 염분 16ppt에서 산소소비량과 암모니아 질소배설량을 25시간 관찰하였다. 산소소비 경향은 대체로 염분이 낮을수록 높은 산소소비 경향을 보였는데, 실험염분 32ppt에서는 2~32ppt 사육군 모두 다른 실험염분(22, 12, 2 ppt) 사육군보다 안정되고 낮은 산소소비량을 나타내었다. 그리고 실험염분 22, 12 및 2 ppt에서는 각기 실험염분과 동일한 염분 사육군에서 가장 높은 산소소비량을 나타낸 반면 32 ppt 사육군은 모든 실험염분에서 가장 낮은 산소소비량을 나타내었다. 암모니아 질소의 배설경향은 실험염분 2 ppt를 제외하고는 2 ppt 사육군에서 가장 높은 질소 배설율을 보였으나, 실험염분 2 ppt에서는 오히려 현저히 낮은 암모니아 질소 배설율을 나타내었다. 저 염분(2, 12 ppt)에서 사육한 황복은 22 및 32 ppt에서 사육된 황복에 비해 저염분에 노출될 경우 삼투조절을 위해 암모니아 배설에 상당한 차이를 보였다. 사육 시스템내에서 조사된 산소소비와 암모니아 질소 배설경향은 섭이와 관련하여 일간리듬을 보였는데, 산소소비는 섭이 3시간 후에 최고를 보였으며 암모니아 질소 배설은 섭이 4시간 후에 각각 최고를 나타내었다.
