Development of an Efficient Bioassay Method to Evaluate Resistance of Chili Pepper Cultivars to Ralstonia solanacearum (고추 풋마름병에 대한 효율적인 저항성 검정법 확립)

  • Bacterial wilt caused by Ralstonia solanacearum is an important disease in cultivation of chili pepper, causing plant death and significant yield losses. Cultivation of disease-resistant varieties is the most suitable measure to control bacterial wilt of chili pepper. To establish an efficient screening method for resistant chili pepper to R. solanacearum, six resistant or susceptible cultivars to the R. solanacearum were selected and the development of bacterial wilt on the cultivars according to several conditions was investigated. Drenching bacterial suspension into the cut roots using a scalpel was more simple and effective to distinguish resistant and susceptible cultivars than inoculation methods of root-dipping or soil-drenching without wounding. A resistant pepper, 'MC4' to R. solanacearum showed high resistance under the developed conditions which were 21- to 28-day-old pepper inoculated with $1{\times}10^8cfu/ml$ of bacterial suspension. On the other hands, the susceptible cultivars represented high disease severity under the conditions. These results indicated that we developed an efficient method to evaluate resistance of chili pepper cultivars against bacterial wilt. In addition, we successfully evaluated resistance degree of 140 commercial chili pepper cultivars to R. solanacearum using the developed method.

Efficient Screening Method for Resistance of Cucumber Cultivars to Fusarium oxysporum f. sp. cucumerinum (오이 덩굴쪼김병에 대한 효율적인 저항성 검정 방법)

  • The study was performed to establish an efficient screening method for resistant cucumber to Fusarium oxysporum f. sp. cucumerinum. The isolate KR5 was identified as F. oxysporum f. sp. cucumerinum based on molecular analyses of ITS and TEF genes and host-specificity test on cucurbits including melon, oriental melon, cucumber, and watermelon. Then four cucumber and two rootstock cultivars showing different resistance degrees to the Fusarium wilt pathogen KR5 were selected. And development of Fusarium wilt of the six cultivars according to several conditions, including incubation temperature after inoculation, inoculum concentration, root wounding, and growth stages of seedlings, was investigated. Disease severity of Fusarium wilt on the resistant cultivars was changed with incubation temperatures after inoculation. The resistant cultivars showed the higher resistance when inoculated plants were kept at 25 or $30^{\circ}C$ than at $20^{\circ}C$. Among four different growth stages of the seedlings, seven-day-old seedling represented the most difference of resistance and susceptibility to Fusarium wilt. From above results, we suggest that an efficient screening method for resistant cucumber to F. oxysporum f. sp. cucumerinum is to dip the non-cut roots of seven-day-old seedlings in spore suspension of $1.0{\times}10^6-1.0{\times}10^7$ conidia/ml and to transplant the seedling into a non-infected soil, and then to incubate the inoculated plants in a growth room at $25^{\circ}C$ for 3 weeks to develop Fusarium wilt.

Determination of paraquat-resistant biotype on Conyza canadensis and the resistant mechanism (Paraquat 저항성 생태형 망초의 선발과 저항성 기작)

  • Paraquat-resistant biotype of Conyza canadensis (L.) Cronq. was determined by chlorophyll loss and random amplified polymorphic DNA (RAPD) analysis and the resistant mechanism was investigated with respect to absorption, translocation, and binding constant. RAPD analysis for paraquat resistant (R) and susceptible (S) biotypes found in a pear orchard revealed that the biotypes possessed remote genetic relationship. Chlorophyll loss, as an indication of paraquat toxicity, of S biotype was 7.8-fold greater than that of R biotype. There were no differences in contents of epicuticular wax and cuticle and amounts of [14C]paraquat penetrating the cuticle between the two biotypes. Little translocation of the herbicide out of the treated leaf was observed in either biotype. Binding constants of paraquat to the cell wall and thylakoid membrane were 7.4-fold and 16.9-fold, respectively, higher in R biotype than in S biotype. The results suggest that the resistance mechanism of C. canadensis biotype is due partly to high binding affinity of paraquat to the cell wall and thylakoid membrane.

Development of Effective Screening Method for Resistance of oyster mushroom to Trichoderma disease in vitro (느타리버섯 푸른곰팡이병 저항성 실내검정 방법의 개발)

  • Trichoderma disease of oyster mushroom has not been effectively detected in the field for testing its resistance against the disease with its varieties. In this study, we investigated the methods to detect its resistance in the laboratory by using media, which enables us to understand the relevant characteristics (e.g., lysis, toxin enzyme, mycelial growth rate). In coculturing with strains of Trichoderma and oyster mushroom, it is possible to observe the difference in the resistance of oyster mushroom against Trichoderma with the phenomena of barrage reaction, overgrowth and lysis. We also observed the inhibition of mycelial growth of oyster mushroom using the dilution method with 48-well plate, but could not observed the inhibition of mycelial growth using the filter paper method of cultural supernatant. In simultaneously culturing both Trichoderma and oyster mushroom, it was possible to detect the inhibition of the mycelial growth of oyster mushroom, but Trichoderma mycelium did not overgrow against oyster mushroom. We found that the pathogenicity was efficient in using solid medium with the phenomena of overgrowth and lysis by inoculating Trichoderma on top of mycelia of oyster mushroom. In conclusion, the methods (e.g., coculture method, dilution method with 48-well plate, post-inoculation method) are recommended to detect the resistance of oyster mushroom against Trichoderma disease.

  • This study was conducted to establish the efficient screening method for resistant cabbage to Fusarium wilt caused by Fusarium oxysporum f. sp. conglutinans. The resistance degrees of nine commercial cabbage cultivars to the disease were evaluated. Among them, five cultivars (YR-honam, Ogane, Greenhot, Redmat, and Ccoccoma) showing different resistance to the fungus were selected. Then development of Fusarium wilt of the cultivars according to several conditions including root wounding, dipping period of roots in spore suspension, inoculum concentration, and incubation temperature to develop the disease was investigated. Highly resistant cultivars such as 'YR-honam' and 'Ogane' hardly showed change of resistance to the disease by root wounding, dipping period, and inoculum concentration, while disease severity of Fusarium wilt on the cultivars was changed with incubation temperatures ($20^{\circ}C$, $25^{\circ}C$ and $30^{\circ}C$). When the cabbage cultivars were incubated at $25^{\circ}C$, they represented the most difference of resistance and susceptibility to Fusarium wilt. From above results, we suggest that an efficient screening method for resistant cabbage to F. oxysporum f. sp. conglutinans is to dip the non-cut roots of 14-day-old seedlings in spore suspension of $1{\times}10^7$ conidia/ml for 0.5 hr and to transplant the seedlings to plastic pots with a fertilized soil, and then to cultivate the plants in a growth chamber at $25^{\circ}C$ for 3 weeks to develop Fusarium wilt.

Mapping of RFLP Markers Linked to Bacterial Blight Resistant Genes (Xa-1, Xa-3) in Rice (벼 흰잎마름병 저항성 유전자(Xa-1, Xa-3)연관 RFLP 마커 탐색)

  • Bacterial blight caused by Xantomonas oryzae pv. oryzae is one of the most serious diseases of rice especially in southern area of Korea. Three races, $\textrm{K}_1$, $\textrm{K}_2$ and $\textrm{K}_3$, are the most dominant species. lo improve rice breeding efficiency using marker assisted selection, some RFLP markers were surveyed for polymorphism between resistant and susceptible to $\textrm{K}_1$ and $\textrm{K}_3$. And, 127 doubled-haploid (DH) lines derived from Milyang121/HRl1650-1-4-2 and 131 DH lines derived from Milyang123/HR10624-AC5 were evaluated to bacterial blight ($\textrm{K}_1$ and $\textrm{K}_3$). Milyang121 and HR10624-AC5 have Xa-1, resistant to $\textrm{K}_1$ race, and Milyang123 has Xa-3, resistant to $\textrm{K}_1$ and $\textrm{K}_3$ race. Three markers, RZ590, RZ536 and RG303, showing polymorphism between parents and resistance gene, Xa-1 and Xa-3, were analysed in the two combinations of DH lines. The segregation pattern of resistant DH population of Milyang123/HR10624-AC5 to susceptible showed 3:1 and 1:1 in $\textrm{K}_1$ and $\textrm{K}_3$ race. In three RFLP markers, RZ590 was linked to Xa-1 on chromosome 4, and RZ536 and RG303 were linked to Xa-3 on chromosome 11. The map distance between Xa-1 and RZ590 was 3.1cM on chromosome 4, and Xa-3 and RZ536/RG303 were 7.6/16.0cM on chromosome 11, respectively. The results of RFLP mapping will be useful for the selection and pyramiding of bacterial blight resistant genes.

Effect of Rice Downy Mildew (Sclerophthora macrospora) on Rice Growth and Screening of Disease Resistance of Cultivars (벼 누른오갈병(Sclerophthora macrospora) 발생이 벼 생육에 미치는 영향 및 병 저항성 품종 검정)

  • It was conducted to investigate the effect of rice downy mildew (RDM) infection to plant growth and yield components in water seeding stage, and to screen of varietal resistance to downy mildew. Being infected by rice downy mildew, chlorotic spot appeared in the leaf and leaf length was shortened. As the infected rice was growing, internode was not elongated properly and was deformed, and then panicle was not arised or mal-formed. Plant height of infected rice was shortened at all growth stage, and while the number of tillers of infected rice was more decreased than that of healthy plant before maximum tillering stage, and that of infected rice was more increased after heading stage. While the number of internode of infected tiller was much increased than that of healthy tiller internode length of infected tiller was shorter. As the rice infected by RDM severely, the number of panicles per square meter and ripening of rice was more decreased and yield of rice was extremely much decreased. As result of the varietal resistance screening with rice seedling, Geyh-wabyeo and Donjinbyeo were resistant varieties to downy mildew.

Breeding of Resistant Cabbage 'CT-171' to Fusarium Wilt (시들음병 저항성 양배추 품종 'CT-171' 육성)

  • A new resistant cabbage variety 'CT-171' to Fusarium wilt was bred by crossing $A337MSBC_5$ with resistance to the disease and 397-$G_6$ with good density and color. 'CT-171' was selected after combining ability, seed gathering and regional adaptability test in 2008. For breeding of resistant varieties, we investigated the development of Fusarium wilt on cabbage seedlings inoculated with Fusariumoxysporum f. sp. conglutinans by root dipping inoculation method. As a result, 'CT-171' showed higher resistance to Fusarium wilt than 'Asiaball' used as control. The maturity of new variety was 58 days and was faster than control and well suited for autumn cultivation because of cold resistance. The anthocyanin pigment of plant was not revealed. The weight, height and width of head were 1.5 kg, 14 cm and 15 cm, respectively and the core size was 5.7 cm and stable in various cultivation environments. 'CT-171' which showed good agricultural character and resistance to Fusarium wilt filed for variety protect right in Korea Seed & Variety Service on February 2013. The new variety will be appropriate for export and domestic consumption.

CMV-P1 Resistance Evaluation Using Enzyme-linked Immunosorbent Assay of Pepper Genetic Sources (Capsicum spp.) (Enzyme-linked Immunosorbent Assay를 이용한 CMV-P1 저항성 고추 유전자원 평가)

  • Recently, Cucumber mosaic virus (CMV)-P1 infection in pepper cultivation is very serious problem, which causes low marketability and yield. In this research, 56 domestic pepper cultivars including 20 PR (Phytophthora resistance) cultivars, 31 foreign pepper lines collected from USA and 112 genetic pepper resources form RDA Genebank were used for CMV-P1 resistance evaluation. Resistance evaluation was performed at 24 days and 51 days after artificial inoculation of peppers by enzyme-linked immunosorbent assay (ELISA). Among the 36 domestic cultivars 'Muhanjilju' were resistant to CMV-P1. All 20 PR cultivars and 29 foreign peppers except 'NuMex Twilight' and 'Chainese Giant' were susceptible. Among 112 pepper lines from RDA Genebank, nine pepper lines were resistant, and 17 pepper lines were moderately resistant, and 86 pepper lines were susceptible. Almost all domestic peppers on the market were highly susceptible to CMV-P1, whereas 17.2% of foreign pepper cultivars and genetic pepper lines from RDA Genebank were resistant or moderately resistant. Resistant pepper lines selected in this study can be used as genetic sources for breeding CMV-P1 resistant pepper.
