The concentration of polyamine and the relationship between the concentration of PA and cold resistance were analysed during the cold stress in the cotyledons and hypocotyls of young spring radishes. The concentration of PUT was increased during the cold stress in the cotyledons and the concentration of PUT and SPD was changed significantly all the stress time. From the results, we suggested that PUT and SPD were synthesized against cold stress or separated from the bound PA during the cold stress. But, the concentration of SPM was not significantly changed during the cold stress. Only SPD existed in the control hypocotyls. The concentration of SPD was increased during the cold stress, but was decreased dramatically during continusly cold stress. The reactivity against stress of hypocotyls was generally more sensitive during the cold treatment than that of the cotyledons because of the lower concentration of PA in the hypocotyls. PUT and SPD were responsible for the resistance of cold stress in the cotyledons and PA did not play an important role on the resistance of cold stress in the hypocotyls. In this study we suggest that PUT and SPD were responsible for the increase of the resistance against the cold stress.
Plants can respond and adapt to cold stress through regulation of gene expression in various biochemical and physiological processes. Cold stress triggers decreased rates of metabolism, modification of cell walls, and loss of membrane function. Hence, this study was conducted to construct coexpression networks for time-based expression pattern analysis of genes related to cold stress in Chinese cabbage (Brassica rapa ssp. pekinensis). B. rapa cold stress networks were constructed with 2,030 nodes, 20,235 edges, and 34 connected components. The analysis suggests that similar genes responding to cold stress may also regulate development of Chinese cabbage. Using this network model, it is surmised that cold tolerance is strongly related to activation of chitinase antifreeze proteins by WRKY transcription factors and salicylic acid signaling, and to regulation of stomatal movement and starch metabolic processes for systemic acquired resistance in Chinese cabbage. Moreover, within 48 h, cold stress triggered transition from vegetative to reproductive phase and meristematic phase transition. In this study, we demonstrated that this network model could be used to precisely predict the functions of cold resistance genes in Chinese cabbage.
An accumulation levels of osmolytes in chilling-tolerant and chilling-sensitive cultivar of Cucurbits against chilling stress were determined during chilling stress. Total soluble sugar contents in tolerant cultivar did not changes fur 10 days after chilling stress, but then slightly increased 20 days after chilling stress. In sensitive cultivar, it was increased rapidly in the beginning of chilling stress, and increased 3.4 times as much 20 days after chilling stress as compared with unstressed plants. Proline contents in tolerant cultivar was rapidly increased by the beginning of chilling stress, and then increased 26.6 times 20 days after chilling stress as compared with unstressed plants. In sensitive cultivar, it was increased 22.0 times 20 days after chilling stress as compared with unstressed plants. A levels of glycine betaine (GB) in tolerant cultivar increased 1.9 times as much during the 20 days of chilling stress. However, concentration of GB in sensitive cultivar did not change during the chilling stress. When plants were treated exogenous GB as a foliar spray, chilling tolerance was significantly enhanced in both cultivars. The foliar application of exogenous GB was induced chilling tolerance by accumulation of GB in the plant organs. However, it does not accumulate endogenous proline.
Several sensors have been developed for soil and plants to assess plant stress due to climate change. Therefore, the objective of the study is to nondestructively evaluate temperature stress on plant by monitoring climatic and soil conditions and plant responses using various sensors. Plant responses were monitored by electrical conductivity in plant stem and sap flow rate. Electrical conductivity in plant stem reflects the physiological activity of plants including water and ion transport. Fully grown Brassica oleracea var. italica was exposed to 20/15 ℃ (day/night) with 16 h photoperiods as a control, low temperature 15/10 ℃, and high temperature 35/30 ℃ while climatic, soil, and plant conditions were monitored. Electrical conductivity in plant stem and sap flow rate increased during the day and decreased at night. Under low temperature stress, electrical conductivity in plant stem of Brassica oleracea var. italica was lower than control while under high temperature stress, it was higher than control indicating that water and ion transport was affected. However, chlorophyll a and b increased in leaves subjected to low temperature stress and there was no significant difference between high temperature stressed leaves and control. Free proline contents in the leaves did not increase under low temperature stress, but increased under high temperature stress. Proline synthesis in plant is a defense mechanism under environmental stress. Therefore, Brassica oleracea var. Italica appears to be more susceptible to high temperature stress than low temperature.
Is obtain cell lines showing high level of rice cold tolerance, direct in vitro selection through cold stress on primary pollen callus derived from anther culture was carried out Genotypic difference in callus formation and plant regeneration was recognized Rates of albino was increased along the duration of cold stress. Reciprocal effects were not noticed in anther culturability There was no variants related to rice leaf discoloration in pollen derived lines from parental varieties, regardless of days of cold stress. The regeneration and recombination of rice leaf discoloration in 146 pollen-derived lines, 70 pollen-derived lines from cold stress at $0^{\circ}C$ for 10 days, and 830 F$_2$ plants presented normal distribution curves with skewness in tolerance and no significant difference among 3 populations. Direct in vitro selection for rice cold tolerance through cold stress on primary pollen callus derived from anther culture, therefore, was revealed ineffective as a in vitro technology.

As rice originates from tropical regions, low temperature stress during the germination stage in temperate regions leads to serious problems inhibiting germination and seedling establishment. Identifying and characterizing quantitative trait loci (QTLs) for low-temperature germination (LTG) resistance help accelerate the development of rice cultivars with LTG tolerance. In this study, we identified QTLs for LTG tolerance (qLTG5, qLTG9) and germination coefficient of velocity under optimal conditions (OGCV) (qOGCV7, qOGCV9) using 129 recombinant inbred lines (RILs) derived from the cross between a low-temperature sensitive line Milyang23 and a low-temperature tolerant variety Gihobyeo. qLTG9 and qOGCV9 were detected at the same location on chromosome 9. At both LTG QTLs (qLTG5 and qLTG9), the alleles for LTG tolerance were contributed by the japonica variety Gihobyeo. At qOGCV7 and qOGCV9, the alleles for low temperature tolerance were derived from Milyang23 and Gihobyeo, respectively. The RILs with desirable alleles at two or more QTLs, i.e., GroupVII: qLTG5+qLTG9 (qOGCV9) and GroupVIII: qLTG5+qOGCV7+qLTG9 (qOGCV9), showed stable tolerance under low-temperature stress. Our results are expected to contribute to the improvement of tolerance to low-temperature and anaerobic stress in japonica rice, which would lead to the wide adoption of direct-seeding practices.
To investigate the relationship between cold stress and the activity of ascorbate peroxidase(APX), dehydroascorbate reductase (DHAR), mRNA expression level of two enzymes, hydrogen peroxide content was studied in lettuce leaves under stress condition imposed by cold stress at $4^{\circ}C$ for 24 hr in the dark and following recovery at $20^{\circ}C$ from cold stress. Hydrogen peroxide content increased gradually in lettuce leaves during cold stress, but decreased slightly following recovery from cold stress. Soluble protein content, however, decreased gradually during cold stress, and then rapidly returned to normal levels following recovery. Total chlorophyll content decreased gradually during cold stress, and then keep constant following recovery. The patterns of chlorophyll a and b content similar to that of total chlorophyll content, and carotenoid content didn't change. The ratio of chlorophyll a and total chlorophyll was increased during cold stress, but decreased with rapid during cold stress, and then the ratio returned to normal levels following recovery. During cold stress, the activity of APX and DHAR in the lettuce leaves increased dramatically, and also transcript levels of mRNA of APX and DHAR, as determined by probing 32P-labeled single stranded RNA of APX and DHAR, highly increased and returned to normal levels following recovery, respectively. Relationship between APX and DHAR activity and hydrogen peroxide highly related ($R^2$=0.8715 and 0.8643), whereas between hydrogen peroxide and total chlorophyll content and soluble content related reversely ($R^2$=0.5021 and 0.8915).
Bradyrhizobium japonicum is a soil bacterium with a unique ability to infect the roots of leguminous plants and establish a nitrogen-fixing symbiosis, which has been used as a microbial manure. In this study, we examined the stress response after pretreatment of cells with cold temperature. When pre-treated with cold temperature ($4^{\circ}C$) for 16 hr, B. japonicum increased the viability in subsequent stress-conditions such as alcohol, $H_2O_2$, heat, and dehydration. For cold adpatation, cultured B. japonicum was exposed to $4^{\circ}C$. Upon subsequent exposure to various conditions, the number of adapted cells pretreated by cold adaptation was 10-1000 fold higher than that of non-adaptated ones. It appeared de novo protein synthesis occurred during adaptation, because a protein synthesis inhibitor, chloramphenicol abolished the increased stress tolerance. By using a degenerate PCR primer set, a csp homolog was amplified from B. japonicum genome and sequenced. The deduced partial amino acid sequence of the putative Csp (Cold shock protein) shares a significant similarity with known Csp proteins of other bacteria.
Rapeseed is a typical winter crop, and its freezing stress tolerance is a major feature for winter survival. Therefore, it is important to comprehend clearly the physical and molecular mechanisms of rapeseed under freezing stress conditions. This study investigates the physical and transcriptome changes of two rapeseed lines, 'J8634-B-30' and 'EMS26', under cold acclimation and freezing temperature treatments. The proline content of 'J8634-B-30' at 5 ℃ increased 8.7-fold compared to that before treatment, and there was no significant change in that of 'EMS26' RNA-sequencing analysis revealed 5,083 differentially expressed genes (DEGs) of 'J8634-B-30' under cold acclimation condition. Among the genes, 2,784 (54.8%) were up-regulated and 2,299 (45.2%) were down-regulated. The DEGs of 'EMS26' under cold acclimation condition were 5,831 genes, and contained 2,199 up-regulated genes (37.7%) and 3,632 down-regulated genes (62.3%). Among them, only DEGs annotated in the cold response-related signaling pathways were selected, and their expression in the two rapeseed lines was compared. Comparative DEGs analysis indicated that cold response related signaling pathways are proline metabolism and ABA (Abscisic acid) signaling. And ICE (Inducer of CBF expression) - CBF (C-repeat-binding factor) - COR (Cold-regulated) signaling were the significantly differentially expressed transcripts in the two rapeseed lines. The major induced transcripts of 'J8634-B-30' induced P5CS (Δ'-pyrroline-5-carboxylate synthetase), which is related to proline biosynthesis, PYL (pyrabactin resistance-like protein, ABA receptor) and COR413 (cold-regulated 413 plasma membrane 1). In conclusion, these result provide a foundation for understanding the mechanisms of freezing stress tolerance in rapeseeds. Further functional studies should be performed on the freezing stress-related genes identified in this study, which can contribute to the transgenic and molecular breeding for freezing stress tolerance in rapeseed.
Calcium-dependent protein kinases (CDPKs) are known to be involved in regulating plant responses to abiotic stresses such as salinity, cold temperature and dehydration,. Although CDPKs constitute a large multigene family consisting of 31 genes in rice, only a few rice CDPKs' functions have been identified. Therefore, in order to elucidate the functions of OsCPK11 in rice, this study was intended to focus on the expression pattern analysis of OsCPK11 in wild type and ND0001 oscpk11 mutant plants under these abiotic stresses. For the salt, cold and drought stress treatment, seedlings were exposed to 200 mM NaCl, 4℃ and 20% PEG 6,000, respectively. RT-PCR and quantitative real-time PCR were performed to determine the expression patterns of OsCPK11 in wild type and ND0001 mutant plants. RT-PCR results showed that OsCPK11 transcripts in the wild type and heterozygous mutant were detected, but not in the homozygous mutant. Real-time PCR results showed that relative expression of OsCPK11 of wild type plants was increased and reached to the highest level at 24 hr, at 6 hr and at 24 hr under salt, cold and drought stress conditions, respectively. Relative expression of OsCPK11 of ND0001 homozygous plant was significantly reduced compared to that of wild type. These results suggested that oscpk11 homozygous mutant knocks out OsCPK11 and OsCPK11 might be involved in salt, cold and drought stress signaling by regulating its gene expression.
