Effects of Vernalization and Preceding Dark Treatment on Floral Induction of Naked Barley (라맥품종의 개화유기에 관한 암흑처리와 춘화처리의 효과)

  • The effects of vernalization and darkness on flower initiation were reviewed by using naked barley varieties. The dark effects were pronounced on flower induction even at 25$^{\circ}C$. There were no differences in flowering responses among varieties. The effects of vernalization and darkness combined with 25$^{\circ}C$were 100% and 60% respectively which showed a significant difference. When the alternation of darkness with light was taken as first and last the flowering rate was 92 percent. When it was intercalated by light treatments the flowering rate was 91 per cent. Thus the effects of vernalization and darkness seems to differ.

사과의 품위개선을 위한 열처리 임계조건 설정 연구

  • 저장, 유통 중 부패미생물 및 해충류 등으로 인한 사과의 질적, 양적 손실발생을 줄이기 위해 열처리 시 사과가 생명체로서 기능을 지닐 수 있는 임계온도 및 시간에 관한 연구를 수행하였다. 후지 사과를 40~$65^{\circ}C$ 범위의 물에 침지하여 일정 시간 간격으로 시료를 취한 후 $0^{\circ}C$에서 냉각, 저장하면서 처리조건에 따른 외관, 호흡특성 및 품질을 비교하였다. 처리온도 및 시간에 따라 사과의 외관상 품질은 차이를 보였는데 4$0^{\circ}C$에서 3시간, 45$^{\circ}C$에서 1시간, 5$0^{\circ}C$에서 25분, 55$^{\circ}C$에서 3분, 6$0^{\circ}C$에서 1분, $65^{\circ}C$에서 15초 처리 시까지는 과피의 갈변발생 등 이상증세를 보이지 않았다. 이와 같은 각각의 임계조건에서 사과를 열 처리한 후 $0^{\circ}C$에 저장하면서 방치 1일 및 7일 후 사과 조직내부의 가스조성을 조사한 결과, 탄산가스 농도는 저온 장시간 처리(4$0^{\circ}C$, 45$^{\circ}C$, 5$0^{\circ}C$)한 직후 급격한 증가 경향을 보인 후 다시 감소하였다. 고온 단시간 처리구(55$^{\circ}C$, 6$0^{\circ}C$, $65^{\circ}C$)의 경우 저온 장시간 처리한 경우에 비하여 낮은 수준이지만 약간의 증감 경향을 보이다가 고른 평형 상태를 유지하였다. 산소 농도는 저온 장시간 처리 직후 급격히 저하된 후 다시 증가하였으며 저장 7일 후에는 4$0^{\circ}C$ 3시간 처리구를 제외하고는 평형 상태를 유지하였다. 고온 단시간 처리구의 경우 처리 직후부터 저장 7일 후까지 고른 평형 상태를 유지하였다. 에틸렌의 경우 저온 장시간 처리구는 처리 직후 그 농도가 급격히 상승하였다가 저장 1일 후부터 대조구보다 낮은 값을 나타내었다. 고온 단시간 처리구의 경우 저온 장시간 처리구와는 달리 저장 1일 후부터 그 농도가 다소 상승되었다가 다시 감소는 경향을 보였다. 한편 열처리하여 냉각시킨 직후 및 저장1주 후 사과의 pH, 산도, 당도, 경도, 과육의 갈변도 등을 조사하였던 바 대조구 및 처리구간에 일부 항목에 있어서는 약간의 차이를 나타내었다.

Changes of Methanogenic Pathway with Incubation Temperatures in the Littoral Sediment of Reservoir Paldang, Korea (팔당호 연안대 저질토에서 배양온도에 따른 메탄발생 경로의 변화)

  • Changes in methanogenic pathway at low temperature were studied by incubation experiments of sediment slurries from the littoral zone of Reservoir Paldang. Methane production rates in sediment slurries increased exponentially between $5^{\circ}C$and $45^{\circ}C$, reached a maximum rate of $7.4\;nmol\;{\cdot}\;g^{-1}\;{\cdot}\;h^{-1}$ at $45^{\circ}C$, and then declined to low rate. The shift of incubation temperature from high temperature ($30^{\circ}C$) to lowtemperature ($15^{\circ}C$) resulted in a decrease of methane production rate and of hydrogen accumulation rate, and the transient accumulation of acetate concentration. Chlorofarm inhibited perfectly methanogenesis and resulted in the accumulation of hydrogen and acetate as immediate precursors for metltane formation at both incubation temperatures of $15^{\circ}C$ and $30^{\circ}C$. In terms of equivalent methane which was calculated from the two intermediary metabolites accumulated in absence of methanogenesis, methane production from acetate was accounted for 14% of total methanogenesis at $30^{\circ}C$ and 75% at $15^{\circ}C$, respectively. When the high acetate concentrations above 19 mM were added to sediment slurries, methane production was inhibited at the low temperature ($15^{\circ}C$) . Our results demonstrate that contribution of acetate on methanogenesis increases at low temperature, but this pathway is inhibited by high concentration of acetate. Therefore acetate-utilizing methanogensis appears to be a key reaction at low temperature, and seems to be one of bottlenecks of the low temperature anaerobic degradation of organic matter in littoral sediments of the reservoir.

The Inhibition of Green Discoloration in Garlic by Conditioning (Conditioning에 의한 마늘의 녹변억제)

  • Garlic was stored at 4, 8, and $12^{\circ}C$ to investigate the development of green discoloration. Green discoloration developed after 7 day of storage ar $4^{\circ}C$, while it developed after 15 day of storage at 8 and $12^{\circ}C$. The effect of maleic hydrazide fertilization on green discoloration of garlic was not observed. Green discoloration of garlic was accelerated by gamma-radiation treatment. The addition of cysteine did not prevent green discoloration, which decreased the commercial value of the garlic due to the presence of white specks on the surface. When 3% ascorbic acid was added to the garlic, green discoloration developed in 6 and 24 hr at room temperature and $4^{\circ}C$, respectively. The tendency of garlic to discolor was also investigated at various storage temperatures. Discolored garlic stored for 30 day at low temperatures was conditioned at $20{\sim}45^{\circ}C$ for 20 day. The green discoloration of garlic conditioned at 20 and $25^{\circ}C$ did not disappear in 20 day, but disappeared in 20 day when conditioned at $30^{\circ}C$. The L, a, and b values of garlic conditioned at 35, 40, and $45^{\circ}C$ for 4 day were similar to those of normal garlic. Conclusively, our results indicated that the best method for suppressing green discoloration was conditioning discolored garlic at $35^{\circ}C$ for 4 day.

Effect of Cold Temperature Dry and Elevated Temperature Wet on Mechanical Properties of CFRP Composites (냉각($-55^{\circ}C$) 및 고온다습 조건($82.2^{\circ}C$)이 탄소섬유강화 복합재의 기계적 특성에 미치는 영향 연구)

  • The mechanical behavior of carbon fiber reinforced polymeric (CFRP) composites was investigated. Both strength and modulus were measured at room temperature dry, cold temperature dry, $-55^{\circ}C$, and elevated temperature wet, $82.2^{\circ}C$ on seven different laminate configurations consisting of $[0_6]_T$, $[90_{12}]_T$, $[0_{16}]_T$ and $[90_{16}]_T$ unidirectional laminates, $[{\pm}45]_{5S}$ angle-ply laminate, $[0/90_{12}/0]_T$ cross-ply laminate, a 36-ply laminate $[0/45/-45/45/-45/0]_{3S}$. Based on the experimental data presented, it is shown that the strength at cold temperature dry, $-55^{\circ}C$ is increased with the brittleness of fiber or matrix. Moreover, it is shown that both shear strength and modulus at elevated temperature wet, $82.2^{\circ}C$ are decreased by the cause of interfacial deterioration between fiber and matrix with moisture absorption.

Varietal Differences of Germination and Emergence Capacity under Low Temperature Conditions in Barley (대맥에서 저온 발아 및 출현력의 품종간 차이)

  • Germination and emergence of 16 barley cultivars bred in Korea were studied under different low temperature conditions in laboratory at Suweon. This study revealed that the barley cultivars differed markedly in their germination rate index (GRI), emergence rate index (ERI), and dry matter weight. The seeds of Baekdong, Namhaebori, Gangbori and Buheung germinated better than others in $4^{\circ}C$ plot. Yeongsanbori, Kwangseong and Baekdong had very poor ERI values, but Buhobori, Namhaebori, Oweolbori, Albori and malting barley, Sacheon 6 had very good ERI values. Germination rate index were not correlated with emergence rate index under any temperature conditions. The correlation coefficient between ERI of $4^{\circ}C$ and $7^{\circ}C$ plots was very high. Those results showed that for screening the varieties or lines with good emergence capacity under low temperature conditions ERI, FS and dry matter weight would be available.

Inhibition of Vibrio parahaemolyticus by Ethanol in Tryptic Soy Broth and Some Fish Homogenates (Tryptic Soy Broth와 생선 Homogenate에 첨가한 Ethanol이 Vibrio parahaemolyticus의 증식과 생존에 미치는 영향)

  • The survival and growth of Vibrio parahaemolyticus in tryptic soy broth(TSB), flounder homogenate and oyster homogenate with 0 or 5% of ethanol was tested at -20, 5, 35, 45 and 50$^{\circ}C$. Growth pattern of V. parahaemolyticus was similar in TSB and flounder homogenate but slightly poor in oyster homogenate at 35$^{\circ}C$. Growth occured at 5% ethanol, in TSB and flounder homogenate after a prolonged lag period but decreased in oyster homogenate during incubation at 35$^{\circ}C$. TSB and fish homogenates containing 0 or 5% of ethanol were inoculated with 10$\^$6/-10$\^$7/ cells/ml of V. parahaemolyticus and cold or heat resistance of the cells were determined at -20, 5, 45 and 50$^{\circ}C$. At 5$^{\circ}C$, the viability in culture broth with 5% of ethanol or without ethanol was not vary with the culture broth. In the presence of 5% of ethanol at -20$^{\circ}C$, cells of V. parahaemolyticus in flounder homogenate and oyster homogenate were more significantly inhibited than in TSB. The D-valves for V. parahaemolyticu at 45 and 50$^{\circ}C$ was significantly lower in oyster homogenate than in TSB and flounder homogenate with 5% of ethanol or without ethanol. The D-values in each culture broth without ethanol were 1.9-3.5 times of that value in each culture broth containing 5% of ethanol at 45 and 50$^{\circ}C$.

Inhibition of Listeria monocytogenes by Low Concentrations of Ethanol (저농도의 Ethanol에 의한 Listeria monocytogenes의 증식억제)

  • The effect of low concentrations of ethanol (3-7%, v/v) in tryptic soy broth (TSB) as an antibacterial agent against Listeria monocytogenes was tested at -20, 5, 35, 45, 50 and 55$^{\circ}C$. Increasing concentrations of ethanol progressively inhibited initial growth of L. monocytogenes at 35$^{\circ}C$. Growth occured at 5% ethanol, but only after a prolonged lag period. The number of viable cells of L. monocytogenes declined during incubation at 7% ethanol. TSB containing 3-7% ethanol was inoculated with 10$\^$5/-10$\^$6/ cells/$m\ell$ or L. monocytogenes and incubated at low temperatures (5$^{\circ}C$, -20$^{\circ}C$). In the presence of 3% of ethanol at 5 or -20$^{\circ}C$, bacterial growth was inhibited more than 90% of control cells. TSB containing 3-7% ethanol was inoculated with 10$\^$6/-10$\^$7/ cells/$m\ell$ of L. monocytogenes and incubated at high temperatures (45$^{\circ}C$, 50$^{\circ}C$, 55$^{\circ}C$). Decrease in viability of the cells incubated at 45 or 50$^{\circ}C$ was slow and the survival of L. monocytogenes was not affected so much in the presence of 3% of ethanol. The viability of L. monocytogenes was decreased with increasing concentration of ethanol and temperature. Decimal reduction times (D-values) based on tryptic soy agar plates at 55$^{\circ}C$ were 20.1, 12.6, 7.4 and 4.2 min in 0, 3, 5 and 7% ethanol, respectively.

  • The critical low temperature of death under submerged soil was -5^\circ C for the underground tuber of Cyperus serotinus while the vegetative organ (tuber or bulb) of Sagittaria pygmaea and Potamogeton distinctus -6^\circ C, -7^\circ C respectively. On the critical high temperature of death in water bath with 45^\circ C. Potamogeton distinctus and Sagittaria pygmaea were 1 hour, 24 hours, but Cyperus serotinus did not die. When vegetative organs of perenivial weeds setted on the soil surface, the propagation organ of Potamegeton distinctus did not die by desication in 17% of soil moisture, but Cyperus serotinus and Sagittaria pygmaea were dead within 2 to 3 hours in 25^\circ C of soil moisture.

Effect of Low Temperature for the Breaking Dormancy of Primula sieboldii, P. modesta var. fauriae and Disporum smilacinum (저온처리 기간이 앵초, 설앵초 및 애기나리의 휴면타파에 미치는 영향)

  • This study has been conducted to find out the possibility of the winter production for pot cultured plants, such as P. sieboldii, P. modesta var. fauriae and D. smilacinum in highland, 800 meter above sea level by early breaking dormancy. Low temperatures for breaking dormancy were treated for 300, 500, 700 and 900 hours under $5^{\circ}C$ in incubators and, then these plants were moved to greenhouse controlled at $10^{\circ}C$ of night temperature. Results showed that P. modesta var. fauriae and D. smilacinum, and P. smilacinum required more than 500 hours and 700 hours of low temperature $5^{\circ}C$ for breaking dormancy, respectively.
