The Cultural Conditions Affecting the Mycelial Growth of Grifola umbellata (저령의 균사생장에 영향을 미치는 배양조건)

  • This study was carried out to obtain the basic data for artificial culture of Grifola umbellata. The optimal condition for the mycelial growth was obtained at $20^{\circ}C$ and pH 4, respectively. G. umbellata showed the most favorable growth on the Hoppkins media. Carbon sources such as glucose, fluctose and manitol were favorable for stimulating a mycelial growth of G. umbellata. Valine, one of nitrogen sources also appeared to be favorable to a mycelial growth. The optimum C/N ratio was about 30:1 in case that 1% glucose as carbon source was mixed with the basal media. Lactic acid as organic acid was most favorable to the mycelial growth. Also, thiamine-Hcl as vitamin source was favorable. The mineral nutrient of $FeSO_4$ or $MgSO_4$ was most favorable to G. umbellata, and its optimal concentration was about 0.01% in $FeSO_4$ and 0.1 % in $MgSO_4$ respectively. Among 4 different cereal extract media, polished rice extract medium which was mixed with silkworm pupae was most suitable for a favorable growth of G. umbellata.

The Effects of Inhibited on Superoxide radical to Grifola umbellata Extract on Mice (생쥐에서 저령 추출물에 의한 Superoxide radical 제거 효과)

  • In this study, the inhibited effects on superoxide radical of Grifola umbellata hot water extracts (Gu-extract) on mice were investigated. The intraperitoneal administration of Gu-extract for 2 days increased swimming time by 1.8 fold, indicating that the extract might contained some anti-fatigue compound(s). The extract inhibited superoxide radical formation by 22% as compared to control, but not lipid peroxidation.

  • Three alkaloids and two steroids were isolated from the sclerotia of Grifola umbellata. Structures of the isolated compounds were determined as 9-$\beta$-D-ribofuranosyladenine (adenosine), 1-$\beta$-D-ribofuranosyluracil (uridine), 2,4-pyrimidinedione (uracil), ergosta-4, 6, 8 (14), 22-tetraen-3-one and ergosta-5, 7, 22-tritene-$3\beta$-ol (ergosterol) respectively on the basis of spectroscopic data and chemical correlations.

  • Grifola umbellatus was extracted using methanol, and the extract was further fractionated by water and ethyl acetate. Assay of each fraction with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium-bromide] revealed significant cytotoxicity effect of the methanol extract of Grifola umbellatus against human gastric cancer cell but not normal human lymphocytes. The methanol extract showed the highest antioxidant activity as well. Antimicrobial activity of Grifola umbellants against Helicobacter pylori was higher in method extract than in other fractions. Grifola umbellatus had a significant inhibitory effect on Helicobacter pylori reducing both its growth and urease activity. These results show that the methanol extract of Grifola umbellatus possesses therapeutic potential on gastric diseases.

  • This study was carried out to obtain the basic data for artificial production of Polyporus umbellatus. Among logs consisted of 16 tree species, the log of Stylax japonica inoculated with Polyporus umbellatus not only produced compact sclerotia of $3{\sim}5\;mm$ in their diameters but resulted in the good mycelial density. Also, the sclerotia were observed on 6 different kinds of logs inoculated with Polyporus umbellatus.

  • The effect of gradient eluted fractions from Et$_2$O extracts of Polyporus umbellatus screlotium was investigated on the viability of leukemia cell lines, K-562, L-1210, HL-60 and U-937 cells. Among those fractions, fraction 2 showed mild cytotoxic effect on L-1210 and HL-60 cells. Fraction 3 showed cytotoxic effect on 4 cell lines, and cytotoxic effect was the most potent on L-1210. The hallmark of apoptosis, DNA fragmentation, also appeared by fraction 3 on L-1210 after 48 hr treatment. Furthermore, this fraction was shown to be able to induce cell death on L-1210 cells by the inhibition of DNA synthesis in [$^3$H]thymidine incorporation test. From these results, P. umbellatus involves a potent chemical component that inhibits the viability of leukemia cell lines, L-1210. Further studies about the components of fraction 3 to function as a apoptosis inducer are necessary.

The Radioprotective Effects of Grifola umbellata Hot Water Extract on Mice (저령(Grifola umbellata)의 열수 추출물이 생쥐에 미치는 방사선 방호 효과)

  • In this study, the radioprotective effects of Grifola umbellata hot water extracts (Gu-extract) on mice were investigated. Single pre-administration of Gu-extract increased the 40-day survival ratio of irradiated mice from 65.5% to 78.6%. The growth of 3 week old male mice in the irradiated group was slightly retarded as compared to those of the control and Gu-extract treated mice. The average spleen and thymus weights of the irradiated mice were lower than those of the control and Gu-extract treated mice. The weight reduction of testis in the irradiated mice was significant. While it was relatively slight in the Gu-extract treated mice as compared to that of control mice. No significant difference in the weight was observed in heart, kidney or liver among three groups. The leukocytes of the Gu-extract treated mice did not decrease dramatically as in the irradiated group, but recovery patterns were similar in both groups. Reduction of erythrocytes were similar in both groups but its recovery occurred more rapidly in the extract treated group. The glucose level of the Gu-extract treated group did not change during the period examined, while it was still higher in the irradiated group than the level in the control group in two weeks. The cholesterol levels in the irradiated and the Gu-extract treated groups were higher than that of control group on day 7, but decreased to the level of the control group on day 14. No difference was observed in total protein amount of the serum among the three groups. SDS-polyacrylamide gel electrophoresis of the soluble proteins extracted from various organs did not reveal differences to any extent in all groups except in the livers of the irradiated and extract treated groups, in which some proteins were missing or less present.

Radioprotective Effects of Grifola umbellata Extracts on ${\gamma}$ - Irradiated Mice (저령(猪笭) 추출물이 감마선에 조사된 생쥐에 미치는 방사선 방호효과)

  • 전신 ${\gamma}$-선 조사전 Grifola umbellata(Gu) 추출물의 단일 투여가 생쥐의 생존율, 체중, 기관 무게 및 혈액세포에 미치는 효과를 조사하였다 ${\gamma}$-선 조사 24시간 전에 Gu 추출물을 복강 투여하였을시, ${\gamma}$-선을 조사한 생쥐의 40-day 생존율을 65.5%에서 78.6%로 증가시켰다. Gu 추출물의 투여는 ${\gamma}$-선 조사에 의한 비장과 흉선의 무게 감소를 완전히 막아주었다(P<0.05, P<0.01) 또한 이와 유사하나 다소 적은 방사선 방호효과가 Gu 투여군의 고환에서도 관찰되었다(P<0.05). Gu 추출물의 투여는 ${\gamma}$-선 조사시 나타나는 조사 후 7일째까지의 백혈구와 림프구수의 현저한 감소를 지연시켰고, 회복속도를 촉진시켰다. 또한 21일째후의 적혈구수의 회복을 촉진시켰다. 이와 같은 결과에서 Gu 추출물의 방사선 방호효과를 알 수 있다.
