Investigation of the Gene Encoding Isotocin and its Expression in Cinnamon Clownfish, Amphiprion melanopus (Cinnamon clownfish Amphiprion melnaopus의 이소토신 유전자 구조와 삼투압 조절이 미치는 영향)

  • Isotocin (IT), a nonapeptide homolog of oxytocin in mammals, has been suggested to be involved in physiological processes including social behaviors, stress responses, and osmoregulation in teleost fish. To study its structure and function, the gene encoding the IT precursor was cloned from the genomic DNA and brain cDNA of the cinnamon clownfish, Amphiprion melanopus. The IT precursor gene consists of three exons separated by two introns, and encodes an open reading frame of 156 amino acid (aa) residues, comprising a putative signal peptide of 19 aa, a mature IT protein of 9 aa, a proteolytic processing site of 3 aa, and 125 aa of neurophysin. Tissue-specific analysis of the IT precursor transcript indicated its expression in the brain and gonads of A. melanopus. To examine its osmoregulatory effects, the salinity of the seawater (34 ppt) used for rearing A. melanopus was lowered to 15 ppt. Histological analysis of the gills indicated the apparent disappearance of an apical crypt on the surface of the gill lamella of A. melanopus, as pavement cells covered the surface upon acclimation to the lower salinity. The level of Na+/K+-ATPase activity in the gills was increased during the initial stage of acclimation, followed by a decrease to its normal level, suggesting its involvement in osmoregulation and homeostasis. The only slight increase in the level of IT precursor transcript in the A. melanopus brain upon low-salinity acclimation suggested that IT played a minor role, if any, in the process of osmoregulation.

  • 어떤 산업이든 기계공업이 관여되지 않는 분야가 없듯이 공작기게가 관여되지 않는 기계공업은 생각할 수 없다. 이와같이 공작기계는 <기계를 만드는 기계> 라는 점에서 모든 산업분야의 근간을 형성하고 있으므로 공작기계의 품질과 성능은 곧 한나라의 기술수준을 가늠하는 척도로서 대표된다고 하여도 과언은 아 니다. 오늘날 선진국들은 수요자가 요구하는 다양한 초정밀공작기계까지도 만들고 있다. 크게는 우주개발에 서 항공기, 선박 및 자동차생산을 위시하여 건설장비, 전자 및 광학, 정밀화학, 각종 시험기기 등 공작기계의 손이 미치지 않은 곳이 없다. 이렇게 공작기계는 전산업에 걸쳐 크게 영향을 주고 있으며, 특히 최근의 산업 화 추세가 점차 자동화, 무인화, 초정밀첨단산업으로 비약함에 따라 공작기계의 역할은 더욱더 중요시되고 있 다. 우리나라 공작기계공업은 1960 년대의 이전 극히 영세한 중소기업중심의 보수공작기계부품 생산형태에서 1960 년대 후반부터 일부 범용기종의 생산이 시작되었으나 보다 본격화한 것은 1973 년 정부의 중화학공업육성 시책에 따라 대기업의 대거 참여로 공작기계제조업은 시설, 자산, 경영면에서 대형화 하였고, 동시에 선진국의 최신기술과 시설을 적기에 도입하므로써 품질향상, 신제품개발, 수출참여 등 공작기계공업 발전에 일대 전기를 마련하였다. 특히 '85 년에 들어서면서부터 3 저의 여파로 국내 자동차산업의 호황, 전자, 기계 및 관련수요 산업의 설비투자와 시설개체에 힘입어 공작기계생산도 활발하기 시작하여 그 어느때보다 생산기반혁신을 위한 구조적 대응책이 절실해지고 있다. 따라서 공작기계공업의 당면과제는 지속적인 기술개발과 품질향상으로 국산 화율을 제고시켜야 함은 물론, 이를 양산화하여 명실공히 선도산업으로서의 위치를 굳히고, 적어도 기계류의 수입국에서 기계류의 수출국으로 기치를 든 탈바꿈을 할때 비로소 불균형이 없는 흑자시대는 도래할 것이다.

Development of Exposure Biomarkers for Endocrine Disrupting Chemicals Using DNA Microarray (DNA 마이크로어레이를 이용한 내분비장애물질 노출지표 개발)

  • 장기간 노출 시 발암 등 인체 유해성을 갖는 환경유래 내분비장애물질(endocrine disrupting chemicals, EDCs)에 대한 선택적이고 민감한 노출지표를 개발하기 위하여 본 연구에서는 DNA microarray를 이용하였다. 피험자는 아직 특별한 질환을 갖지 않는 18세 이상 연령, 성을 맞춘 EDCs고농도 노출군(N = 16)과 저농도군(N = 16)으로 구성되었다. 노출정도 구분은 10년 이상 거주지가 K산업폐기물 소각장과 2.5 km 반경 내, 외 인지에 따라 고노출군,저노출군으로 구분하였다. 피험자의 말초혈에서 total RNA를 분리, 각 군당 B인씩 pool로 cDNA를 합성하여 oligonucleotide DNA 칩에 적용하였다. 유전자발현의 차이를 GenePixPro 4.0 software를 이용하여 분석하였다. 총 3장의 칩을 이용하여 공통적으로 저노출군보다 고노출군에서 2배 이상 발현의 증가를 보인 유전자는 plasminogen activator(PLAT)등 12종이 관찰되었고, l/2이하로 발현의 감소를 보인 유전자는 kallikrein 3 (KLK3)등 29종이었다. 이 들 유전자는 PLAT등 면역계 반응에 관여하는 유전자 및 apoptosis, transport, G protein, chromatin, 암화, 발생 (development), 대사 등에 관여하는 유전자들이었다. 그러므로 KLK3등 본 연구에서 발굴한 유전자는 향후 확대된 인구에서 본 연구 결과의 확인을 통하여 EDCs특이적 노출지표로써, 나아가 암 등 EDCs관련 질병의 기전 및 병인학을 구명하는데 이용가치가 높다고 사료된다.

Identification of Amino Acid Residues Involved in Xylanase Activity from Bacillus Pumilus TX703 (Bacillus Pumilus TX703 유래 Xylanase의 활성에 관여하는 아미노산 잔기의 확인)

  • The purified xylanase from Bacillus pumilus TX703 was modified with various chemical modifiers to determine the active sites of the enzyme. Treatment of the enzyme with group-specific reagents such as carbodiimide or N-bromosuccinimide resulted in complete loss of enzyme activity. These results assumed that these reagents reacted with glutamic acid or aspartic acid and tryptophan residues located at or near the active site. In each case, inactivation was performed by pseudo first-order kinetics. Inhibition of enzyme activity by carbodiimide and W-bromosuccinimide showed non-competitive and competitive inhibition type, respectively. Addition of xylan to the enzyme solution containing N-bromosuccinimide prevented the inactivation, indicating the presence of tryptophan at the substrate binding site. Analysis of kinetics for inactivation showed that the loss of enzyme activity was due to modification of two glutamic acid or aspartic acid residues and single tryptophan residue.

Family Involvement and The Entry Mode into Entrepreneurship: The Contingent Role of National Culture (가족관여와 창업방식간의 관계에 대한 연구: 국가문화의 조절효과를 중심으로)

  • This study examines the role of family involvement and national culture in entrepreneurs' choice of mode of entry into entrepreneurship. Mode of entry consists of two choices: starting a new venture or taking over an existing firm. Specifically, we investigate how cultural values moderate the relationship between family involvement and their choice of business takeover. Using a cross-national sample of 3,831 respondents from 33 countries in Flash Eurobarometer data, we develop key variables and test our hypotheses employing hierarchical linear modelling. Our results show that the relationship between family involvement of entrepreneurs and their choice of business takeover is moderated by individualism, short term orientation, and power distance. This research contributes to understanding comparative international entrepreneurship based on national culture. Implications for policy makers and educators are discussed.

  • Here, we evaluated the effect of thrombin on the interleukin-6 production induced by tumor-necrosis-factor-$\alpha$ in endothelial cells. It is well known that tumor-necrosis-factor-$\alpha$ mediates inflammatory responses by activation of nuclear factor-kappa-B in endothelial cells. Here, we showed that lower concentration of thrombin decreased the production of interleukin-6 induced by tumor-necrosis-factor-$\alpha$ and this inhibitory effect of thrombin on interleukin-6 production was mediated by interacting with protease-activated-receptor-1. In addition, phosphoinositide-3-kinase was also involved the anti-inflammatory responses by lower concentration of thrombin in endothelial cells. These results suggested that lower concentration of thrombin mediated anti-inflammatory responses by interacting with protease-activated-receptor-1 on the cell membrane and phosphoinositide-3-kinase in the cell. These findings will provide the important evidence in the development of new medicine for the treatment of severe sepsis and inflammatory diseases and good clue for understanding unknown mechanisms by which thrombin showed the pro-inflammatory or anti-inflammatory activities in endothelial cells.

Biological Activities of Substance Extracted from the Fruit Body of Formitopsis rosea (잔나비버섯 자실체로부터 분리한 물질의 특성 및 생리활성)

  • This study was conducted to investigate the characteristics and biological activities of the substance extracted from the fruit body of Formitopsis rosea. The substance was extracted by hot water and then it was separated high and low molecular weight fraction by ethanol precipitation, The high molecular weight fraction was found to be a proteoglycan composed of glucose, mannose, galactose, xylose, fructose, ribose and amino acids. The amino acids of proteoglycan were mainly threonine, isoleucine, glycine, aspartic acid. Anticomplementary activity of the high molecular weight fraction was higher than that of the low molecular weight fraction. And also, the high molecular weight fraction enhanced phagocytic activity and nitric oxide production of macrophage. In enzymatic lipid peroxidation reaction with $ADP-FeCl_3-NADPH$, the lipid peroxidation was inhibited 82.4% and 86.6% by high and low molecular weight fractions, in nonenzymatic lipid peroxidation reaction, it was 83.0% and 84.6%, respectively.

공생별 Z And의 특이 분광선 연구

  • 두 개의 별로 구성되었을 것이라고 여겨진 공생별은 밝기변화는 궤도운동에 따라 밝기가 변하는 것으로 알려졌다. 분광 관측 자료에는 이러한 궤도 변화 외에도 다양한 요소가 밝기 변화에 관여하는 것이 특성으로 나타난다. 또한 공생별은 밝기가 급격하게 증가하고 혹은 감소하기도 하는데, 이는 폭발에 기인하는 것으로 판단된다. 이러한 변화, 궤도 운동에 따라 기하학적 변화와 폭발 현상을 모두 볼 수 있는 대표적인 공생별이 Z And이다. 우리는 선행연구(MIKOLAJEWSKA & KENYON, 1996)에서 발표한 저분산 분광 자료를 이용하여 위상별로 변하는 상대적 선세기 변화를 조사하였다. MIKOLAJEWSKA & KENYON (1996)의 자료는 저분산 기기로(${\Delta\lambda\sim}3{\AA}$), HeI, HII, [OIII]5007, [NeV]등이 관측되었는데, 이러한 선들의 세기를 광이온 모델을 이용하여 예측한 후, 공생별 가스를 이온화시킨 중심별(WD)의 물리적 특성을 연구하였다. 또한 Hyung & Aller가 2002년 8월 12일 Lick Observatory에서 Hamilton Echelle Spectrograph (HES)를 사용하여 3600초 노출 관측한 고 분산 분광자료(${\Delta\lambda\sim}0.1{\AA}$)도 분석하였다. HES 관측 자료는 공생별의 위상이 $\Phi$=0.22이며, HES자료는 $3470{\AA}-9775{\AA}$에서 HI, HII, HeI, HeII, NII, NIII, OII, [OI], [OII], [OIII] 등이 있었다. 이 선들의 선 윤곽을 IRAF와 StarLink/Dipso를 이용하여 분석하고, 각 성분이 위상($\Phi$=0.22)인 상태에서의 관측자에 대해 어떠한 기하학적인 구조를 가지고 있는지 연구하였다. CLOUDY를 사용하여 광 이온 모형성운을 만들어 화학원소 및 성운가스의 물리적 조건을 연구하였다. Z And의 수소의 수밀도($N_H$)는 $10^{8.5}/cm^3$으로 가정하였다. 중심별 온도는 약 110,000K, 광도는 태양의 2000배로 추정되었다.

Korean vs. American Corporate Websites: Interactivity, Comparative Appeals and Use of Technology

  • This study is a cross cultural examination of interactivity, comparative advertising, and technology use in Korean and U.S. corporate websites. Utilizing cultural analysis criteria of high vs. low context, power distance and individualism-collectivism, we compared interactivity in U.S. and Korean websites. Our findings indicate that U.S. corporate websites tend to emphasize customer-message and customer-advertiser interactivity, while Korean websites highlight customer-customer interactivity. Findings on comparative advertising strategy indicate that cultural bias represented in U.S. individualism and Korean collectivism leads U.S. web advertisers to utilize direct and indirect comparative advertising more frequently than their Korean counterparts. The results of our technology analysis unexpectedly show that there is no significant difference between the two countries.

Neonatal Rat Necrotizing Enterocolitis Model Adopting Oral Endotoxin and Hypoxia Exhibits Increased Apoptosis through Caspase-3 Activation (경구 내독소와 저산소로 유발된 신생쥐의 괴사성 장염모델에서 caspase-3 활성화를 통한 세포자멸사의 증가)

  • Purpose : The aim of this study was to develop a model for necrotizing enterocolitis (NEC) in the neonatal rat using endotoxin and hypoxia, a plausible insult in a neonatal intensive care and to investigate the role of apoptosis as the underlying mechanism. Methods : Newborn rats were given oral endotoxin and intermittent 8% hypoxia$\pm$caspase inhibitor. The intestinal histology was evaluated using hematoxylin-eosin staining. Apoptosis was analyzed with TUNEL staining and by measuring the caspase 3 activity in the intestinal lysates. IEC-6 cells were assessed for apoptosis and the expression of Bax, Bcl-2, Fas and FasL was measured after treatment with endotoxin and hypoxia. Results : Oral endotoxin (5 mg/kg) and exposure to 8% hypoxia of 60-min duration twice induced human NEC-like lesions in the rat intestine. Intestinal tissue revealed increased apoptosis and caspase-3 activity. After caspase inhibitor treatment, the grades of both apoptosis and NEC were significantly reduced. IEC-6 cells exhibited increased apoptosis and caspase 3 activity after endotoxin and hypoxia treatment and significantly increased Bax/Bcl- 2 ratio compared to control cells. Conclusion : This neonatal rat model of NEC which was induced by oral endotoxin and intermittent hypoxia showed increased apoptosis of intestinal epithelial cells that was mediated by caspase 3 activation. Our model has a advantage in the study of NEC because the use of much more clinically plausible insults may provide a suitable model for the investigation of its pathophysiology and therapeutic trials.
