Oxidation of Primary Alcohol Groups of Polysaccharides with 2,2,6,6-Tetramethyl-1-Piperidine Oxoammonium Ion (2,2,6,6-Tetramethyl-1-Piperidine Oxoammonium Ion에 의한 다당류내 1차 알코올의 특이적 산화)

  • The primary alcohol groups of four kinds of polysaccharides (com starch, rice starch, sweet potato starch, and cellulose), with different structures and water solubilities, were oxidized to carboxyl groups using 2,2,6,6-tetramethyl-1-piperidine oxoammonium ion (TEMPO) at $25^{\circ}C$. The optimum pH, TEMPO content, and NaBr content for the TEMPO/hypobromite-catalyzed oxidation of the polysaccharides were $10.5{\sim}11.0$, 10 mmol/mol primary alcohol, and 0.49 mmol/mol primary alcohol, respectively. The oxidation degree for the primary alcohol group was more than 90% for all four kinds of the polysaccharides. The oxidation process greatly increased the water solubility of the polysaccharides. Water-insoluble polysaccharide such as cellulose became water-soluble to the extent of 8.42% (w/v). And also, the polysaccharides with very low water solubility (less than 0.10% (w/v)) such as com starch, rice starch, and sweet potato starch had high water solubility of approximately 45%(w/v). The gel-forming abilities with calcium ion were determined. The oxidized polysaccharides are new anionic polymers with unique structures that could have application as gums, gels, and films.

Effects of Riboflavin-photosensitized Oxidation on the Formation of Volatile Compounds in Oleic Acid Model Systems (리보플라빈 광산화에 의한 올레산 모델시스템에서 휘발성 물질 발생연구)

  • Formation mechanisms of light-induced volatile compounds were studied using riboflavin-photosensitized oleic acid model systems. Volatile compounds in model systems with 4000 ppm riboflavin at $35^{\circ}C$ under light or in the dark for 39 hr were isolated and identified by a combination of solid phase microextraction (SPME), gas chromatography (GC), and mass spectrometry (MS). Total volatiles in oleic acid with riboflavin under light for 13, 26, and 39 hr increased by 90, 190, and 270%, respectively, compared to those in oleic acid without riboflavin under light. Total volatiles in samples without riboflavin under light or samples with riboflavin in the dark were not significant in tested conditions (p>10.05). Riboflavin did not act as a photosensitizer when it was dispersed in oleic acid. Heptane, octane, heptanal, octanal, nonanal, and 2-nonenal were significantly increased in riboflavin-photosensitized samples compared to those in samples without riboflavin (p<0,05). Light-induced volatile compounds including heptanal and 2-nonenal from oleic acid could be explained using singlet oxygen oxidation, the formation mechanism of which were not understood using triplet oxygen oxidation. These results will help to understand volatile formation in oleic acid containing foods stored under light.

Isoflavone Distribution and $\beta$-Glucosidase Activity in Home-made and Factory-produced Doenjang (재래식 및 개량식 된장의 아이소플라본 분포 및 $\beta$-glucosidase 활성 연구)

  • Isoflavone distribution and $\beta$-glucosidase activity in 16 commercially available doenjang samples were determined. Twelve of the samples were home-made doenjang (HMD) with a relatively long fermentation period and 4 of the samples were factory-produced doenjang (FPD) from four different manufactures. Total isoflavones (TI) in the HMD ranged from $370-723\;{\mu}g/g$, while those in FPD ranged from $179-537\;{\mu}g/g$. The isoflavone distribution in HMD was different from those in FPD. Generally, the TI in HMD was higher than those in FPD. The major isoflavone was aglycones, which ranged from 42.98 to 89.96% in HMD and from 35.51 to 93.48% in FPD. Isoflavones in the $\beta$-glucoside forms were not detected in tested FPD samples. Principal components analysis (PCA) of the isoflavone profiles showed that HMD were differentiated from FPD. First principal component (PC1) and second principal component (PC2) expressed 43.6 and 22.9% of the data variability, respectively. $\beta$-Glucosidase activity in doenjang was lower than that in raw soybeans. The results of this study can be used to understand the differences in the isoflavone distribution in traditionally manufactured and factory produced doenjang.

Enhancement of ${\beta}$-1,3-Glucan Functionality by Batch and Semi-continuous Typed Specific Carboxylation (회분식 및 반연속식 특이적 카르복실화 반응에 의한 ${\beta}$-1,3-glucan의 기능성 향상)

  • In this study, batch or semi-continuous reactions, introducing site-specific carboxylic acids in ${\beta}$-1,3-glucan structures, were performed to increase water solubility and gel forming ability, using TEMPO/hypobromite with or without NaBr as catalysts. Regio-selective carboxylic acid formations were determined with infrared (IR) and $^{13}C$ nuclear magnetic resonance (NMR) spectroscopic analyses. The regio-selective reactions with and without NaBr gave oxidation yields of 92.5 and 85.6%, respectively, in the batch type, and yields of 93.9 and 86.4%, respectively, in the semi-continuous type. The reaction times in the batch and semi-continuous reactions without NaBr were delayed by 100 and 150%, respectively, as compared to those with NaBr. A combination of IR and $^{13}C$ NMR analyses were used to confirm the formation of carboxylic acids in ${\beta}$-1,3-glucan. From the batch reactions with and without NaBr, the water solubilities of oxidized products were 50.0 and 55.6%, respectively, and in the semi-continuous reactions they were 52.6 and 53.5%, respectively; while the water solubility of the native ${\beta}$-1,3-glucan was less than 1.0%. Finally, as compared to the native ${\beta}$-1,3-glucan, the gel forming ability of the reaction products was greatly increased irrespective of the presence of NaBr or the reaction type.

Optimization of Onion Oil Microencapsulation by Response Surface Methodology (반응표면분석법에 의한 양파유 미세캡슐화 공정의 최적화)

  • Using agar and gelatin as wall materials, onion oil was microencapsulated using the extrusion spraying technology. A sensitive methodology was developed for quantitative determination of the microencapsulation yield through ethyl acetate extraction and gas chromatographic analyses. Optimal conditions for the microencapsulation process consisting of the ratio of [core material, Cm] to [wall material, Wm] ($X_1$), temperature of dispersion fluid ($X_2$), detergent concentration in dispersion fluid ($X_3$), and concentration of emulsifier $(X_4)$ were determined using response surface methodology. The regression model equation for the yield of microencapsulation (Y, %) of onion oil could be predicted as $Y\;=\;97.028571-0.775000\;(X_1)-0.746726\;(X_1){\cdot}(X_1)\;-\;1.100000\;(X_3){\cdot}(X_2)$. The optimal conditions for the microencapsulation of the onion oil were determined as the ratio of [core material] to [wall material] of 4.5 : 5.5 (w/w), the temperature of dispersion fluid of $17.1^{\circ}C$ detergent concentration in dispersion fluid of 0.03%, and the concentration of emulsifier of 0.42%. Results revealed the most stable microcapsule of onion oil could be formed with the highest yield of microencapsulation (more than 95%) under optimal conditions.

Arthscopic Treatment of Osteochondritis Dissecans in Femoral Condyle (관절경을 이용한 대퇴골과 박리성 골연골염에 대한 치료)

  • Purpose : We analyzed clinical and radiological results of the treatment of osteochondritis dissecans in the femoral condyle under arthroscopic guidance. Materials and Methods : The study group consists 19 cases in 17 patients. Average follow up period was 34 months and average age was 16 years. The cases were classified by 4 different groups, using the fellowing system: Group 1-stable lesion and no specific treatment after arthroscopic examination; Group 2-early separation and multiple drilling; Croup 3-unstable lesion and Herbert screw fixation; Croup 4-loose body removal and/or crater curettage. The results were analyzed by the criteria of Hughston which including clinical and radiologic outcomes. Results : There were 14 cases$(74\%)$ of good and excellent results in 19 knees in which, $75\%$(3/4) in Group 1, $75\%$(3/4) in Group 2, $86\%$(7/8) in Group 3 and $33\%$(1/3) in Group 4. The result of Herbert screw fixation group was better than that of other groups with statistically significant differences. Conclusion : In the treatment of osteochondritis dissecans of skekletally immature patients, arthroscopic finding was reliable guidance in decision of treatment method and active fixation was recommended in patients with large, unstable lesion.

Oil Fence Durability Enhancement for Marine Environmental Protection : Improvement of Inspection Process (해양환경 보호를 위한 오일펜스의 내구성 향상 : 검정제도 개선 방향)

  • Oil fences effectively prevent the spread of oil spilled in the sea, thereby reducing the damage to the marine environment. However, the fence is damaged by oil and structures at the accident site and is discarded. When incinerated disposal method for discarded fences, fine dust, and harmful materials are generated. Moreover, as a part of the damaged fence is dumped into the sea, it may cause secondary environmental pollution, such as microplastics. Therefore, in this study, durability was measured using the most common solid foam type oil fences. As a result, the reduction rate of after five days of contact was 13 % in seawater and 3 % in oil, affected by temperature changes. Thus, the durability of the fence should be improved because it is exposed to seawater and oil and affected by wind, light, and waves depending on the weather conditions. Therefore, we suggest a method to improve the oil fence inspection to strengthen the durability of the fence's fabric part.

Analysis of Heating and Cooling Energy Consumption in Rental and Sales Apartments in Busan (부산시 임대아파트 및 분양아파트의 냉난방에너지 소비량 분석)

  • This study analyzed the energy consumption differences between rental and owner owned (purchased) apartments in Busan, South Korea during the cooling and heating seasons. Analysis revealed that the average electricity consumed for cooling was 2.5 kWh/m2·yr for rental apartments and 2.3 kWh/m2·yr for purchased apartments, a difference of 0.2 kWh/m2·yr. The average electrical heating energy consumption was 3.3 kWh/m2·yr for rental apartments and 2.2 kWh/m2·yr for purchased apartments, a difference of 1.1 kWh/m2·yr. It was estimated that the use of electric blankets and heaters was higher in rental apartments than purchased apartments resulting in higher electrical heating energy consumption. The average gas heating energy consumption was 7.0 kWh/m2·month for rental apartments and 6.8 kWh/m2·month for purchased apartments. When electricity and gas usage was combined for heating, the average total heating energy consumption was 10.3 kWh/m2·month for rental apartments and 9.0 kWh/m2·month for purchased apartments. This indicates that rental apartments consume 1.3 kWh/m2·month more energy than purchased apartments during heating season. Overall, rental apartments consume more energy than purchased apartments during both the cooling and heating seasons.

A Study on the Linoleum of the Deoksugung and Changdeokgung Palaces in the Early 20th Century: focusing on its manufacturing process, characteristics, and usage (20세기 초 덕수궁·창덕궁에 유입된 리놀륨(Linoleum) 바닥재 연구: 리놀륨의 제작 방식과 특성 및 사용을 중심으로)

  • Linoleum is a resilient, hygienic, and eco-friendly floor covering. It was developed in England by Frederick Walton in 1863. Made of cork flour, linseed oil, and burlap as its main materials, the flexible and waterproof linoleum became globally popular in the early 20th century. Unlike the vinyl coverings, the burlap-backed linoleum was used not only in commercial spaces but also in household areas like kitchens, bathrooms, and even living rooms. As a global product, linoleum was imported and used in Korean palaces like Deoksugung and Changdeokgung in the early modern period. According to the record Deoksugung Won-Ahn, linoleum was applied to the major buildings, including Hamnyeongjeon, Deokhongjeon, and Jeonggwanheon, and various other venues. The linoleum used in these places are mainly monochrome blue and brown color, which probably means that they are from England. The trade records in the early 20th century show that linoleum was imported mainly from England and America. The Ewangjik building floor plan in the Changdeokgung Palace shows that linoleum was used extensively. There are even some originals, which were laid in 1920 and left in the Changdeokgung Palace. When Daejojeon and Huijeongdang were rebuilt in 1920, the interior was outfitted with western features and linoleum was used in areas such as bathrooms, the tonsorial parlor, and one of the rooms on the west side of Huijeongdang. In situ in the Daejojeon and Huijeongdang areas in the Changdeokgung Palace are monochrome, patterned black, and stylized floral tile patterned, which are closely similar to American products made by ALC and Armstrong company. This study will help us better understand linoleum's characteristics, its uses and the material itself. It will also form the basis for the restoration of Changdeokgung Palace as well as other modern interiors with linoleum flooring in the future.

Optimization of Conditions for the Microencapsulation of ${\alpha}-Tocopherol$ and Its Storage Stability (${\alpha}-Tocopherol$ 미세캡슐화의 최적화 및 저장안정성 규명)

  • We have produced the microcapsule composed of ${\alpha}-tocopherol$ as a core material (Cm) and the gelatinized polysaccharide as a wall material (Wm). Firstly, we have developed a simple, sensitive, and quantitative analysis method of the microencapsulation product using 5% cupric acetate pyridine solution. We could then optimize all the conditions for the microencapsulation process such as the ratio of [Cm] to [Wm], the temperature of dispersion fluid, and the emulsifier concentration using response surface methodology (RSM). As for the microencapsulation of ${\alpha}-tocopherol$, the regression model equation for the yield of microencapsulation (YM, %) to the change of an independent variable could be predicted as follows : YM=99.77-1.76([Cm]:[Wm])-1.72$([Cm]\;:\;[Wm])^2$. From the ridge of maximum response, the optimum conditions for the microencapsulation of ${\alpha}-tocopherol$ were able to be determined as the ratio of [Cm] to [Wm] of 4.6:5.4(w/w), the emulsifier concentration of 0.49%, and dispersion fluid temperature of $25.5^{\circ}C$, respectively. Finally, the microcapsules produced under the optimal conditions were applied for the analysis of storage stability. The optimal conditions for the storage were found to be the values of pH 9.0 and $25{\sim}35^{\circ}C$. And the storage stability of the microcapsules containing ${\alpha}-tocopherol$ were higher than 99% for a week at pH 9.0 and $25^{\circ}C$.
