The Volatile Flavor Components of Fresh Codonopsis lanceolata cultivated on a wild hill (야산 재배 더덕의 휘발성 향기성분에 관한 연구)

  • Flavor components in fresh Codonopsis lanceolata cultivated on a wild hill were detected by headspace sampling(HSS) method and GC-MS equipped with a VB-5(5% phenylmethyl polysiloxane) column. The 167 volatile compounds that were detected, consisted of 28 terpenes and terpene alcohols, 34 hydrocarbon, 31 alcohols, 13 aldehydes and ketones, 25 esters, 6 acids, 10 ethers and 20 miscellaneous components. The ten major volatile flavor components, comprising about 58% of the total, were dl-limonene (10.2%), ${\alpha}$-guaiene (9.0%), 2,2,6-trimethyl-octane (8.6%), hexadecane (8.0%), isolongifolan-8-ol (4.2%), 2,4,4-trimethyl-1,3-pentanediol diisobutyrate (4.1%), ${\beta}$-selinene (3.9%), 2,2,3-trimethylnonane (3.6%), 3-methyl-5-propyl-nonane (3.1%), and ledene (3.1%). The unique aroma of fresh Codonopsis lanceolata described by sensory evaluation was green, earthy, camphoraceous and aldehydic. The components attributed to green or camphoraceous flavor such as 1-hexanol, 2-methylhexan-3-ol, 3-hexen-1-ol, cis-3-hexenyl butyrate, ethylhexanol, hexyl acetate, trans-2-hexen-1-ol, camphor, longiborneol and menthol were not included in the ten or twenty major volatile components which had the largest peak area in descending order. We concluded that the intensity of green and camphoraceous flavor might be used as an indicator of the freshness of Codonopsis lanceolata.

  • 한국산 장뇌산삼의 열매, 잎, 줄기 및 뿌리를 -7$0^{\circ}C$ 동결건조 분말화시킨 시료에 대한 추출용매와 농도에 따른 유용성분의 함량을 비교, 분석하였다. 추출액의 당도는 잎과 줄기의 80% 에탄올추출액이 각각 22.58%와 22.53%로 가장 높았고, pH는 4.43-7.41 수준이었고, 갈변도는 흡광도가 잎과 뿌리 80% 에탄올추출액이 1.803과 1.085로 가장 높았다. 색도의 경우 L값는 줄기 100% 증류수추출액이 24.56, 열매 80% 메탄올추출액이 24.35로 가장 높았고, 뿌리와 잎 80% 에탄올추출액이 각각 17.47과 19.59로 가장 낮은 값을 나타내었다. a값은 잎100% 증류수추출액이 0.41로 가장 높았고, 줄기 80%메탄올추출 액이 -0.49로 가장 낮은 값을 보였다. b값은 줄기 80% 메탄올추출액이 3.69로 가장 높았고, 열매 100% 증류수추출액 이 0.45로 가장 낮은 값이었다. 주요 유리당은 sucrose, glucose 및 fructose 이었고, 유리당 총함량은 열매와 줄기 100% 증류수추출액이 각각 6733 mg/100g과 6142 mg/100g으로 가장 많은 량을 함유하였고, sucrose는 뿌리 80% 메탄올추출액 이 3673 mg/100g으로, glucose 및 fructose는 줄기 80% 에탄올추출액과 잎 80% 메탄올추출액에 각각 4283 mg/100g과 1897 mg/100g으로 높은 함유량을 나타내었다. 또한 잎과 줄기에는 xylose가 304-524 mg/100g 수준으로 함유되어 있었고, 뿌리에는 maltose가 소량 함유하고 있었다. 주요 유기산으로는 citric acid, tartaric acid 및 malic acid가 확인되었고, citric acid는 뿌리 80% 메탄올추출액이 1849 mg/100g으로 가장 높은 함량이었고, tartaric acid는 잎 80% 메탄올추출액이 3263 mg/100g으로 가장 높은 함량이었고, malic acid는 뿌리 80% 메탄올추출액이 1856 mg/100g으로 가장 높은 함량이었으며, 뿌리 80% 메탄올추출액에는 succinic acid, malonic acid 및 oxalic acid 등이 확인되었다. 유리아미노산은 L-Arginine, ${\gamma}$-Amino-n-butyric acid, Ethanolamine, L-Proline, $\beta$-Alanine 및 L-sarcosine 등 총 35종이 확인되었고, L-Arginine은 251-7379 mg/100g 수준으로 특히, 뿌리 80% 메탄올추출액에는 1319 mg/100g으로 총함유량의 79.13%로 매우 높은 함유량을 나타내었다. P, K, Na, Ca 및 Mg 등이 주된 무기질로 확인되었고, 그 중 P는 줄기 100% 증류수추출액에 15563 mg/100g으로 가장 높았고, K은 잎 80% 메탄을 추출액에 4952 mg/100g, Ca과 Na은 잎과 열매 100% 증류수추출액에 각각 3052 mg/100g과 1798 mg/100g, Mg은 잎 100% 증류수추출액에 950 mg/100g으로 매우 높은 함유량을 보였고, 또한 미량원소로는 Zn, Cu, Cr, Mn, Co, Mo, Fe 등이 함유되어 있었다.

Differential Anti-Carcinogenic Effect of Mountain Cultivated Ginseng and Ginseng on Mouse Skin Carcinogenesis (마우스 피부암에 대한 장뇌삼과 인삼의 특이적 항암 효능)

  • Mountain cultivated ginseng (MCG) is a type of Panax ginseng C. A. Mayer, grown in the mountains by artificial seeding. In general, it has been known that the biophysical activities of MCG is greater than that of ginseng. However, the in vivo efficacy of MCG on cancer has not been studied. In this study, we investigated the anti-carcinogenic effect of MCG and ginseng using the 7,12-dimethylbenz[a]anthracene (DMBA)-12-O-tetradecanoylphorbol- 13-acetate (TPA) two stage mouse skin carcinogenesis model. Six weeks of female ICR mice were divided into control, MCG, and ginseng diet groups and were subjected into two different experimental protocols. In the first study, each experimental diet was fed with TPA promotion for 24 weeks. The result showed that supplementation of MCG reduced tumor incidence, tumor multiplicity, and tumor size compared to those of the control and ginseng groups. In the second study, 3 groups of mice were supplied with each diet 4 weeks before DMBA tumor initiation, until the end of experiment. The result showed that tumor incidence, tumor multiplicity, and tumor size were reduced in the ginseng diet group compared to those of the control and MCG groups. TPA-induced BrdU incorporation was also significantly reduced in the ginseng diet group. Taken together, these results suggest that MCG is chemotherapeutic, whereas ginseng has a chemopreventative effect on mouse skin cancer.

Wild Ginseng Exerts Anti-inflammatory Effects via $NF-{\kappa}B$ inactivation in RAW 264.7 Cells (장뇌산삼의 $NF-{\kappa}B$ 억제를 통한 RAW 264.7 세포에서의 항염증 효과)

  • Inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 are important inflammatory mediators that have been implicated in pathogenesis of inflammation and certain types of human cancers. The present study was designed in order to determine whether Wild ginseng (Panax ginseng C. A. Mayer) could modulate $I{\kappa}B$-kinase (IKK), iNOS and COX-2 gene expression and its immune responses in RAW 264.7 macrophages stimulated with lipopolysaccharide (LPS, 1 ${\mu}/m{\ell}$). Wild ginseng extract dose-dependantly (*0.5 - 2 ${\mu}/m{\ell}$) decreased the LPS-induced IKK, iNOS and COX-2 mRNA expression and its immune responses. Moreover, it inhibited unclear factor (NF)-${\kappa}B$ immune response by LPS. These data be likely to indicate that Wild ginseng may acts as inflammatory regulator and may be possible to develope a useful agent for inflammatory diseases.

  • Today, in both domestic and foreign markets, various ginseng products are collected. However, the discrimination of kinds of ginseng is not working well because of improper technical management, which has a bad effect on the normal trade of ginseng. Therefore, the author publishes the text, based not only on many years of real experience of ginseng product mangement but also on the study of the features of wild ginseng, Jang Roy and cultivated ginseng.

Differences in Free Amino Acids between Korean Ginsengs and Mountain Ginsengs (고려인삼과 장뇌삼의 유리 아미노산 비교)

  • Free amino acids were extracted and analyzed from Korean mountain ginsengs. Chinese mountain ginsengs and Korean white ginsengs by HPLC. The highest total free amino acid content was 12.46 mg/g in Korean white ginseg(P) and the lowest total free amino acid content was 6.86 mg/g in Korean mountain ginseng (Kㅡ6) The content of arginine in a Korean white ginseng(Y) was 8.77 mg/g Arginine was 77.80% of total free amino acids in a Korean mountin ginseng (KM2) The amount of histidine and methionine in Korean mountain ginsengs were higher than any other ginsengs. The highest amount of threonine and lysine were observed in Korean white ginseng and Chinese mountain ginseng respectively, The contents of glycine in Korean mountain ginseng and korean white ginseng were higher than Chinses mountain ginseng. There is no significant difference between two mountain ginsengs and Korean white ginseng.

Immunohistochemical Study of Wild Ginseng on Benzopyrene Induced $TNF-{\alpha}$ and COX-2 Expression in Rats (장뇌산삼이 Benzopyrene으로 유도된 간조직의 $TNF-{\alpha}$와 COX-2의 면역조직학적 분포에 미치는 영향)

  • Polycyclic aromatic hydrocarbon (PAH), such as benzo(a)pyrene (B(a)P), are toxic environmental contaminants known to enhance oxidative stress, production of pro-inflammatory and inflammatory cytokines. The present study was designed in order to determine whether wild ginseng (Panax ginseng C. A. Meyer) protect PAH-induced oxidative stress and inflammation. B(a)P (0.5 mg/kg, i.p.) treatment increased the distribution of immunoreactive cells for tumor necrosis factor $(TNF)-\alpha$ and cyclooxygenase (COX)-2 in peri-portal triad region and immunoreaction was shown in the cytoplasm of macrophage. Pre-treatment with wild ginseng significantly decreased immune responses in the rats treated with B(a)p. The rats given 50 mg/kg/day for 4 weeks before B(a)P treatment had 1.39-fold and 1.5-fold inhibition of $TNF-\alpha$ and COX-2 positive reaction, respectively. Wild ginseng extract alone had no effect on the distributional changes. The SOD activity as scavenger enzymes after wild ginseng administration dose-dependantly increased compared with butylated hydroxytoluene, a general radical scavenger. These data likely indicate that wild ginseng extract may act as inflammatory regulator in conjunction with inhibition of oxidant dependent metabolic activation in environmental contaminants-induced hepatic inflammation.

Analysis of volatile compounds and metals in essential oil and solvent extracts of Amomi Fructus (사인으로부터 추출한 정유와 용매 추출물의 휘발성 물질 및 금속성분 분석)

  • Amomi Fructus with anti-oxidative activity was chosen and essential oil was obtained by SDE (simultaneous distillation extraction), and 39 constituents were determined by GC-MS (gas chromatography-mass spectrometry). Major components were camphor, borneol acetate, borneol, D-limonene and camphene. Three solvent extracts such as hexanes, diethyl ether and methylene chloride from Amomi Fructus were obtained. These were analyzed by GC-MS and 4 more constituents were identified in addition to 39 components discovered in essential oil. Five major components such as camphor, borneol acetate, borneol, D-limonene and camphene were also detected, however the relative peak percents of those components were different from those of constituents in essential oil. To estimate the kind and the amount of materials evaporated at certain temperature and conditions from essential oil and solvent extracts, dynamic headspace apparatus was used and materials evaporated and trapped at certain conditions were analyzed by GC-MS. Recovery yield of SDE method from Amomi Fructus was measured by using camphor and standard calibration solution of camphor methanol solution and, the yield was 82.0%. Content of Hg was measured by mercury analyzer and contents of Cd, Pb, Cr, Mn, Co, Ni, Cu and Zn in Amomi Fructus, essential oils and solvent extracts were determined by ICP-MS (Inductively coupled plasma-mass spectrometer). Pb, Cd and Hg were measured in the concentration of 0.72 mg/kg, <0.10 mg/kg and 0.0023 mg/kg, respectively and these were below permission level of purity test. Contents of Mn, Cu and Zn in Amomi Fructus were 213 mg/kg, 8.29 mg/kg and 31.0 mg/kg, respectively and which were relatively higher than other metals such as Cr, Co and Ni. Metals such as Mn (0.65 ~ 9.08 mg/kg), Cu (1.16 ~ 4.40 mg/kg) and Zn (1.10 ~ 3.80 mg/kg) in essential oil and solvent extracts were detected. At this point it is not clear that the metals were cross-contaminated in the course of treating Amomi Fructus or metals were contained in Amomi Fructus. The influence evaluation toward biological model study of these metals in essential oil and solvent extracts will be needed.

Scientific Conservation Treatment of the Celadon Jar with the Inscription of 'the Fourth Sunhwa Year'(National Treasure No.326) (국보 제326호 청자 '순화4년'명 항아리의 과학적 보존처리)

  • The celadon jar with inscription of 'the Fourth Sunhwa Year' is an important chronology that shows the conditions of production of the early celadon due to the inscription on the bottom including its purpose, application, and the producer. This celadon jar has been restored several times in the past. However, concerns over the structural stability, such as the separation and level differences in the joined cracks, have risen because of the aging of the repair materials, which were subjected to various environmental changes over a long time. By examining the conditions of preservation, the major damage was identified as the '入' shaped crack on the front, the 'V' shaped restored part and the crack on its left side, and the 'J' shaped crack on the back side. In the past, the cracks were found to be joined using a refined lacquer containing camphor, drying oil, rosin, etc. mixed with soil powder. The joint line was repainted with the refined lacquer and covered with gold powder. The missing parts were restored with gypsum and colored with acrylic color. After that, the repair materials were aged and emergency treatment was performed at the National Museum of Korea in 1981. At that time, Cemedine C or Cemedine C mixed with microballoons was used for reinforcing the cracks. Conservation treatment focused on removing the past repair materials and reinforcing the physically fragile parts by joining and restoring them based on the examination of the preservation condition. in addition, the area around the restored part was colored for future exhibition.

Studies on the Food Preservation by Antimicrobial Action of Medicinal Herbs -Part I. Repression of Growth of Film Yeasts in Soy Sauce by Some Medicinal Herbs- (생약재(生藥材)에 의(依)한 식품보존(食品保存)에 관(關)한 연구(硏究) -(제 1 보) 몇 가지 생약재(生藥材)의 간장 방부효과(防腐效果)-)

  • Several herbs were tested for their antimicrobial activities during preservation of soy sauces. The herbs which resulted in retarded growth of film yeasts were extracted and added to various media such as koji-extract agar, buion-peptone agar to elucidate effects on the growth of film yeasts, Aspergillus oryzae and Bacillus subtilis. The results obtained were as follows: 1) Addition of phellodendron powder resulted in retarded formation of film in fermented soy sauces. Film was produced after 2 days without addition of the phellodendron powder. However, formation of film was observed after 8 days when 0.5 per cent of the phellodendron powder was added, and after 12 days when 1.0 per cent of the powder was added to fermented soy sauce. 2) For the amino acid soy sauce, formation of film was retarded for 6 days by 0.5 per cent of the pow der and for 8 days by 1.0 per cent of the powder. 3) Repression of film yeasts by addition of helenii and camphora powder was recognized respectively, but other herbs were appeared to have no detectable effects. 4) Growth of film yeast and Asp. oryzae way, retarded for 5 days on koji-extract agar plates which contain 0.5 per cent of phellodendron extract. Bac. subtilis also showed retarded growth for 5 days on buion-peptone agar plates containing the same amount of the extract. 5) Retarded growth of film yeast for 5 days was obtained when grown on koji-extract agar plates containing 15 per cent of sodium chloride and 0.3 per cent of phellodendron powder, however, no growth of Asp. oryzae was obtained on this concentration of sodium chloride. Growth of Bac. subtilis was repressed for 5 days on buion-peptone agar plates which contain 15 per cent of sodium chloride and 0.1 per cent of phellodendron powder.
