Pathogenicity of Anastomosis Groups and Cultural Types of Rhizoctonia solani on Crops (Rhizoctonia solani의 균사융합군 및 배양형별 작물에 대한 병원성)

  • Rhizoctonia solani 균주들의 각 균사융합군 및 배형별 72종의 기주작물에 대한 병원성을 접종실험에 의해 조사하였다. 33종의 작물에서 단독으로 분리된 균사융합군 및 배양형은 그 기주작물에 병원성이 강하였으며, 8종의 작물에서 단독으로 분리된 것들은 병원성이 약하거나 강하였고, 6조의 작물에서 단독으로 분리도니 것들은 병원성이 약하거나 없었다. 26종의 작물에서 같이 분리된 균사융합군 및 배양형은 그 기주작물에 대한 병원성이 대부분 다르거나 간혹 비슷했다. AG-1(IA)는 1종을 제외한 기주작물에 잎마름병 혹은 잎썩음병과 잎집무늬마름병을 일으켰다. AG-1(IB)는 19종의 기주작물에 잘록병과 잎마름병 혹은 잎썩음병을 일으켰으나, 9종의 기주작물에는 병징을 약하게 유발시키거나 혹은 유발시키지 못했다. AG-1(IC)는 2종의 유채속작물에 잘록병과 밑둥썩음병 혹은 잎썩음병을 일으켰다. AG-2-1은 1종을 제외한 기주작물에 잘록병, 밑둥썩음병, 관부썩음병, 눈마름벼, 뿌리썩음병, 잎마름병을 일으켰다. AG-202(IIIB)는 기주작물에 뿌리 및 줄기썩음병, 잎마름병, 잘록병, 잎자루썩음병을 일으켰다. AG-2-2(IV)는 기주작물에 잘록병, 관부썩음병, 줄기썩음병, 라이족토니아마름병을 일으켰다. AG-3은 감자에 검은무늬썩음병을 일으켰다. AG-4는 42종의 기주작물에 여러 가지 병을 일으켰으나, 3종의 기주작물에는 잘록병, 눈마름병, 잎집무늬마름병을 일으켰으나, 9종의 기주작물에는 병징을 약하게 유발시키거나 혹은 유발시키지 못했다.

  • Korean ginseng (Panax ginseng) possesses various biological and pharmacological properties. Damping-off is a critical disease on ginseng seedlings, which is caused by the fungal pathogens Rhizoctonia solani and Pythium sp.. This disease is generally controlled by the application of fungicides, but also biological control is an efficient and environmentally friendly way to prevent ginseng damping-off. In a previous study, we screened soil-borne bacteria with potential applications as biological control agents for ginseng damping-off and selected the bacterial strain Streptomyces sp. A3265, producing antifungal substances guanidylfungin and methylguanidylfungin. In this study, we investigated control efficacy of Streptomyces sp. A3265 against ginseng damping-off in the field. As a result, the incidence of damping-off was significantly reduced when soaking ginseng seeds in the culture broth of Streptomyces sp. A3265.

Control of Ginseng Damping-Off Disease Using Chitinolytic Bacterial Mixtures (키틴분해미생물을 이용한 인삼 잘록병 방제)

  • An effective bioformulation of mixtures of chitin-degrading bacteria has been used successfully to control plant diseases and nematodes. In this study, the bioformulation approach was assessed to control damping-off disease of ginseng. In pot experiments with soils infested with dapming-off pathogens of ginseng, root-drenchings of Chrobacterium sp. C-61, Lysobacterium enzymogenes C-3, and mixture of two bacterial strains grown in chitin minimal medium were signficantly increased emergence of seeds and reduced damping-off disease incidence of seedlings. Efficacy of the bioformulated product depended on the dose and timing of application. In two-year-old ginseng field, the high control efficacies were achieved by soil drenching of two times with an undiluted product or three times with a 10-fold diluted product. In a To-jik nursery (self soil nursery), biocontrol efficacy of the undiluted product against damping-off disease were similar to that of a seed dressing with fungicide, Tolclofos-methyl WP. These results suggest that the bioformulated product containing Chromobacterium sp. C-61 and L. enzymogenes C-3 could be an effective approach to control of ginseng damping-off disease.

Control of Ginseng Damping-off by Streptomyces sp. A75 and A501 (Streptomyces sp. A75와 A501 균주의 인삼 잘록병에 대한 방제효과)

  • Streptomyces sp. A75 and A501 inhibited the mycelial growth of pathogenic Rhizoctonia solani and Pythium sp., which cause the ginseng disease known as damping-off. Three methods were evaluated for the control of these pathogens, using a mixture of the culture broths from Streptomyces sp. A75 and A501. The methods tested were seed dipping with 50-fold diluted broth, drenching of soil with 100-fold diluted broth after sowing, and combined seed dipping and drenching. These methods reduced the incidence of ginseng damping-off caused by R. solani by 81.3%, 84.8%, and 32.2% and that caused by Pythium sp. by 51.0%, 52.1%, and 75.3%, respectively. Based on these results, the combination of seed dipping and soil drenching after sowing using a mixture of the culture broths from Streptomyces sp. A75 and A501 effectively reduced the incidence of damping-off in ginseng.

First Report of Fusarium oxysporum Causing Damping-off on Paprika in Korea (Fusarium oxysporum에 의한 파프리카 잘록병)

  • In February 2019, a damping-off disease occurred at the seedling stage of paprika in a commercial nursery located in Cheorwon, Korea. A species of Fusarium was isolated from the diseased plant and it was identified as Fusarium oxysporum based on morphological characteristics and nucleotide sequence data of translation elongation factor $1-{\alpha}$ and the largest subunit of RNA polymerase. The isolate obtained was revealed to be pathogenic to the host plant through pathogenicity tests, and the reisolation of the pathogen confirmed Koch's postulates. This is the first report of damping-off caused by Fusarium oxysporum on paprika in Korea.

Development of Expert System for Diagnosis of Cucumber's Diseases and Nutritional Disorders (오이의 주요 병 및 영양 장애 진단 전문가 시스템 개발)

  • An expert system to diagnose cucumber's diseases and nutritional disorders(ESCD) was developed using HCLIPS. The knowledge base was made through interviews with farmers who had cultivated cucumbers for years and experts who had a specialty of cucumber's diseases and disorders. The photographs of cucumber's diseases and nutritional disorders were used to assist users to comprehend queries of the ESCD. The ESCD was able to diagnose 11 fungal and bacterial diseases and 10 nutritional disorders, which could be frequently found in the fields and greenhouses. Diagnosis accuracy of the developed ESCD was validated by the farmers and the expert who joined the development.

Effect of Fludioxonil, Flutolanil, and Thifluzamide on Suppression of Damping-off Caused by Rhizoctonia solani on Panax ginseng (인삼 잘록병(Rhizoctonia solani) 발생억제에 미치는 Fludioxonil, Flutolanil 및 Thifluzamide의 효과)

  • In vitro and in vivo effectiveness of fungicides were evaluated for the control of damping off caused by Rhizoctonia solani on Panax ginseng. Fludioxonil(67 mg a. i./L), flutolanil(75 mg a. i./L), thifluzamide(35 mg a. i./L), and mepronil (750 mg a. i./L) were selected from 9 fungicides, which were based on inhibition of mycelial growth of R. solani (isolate Rh 9801) and duration of fungicidal effectiveness against the pathogen in vitro. Field trials were made twice in the year of 2003 and 2004. Experimental plots $(54m{\times}0.9m)$ of 4-year-old ginseng fields were artificially infested with 5kg and 14 kg in fresh weight of inoculum in 2003 and 2004, respectively. The fungicides were drenched at a volume of 8l in $3.6m{\times}0.9m$ with 3 replications. Fludioxonil, flutolanil, thifluzamide and mepronil reduced the incidence of damping off by $73\%,\;69\%,\;69\%\;and\;43\%$, respectively. In the 2004 trial, fludioxonil, flutolanil, and thifluzamide showed similar result as reducing the incidence by $85\%,\;84\%,\;and\;82\%$, respectively, in the plot where the inoculum was applied 2.8 times more than the 2003. The disease incidences in untreated control were $12\%$ in 2003 and $47\%$ in 2004.

Fungicide Screening for Control of Summer Spinach Damping-off Caused by Rhizoctonia solani (Rhizoctonia solani에 의한 여름 시금치 잘록병의 방제를 위한 살균제 선발)

  • Damping-off of summer spinach caused by Rhizoctonia solani AG-4 has become a very important disease. For the control of summer spinach damping-off, antifungal activity of thirteen fungicides (pencycuron, trifloxystrobin, pyraclostrobin, azoxystrobin, kresoxim-methyl, validamycin, fluazinam, Benlate-T, flutolanil, cyazofamid, hexaconazole, tebuconazole, prochloraz) were evaluated in vitro and in vivo. Pencycuron, pyraclostrobin, validamycin, fluazinam, Benlate-T, hexaconazole, tebuconazole, and flutolanil significantly suppressed the mycelial growth of the pathogenic fungus. However, trifloxystrobin, azoxystrobia kresoxim-methyl, cyazofamid, and prochloraz did not represent good inhibition on the growth of R. solani. When applied by soil drenching (2,000 mg/L), pencycuron, pyraclostrobin, validamycin, fluazinam, Benlate-T, and flutolanil provided spinach survival ratios of 97.8%, 84.4%, 93.3%, 95.6%, 91.1%, and 86.7%, respectively. Also when treated in seed at 2,000 ing/L, pencycuron and pyraclostrobin displayed survival ratios of more than 85.1%.

A Multi-microbial Biofungicide for the Biological Control against Several Important Plant Pathogenic Fungi (진균성 식물병해 방제를 위한 항생물질 생산 길항미생물의 복합제제화)

  • In order to develop a multi-microbial biofungicide against several important plant pathogenic fungi, strains were isolated from the phtophthora blight suppressive red-pepper field soil of Gyeongsangbuk-do, Korea. Strains AY1, AY6, AB1, BB2 and F4, which had strong antagonistic ability against Phytophthota capsici and Fusarium oxysporum, were selected for their involvement with strains of biocontrol fungicide. There were no antagonism among the selected strains and were compatible for making the biofungicide. Their antagonistic mechanisms, except for strain BB2, were an antibiosis by the production of antibiotic, while BB2 produced not only an antibiotic but also cellulase as an antagonistic mechanism against blight causing P. capsici. They were identified as Halobacterium sp. AB1, Xenorhadus sp. AY1, Bacillus sp. AY6, Bacillus sp. BB2, Zymomonas sp. F4 by various cultural, biochemical test and $Biolog^{TM}$ System 4.0. The highest levels of antifungal antibiotic could be produced after 48 hrs of incubation under the optimal medium which were 0.1% galactose, 0.1% $NaNO_2$, 5 mM $Na_2{\cdot}HPO_4$ (pH 5.5). The cultured multi-microbial biofungicide showed strong biocontrol activity against bacterial wilt disease and fusarium wilt disease in cucumber and tomato fields.
