Genetic Studies of Korean Population 29. Sister Chromatid Exchange Frequency in Homicidal Schizophrenic Patient Treated with Antipsychotic Drugs (한국인 집단의 유전학적 연구 29. 약물치료를 받은 정신분열 살인환자의 자매염색분체 교환 빈도)

  • 정신분열 살인환자로 격리 치료를 받고 있는 집단을 대상으로 이들을 치료하고자 복용시키는 Haloperidol, Perphenazine, Lithium carbonate 등과 같은 항정신질환 치료제가 자매염색분체 교환(Sister Chromatid Exchange SCE)에 미치는 영향을 조사하였다. 항정신질환 치료제를 계속적으로 복용하고 있는 환자 100명(남자: 76명, 여자: 24명)과 치료제를 전혀 복용하지 않은 남자 환자 10명을 대조군으로 하여 SCE의 빈도를 분석한 결과 항정신질환 치료제를 복용한 환자군에서의 SCE 평균빈도는 세포당 12.24$\pm$0.20으로 항정신질환 치료제를 복용하지 않은 대조군에서의 평균빈도인 세포당 8.77$\pm$0.20보다 높아 유의한 차이를 볼 수 있었다 그러나 항정신질환 치료제를 복용하지 않은 대조군에서의 평균빈도는 이미 보고된 바 있는 정상인 한국인 집단에서의 평균빈도인 세포당 8.78$\pm$0.24(Park et al. , 1992)와 별 차이가 없었다 한편 항정신질환 치료제를 장기간 복용하는데 따른 SCI 빈도의 차이가 있는지의 여부를 보기 위하여 1년 미만에서 6년 정도까지의 치료제 복용기간에 따른 SCE의 평균빈도를 비교 분석한 바 복용기간에 따른 SCE 평균빈도의 유의한 차이는 볼 수 없었다. 정신분열환자들이 복용하고 있는 항정신질환 치료제가 SCE에 영향을 줄 수 있다는 결과를 얼었다.

Sister chromatid exchange in peripheral lymphocytes of radiation exposed workers in a hospital (방사선 직업 종사자의 자매염색분체교환)

  • This study is being carried out, in two different random sample groups, between 20 men who were radiation exposed workers in the two general hospitals located in "T" city as a experimental group and 20 healthy men who were non-radiation exposed workers as a control group. The occurring frequency of the sister chromatid exchange as a biological dosemeter of radiation were studied. And the age, duration of employment and smoking were used as variable for the experiment. The results are as follows : The frequency of SCE were noticed respectively by each variable : 1) by age as a variable, the frequency were increased notably in radiation exposed workers group rather than a control group(p<0.05). 2) by duration of employment, the difference of the frequency were not recognised significantly in statistical among radiation exposed workers. 3) in smoker the frequency were increased notably in a radiation exposed workers than a control groups(p<0.05). Taking into consideration the above results, the age and smoking could affect the frequency of SCE, however, the size of sample were too small to generalize. Therefore, the following suggestions are recommended to get more accurate result. 1) In order to clarify the correlation in a smoking as variable, finding the volume of smoking and its related factor are necessarily required. 2) In order to confirm the correlation in each variable, adopting of a bigger-sized sample are needed and the study itself also be carried out repeatedly.

Selenium Effect on the Frequency of SCEs Induced by Heavy Metals in Human Lymphocytes (Selenium이 mercury, cadmium 및 chromium에 의한 자매염색분체교환(姉妹染色分體交換)의 빈도(頻度)에 미치는 영향(影響))

  • The protective effect of sodium selenite($Na_2SeO_3$) against the cytogenetic toxicity of heavy metals was investigated on human whole-blood cultures in relation to induction of sister chromatid exchange (SCE) in secondary metaphase chromosome. Methylmercury chloride($CH_3HgCl$), cadmium chloride($CdCl_2$), potassium dichromate($K_2Cr_2O_7$), and sodium selenite caused to the typically dose-dependent increase in sister chromatid exchanges (SCEs) by the concentrations ranging from $0.3{\mu}M\;to\;10{mu}M$. However, the inductions of sister chromatid exchanges by methylmercury chloride or cadmium chloride were inhibited by the simultaneous addition of sodium selenite $1.2{mu}M$. The frequencies of SCE were decreased to the level of control in the molar ratios as 2:1, 1:1, 1:2, and 1:4 of selenium selenite vs. methylmercury chloride, and as 1:1 and 1:2 of selenium selenite vs. cadmium chloride, while the frequencies of SCE induced by potassium dichromate were not changed by the addition of sodium selenite in culture condition. Mitotic indices were decreased in the higher concentrations of chemicals and not significantly changed by the simultaneous addition of sodium selenite to the culture condition containing each chemicals.

  • Endocrine disruptors have been implicated in carcinogenesis in animal studies, but carcinogenetic effects on human remain controversial. In order to examine the genotoxicity of two common endocrine disruptors, Bisphenol A and Diethylstilbestrol, cytogenetic endpoints including chromosome aberration (CA), sister chromatid exchange (SCE), micronuclei (MN) analyses and DNA damage by single cell gel electrophoresis (SCGE) were assessed. The effects of Bisphenol A and Diethylstilbestrol on the frequencies of CA and MN were increased in a dose-dependent manner and that of Bispheol A was more significant by Kendall'$\tau$test. Bisphenol A and Diethylstilbestrol also increased the frequency of SCE. Bisphenol A and Diethylstilbestrol induced DNA damage in a dose-dependent manner and the DNA damage induced by Diethylstilbestrol in human blood lymphocytes was more significant.

  • Reciprocal exchanges of DNA in sister chromatids (SCEs) are induced by various carcinogens and mutagens, although the quantitative relationship between the number of mutations and SCEs induced varies among chemicals. Nevertheless, the analysis of SCEs production by various agents often proposed as a sensitive and quantitative assay for mutagenicity and cytotoxicity. Mercury, even if which has no evidences for mutagenicity and carcinogenicity, is reported to exert some cytotoxic effects, such as chromosomal aberrations or bad influences to ovulation and reproduction in experimental animals, etc.. In this study, tests for sister chromatid exchanges have been carried out on normal human lymphocytes in whole blood culture to add mercury chloride ($HgCl_2$) or methylmercury chloride ($CH_3\;HgCl$) for 72 hr. The results indicate the dose-dependent relationship between the frequencies of SCEs and the concentrations of $HgCl_2,\;CH_{3}HgCl$ and 5-bromo-2'-deoxyuridine (BrdU). Lymphocyte proliferation has depressed in the higher concentration of mercury.

A Study on the Induction of Sister-Chromatid Exchanges in Chinese Hamster Ovary $K_1$ Cells by Exposure to Cadmium (Chinese Hamster Ovary $K_1$ 세포의 자매염색분체교환에 미치는 카드뮴의 영향에 관한 연구)

  • In order to examine the mutagenicity of cadmium dichloride the author studied the induction of sister chromatid exchanges in chinese hamster ovary $K_1$ cells which treated with cadmium dichloride at various concentrations. The results obtained were as follows : 1. In cells treated with $10^{-4}M$ cadmium dichloride, a small number of cells were viable but no mitosis was bound. 2. The frequencies of sister chromatid exchanges in cells treated with $10^{-5}M\;and\;10^{-6}M$ cadmium dichloride as $10.7{\pm}1.9\;and\;8.3{\pm}2.1$, respectively, were significantly increased for control ($6.0{\pm}2.3$). (P<0.05). 3. There were dose-dependent relationship between the concentration of cadmium dichloride and frequency of sister chromatid exchanges in cells treated with cadmium dichloride at concentration ranging from $10^{-5}\;to\;10^{-7}M$.

Sister Chromatid Exchanges(SCE) in Cultured Human Lymphocytes Induced by Cadmium, Selenium and Zinc (배양임파구에서 카드뮴, 셀레늄 및 아연 투여가 자매염색분체교환에 미치는 영향)

  • To evaluate the cytogenetic toxicity, of cadmium and the reducing effect of selenium or zinc on cadmium toxicity, the induction of SCEs in cultured human lymphocytes by the concentraion of 0.5 $\mu$M to 16.0 $\mu$M of cadmium chloride and those of cadmium chloride combined with sodium selenite or zinc chloride 1.2 $\mu$M, respectively was investigated. The induction of SCEs by cadmium chloride in the range of 0.5 $\mu$M to 16.0 $\mu$M increased in a dose-dependent manner. A notable increase in SCEs by sodium selenite as well as zinc chloride was also observed. However, the frequency of SCEs by cadmium chloride was inhibited by the simultaneous addition of sodium selenite and zinc chloride 1.2 $\mu$M, respectively. The mitotic index significantly decreased in higher concentration of cadmium chloride but not was significantly different in any concentration of cadmium chloride with the simultaneous addition of sodium selenite or zinc chloride. The results showed that the decreased additive SCE effect was observed when induced by the combined treatment which could suggest that sodium selenite and zinc chloride have a protective effect on cadmium chloride.
