Enhanced Berberine Production in Sucrose Solution by Plant Cell Suspension Cultures of Thalictrum rugosum (자당 수용액을 이용한 Thalictrum rugosum 식물세포배양에서의 berberine 생산성 증진)

  • The effects of sucrose solution on cell growth and berberine production were studied in Thalictrum rugosum cell cultures. Application of sucrose solution without any other components enhanced berberine accumulation significantly in spite of slower cell growth. At high sucrose concentration, cells became very compact and fourfold increase in specific berberine content was achieved. Optimum concentration of sucrose in plain water to maximize the productivity was found to be 8%. Time course experiment in 8% sucrose solution showed that more than 5 days were required to utilize the advantage of this system efficiently. Addition of vitamins, growth regulators, and inorganic salts into the solution was not effective in increasing berberine productivity.

  • Plant growth is regulated by a variety of factors, including organic matter availability. Organic nutrients are carbohydrate molecules from photosynthetic products produced by tissues associated with carbon and energy fixation called "sources". These compounds flow through plant vascular bundles into non-photosynthetic or growing tissues called "sinks". Among these possible compounds, the disaccharide fructosyl glucose, sucrose, is the most representative. During the transport of sucrose, the pathway from the source to the sinks can include hydrolysis of sucrose into glucose and fructose derivatives or direct transfer of sucrose. Among the enzymes involved in this, β-D-fructofuranosidase is the most important. Soluble neutral β-D-fructofuranosidase, one of several isoenzymes, is located in intracellular protoplasts and helps plant cells metabolize sucrose to produce energy. In order to track the activity of this enzyme during the course of plant growth, histological methods were used for the most effective immunolocalization. As a result, the activity was higher in the phloem and epidermis than in the mesophyll tissue in the leaf. In the growing stem, activity was high in the phloem, epidermis, and cortex. The activity of the root, which is a sink tissue, was high in all parts, but especially the highest in the root tip part. It is thought that this is because it helps unloading of sucrose in sink tissues that require sucrose degradation and plays a role in hydrolysising sucrose.

AN EFFECT OF XYLITOL ON THE ADHESIVENESS OF STREPTOCOCCUS MUTANS TO SYNTHETIC HYDROXYAPATITE; AN IN VITRO STUDY (자일리톨 함유 식품이 합성 수산화인회석에 대한 Streptococcus mutans의 부착에 미치는 영향에 관한 실험적 연구)

  • There have been efforts that inhibit development of dental caries by sugar substitution. But, it is controversial if xylitol has anticariogenic effect in the presence of sucrose. And there are few papers dealing with the combined action of xylitol and sucrose. For the purpose of resolving this controversy, the author investigated the effect of xylitol on enamel demineralization and on adhesiveness of S. mutans to hydroxyapatite in the presence of sucrose. Five experimental solutions were prepared as follows: (S: sucrose, X: xylitol) Group 1: BHI broth Group 2: BHI+1% S Group 3: BHI+0.75% S+0.25% X Group 4: BHI+0.5% S+0.5% X Group 5: BHI+0.25% S+0.75% X Group 6: BHI+1% X Each solution was inoculated with $100{\mu}l$ of S. mutans JC-2. And saliva coated hydroxyapatite beads were put into each experimental solution. And then each solution was incubated at $37^{\circ}C$ under anaerobic condition. After incubation, the adhesiveness of S. mutans on hydroxyapatite was evaluated. The Vickers hardness numbers were measured on extracted human primary teeth, and these teeth were dipped into the same experimental solution and incubated at $37^{\circ}C$ under anaerobic condition for 48hours. Surface microhardness were measured again after incubation. The obtained results were as follows; 1. In the presence of sucrose, xylitol can reduce the adhesiveness of S. mutans on hydroxyapatite surface from the ratio of 25% sucrose to 75% xylitol(P<0.05). 2. In the presence of sucrose, xylitol can reduced demineralization of primary teeth enamel surface from the ratio of 50% sucrose to 50% xylitol(P<0.01).

  • 순무(Brassica rapa L., Brassica campestris L.)는 십자화과에 속하는 채소로 무의 일종이다. 모양은 팽이와 유사하고 껍질은 적자색을 띄고 있다. 이를 원료로 한 연구로 피클, 젖산발효, 식이섬유소의 함량, 성분분석에 관한 연구 등이 보고되어 있다. 본 연구에서는 순무를 활용한 농가형 음료 및 식품 소재 개발의 일환으로 자당 첨가량과 발효온도를 달리하여 순무 발효액을 제조하고 이화학적 특성을 검토하였다. 순무는 강화군 농가에서 직접 구입하였다. 순무를 $4{\times}5{\times}1cm$의 크기로 세절하고 중량대비 40%, 60%, 80%, 100%의 자당을 첨가한 다음 $24^{\circ}C$에서 15일간 발효하였다. 또한 발효온도에 따른 변화는 60%의 자당을 첨가하여 $18^{\circ}C,\;24^{\circ}C,\;30^{\circ}C$에서 21일간 발효하였다. 시료는 3일 간격으로 채취하여 수율, 당도($^{\circ}Brix$), 산도(acetic acid, %), pH를 조사하였다. 자당 첨가량이 추출 수율에 미치는 영향은 40%, 60%, 80%, 100% 순으로 각각 39.14%, 55.38%, 53.56%, 55.72%로 40%를 첨가한 군을 제외하고 차이가 없었다. 당도와 pH는 자당 첨가량이 높을수록 증가하였고 총 산도는 각각 0.415, 0.446, 0.242, 0.194 (acetic acid, %)로 나왔다. 발효온도에 따른 당도와 pH의 변화는 시간이 경과할수록 감소하였으나 산도는 발효 15일 이후에는 $30^{\circ}C$$24^{\circ}C$보다 점차 높아졌다. 총산도의 변화는 $24^{\circ}C$$30^{\circ}C$에서 차이가 없었다. 본 실험의 결과에서 볼 때 순무발효액을 추출하기 위한 가공조건은 60~80%의 자당을 첨가하여 $24^{\circ}C$에서 15일간 발효하는 것이 적당한 것으로 사료된다.

Effect of Saccharides and Incubation Temperature on pH and Total Acidity of Fermented Black Tea with Tea Fungus (배양액 제조에 사용된 당의 종류와 농도 및 배양 온도가 Tea Fungus발효 홍차의 pH 변화와 Total Acids생성에 미치는 영향)

  • The fermented black tea with tea fungus (FBTF) known to cure various diseases was prepared by culturing tea fungus biomass in black tae with different kinds (sucrose, glucose, fructose and corn syrup) and concentrations (10-60% m/v) of saccharides. pH changes and total acid production of FBTF were investigated during 14 dats incubation at $5-45^{\circ}C$ The patterns of acid production in sucrose and fructose black tea solution were similar each other, and those in glucose and corn syrup black tea solution were similar each other showing that tea fungus biomass utilizes fructose than glucose more efficiently. The optimum incubation temperature for the formation and growth of tea fungus Biomass, and the acid production was $30^{\circ}C$ Low contents of total aids (0.1%-0.2%) were produced in 20% of higher concentrations of sucrose and fructose black tea solution, while 1.8% and 0.68% were produced in the 10% of sucrose and fructose black tea solutions, respectively, pH of FBTF dropped to 2.74-3.56 after 2 days of incubation of all the culture solution and Kept this level to 14 days incubation.

  • Xylitol, a five-carbon natural sugar alcohol, is widely used non-cariogenic sugar substitute. In present study, the effects of xylitol on the expression of mRNA for glucosyltransferase which synthesizes glucan from sucrose were detected by Fluorescent in situ hybridization (FISH) and flow cytometry. FITC fluorescences for mRNA of gtfB, gtfC and gtfD were decreased further with increasing concentration of xylitol from 1% to 10% when detected by FISH. Flow cytometric analysis also showed that the expression of gtfB, gtfC and gtfD was increased by the addition of sucrose and decreased by the addition of xylitol to BHI broth containing 1% sucrose. In conclusion, the expression of gtfB, gtfC and gtfD mRNA was decreased by the addition of xylitol.

IN VITRO STUDY OF CARIOGENIC POTENTIAL OF INFANT FORMULAS (이유식의 우식유발능에 관한 생체외 연구)

  • The purpose of this study was to assess the cariogenic potential of five infant formulas in vitro. as compared with 10% sucrose solution and whole bovine milk. Buffering capacities were determined by the amount of 0.1 N lactic acid consumed to titrate the 50ml specimen solutions to pH 4.0. The pH of the specimen solution inoculated by Streptococcus mutans was measured by pH meter and the surface microhardness of primary tooth enamel immersed in the specimen solution was measured by the microhardness tester, before and after 48 hours incubation. Also, the solubility of calcium from synthetic hydroxyapatite was evaluated by atomic absorption spectrophotometer The buffering capacity of infant formulas was higher than that of sucrose solution and lower than that of milk, and there were significant differences among infant formulas(P<0.01). The average pH of infant formulas after 48 hours incubation was lower than that of sucrose solution and milk, and there was significant difference between infant formulas and milk(P<0.05). There were no signifiant differences among oops in the microhardness change of primary tooth enamel and in the amount of dissolved calcium ion from synthetic hydroxyapatite after incubation with Streptococcus mutans. In conclusion, infant formulas seemed to fulfill the basic requirments to cause dental caries in primary teeth, and there were significant differences of cariogenic potential among infant formulas. Cooperative efforts of dentistry and manufacturers to reduce the cariogenic potential of infant formulas would be necessary to prevent the early childhood caries in children.

Effect of Preventing the Maillard Reaction between Casein and Glucose with Corn Starch and Sucrose (옥수수 전분 및 자당에 의한 Casein 과 포도당 간의 갈변반응 저지효과)

  • To estimate the effect of preventing the Maillard reaction between casein and glucose coated by freeze drying with corn starch slurry or mixture slurry of corn starch and sucrose, the reactants were treated into five groups of nonbrowning material(control), uncoated browning material, browning material coated casein alone with starch slurry, browning material coated both of reactants with starch slurry and browning material coated both of reactants with mixtrue slurry. All samples had a moisture content of about 15%. Browning reaction was carried out by storage for 30 days at $37^{\circ}C$, 67% RH. In vitro available lysine contents were decreased by the browning regardless of coating the reactants and were higher about 20.5% in the browning materials coated with mixture slurry than in the uncoated browning materials. Fructosyl-lysine contents were increased about tenfold by the browning regardless of coating and were decreased about 15.8% in the browning materials coated with mixture slurry as compare with the uncoated browning materials. The materials showing the greatest resistance to the browning reaction in the coated materials were those in which both of reactants were coated with the mixture slurry of corn starch and sucrose.

Effects of Carbon and Nitrogen Sources on the Shoot Formation in bioreator culture of Scrophularia buergeriana Miquel (현삼에서 탄소원과 질소원의 종류와 농도가 기내 식물체 분화에 미치는 영향)

  • To determine the proper carbon and nitrogen sources and their proper levels for mass micro propagation of Scrophularia buergeriana Miquel, tonic and curing cough experiment were applied and a method for mass cultivation by using bioreactors (2.5 L) was expinined. Proper ratio of $NH_4NO_3\;:\;$KNO_3$ was 413 mg/L : 1900 mg/L for multiple shoot production. Sucrose was more effective than glucose or fractose as carbon source and 3% concentration was good for shoot formation. Total nitrogen was not detected after six weeks both in 500 ml flask and bioreactor culture. Sucrose was decreased sharply after two weeks and there was no sucrose left after three weeks both in 500 ml flask and bioreactor culture. The stirrer in bioreactor caused shear stress to shoots severely. The sphere type bioreactor was better than the cylinder type and removal of inner loop in sphere type was more effective to avoid shear stress.
