Studies on the Tissue Culture of Some useful woody species (유용수종(有用樹種)의 조직배양(組織培養)에 관(關)한 연구(硏究))

  • In order to substract the time and cost of propagation for inducing the haploid plants per each species. 500 anthers of late uninucleate microspore on early binucleate microspore stage of Robinia pseudoacacia (Fuel tree) Punius granatum (Ornamental tree). Aleurites fordii (Faty tree) and Styrax japonica (Silvicultural tree) were cultured on the modified Murashige and Skoog's medium supplemented with Kinetin, 2.4-D and NAA as growth regulators. And I observed the samples of cultured anthers under the microscope which were made by Microtoming method and Paraffin method. The results were summarized as follows: 1) Among 500 cultured anthers per each species, anther numbers inducing the diploid callus were as follow: Styrax japonica 20 (4% for the species total); Aleurites fordii 10 (2% for the species, total) and Punica granatum 45 (9% for the species total) were showed. 2) 2n Callus were induced from anther wall. but haploid callus were induced from anther locule. 3) Haploid callus were induced only in 25 anthers (5% for the species total) of Robinia pseudoacacia. 4) These haploid callus were not originated from body cell of anther wall tissue, but from reduced microspores, 5) Since already reported many herbaceous haploid plants were induced from the callus which were originated from reduced microspores, I conclude that the anther of woody plant which induced the haploid callus also will be cultured haploid plant.

Changes in Sleep Patterns and Mood States of Shift Workers Following Nocturnal Light Exposure (교대근무자에서 야간 광 노출에 따른 수면양상 및 기분상태 변화)

  • Objectives: We intended to observe changes in sleep patterns and mood states of night-shift workers following light exposure. We also estimated the degree of tolerance of light exposure. By studying these, we investigated the possibility of applying light therapy to night-shift workers for improving their adaptation. Methods: Twelve night-shift nurses working at Yong-In Mental Hospital volunteered to participate in this study. The study consisted of 3 parts: 1) night-shift control study; 2) light exposure study; 3) day-shift control study. All the nurses accomplished 3 parts of the study, each of which continued for 3 days, except one nurse who did not participate in day-shift control study. During light exposure study, nurses were exposed to bright light for 4 hours from 1AM to 5AM. Sleep patterns were evaluated with wrist actigraphy and automatic sleep analysis program. Mood states and side effects of light exposure were assessed with self-report scales. Results: Sleep period time, total sleep time, and sleep efficiency were increased following light exposure compared with night-shift control study. Light exposure study showed no difference from day-shift control study in above-mentioned sleep parameters. Daily fluctuation of sleep efficiency was less prominent during light exposure study than during night-shift control study. During light exposure study, the subjects felt more elated and energetic in the evening after daytime sleep than during night-shift control study. None of the subjects complained of severe side effects related to light exposure on the third day of light exposure. Tolerance of side effects was noted to develop with the repetition of light exposure. Conclusion: Light exposure improved the daytime sleep of night-shift workers to the level of normal nighttime sleep, making the subjects more elated and energetic. Side effects of light exposure were found to be tolerable. Light exposure seems to be safely applicable to night-shift workers for their adaptation.

  • Policemen judge the situations rationally and use their equipment such as handcuffs and rope within the purview, finding them needed to arrest criminals in the act who commit crimes which conforms to death penalty, life imprisonment or long imprisonment for over 3 years in accordance with Clause 10-2, Article 1 of the Police Mandate Law and prevent fleeing from them, defend their and others' lives and bodies, or if there are probable causes to be recognized that using equipment is necessary to restrain the interference with government officials in the execution of their duties. However, as the cases which the criminals run away in handcuffs or with both hands tied occur, it results in the waste of police force, distrust and enormous trouble in the pursuit of their duties. Therefore, if the way to perceive fleeing of criminals who have already worn the police equipment by some simple assistive devices without developing other new equipment, it will be very effective for police duties. This study is about the combination apparatus for fugitive prevention attached to the existing handcuffs and rope whose alert sounds let the staffs working inside the office perceive the fleeing of wanted criminals and examined suspects who wear the handcuffs or are tied up with rope, providing that they go through the exit where a transmitter and a receiver were set. The combination apparatus for fugitive prevention which the study introduces contains the connecting parts which connect a flexible tube(cognition tags inside of the tube) of connector equipped with the police equipment with the ends of the tube and the part where these two meet and which connect them inside of the tube. The connecting parts are easy to be attached to the police equipment such as handcuffs and rope, but hard to be dismantled by the people tied up with the equipment. It enables watchers to perceive the fleeing of wanted criminals and examined suspects who wear the handcuffs or are tied up with rope, providing that they go through the exit where a transmitter and a receiver were set. Plus, if it is combined together with the portable receiver, it can be installed on the patrol cars and easily adopted to supervise illegally accessing of evidences. It is also avaliable to be adjunctively utilized for the handcuffs provided and the cost is so reasonable. Owing to its snap-on way to the cuffs, it can clear up any invasion of privacy and it can not be used as a self-injury tool because of the soft tube. Using AM Tag minimizes the lack of malfunction.

Analysis on the Effect of the Dental Health Characteristics of Adult on the Status of Recognition and Practical Application of Dental Hygiene Devices (성인의 구강건강 특성이 구강위생용품의 인지도와 사용 실태에 미치는 영향)

  • The purpose of this study, which was processed from March $22^{nd}$ to April $9^{th}$ of 2010, was to figure out recognition, well-formed instructions, and Practical application about dental hygiene devices based on the dental health characteristics of 350 adults who dwell in Busan and Gyeongnam regions. Also, this study recommended the adults to use dental hygiene devices and provided such devices to help individuals take care of their dental health at home. The collected data was statistically processed with a statistics SPSS (Statistical Packages for Social Science 14.0. SPSS Inc. USA) program. First of all, in order to demonstrate the characteristics of the sample population statistics and that of the descriptive data, frequency analysis was performed and to find out the relationship between variations of the dental health, Chi-squared test through Crosstabs was operated. According to the study, recognition and Practical application of dental hygiene devices were very low. Therefore, public announcement about necessity and effectiveness of such devices should be reinforced nationwide and instructions on choosing the right device and using of the dental hygiene devices based on patients' conditions by dental hygienic human efforts gathered from dental health organization should be carried out.

Different Influence of Environmental Interpretation for Increasing Visitors' Interests -The Case of World Cup Park- (사전환경해설이 공원이용객의 환경관심에 미치는 영향의 차이 -월드컵공원을 대상으로-)

  • The goal of the study is to provide basic information that can be used to come up with environmental interpretation methods that are appropriate to the conditions of urban parks. For this matter, research was conducted to find out whether the environmental interpretation that was provided to the visitors of the World Cup Park Visitor Center encouraged them to become interested in the environment and to understand how such environmental interest was related to the level of satisfaction that the visitors were experiencing from their visits to the park. At the same time, the research was designed to find out whether the park's visitors were affected differently in terms of their environmental interests and their experience depending on the two different types of environmental interpretations; Interpreter's Interpretation and Self-guiding Interpretation. Experimental result showed no statistically meaningful correlation between environmental interpretation and environmental knowledge, though the experimental group subjected to environmental interpretation was found to have higher environmental knowledge than that of the control group, which was not subjected to environmental interpretation. As for the correlation between environmental knowledge, which was acquired through environmental interpretation, and environmental interest, the group, which showed big changes in terms of the volume of environmental knowledge, was found to have higher environmental interest than that of the group, which recorded a low level of changes in the volume of environmental knowledge. Also, the difference in their level of environmental interest was big enough to be acknowledged statistically. Also environmental interest, which was created thanks to environmental interpretation, was found to affect the level of satisfaction visitors feel when visiting the park. Even though the study was intended to find out how environmental interpretation affects park visitors by means of analysis that can be proved based on facts, it was difficult to control some of the variables due to the circumstances under which the experiments were conducted. Despite this, the study can be considered meaningful in the fact that the researches and experiments were conducted at a park that is actually visited by people. It is also believed that as one of the first studies done on the World Cup Park, it could serve as a basis upon which future studies could be carried out.

Need Assessment for Enlargement of Oriental Medical Care Service Room in Rural Community Health Center (농촌지역 보건소 한방진료실 확대설치 요구도 및 관련요인 - 일개 군 보건소 한방진료실 내소자를 대상으로 -)

  • Objectives: The oriental medical care has been getting popular in community health centers because of uniqueness, changing of disease patterns, and increasing of elderly population. From 1998, oriental medical doctors has been working in several rural community health centers for their military obligations. At this point of time, it is necessary to evaluate the oriental public health doctors system. This study was performed to investigate the utilization patterns, the degree of satisfactions, needs of oriental medical care service provided by community health center in a designated Gun area. This study focused on the need for extending over Myun area of community health center's oriental medical care services. Methods: Person-to-person interview survey method through a structured questionnaire was done by a personnel at a oriental medical care service room in a Gun community health center. The major statistical method used for the analysis were the t-test, ANOVA, and logistic analysis. Results: The total number of responded subjects in this study was 163 residents. Among these 65.0% were aged 61 or over, and only 13.5% recognized themselves were healthy. 73.7% of the respondents demanded establishment of more oriental medical care services provided by community health center to other Myun area. Factors affecting the need for enlargement of oriental medical care service room were education level, subjective awareness of access to community health center, and cost satisfaction of oriental medical service provided by community health center. Thus, a resident who had graduation of middle school achievement or above(OR=3.35), had a long way to center(OR=2.47), satisfied with oriental medical service cost(OR=2.78) had demonstrated increased chance of need by logistic regression analysis.

The comparison of Patient Hygiene Performance(PHP) Index according to the number of Oral Health Care worker with Disabled (장애인 구강건강관리인력에 따른 구강환경관리능력 지수 비교)

  • Objectives: Currently, oral health of the disabled is taken care of by the social workers, not by dental hygienists, who are the oral health professional in this area. Therefore, we aim to enhance the equity of oral health for the disabled by providing the correct oral health care method to social workers residing in the welfare facility for the disabled. Methods: Four dental hygienists and four social workers were given the class I intellectual disabilities living in 'o' welfare facilities for disabled people in Songpa-gu, Seoul from April 13, 2019 to April 20, 2019. Patient Hygiene Performance(PHP) Index were measured and compared. In advance, the social workers were taught brushing (Rolling method), and the method of brushing and measuring tools were integrated. Results: Twice a total of dental hygienists and social workers practiced toothbrushing(Rolling method) for the class I intellectual disabilities who is a person to be brushed. When comparing the Patient Hygiene Performance(PHP) Index after the second round, the result shows that both the first and second dental hygienists' Patient Hygiene Performance(PHP) Index is lower. Conclusions: Comparing oral health knowledge level and Patient Hygiene Performance(PHP) index of dental hygienist and social workers, the result shows that dental hygienist has higher oral health care ability. Therefore, the dental hygienist should be placed in welfare facility for the disabled as an expert in oral health management to create an environment in which the disabled and social workers can be trained. In addition, the curriculum of the college that nurtures the dental hygienists should have a course to understand the characteristics of the disabled person in order to enhance the professionalism of dental hygienists.

Mutations of p53 Tumor Suppressor Gene in Human Lung Cancer Cell Lines (사람 폐암세포주에서 p53 종양억제유전자의 변이)

  • Background: Recent advancement of molecular genetics has revealed that malignant transformation of a cell may be a complex multistep process and this process is grouped, in general, into two distinct categories, activation of protooncogenes and inactivation of tumor suppressor genes. This study was focused on the mutation of p53 tumor suppressor gene, because p53 gene mutation is now generally accepted to be one of the most frequent genetic changes in a variety of human cancers. Although lung cancer is one of the common cancers in Korea, the genetic change in the carcinogenesis process is not yet known clearly. To investigate the role of p53 gene mutation in lung cancer, we examined the mutations of exon 4-8 of the p53 gene in humna lung cancer cell lines, because most of the mutations of p53 gene have been reported to develop in exon 4-8. Method: Genomic DNA was obtained by the digestion of proteinase K and the extraction by phenol-chloroform-ethanol method from two human pulmonary adenocarcinoma cell lines, PC-9 and PC-14, and one human small cell lung cancer cell line, H69. To detect the mutations of exon 4-8 of the p53 gene, polymerase chain reaction single-strand conformation polymorphism(PCR-SSCP) analysis was performed with the DNA extracted from the cells. Results: The mutation of p53 gene was present in all three cell lines tested. In PC-9, PC-14 and H69, the altered mobility was detected in exon 7, 7 and 5, respectively. Conclusion: These results suggest that p53 gene mutation plays an important role in certain steps of the carcinogenesis of human non-small cell and small cell lung cancer.

A Prediction Model for the Radiation Safety Management Behavior of Medical Cyclotrons (의료용 Cyclotron의 방사선안전관리 행위 예측모형)

  • This study attempted to provide reference materials for improving the behavior level in radiation safety managements by drawing a prediction model that affects the radiation safety management behavior because the radiation safety management of medical Cyclotrons, which can be used to produce radioisotopes, is an important factor that protects radiation caused diseases not only for radiological operators but average users. In addition, this study obtained follows results through the investigation applied from January 2 to January 30, 2008 for the radiation safety managers employed in 24 authorized organizations, which have already installed Cyclotrons, through applying a specific form of questionnaire in which the validity was guaranteed by reference study, site investigation, and focus discussion by related experts. The radiation safety management were configured as seven steps: Step 1 is a production preparation step, Step 2 is an RI production step, Step 3 is a synthesis step, Step 4 is a distribution step, Step 5 is a quality control step, Step 6 is a carriage container packing step, and Step 7 is a transportation step. it was recognized that the distribution step was the most exposed as 15 subjects (62.5%), the items of 'the sanction and permission related works' and 'the guarantee of installation facilities and production equipments' were the most difficult as 9 subjects (37.5%), and In the trouble steps in such exposure, the item of 'the synthesis and distribution' steps were 4 times, respectively (30.8%). In the score of the behavior level in radiation safety managements, the minimum and maximum scores were 2.42 and 4.00, respectively, and the average score was $3.46{\pm}0.47$ out of 4. Prosperity and well-being programs in the behavior and job in radiation safety managements (r=0.529) represented a significant correlation statistically. In the drawing of a prediction model based on the factors that affected the behavior in radiation safety managements, general characteristics, organization characteristics, and selfefficacy didn't show a significant path statistically in which the prosperity and well-being programs in job characteristics affected the behavior in radiation safety managements. Therefore, it is necessary to establish a strategy that improves the level of prosperity and well-being levels in job characteristics in order to increase the behavior in radiation safety managements. Thus, this study provides basic materials for the radiation safety management of Cyclotron through the full-scale investigation that is first applied in Korea.

Three-Dimensional High-Frequency Electromagnetic Modeling Using Vector Finite Elements (벡터 유한 요소를 이용한 고주파 3차원 전자탐사 모델링)

  • Three-dimensional (3-D) electromagnetic (EM) modeling algorithm has been developed using finite element method (FEM) to acquire more efficient interpretation techniques of EM data. When FEM based on nodal elements is applied to EM problem, spurious solutions, so called 'vector parasite', are occurred due to the discontinuity of normal electric fields and may lead the completely erroneous results. Among the methods curing the spurious problem, this study adopts vector element of which basis function has the amplitude and direction. To reduce computational cost and required core memory, complex bi-conjugate gradient (CBCG) method is applied to solving complex symmetric matrix of FEM and point Jacobi method is used to accelerate convergence rate. To verify the developed 3-D EM modeling algorithm, its electric and magnetic field for a layered-earth model are compared with those of layered-earth solution. As we expected, the vector based FEM developed in this study does not cause ny vector parasite problem, while conventional nodal based FEM causes lots of errors due to the discontinuity of field variables. For testing the applicability to high frequencies 100 MHz is used as an operating frequency for the layer structure. Modeled fields calculated from developed code are also well matched with the layered-earth ones for a model with dielectric anomaly as well as conductive anomaly. In a vertical electric dipole source case, however, the discontinuity of field variables causes the conventional nodal based FEM to include a lot of errors due to the vector parasite. Even for the case, the vector based FEM gave almost the same results as the layered-earth solution. The magnetic fields induced by a dielectric anomaly at high frequencies show unique behaviors different from those by a conductive anomaly. Since our 3-D EM modeling code can reflect the effect from a dielectric anomaly as well as a conductive anomaly, it may be a groundwork not only to apply high frequency EM method to the field survey but also to analyze the fold data obtained by high frequency EM method.
