Effects of Grape Extracts on Free Radical Scavenging Activity and Inhibition of Pro-Inflammatory Mediator Production in Mouse Macrophage Cells (포도 추출물들의 자유 라디칼 소거 작용 마우스 대식세포주의 염증 발현 매개 인자들에 대한 생성 억제 효과)

  • Antioxidant and anti-inflammatory potentials of various grape extracts were evaluated. Extracts from Kyho seed, Kyho stem, and Campbell seed showed potent 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activities compared to resveratrol $(IC_{50}=16.9,\;21.5,\;21.9,\;34.6\;{\mu}g/mL,\;respectively)$, among which, antioxidant effect of Kyho seed extract were similar to that of vitamin C $(IC_{50}=12.2\;{\mu}g/mL)$. These extracts also exhibited inhibitory activities on lipopolysaccharide (LPS)-induced prostaglandin $E_2$ production and nitrite formation in mouse macrophage RAW 264.7 cells at $50\;{\mu}g/mL$. Kyho stem and seed extracts showed growth inhibitory activities in human lung and colon cancer cells. These results suggest the potential roles of grape extracts as antioxidants and anti-inflammatory agents.

Effect of Complex Extract Including Cornus officinalis on the Cimetidine/Ethanol-induced Erectile Dysfunction Model in Rats (산수유를 포함한 복합추출물이 cimetidine으로 발기부전을 유도한 동물모델에서 성기능 개선 효과)

  • Erectile dysfunction (ED), also known as impotence, is the inability to attain and sustain an erection firm enough to have sexual intercourse. Frequent ED may be a symptom of health problems including heart disease, obesity, alcoholism, stress, smoking, and depression, that need treatment. This study aimed to effect of complex extract (CPL) including Cornus officinalis on sexual function factor in the erectile dysfunction rat model. The erectile dysfuction rat model was induced by cimetidine (500 mg/kg in 5% ethanol, oral injection 2 weeks). Rats were oral administered with different concentration of CPL in rat erectile dysfunction model. As a results, sexual function factors (NO, cGMP) significantly improved in CPL treated groups (CPL-300, 600, 900 mg/kg) compared to CON group. Serum testosterone was increased in a dose-dependent manner after CPL treatment. Furthermore, administrations of CPL restored lumen areas of the prostate in the erectile dysfunction rat model. These results indicated that CPL alleviated erectile dysfunction by increasing sexual function factor and testosterone in rat model. CPL could be used to natural treatement for erectile dysfunction. However, further study is required to identify active ingredient and its mechanism of erectile dysfunction.

A Study on Gesture Recognition Using Principal Factor Analysis (주 인자 분석을 이용한 제스처 인식에 관한 연구)

  • In this paper, we describe a method that can recognize gestures by obtaining motion features information with principal factor analysis from sequential gesture images. In the algorithm, firstly, a two dimensional silhouette region including human gesture is segmented and then geometric features are extracted from it. Here, global features information which is selected as some meaningful key feature effectively expressing gestures with principal factor analysis is used. Obtained motion history information representing time variation of gestures from extracted feature construct one gesture subspace. Finally, projected model feature value into the gesture space is transformed as specific state symbols by grouping algorithm to be use as input symbols of HMM and input gesture is recognized as one of the model gesture with high probability. Proposed method has achieved higher recognition rate than others using only shape information of human body as in an appearance-based method or extracting features intuitively from complicated gestures, because this algorithm constructs gesture models with feature factors that have high contribution rate using principal factor analysis.

Water Quality Elements Extraction of Lake by the Landsat TM Images (Landsat TM 영상에 의한 호수의 수질인자 추출)

  • It is necessary to check the water quality of the lake on a continuous basis to determine the appearance of water pollution; however, it not only takes much time and expenses but it is considerably difficult to investigate the wide range of the area. If we use the remote sensing technique through the use of satellites, the status of water quality can be checked covering many wide areas simultaneously; and because the same area can be measured on a periodic basis, it is extremely effective in investigating the water quality. Furthermore, as some of the Landsat sensors carry characteristics which sense objects according to wave length, the distribution of water quality can be checked relatively accurately within a short period of time, while its image can be displayed in color. Hence, this research has attempted to extract water quality elements, such as transparency, water depth, and surface water temperature by utilizing the satellite data, and has prepared the water quality distribution image map of the Lake Hwajinpo by presenting the related empirical formula of the water quality elements. If the water quality distribution image map is prepared after extracting the water quality elements from the DN of the Landsat TM image and then carrying out TIN analysis through the use of GIS, relatively more accurate pattern can be learned covering a wide rage of area than the pattern presented based on the value obtained from actual observation.

Suppressive Effects of Ethyl Acetate Fraction from Green Tea Seed Coats on the Production of Cell Adhesion Molecules and Inflammatory Mediators in Human Umbilical Vein Endothelial Cells (Human Umbilical Vein Endothelial Cells에서 녹차씨껍질 에틸아세테이트 추출물의 세포부착물질 및 염증매개인자 생성 억제효과)

  • Anti-atherogenic effects in tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$)-stimulated human umbilical vein endothelial cells (HUVEC) are involved with suppressed oxidative stress, cell adhesion molecules, and pro-inflammatory factors. The aim of this study was to determine whether green tea seed coat ethyl acetate fraction (GTSCE) could modulate cell adhesion molecules and inflammatory mediators in HUVEC stimulated with TNF-${\alpha}$. Nitric oxide (NO) production was significantly increased in TNF-${\alpha}$-stimulated HUVEC compared to TNF-${\alpha}$ only treated cells. The NO that is produced by endothelial nitric oxide synthase dilates blood vessels and has protective effects against platelet and leucocyte adhesion. GTSCE at 25, 50, 75, and $100\;{\mu}g$/mL significantly (p<0.05) reduced TNF-${\alpha}$ production. GTSCE significantly (p<0.05) inhibited soluble vascular cell adhesion molecule-1 level, in a dose-dependent manner. Monocyte chemoattractant protein-1 level was also significantly (p<0.05) inhibited by GTSCE treatment at $75\;{\mu}g$/mL compared to the TNF-${\alpha}$-only treated group. Total antioxidant capacity by GTSCE was significantly (p<0.05) enhanced compared to the TNF-${\alpha}$-only treated group. These results suggest that GTSCE can inhibit the production of cell adhesion molecules and inflammatory mediators and could be used as a candidate bioactive material to prevent the development of atherosclerosis.

Effects of rhubarb extract on osteoclast differentiation in bone marrow-derived macrophages (대황 추출물이 골수유래 대식세포의 파골세포 분화에 미치는 영향)

  • Objectives: This study aimed to investigate the effects of rhubarb extract on osteoclast differentiation in bone marrow-derived macrophages (BMMs). Osteoclasts are vital for bone resorption and remodeling. Osteoclast dysregulation can contribute to various bone-related disorders that directly affect oral health. Rhubarb, a medicinal plant with anti-inflammatory properties, has been shown to modulate bone metabolism. Methods: BMMs were isolated from the femurs and tibias of 5-week-old C57BL/6 mice and cultured in the presence of mouse macrophage colony-stimulating factor (M-CSF) for 3 days. Subsequently, BMMs were treated with M-CSF and receptor activator of nuclear factor-κB ligand (RANKL) to induce osteoclast differentiation. Results: Rhubarb extract effectively suppressed osteoclast differentiation in BMMs. Furthermore, rhubarb extract inhibited the mRNA expression of tartrate-resistant acid phosphatase (TRAP) and cathepsin K (CTSK), which are essential for osteoclastogenesis. Moreover, it inhibited the RANKL-induced expression of nuclear factor of activated T cell c1 (NFATc1), a crucial transcription factor in osteoclast differentiation. Conclusions: These results suggest that rhubarb extract promotes oral health by inhibiting osteoclastogenesis in BMMs. Thus, rhubarb extract shows promise as a therapeutic agent for bone-related disorders that directly affect oral health, particularly those associated with abnormal osteoclast activity. Further research and exploration of the underlying mechanisms are warranted to fully understand their potential clinical applications.

Effect of Cnidium japonicum Miq. Crude Extracts on UVB-induced Photoaging Damage in Human Keratinocytes (HaCaT 세포에서 UVB로 유도된 광노화에 대한 갯사상자 추출물의 효능)

  • Cnidium japonicum (C. japonicum) is a type of halophyte that inhabits soil of a high salinity, and according to previous studies, it is known to have antitumor effects. However, the skin's protective effect, particularly against UVB irradiation, has not been revealed. In this study, C. japonicum crude extract was studied to determine its effect on damage to human keratinocytes (HaCaT) induced by UVB irradiation, and ROS assays were performed, the results of which showed that C. japonicum crude extract affects UVB-induced photoaging damage in human keratinocytes. To examine inhibitory effects against the expressions of MMPs, RT-PCR and Western blot assay were performed by treating the crude extract at concentrations of 10, 50, and 100 ㎍/ml by irradiating UVB at 15 mJ/cm2. As a result, it was confirmed that the mRNA and protein expression levels of MMP-1, MMP-3, and MMP-9 decreased in the group treated with C. japonicum crude extract, which also effectively regulated the antioxidant defense mechanism pathway by activating JNK, ERK, and p38. In conclusion, the current study suggested the possibility that C. japonicum could be used as a raw material for anti-photoaging cosmeceuticals in the future.
