Effects of Sound Stress on Physiological Processes of the American Leafminer, Liriomyza trifolii, and Proteomic Analysis (스트레스 음파 처리에 따른 아메리카잎굴파리(Liriomyza trifolii)의 생리 변화와 프로테오믹 분석)

  • This study investigated the adverse effects of sound treatment on physiological processes of the American leafminer, Liriomyza trifolii, during several developmental stages. Larval feeding activity was analyzed by measuring feeding tunnel length. It was significantly suppressed by sound treatment (5,000 Hz, 95 dB). Sound treatment delayed the pupal period at 315 - 5,000 Hz and prevented adult emergence at 1,000 - 5,000 Hz. Female oviposition was also inhibited by the stress sound treatments. However, phototactic adult movement was not affected by sound treatment. Pupae treated with 5,000 Hz showed marked changes in protein patterns analyzed by two dimensional electrophoresis. MALDI-TOF analysis of specific protein spots indicated that trafficking protein particle complex I, triosephosphate isomerase, hypothetical protein TcasGA2_TC013388, polycystin-2, paraneoplastic neuronal antigen MA1, and tropomyosin I (isoform M) were predicted in the control insects and disappeared in the insects treated with sound. By contrast, DOCK9, cytoskeletal keratin II, and F0F1-ATP synthase beta subunit were predicted only in the sound-treated insects. Furthermore, stress sound significantly increased the susceptibility of L. trifolii to insecticides. These results suggest that physiological processes of L. trifolii are altered by sound stress, which may be exploited to develop a novel physical control tactic against L. trifolii.

Study on the Juvenile Hormone Binding Protein in the Hemolymph of the Silkworm Larva, Bombyx mori. (누에 체액의 유약호르몬 결합단자질(Juvenile hormone hinding protein)에 관한 연구)

  • In order to examine a physiological role of juvenile(JH) binding proteins in the hemolymph of the silkworm larva, Bombyx mori, [3H] JH I incubated hemolymph was separated by polyacrylamide gel electrophoresis in the fifth-instar larva and the activity of the binding protein was analyzed using charcoal binding assay. The results obtained were as follows; 1. The JH was bound by two protein fractions in the hemolymph of the fifth-instar larva; One was JH binding lipoprotein(JH-LP), the other was JH speific binding protein(JHBP). Their relative mobility values(Rm) were 0.3∼0.33 and 0.81∼0.84, respectively. There were no valid differences in those values from developmental stages of both male and female silkworms. 2. Total protein contents of the hemolymph were gradually increased during the fifth-instar larva, while at the prepupa decreased. The maximum ones were observed at the spinning period and the contents from female were much higher than those from the male. 3. JH binding activity per ml of the hemolymph was low in the early stage of the fifth-instar larva and its activity was maximized at the psinning period and at the prepupa slightly decreased. 4. There was a similar pattern between changes of the JH binding activity per ml of the hemolymph and of the total protein contents of the hemolymph. 5. The JH binding activity per mg of the hemolymph proteins was high in the early stage of the fifth-instar larva, while from the 6th day of the fifth-instar larva to the prepupa its activity showed the lowest levels.

Host plants and Biological Characteristics of Illeis koebelei Timberlake (Coleoptera: Coccinellidae: Halyziini) in Gyeonggi-do (노랑무당벌레의 발생기주 및 생물학적 특성)

  • We investigated mycophagous ladybird, Illeis koebelei from 12 species of plants infected with powdery mildew in Gyeonggi-do, Korea. The pear tree, Pyrus ussuriensis var. macrostipes (Nakai), was most preferred by I. koebelei. This species was found from early July to early November in pear orchards. There was no entomophagous trace in the gut of I. koebelei without powdery mildew spores in a microscope. All stages except egg and pupa are obligate mycophagous, and the feeding potential is ranked as follows: fourth instar, adults, third instar, second instar, and first instar. Feeding amounts of each stage of I. koebelei were 45.6, 144.4, 372.2, 628.1, and $473.7mm^2$ of cucumber powdery mildew per day. Fourth instar larvae showed highest consumption of cucumber powdery mildew. Developmental periods of four larval instars and adults feeding cucumber powdery mildew were 1.2, 2.3, 2.3, 4.6, and 37.7 days, respectively, at $25^{\circ}C$. In this study, we could not determine the feeding potential of I. koebelei against the cucumber powdery mildew; therefore, and further studies are required to elucidate the potential of this species as a biological control agent, e.g., mass rearing, selection of low toxic chemical agents for Integrated Pest Management (IPM), and control techniques against powdery mildew in agro-ecosystems.

Esterase Isozymes of Beet Armyworm, Spodoptera exigua(H bner), with Development and Tissues (발육 및 조직에 따른 파밤나방(Spodoptera exigua (H bner)) 에스테라제 동위효소)

  • The effect of physiological factors of beet armyworm, Spodoptera exigua (Hubner), on esterase variation was analyzed by comparing electrophoretic esterase isozymes. Each esterase isozyme was also characterized by substrate and inhibitor specificities. A total of 28 esterase isozymes were separated on 10% nondenaturing polycarylamide gel electrophoresis (PAGE). These isozymes were denoted from El to E28 according to cathodal migration distances. There was a variation in esterase isozymes among developmental stages. Larvae and pupae had more isozymes than did adults. Eggs had only eight isozymes. The isozymes of El and E2 were specific only in the first instar larvae. Esterases also showed variation according to different tissues. More kinds of esterase isozymes were found in epidermis and gut tissues than in hemolymph and fat body. Some isozymes were specific in epidermis (from El to E6), gut (E10, El 1, E25, E26, and E27), and hemolymph (E18). Among 10 naphthyl esters, a-naphthyl propionate was the most reactive substrate to the esterase isozymes. The isozymes were classified into cholinesterases (El0 and E24), arylesterases (E4, E9, E17, E19, E21, and E23), and carboxylesterases (the others) on the basis of inhibition by the esterase inhibitors-eserine, dichlorovos, moncrotophos, and paraoxon.

The Development Properties of Sericin Jam and Composition of Amino Acid in Hemolymph (세리신잠(Nd-s)의 발육특성과 혈액아미노산 조성)

  • This experiment was to know properties of Sericin Jam that development, growth of silkgland, content of sericin and composition of amino acid in hemolymph. The characters of Sericin Jam can he seen form this experiment. Hatching ratio was 85% in Sericin Jam and 95% in Jam 120. Especially hatching period of Sericin Jam was longer than Jam 120 and also hatching of Sericin Jam was ununiform. The larval duration of Sericin Jam was 20 days and 23 hours, and Jaw 120 was 21 days and 22 hours. In Sericin Jam, middle silkgland contain a great p arts in silkgalod and posterior silkgand is short and no curves. The period of mounting to emergence was 12 days in Sericin Jam. The period of pupa was 7day. It is property of Sericin Jam that the period is short. Cocoon was very thin and light in Sericin Jam. Weight of cocoon shell of Sericin Jam is 2.7cg. The sericin protein quantity was 100% in cocoon shell of Sericin Jam, about 28% in cocoon shell of Jam 120, however the sericin ratio per Sericin Jam cocoon was 34.6% compare to cocoon shell of Jaw 120 in sericin protein quantity. The amino acids in hemolymph of Sericin Jaw was much hidtidine, lysine, glut amic acid. And the amount of almost amino acids were increased depends upon development at t he 5th instar.

Control of Lycoriella ingenua (Diptera: Sciaridae) in Exports of King Oyster Mushroom, Pleurotus eryngii, using Ionizing Radiation (이온화에너지를 이용한 수출용 큰느타리버섯의 긴수염버섯파리 방제)

  • King oyster mushrooms are one of the major fresh agricultural products which their exports are increasing every year in Korea. Lycoriella ingenua, is notorious insect pest in agriculture, especially in mushroom production. Larvae of L. ingenua cause mainly direct crop damage and adults are vectors of several dangerous fungal pathogens. In this study, the effects of electron beam, X-ray, and gamma-ray irradiation on the development and reproduction of L. ingenua were evaluated. In addition, to find the optimal dose to control L. ingenua in a box filled with king oyster mushrooms, an empirical experiment was conducted for each radiation. As a result, the development and reproduction of L. ingenua were inhibited at 50 Gy for all electron beam, X-ray, and gamma-ray irradiation. Additionally, at the top, middle, and bottom of the export box filled with king oyster mushrooms, the development and reproduction of L. ingenua were inhibited by electron beam with 150 Gy, X-ray with 100 Gy, and gamma-ray with 50 Gy. These results can be provided as basic data for establishing an integrated quarantine management system when exporting mushrooms. It will also contribute to the safety of agricultural products and the strengthening of export competitiveness.

  • Laying hens' excrement from eating food wastes was mixed with paper mill sludge, aged for 21 days and then provided to the juvenile earthworms(Eisenia fetida) for 10 weeks. Biomass of earthworm population decreased by 5.7% of initially introduced population. Very few juvenile earthworms developed into the clitellates and clitellated earthworms could not produce cocoons at all, which was supposed to be caused by inhibition effects of salts in laying hens' excrement upon the sexual development of Eisenia fetida. But there was no significant effect on the survivorship of earthworm population.

  • Insecticidal activity, systemic and residual effects, and effects on enzyme activities (esterase, acetylcholinesterase, glutathione S-transferase) of indoxcarb were evaluated against Plutella xylostella. The insecticide was very effective against larvae of P. xylostella. Also, indoxacarb showed only residual effect to P. xylostella when applied to vegetable leaves. It inhibited acetylcholinesterase activity, but didn't do esterase and glutathione S-transferase activities.

  • An aphid predator, Chrysopa pallens Ramber, was reared on the artificial diets containing chicken egg yolk, yeast hydrolysate, brewer's yeast or Vanderzant's vitamin mixture, sucrose andlor bee honey, casein hydrolysate, and cholesterol. On these diets, 20.0 to 70.0% of the 1st instar larvae developed to apparently normal adults depending on diets used. The adults fed on one of these diets which was the most effective laid 230 fertile eggs for her 36 days of adult life span. The nonlipid part of the aphid, Myzus persicae Sulzer was thought to be nutritionally more important than the lipid part for the development of the green lacewing.

  • Adult specific proteins, referred to as ASP-I(adult specific protein of slow mobility) and ASP-II(adult specific protein of slow mobility) at the pharate adult stage of the silkworm, Bombyx mori, were detected by polyacrylamide gel electrophoresis. The adult specific proteins (ASP-I and ASP-II) were defined as a kind of adult hemolymph proteins without sex specificity, and the was no variation in the respective electrophoretic mobility of ASP-I and of ASP-II among the forty-one silkworm varieties. tested.
