  • Molecular phylogenetic studies were conducted to evaluate interspecific relationships in 40 populations of Viola including 35 Korean taxa, four Japanese populations and one outgroup using nuclear ribosomal ITS sequences. The phylogenetic analyses were conducted using parsimony and neighbor-joining methods. Subsection Trigonocarpae of section Nomimium appeared as the most basal clade within the Korean Viola. Section Dischidium and Chamaemelanium was monophyletidbootstrap 100%) and placed between subsect. Trigonocarpae and three other subsections of sect. Nomimium. Sect. Nomimium was paraphyletic. Although each subsectional grouping was in accordance with previous infrageneric classification based on morphological characters, yet discordance remained at the series level. Two evolutionary trends observed in the ITS tree were as follows. First, subsect. Trigonocarpae(x=10) was derived from the outgroup(x=6); Second, subsects. Bilobatae and Vaginatae(x=10 or 12), and subsect. Patellares(x=12) of sect. Nomimium were originated from sects. Dischidium and Chamaemelanium(x=6). Viola albida complex including three very closely related taxa was recognized as independent group within subsect. Patellares in parsimony tree. This result suggested that they should be treated as a taxa in series Pinnatae. Phylogenetic position of a putative hybrid species, Viola woosanensis was not supported with previous morphological hypothesis.

  • Recently, in several regions of Korea, we discovered the occurrence of Sagina micropetala Rauschert and S. procumbens L. All the plants of the genus Sagina growing in Korea are 5-merous and belong to the sect. Spergella. However, the two taxa we discovered belong to the sect. Saginella and are 4-merous. In addition, morphological characteristics such as sepals during the fruiting season and seeds were significantly different from those of species of the genus Sagina that occur in Korea. To clearly identify these species, we compared and observed their primary morphological characteristics using illustrated books and literature. Morphologically, S. micropetala exhibits the following traits: pubescence along the leaf margin, horizontally spreading sepals with red margins, and convex protrusions on the seed surface. In contrast, S. procumbens can be distinguished by its pedicels, which are curved during the flowering season and erect during the fruiting season. This plant also has broadly ovate sepals with white or yellowish-green margins, and flat seed surface protrusions. In terms of molecular analyses, 19 samples of Sagina were divided into two clades (S. micropetala and S. procumbens), confirming the taxonomic identity of the Sagina spp.

  • This study investigated the environmental factors and conducted a RAPD analysis for a better understanding of the environmental characteristics and regional genetic variation in samples from 18 different areas of Viola diamantiaca. The habitats are mostly located on the slopes of mountains facing north at an altitude ranging from 614 m to 1,462 m above sea level with angles of inclination ranging from 3 degrees to 30 degrees. A total of 268 vascular plant taxa are identified in 35 quadrates of 18 habitats. The importance value of V. diamantiaca is 11.58%, and four highly ranked species, Sasa borealis (5.61%), Meehania urticifolia (5.21%), Ainsliaea acerifolia (3.62%), Pseudostellaria palibiniana (3.60%) are considered to have an affinity with V. diamantiaca in their habitats. The degree of their average species diversity is 1.36, while this metric for their evenness and dominance are 0.89 and 0.07, respectively. The average field capacity of the soil is 25.99%, with organic matter at 17.47%, and the pH is 5.19. The soil texture was confirmed as sandy loam of eleven and loam of seven. The result of the RAPD analysis, among 78 bands amplified with a primer, 64 (84.6%) showed polymorphism. Eighteen populations could be classified into five groups with similarity coefficient values ranging from 0.53 to 0.86. The Mt. Jiri population, which is geographically segregated, shows basal branching within the 18 populations. Five populations, including two in the southern district in Gangwon-do and three in Chungcheongbuk-do, form a distinct clade. Four populations in the central district of Gangwon-do and Mt. Bohyeon in the Gyeongsangbuk-do clade form a sister to the clade containing two populations in Gyeonggi-do and five populations of the northern district in Gangwon-do. The Mt. Gariwang population is placed between the southern district and the central district in the Gangwon-do clades.

  • The in vitro organogenesis is one of important issues in plant embryology, and somaclonal variations are existing in calli and/or regenerants induced from a process of the organogenesis with in vitro circumstances. In this study, expressions of organogenesis-related genes were evaluated and genetic stability of regenerants derived from the process of in vitro organogenesis were measured using ISSR markers in Imperata cylindrica 'Rubra', Poaceae. The expressions of organogenesis-related genes were detected all of regenerants at the process of the organogenesis. All ISSR markers produced with an average of 71 bands per in vitro-cultured regenerants, and the scorable bands were varied from two to eight with an average of 5.14 bands per a primer. The polymorphism rates of the in vitro regenerants were higher than that of mother plants (1.4%), showing 4.1% (pot-cultured regenerants), 4.3% (field-cultured regenerants), 4.2% (in vitro-cultured regenerants), 5.6% (calli with green shoots) and 1.4% (calli), respectively. The genetic similarity matrix (GSM) among all accessions ranged from 0.747 to 1.0 with a mean of 0.868. GSM of the regenerants showed differences (from 0.972 to 1.00) compared with that of mother plants (0.991). According to the clustering analysis, two independent groups were divided into; the one is mother plants and regenerants cultured at room and open field, the other is regenerants cultured in vitro. The results give a new insight for understanding the dynamics of organogenesis in monocot plant.
