Biodiversity Research for Baekdudaegan Conservation (백두대간 보전을 위한 생물다양성 연구)

  • 백두대간은 백두산에서 시작된 큰 산줄기로 금강산, 설악산, 덕유산 등을 거쳐 지리산까지 이어지는 우리나라 중심 산줄기이다. 백두대간은 우리 민족 고유의 사회 문화적 가치는 물론 자연 생태적 측면에서 매우 중요한 지역이다. 백두산에서 지리산까지 약 1,400km, 남한 지역은 약 700km가 남북으로 길게 연결되어 우리나라 주요 산들을 연결하는 한반도 중심축을 형성하고 있다. 백두대간은 생물다양성의 보고이자 주요강의 발원지로 중요한 역할을 하고 있다. 이러한 주요 지역에 위치함 국립백두대간수목원은 수목원 정원의 조성 및 진흥에 관한 법률에 근거하여 기후대 및 식생권역별 국가수목원의 확충 계획에 따라 온대 북부 권역에 조성되었다. 백두대간 산림생물 다양성 유지와 산림생물자원의 보전 및 활용을 위한 다양한 연구를 수행하고 있다. 국립백두대간수목원의 시드볼트는 생물다양성 보존과 산림생물유전자원의 지속가능한 활용을 위하여 종자보존 협력 네트워크를 구축 하고 있다. 또한 아시아를 포함한 전세계 야생 종자를 수집하기 위한 기후대 생태권역별 종합계획 수립과 표준화 모델 개발을 통해 체계적인 협력체를 구축 중이다. 종자의 안정적인 장기 보존을 위한 형태학적, 분자생물학적 연구를 통해 종자의 저장특성 연구 및 수명연구를 통해 예측 모델을 연구하며 이를 기반으로 생태계 복원을 위한 종자 활용 연구를 수행하고 있다. 국립백두대간수목원은 백두대간 생물종의 조사, 생태계 보전 복원연구를 위한 현지 조사, 서식지외 보전 기관 역할, 유용자원 발굴 및 연구 등 백두대간 생물다양성 보전을 위한 다양한 역할을 하고 있다.

Draft genome sequence of lytic bacteriophage KP1 infecting bacterial pathogen Klebsiella pneumoniae (병원균 Klebsiella pneumoniae를 감염시키는 용균 박테리오파지 KP1의 유전체 염기서열 초안)

  • Klebsiella pneumoniae is a Gram-negative, rod-shape bacterium causing disease in human and animal lungs. K. pneumoniae has been often found to gain antimicrobial resistance, thus it has been difficult to treat K. pneumoniae infection with antibiotics. For such infection, bacteriophage can provide an alternative approach for pathogenic bacterial infection with antimicrobial resistance, because of its sensitivity and specificity to the host bacteria. Bacteriophage KP1 was isolated in sewage and showed specific infectivity to K. pneumoniae. Here, we report the draft genome sequence of Klebsiella pneumoniae phage KP1. The draft genome of KP1 is 167,989 bp long, and the G + C content is 39.6%. The genome has 295 predicted ORFs and 14 tRNA genes. In addition, it encodes various enzymes which involve in lysis of the host cell such as lysozyme and holin.

  • Parkinson's Disease (PD) is one of the three major old-age onset neurodegenerative diseases, its incidence rate is increasing worldwide as being an aging society. The number of PD patients has increased from 3 million in 1990 to 6.2 million in 2015 and is expected to increase to 12.4 million by 2040. Although many therapeutic candidates have been under development, but not yet been suggested the therapeutics of PD. For analyzing the trends and the status of convergence of technologies in the PD therapeutics, we classified into six sub-categories using global patent information and analyzed the level of technical competitiveness and technology convergence according to year, country, applicant, and technical description. These results can be used as a fundamental understanding for the current technical trend and the status of convergence of PD therapeutics, and establish the direction and strategy of R&D.

  • Software project planning includes several processes for estimating the effort required to complete software project tasks, allocating human resources to tasks, and creating a project plan. Because software planning is becoming more complicated as the size of software projects grow, it is difficult for project managers to decide on a reasonable project plan. To help them, many automatic software project planning approaches have been proposed. The approaches all focus on minimizing project duration. But if the plan is simply to minimize the duration, without considering software quality, the plan can harm the eventual software quality. In our research to create a reasonable project plan, we consider software quality as well as duration of the project, by defining a software quality score. The project manager can plan the project to maximize software quality for a specific project duration.

  • 대부분의 대규모 배수지는 고지대에 위치함으로써 자연유하를 통해 배, 급수지역으로 용수를 공급한다. 이를 위해 배수지 전단에는 가압장이 위치하여 정수장에서 처리된 용수를 고지대에 위치한 배수지로 송수한다. 이때 가압장에서 발생하는 전력소비량이 매우 높은 것으로 알려져 있으며, 효율적인 펌프운영을 통해 상당한 전력비용 절감이 가능할 것으로 판단된다. 일반적인 가압장의 운영은 시스템 운영자의 경험을 토대로 해당 가압장에 연결된 배수지의 수위에 따라 펌프의 작동여부를 결정하는 방법이 주로 사용되고 있다. 이러한 운영방법은 용수공급의 안전성을 우선시함으로써 배수지의 수위를 일정하게 유지하고자 빈번하게 펌프를 작동하게 되고 따라서 가압장에서 소모되는 전력량이 커서 운영효율 측면에서는 바람직하지 않다고 할 수 있다. 또한 빈번한 펌프의 작동으로 인해 펌프의 수명이 단축될 뿐만 아니라, 배수지내 용수의 수질저하 문제도 발생할 수 있다. 본 연구에서는 효율적인 펌프장 운영을 위해 급수지역의 24시간 용수사용량을 예측하고, 그에 따른 펌프장의 가압 유량 및 양정을 파악하여 적정용량의 펌프를 선정하고 운영함으로써 펌프의 운영비용의 최소화 및 안정적인 용수공급을 동시에 달성하고자 한다. 이를 위해, 실시간 최적화 모형을 개발하였다. 개발된 최적화 모형은 상수관망해석 프로그램(EPAENT)을 연계하여 수요절점의 수압조건 및 운영상황을 모의하였다. 최적화 기법으로는 유전자알고리즘을 사용하였으며, 실제 시스템의 운영상황를 반영하기 위한 다양한 제약조건(operational constraints)을 적용하였다. 개발된 모형은 정속펌프(혹은 On/Off 펌프) 뿐만 아니라, 최근 실무에서 널리 사용되고 있는 변속펌프(variable speed pump)를 추가적으로 고려하였다. 개발된 모형은 국내에서 실제 운영되고 있는 송, 배수 시스템에 적용하여 모형의 실무 적용가능성을 검증하였다.

Investigation on the effect of water extracts of Mangifera indica leaves on the hair loss-related genes in human dermal papilla cells (망고 잎 열수 추출물의 모유두 세포에서 탈모 관련 유전자 발현에 미치는 영향 연구)

  • Objectives : Mangifera indica leaves are well known for having a variety of benefits, including anti-inflammatory, anti-tumor, diabetic retinopathy and diabetic vasculosis. However, the effects of Mangifera indica leaves on hair loss inhibition have not been studied. In this study, we investigated to find out the activity of Mangifera indica leaves on hair loss. Methods : 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid(ABTS) analysis was performed to confirm the antioxidant efficacy of the water extract of Mangifera indica leaves (WEML). To examine the effect of WEML on cell viability in dermal papillar (DP) cells, 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetra Zolium (MTS) analysis was performed. The changes in the mRNA expression level of the hair loss and hair growth-related genes in dermal papilla cells by WEML treatment were confirmed by quantitative RT-PCR. Results : In dermal papilla (DP) cells, ABTS analysis and MTS analysis of WEML showed antioxidant efficacy and low cytotoxicity. As a result of gene expression analysis through Quantitative RT-PCR, no changes in hair growth-related genes BMP6 and CTNNB1 was confirmed. but inhibitory activity of WEML on hair loss-related genes EGR1, SGK, DKK1, SRD5A1 and SRD5A2 was confirmed. Conclusion : We confirmed that WEML has excellent antioxidant efficacy and a inhibitory activity of hair loss-related genes including 5α-reductase genes. These results suggest that Mangifera indica leaves have a potential activity as a hair loss treatment for hair loss and hair growth. Biochemical or molecular biological research on hair loss is needed.

Investigation of the effect of Hibiscus sabdariffa L. extracts on tight-junction related genes in human keratinocyte HaCaT cells (히비스커스 추출물이 인간 각질 형성 세포의 밀착 연접 관련 유전자 발현에 미치는 영향 연구)

  • Objectives : Hibiscus (Hibiscus sabdariffa L.) is rich in antioxidants such as flavonoids and anthocyanins and is known to have anti-inflammatory activity and anti-aging function of the skin, but there is no study on its effect on the skin barrier. This study aim to investigate the positive effect on the skin barrier by confirming the effect of water extracts of Hibiscus sabdariffa L. (WEHS) on the tight junction-related gene expression. Methods : The antioxidant efficacy of WEHS was investigated through ABTS and DPPH assays, and 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium was performed to examine the effect on cell viability. quantitative Reverse transcription polymerase chain reaction and immunoblot analysis were performed to confirm the effect of WEHS on the expression of tight junction-related genes, and immunofluorescence microscopy was used to confirm the movement of Claudin 1 protein into tight junctions. Results : WEHS showed strong antioxidant activity and induced an increase in both mRNA and protein expression levels of Claudin 1 among tight junction-related genes. The strong localization of Claudine 1 protein increased by WEHS to the tight junction was confirmed by immunofluorescence microscopy. Conclusions : Hibiscus was confirmed through this study to show antioxidant activity and the function of promoting the expression of the tight junction Claudin 1 gene, suggesting that Hibiscus can be used as a material for the prevention and treatment of skin moisturizing and atopy, which have an important influence on tight junction.

Prediction of Drug Side Effects Based on Drug-Related Information (약물 관련 정보를 이용한 약물 부작용 예측)

  • Side effects of drugs mean harmful and unintended effects resulting from drugs used to prevent, diagnose, or treat diseases. These side effects can lead to patients' death and are the main causes of drug developmental failures. Thus, various methods have been tried to identify side effects. These can be divided into biological and systems biology approaches. In this study, we use systems biology approach and focus on using various phenotypic information in addition to the chemical structure and target proteins. First, we collect datasets that are used in this study, and calculate similarities individually. Second, we generate a set of features using the similarities for each drug-side effect pair. Finally, we confirm the results by AUC(Area Under the ROC Curve), and showed the significance of this study through a comparison experiment.

  • Food safety accidents have been increasing by 2% over 5,000 cases every year since 2009. Most people know that the best method to prevent food safety accidents is a quick inspection, but there is a lack of inspection technology that can be used at the non-analytic level to food production and distribution sites. Among the recent on-site diagnostic technologies, the methods for testing gene-based food poisoning bacteria were introduced with the STA technology, which can range from sample to detection. If food safety information can be generated without forgery by directly inspecting food hazard factors by remote, unmanned, not human, pollution sources can be managed by predicting risks more accurately from current big-data and artificial intelligence technology. Since this information processing can be used on smartphones using the current cloud technology, it is judged that it can be used for food safety to small food businesses or catering services.

Generating Pairwise Comparison Set for Crowed Sourcing based Deep Learning (크라우드 소싱 기반 딥러닝 선호 학습을 위한 쌍체 비교 셋 생성)

  • With the development of deep learning technology, various research and development are underway to estimate preference rankings through learning, and it is used in various fields such as web search, gene classification, recommendation system, and image search. Approximation algorithms are used to estimate deep learning-based preference ranking, which builds more than k comparison sets on all comparison targets to ensure proper accuracy, and how to build comparison sets affects learning. In this paper, we propose a k-disjoint comparison set generation algorithm and a k-chain comparison set generation algorithm, a novel algorithm for generating paired comparison sets for crowd-sourcing-based deep learning affinity measurements. In particular, the experiment confirmed that the k-chaining algorithm, like the conventional circular generation algorithm, also has a random nature that can support stable preference evaluation while ensuring connectivity between data.
