Environmental Monitoring of Herbicide Tolerant Genetically Modified Zoysiagrass (Zoysia japonica) around Confined Field Trials (제초제저항성 유전자변형 들잔디의 시험 격리포장 주변 환경방출 모니터링)

  • The cultivation area and use of genetically modified (GM) crops have been increased continuously over the world. Seed distribution and transgenes to environmental ecosystem is one of the most important factors in risk assessment and risk management of GM crop. Safe management for the development and commercialization of GM crops is required according to The Act on Transboundary Movements of Living Modified Organisms,etc (LMO Act) in Korea. This study was conducted to setup the environmental monitoring system of GM zoysiagrass (event JG21 and JG21-MS). The monitoring was performed in 4 GMO confined fields, Sungwhan, Ochang, Jeju University and Jeju Namwon. In the result of monitoring, we could not found any gene flow and distribution of GM zoysiagrass in the 3 fields, but one spill of JG21 was found in the Namwon field in 2012. These results suggest that continuous monitoring is necessary to detect the occurrence of GM zoysiagrass for preventing genetic contamination in natural environment.

Development of detection methods for six approved LM crops in Korea (신규 수입 승인 6개 유전자변형작물의 검출기법 개발)

  • Living modified crops are genetically modified living organisms and are widely used in biotechnical research and desired goods. As the reliance on LM products, concerns about safety of LMOs have been continuously increased in South Korea. We established the detection methods for unintentional released LMOs in environmental conditions. To detect six LM event genes of 1 canola, 1 maize and 4 soybeans, PCR conditions were based upon consideration of the Joint Research Centre information. Genomic DNAs were isolated from LM samples and PCR analysis were performed using each event-specific primer pair. Event-specific genes of all events were efficiently recognized by our methods. To investigate the insertion site of LM genes in each genome, we verified PCR product sequence by DNA sequencing. These results suggest that the LM event-specific gene amplification can be efficiently developed. In addition, our detection method is fit for monitoring and post-management of LM crops in the environment.

Current status of comparative compositional analysis for GM crop biosafety assessment (유전자변형작물 안전성평가를 위한 영양성분 비교연구 동향)

  • Approvals for cultivation and import of genetically modified (GM) crops have dramatically increased around the world. Comparative compositional studies are an important aspect of safety assessments of products from GM crops and are based on substantial equivalence. Compositional analyses focus on determining similarities and differences between the compositions of the GM crops and their conventional counterparts, and thereby assessing the compositional equivalence of GM crops and their conventional comparators. The analytes, such as major constituents, key nutrients, and antinutrients, are generally determined on a crop-specific basis according to the OECD consensus document. The use of standard methods throughout the processes, such as selection of comparators, field trials, analytical methods, and statistical data analysis, is crucial. In this study, we showed the general framework of compositional studies. Literature for compositional studies of GM crops conducted abroad and in Korea was reviewed to obtain information about analytes, conventional counterparts, cultivation year, location, and statistical methods. The studies conducted abroad assessed for commercial release of GM crops such as soybean, maize, and cotton, while domestic studies were mainly performed for research in rice. In addition, we suggested a guidance for conventional comparators and field trials applicable to the domestic situation.

Primer for the Potato Specific Internal Control DNA and Screening Method for the Genetically Modified Potatoes by Competitive Duplex-PCR (감자 특이 Internal Control DNA 증폭용 Primer와 이를 이용한 유전자 변형 감자의 경쟁적 이중 PCR 검정법)

  • We report the new method for the screening of genetically modified potato by competitive duplex-PCR using the potato specific single oligomer primer for the internal control and CaMV 35S promoter or NOS terminator specific primers. The single oligomer primer (rAGU4A) amplify the potato specific internal control band from the homozygous potato genomic DNA in the RAPD profiles of all analyzed potato varieties. The 530 bp internal control DNA was amplified independently to CaMV 35S promoter or NOS terminator DNA and identified as repetitive or microsatellite DNA of potato (AF541972). With this new technique, the transgenic potatoes which were transformed with vectors contained the different foreign genes are analyzed. In case of the commercialized transgenic potato varieties, 'Hew Leafs', those two genetic factors are used for promoter and terminator respectively So, this new PCR technique should be a promising method of cost effective and accurate screening for the commercialized GM potatoes on market.

Assessment of gene flow from insect-resistant genetically modified rice (Agb0101) to non-GM rice (해충저항성 유전자변형 벼(Agb0101) 유전자 이동성 평가)

  • Genetically modified (GM) crops have been developed worldwide through the recombinant DNA technology and commercialized by global agricultural companies. Until now, GM crops have not been cultivated commercially in Korea. Commercialization of GM crops requires a compulsory assessment of environmental risk associated with the release of GM crops. This study was conducted to evaluate the frequency of pollen mediated gene flow from Bt transgenic rice (Agb0101) to japonica non-GM rice (Nakdongbyeo), indica non-GM rice (IR36), and weedy rice (R55). A total of 729,917, 596,318 and 230,635 seeds were collected from Nakdongbyeo, IR36, and R55, respectively, which were planted around Agb0101. Selection of the hybrids was determined by repeated spraying of herbicide and Cry1Ac1 immunostrip assay. Finally, the hybrids were confirmed by PCR analysis using specific primer. The hybrids were found in all non-GM rice and out-crossing ranged from 0.0005% at IR36 to 0.0027% at Nakdongbyeo. All of hybrids were located within 1.2 m distance from the Agb0101 rice plot. The meteorological elements including rainfall and temperature during rice flowering time were found to be important factors to determine rice out-crossing rate. Consideration should be taken for many factors like the meteorological elements of field and physiological condition of crop to set up the safety management guideline to prevention of GM crops gene flow.

Assessment of Microbial Community in Paddy Soils Cultivated with Bt and Nakdong Rice (Bt 벼의 토양미생물상 영향 비교평가)

  • The cultivation of genetically modified (GM) crops has increased due to their economic and agronomic advantages. Before commercialization of GM crops, however, we must assess the potential risks of GM crops on human health and environment. The aim of this study was to investigate the possible impact of Bt rice on the soil microbial community. Microbial communities were isolated from the rhizosphere soil cultivated with Bt rice and Nakdong, parental cultivar and were subjected to be analyzed using both culture-dependent and molecular methods. The total counts of bacteria, fungi, and actinomycetes in the rhizosphere of transgenic and conventional rice were not significantly different. Denaturing gradient gel electrophoresis (DGGE) analysis of PCR-amplified 16S rRNA genes revealed that the bacterial community structures during cultural periods were very similar each other. Analysis of dominant isolates in the rhizosphere cultivated with Bt and Nakdong rice showed that the dominant isolates from the soil of Bt rice and Nakdong belonged to the Proteobacteria, Cloroflexi, Actinobacteria, Firmicutes, and Acidobacteria. These results indicate that the Bt rice has no significant impact on the soil microbial communities during cultivation period. Further study remains to be investigated whether the residue of Bt rice effect on the soil environment.
