Gas Liquid Chromatographic Studies on Sugars and Organic Acids in Different Portions of Hot Pepper Fruit (Capsicum annuum L.) (Gas Liquid Chromatography에 의한 고추 부위별(部位別) 당(糖) 및 유기산(有機酸) 조성(組成)에 관한 연구(硏究))

  • Sugars and organic acids in Korean hot pepper fruit (C, annuum L.) according to its portions, pericarp, seeds and placenta, were determined by gas liquid chromatography (GLC). ${\alpha}-\;and\;{\beta}-fructoses,\;{\alpha}-\;and\;{\beta}-glucoses$, galactose, sucrose and maltose were identified. Major sugar in pericarp was fructose, while those in seeds and placenta were surose and fructose. Amount of free sugars in seeds was higher than that in pericarp or placenta. Succinic, fumaric, malic, tartaric, citric and quinic acids were found. Citric acid was a major organic acid found in pericarp and placenta. The amount of each identified organic acid, however, was similar in seeds. Quantity of total organic acids identified in pericarp or placenta was higher than that in seeds.

Evaluation of Nutritional Characteristics of Platycodon grandiflorum Seeds (도라지 종자의 영양학적 특성 평가)

  • In this study, proximate composition, crude fiber, reducing sugar, free sugars, organic acids, minerals and amino acids of Platycodon grandiflorum seeds were analyzed to evaluate its nutritional value. Moisture, crude protein, crude fat, crude ash and carbohydrate contents of seeds were 6.97, 26.05, 27.46, 3.78 and 35.74%, respectively. Crude fiber of 6.31% and reducing sugars of 1.54% were also determined. Sucrose(1,661 mg/100 g) and lactic acid(1,224 mg/100 g) were most abundant free sugar and organic acid, respectively. Both phosphorus and potassium were main minerals that contained more than 700 mg in 100 g seeds. Amino acids analysis of 100 g seeds showed that glutamic acid(3.45 g), arginine(2.51 g), aspartic acid(1.66 g), leucine(1.29 g), lysine(1.10 g), alanine(1.05 g) and glycine(1.04 g) were abundantly contained in order, while others were less than 1 g.

Evaluation of Hot Water Treatment for Disinfection of Vegetable Seeds for Organic Farming (채소 종자별 온탕침지 종자소독 효과검정)

  • Hot water treatment that is the most appropriate seed disinfection method for organic vegetable farming was evaluated in this study. Among the leafy vegetable seeds lettuce that was the most sensitive to hot water was suitable to treat at $45^{\circ}C$ for 25 min, while Chinese cabbage and radish seeds were optimally treated at $50^{\circ}C$ for 25 min. The treatments resulted in similar or higher seed germination rate than non-treated seeds and promoted plant growth. In addition, fungi such as Alternaria, Aspergillus, Penicillium, or Mucor grown on the seeds were suppressed over 90% and the bacterial growth on lettuce seeds reduced 98.5% by the treatment. Among the fruit vegetable seeds pumpkin that was vulnerable to hot water was suitable to treat at $50^{\circ}C$ for 15 min, while cucumber and hot pepper seeds revealed optimum treatment at $50^{\circ}C$ for 25 min as chinese cabbage and radish. The treatment also showed similar or higher seed germination rate and growth than non-treated seeds. Furthermore, fungi such as Rhizopus, Aspergillus, Penicillium or Mucor grown on the seeds reduced from 72.0% to 95.4%. The bacterial growth on cucumber and red pepper seeds was suppressed from 65.5% to 86.0% by the treatment. Results indicated that the hot water treatment is practical for disinfection of organic vegetable seeds and the optimum temperature and soaking time varied among the seeds.

Detection of Seed Born Fungi from the Imported Seed and Effect of Fungicide for Them (수입종자에서의 병원균분리와 약제처리효과)

  • Twenty-five species of fungi were deleted and identified from 27 kinds of the intercepted seeds from 5 countries. Eight species of fungi including Fusariun moniliforme were detected from the imported calabash seed (Lagenaria siceraria). These seeds were the most contaminated among the intercepted reeds tested. Six kinds of seeds including larch(Larix kaemferi) were disinfected with Vitathiram, Benlate-T and Captan. Rhynchosporium was detected from seeds treated with Vitathiram whereas Alternaria spp. were detected from seeds treated with Benlate T or Captan. Six genera of fungi were detected from the untreated seeds. In the case of germination, it seem that seed disinfectants do not have any damage on germination of seed.

Status of Pepper Organic Farming and Collection of Pepper Germplasm

  • 고추 유기농재배에 적합한 고추 유전자원을 선발하기 위한 기초자료를 확보하기 위하여 우리나라 고추 유기농가에 대한 현황 분석을 실시하였고 국내외 고추 재래종 유전자원을 수집하였다. 고추 161개의 유기농가에 대한 설문조사 결과 총 재배면적은 64ha 였고 이중 노지재배 38ha, 비닐하우스재배 26ha 였다. 유기농 재배경력은 10년 이하가 124농가(76.9%)로 가장 많았고 11~15년, 16~20년, 20년 이상이 각각 19농가(11.8%), 12농가(7.5%), 6농가(3.8%)였다. 유기농 재배에 사용하는 고추 종자는 157농가(97.6%)가 종묘회사에서 시판되는 F1품종을 이용하고 있었고 종묘회사에서 육성되는 품종을 이용하는 주된 이유는 유기종자의 자가채종이 어렵고(43.4%) 품질이 낮으며(34.6%) 병에 약해서 (9.9%) 기타(12.1%) 순이었다. 품종 선택시 최우선으로 고려하는 사항은 품질(51농가, 31.7%)과 수량 (47농가, 28.2)이 병해 저항성보다(45농가, 28.0%) 높은 수준을 차지하였다. 2008년과 2009년 2년간 국내 재래종을 탐색?수집하여 국내에서 수비초 등 9자원을 수집하였고 우즈베키스탄에서 재래종 및 고정종 자원 19자원을 수집하였다. 국내에서 수집한 재래종 자원들은 현재 몇 농가에서만 재배가 되고 있으며 이들 자원들은 국내 환경에 오랫동안 적응되어 유기농 재배에 적합할 것으로 사료 되었고 우즈벡키스탄에서 수집된 자원들은 단고추가 4자원, 매운고추가 15자원으로 이 중 단고추 1자원은 과중이 200g 이상인 대과 자원이었고 이들 자원 또한 고정종으로 유기농재배에 적합할 것으로 사료되었다.

  • Diseases were surveyed in 5 Rice paddy field areas of Organic paddy field and conventional paddy field. The 3 major diseases, rice leaf blast, bacterial leaf blight and sheath blight in rice were surveyed at duck raising, rice bran and conventional rice paddy field. They were serious in duck raising paddy field, rice bran paddy field more than conventional paddy field. The Ilpum variety were infected seriously more than Chucheong. At the effectivity test of the environment-friendly substance for the rice-seed sterilization, 1000-times diluted brown-rice vinegar showed controlling effect against Bakanae disease, and germination rate also was good. There was no Weeds Control efficacy by cultivating of hairy vetch in rice paddy field. However, extract of hairy vetch Leaf and root repressed the germinating of lettuce seed.

  • Effect of various physical and chemical treatments on dark germination of Oenothera lamarckiana seeds were primarily investigated to find out factors affecting germination. Germination of seeds which did not germinate in the constant temperature and darkness was induced by several physical treatments such as sonification, wetting and drying, freezing and thawing, and removal of seed coat. Pretreatment of chilling ($4^{\circ}C$), freezing ($-10^{\circ}C$) and incubation at high temperature ($80^{\circ}C$) induced dark germination of seeds which did not germinate in the constant temperature ranges of 15 to $40^{\circ}C$ under darkness. Alternating temperature also had a stimulatory effect on dark germination of Oenothera lamarckiana seeds. Sensing of seeds to alternating temperature appeared to be completed during the first two days after imbibition. The minimum difference of temperature required for dark germination was $5^{\circ}C$ in the range of $15-25^{\circ}C$. A thiourea (1.0%) treatment induced dark germination, but GA, IAA, BA and Ethrel failed to do so.

Germination Induction of Some Weed Seeds Stored Under the Dry and Low Temperature Condition (건조저온저장(乾燥低溫貯藏)한 몇가지 잡초종자(雜草種子)의 발아유기(發芽誘起))

  • These experiments were conducted to recognize the germination change in relation to storage period, using 25 kinds of field-collected weed seeds stored under the dry and low temperature ($4{\pm}1^{\circ}C$), and to establish a simple germination induction methods in some kinds of them. No or weak dormancy was observed in Lepidium virginicum, Rumex japonicus, Cirsium japonicum var. ussuriense, Aeschnomene indica and Ludwigia prostrate. The germination of Digltaria sanguinalis, Portulaca oleracea, Amaranthus spp., Echinochloa crux-galli, Polygonum persicaria, Cyperus iria, and Aeschynnmene indica was enhanced by $H_2SO_4$ treatment for 20, 2, 2, 50, 40, 3 or 5 minutes, respectively. Favorable germination in Polygonum persicaria and Echinochloa crus-galli was obtained by soaking in water for 25 days and 50 days, respectively. Moisture treatment for 21 days and 14 days caused the enhancement of germination in Cyperus difformis and Aneilema keisak, respectively. Combination of various treatments in Cyperus iria was most effective when the seeds treated with $H_2SO_4$ for 3 minutes incubated in alternating temp. after soaking in 0.2% $KNO_3$, but their effect on Digitaria sanguinnlis was little.

Screening of Seed Treatment Agents against Leaf Blight and Black Root Rot for Carrot Organic Cultivation (당근 유기재배를 위한 검은잎마름병과 검은무늬병 종자소독제 선발)

  • This study was conducted to control the seed borne pathogens (Alternaria spp.) of carrot and to examine the seed germination rate by using 13 environment friendly agricultural materials instead of conventional chemicals for organic cultivation. The growth inhibiting effects on pathogens showed the different responses according to each agricultural material and effective 7 materials against seed borne pathogens were selected. Among 7 materials, the carrot seeds sterilized with plant extracts, Tanger Stop and Land Saver were not germinated at all. The germination rate of seeds sterilized with other materials showed the similar levels with reference chemical (Benlate-T) and non-treated seeds. Infection rates of seeds sterilized with seaweed extract, Bellopper for controlling A. radicina and plant extract, Ssial-100 for A. dauci were similar or low infection rate compared with reference chemical. The germination rate and root length of seeds sterilized with Bellopper and Ssial-100 was similar or superior to those of chemical sterilized seeds. The optimal condition seemed to be a little bit different depending on the concentration of materials. As a result, the sterilization of carrot seeds by using the environment friendly materials could be effectively utilized as a technology to inhibit the infection of seed-borne pathogens.

Differences of Electrical Conductivity, Organic and Inorganic Constituents in Leakage from Aged and Non-aged Vegetable Seeds (채소 퇴화종자와 건전종자 침지용액의 전기전도도, 유기및 무기성분의 차이)

  • The leakage characteristics of electrical conductivity, inorganic constituents-K, Ca, Mg and Na, total sugar and total amino acid from the exudates of some vegetable seeds of viable and non-viable(artificially aged) were quantified to get basic information about the detection of the non-viable seeds. The crops studied were radish, cabbage, broccoli, onion and carrot. The time course electrolyte leakage was different from viable and non-viable seed of cruciferae but not sensitive in onion and carrot seed In inorganic constituents, potassium leakage was the greatest amount and difference between viable and non-viable seeds, but Ca, Mg and Na leakages were not as much differences as potassium. Total sugar as glucose and total amino acid as glycin leaked a lot more in aged radish, cabbage and broccoli seed than non-aged seed and the large differences were appeared after 4 hour imbibition. As a results, in general the leakages from the aged seeds were greater than from the non-aged seeds in most components tested but they were varied depending on species or varieties and components.
