Method Validation for Monitoring of Agricultural Worker Exposure to Insecticide Fenthion (살충제 Fenthion에 대한 농작업자 노출 측정을 위한 분석/시험방법 검증)

  • Exposure measurement of agricultural worker to pesticide is one of important part of health risk assessment of pesticide. Therefore exposure matrices, apparatus, instruments and methods must be validated in advance to field experiment. In this study, method validation with an organophosphorus insecticide fenthion was carried out for exposure monitoring of agricultural worker. LOD and LOQ were 0.01 and 0.05 ng, respectively. Calibration curve linearity ($R^2$ > 0.999) and reproducibility (C.V. < 3%) were also excellent. Recovery at LOQ, 10LOQ and 100LOQ levels from gloves, socks, mask, patch, solid sorbent, glass fiber filter was 76~113% (C.V. < 3%). Trapping efficiency was 95~105% while no breakthrough was observed. Method validation for the exposure monitoring was established successfully through several experiments. Such method validation can be usually performed in laboratory and not much different for each pesticide so that, this techniques will be applied widely in research for pesticide exposure monitoring by combination with body surface area and respiration rates.

Heavy Metals Contens and Organochlorine Pestieide Residues in Commercial Salted and Fermented Sea Foods (시판 젓갈중의 중금속 및 유기염소 잔유농약의 함량)

  • Heavy metal contents and residues level of organochlorine pesticides were analyzed for commercial salted and fermented sea-food which bought from pusan area. Heavy metal contents ranged from 0.11 to 0.46 ppm of mercury, trace to 1.1 ppm of lead, 0.01 to 0.34 ppm of cadmium, 0.4 to 5.98 ppm of cupper, 0.16 to 4.5 ppm of zinc, trace to 0.49 ppm of manganese and trace to 1.35 ppm of arsenic. Residue level of organochlorine pesticides in salted and fermented sea-foods ranged from 0.3 ppb to 1.825 ppb of total BHC Organochlorine pesticides of DDE, dieldrine, pp'-DDD and pp'-DDT were detected a small amount from Ge jeot and Myeol-Chi jeot. In conclusion. the heavy metal and levels of organochlorine pesticides residues were safety to eat the salted and fermented sea foods.

Physico-chemical stability of pesticide formulations under different storage conditions (저장조건별 농약제품의 물리.화학적 안정성)

  • To confirm physico-chemical stability of formulated products, this study was conducted under different storage conditions; room temperature and accelerated temperature of $54^{\circ}C$. The tested pesticide formulations were dichlorvos 50% EC, acephate 50% WP, hymexazol 4% DP, thiram 80% WP and isoprothiolane 12% GR. The selected formulations were stored for 10 weeks and 5 years under the given temperature in maximum and contents of active ingredients were also analyzed by GLC or HPLC after each time of storage. The degradation rates of 5 active ingredients under the two conditions showed a similar trend except acephate. Acephate was rapidly decomposed at $54^{\circ}C$ but slowly decomposed at room temperature, and the degradation rate under the accelerated condition was 2.4 to 5-fold higher than that under the room temperature. Consequently, the stability test on active ingredients in pesticide formulations was able to recommend to be carried out under the accelerated condition except acephate. And the physical properties of all formulations tested under two storage conditions were good.

Examination of Pesticide Poisoning Deaths Statistics in Korea and Precautionary Measures Against Pesticide-Poisoning (국내 농약중독 사망사고 현황분석과 사고 예방대책)

  • The data on pesticide poisoning death toll during 2000-2005 was obtained from Korean National Statistical Office and analysed to find out any precautionary measures against pesticide poisonings. Pesticide poisoning death toll showed slightly increasing trend with year up to about 3500 in 2005. Most of the pesticide poisoning death cases were intentional self-poisoning accounting for more than 80% of total pesticide poisoning death. The pesticide most often implicated in poisoning death were paraquat dichloride, a non-selective herbicide and organophosphate/carbamate insecticides. The regional distribution of pesticide poisoning death showed urban 21.6% and rural area 78.4%. As for the occupation of the dead, agricultural, forestry and fishery workers accounted for 27.4% and non-agricultural workers was 72.6% of pesticide poisoning death. Therefore, government need to take some precautionary measures to reduce the number of pesticide poisoning cases. It is strongly recommended that pesticide regulatory authorities encourage and train the agricultural workers to use safe storage boxes and enforce strict regulation for the prevention for the prevention of highly toxic pesticides sales to non-agricultural workers.

A Survey for Pesticide Residues in Major Rivers of Korea (주요하천수(主要河川水)에 대(對)한 농약잔류(農藥殘留) 실태조사(實態調査))

  • To obtain basic information for the safe use of pesticides, the aquatic environment of 5 major rivers was surveyed in April and August 1982 for organochlorine and organophosphorus pesticide residues. Regardless of sampling times and sites, pesticide residue levels were very low. Of the organophosphorus pesticides surveyed, only IBP and diazinon were detected in water samples while fenthion, fenitrothion, chlorpyriphos, chlorpyriphos-methyl, chlorfenvinphos, phenthoate, and edifenphos were not detected in any water samples. Residue levels and detection frequencies of IBP or diazinon were much higher in water samples collected in August than those collected in April. Detection frequencies of organochlorine pesticides, on the other hand, were high, but their actual residue levels were very low. BHC was detected nearly in all water samples while aldrin was not found in any samples. Detection frequencies of heptachlor, endosulfan, and o.p`-DDT were considerably high but dieldrin and p.p`-DDT were detected in a few water samples and their residue levels were also very low.

Removal Efficiency of Pesticide Residues on Apples by Ultrasonic Cleaner (초음파 세척기를 이용한 사과의 잔류농약 제거 효과)

  • In order to determine the removal efficiency of pesticide residues on apples by ultrasonic cleaner, apples(Fuji var.) were artificially treated by dipping in water solution of fenitrothion EC, chlorpyrifos EC and phenthoate EC. The treated apples were washed by the ultrasonic cleaner with water, 30% ethanol solution or 0.2% detergent solution, respectively. The removal efficiency of fenitrothion, chlorpyrifos and phent-hoate by ultrasonicated washing in water for 0.5min. was 39.2%, 32.0% and 50.4%, respectively, but there was a tendency to decrease the removal efficiency of the pesticide residues as the duration of ultrasonicated washing period is increased. The maximum removal efficiency of the pesticide residues washed in the detergent solution was observed by ultrasonicated washing the samples for 5min. and it was recorded as 33.5% ; fenitrothion, 30.1% ; chlorpyrifos and 48.3% ; phenthoate, respectively. However it's appeared that the longer the ultrasonicated washing period in 30% ethanol solution the more pesticides from the apples were removed and the maximum removal efficiency of the pesticides was recorded as 66.2%(fenitrothion ; washed for 15min.), 41.7%(chlorpyrifos ; washed for 10min.), 74.2%(phenthoate ; washed for 10min.).

Factors influencing the axes of anterior teeth during SWA on masse sliding retraction with orthodontic mini-implant anchorage: a finite element study (교정용 미니 임플랜트 고정원과 SWA on masse sliding retraction 시 전치부 치축 조절 요인에 관한 유한요소해석)

  • Objective: With development of the skeletal anchorage system, orthodontic mini-implant (OMI) assisted on masse sliding retraction has become part of general orthodontic treatment. But compared to the emphasis on successful anchorage preparation, the control of anterior teeth axis has not been emphasized enough. Methods: A 3-D finite element Base model of maxillary dental arch and a Lingual tipping model with lingually inclined anterior teeth were constructed. To evaluate factors influencing the axis of anterior teeth when OMI was used as anchorage, models were simulated with 2 mm or 5 mm retraction hooks and/or by the addition of 4 mm of compensating curve (CC) on the main archwire. The stress distribution on the roots and a 25000 times enlarged axis graph were evaluated. Results: Intrusive component of retraction force directed postero-superiorly from the 2 mm height hook did not reduce the lingual tipping of anterior teeth. When hook height was increased to 5 mm, lateral incisor showed crown-labial and root-lingual torque and uncontrolled tipping of the canine was increased.4 mm of CC added to the main archwire also induced crown-labial and root-lingual torque of the lateral incisor but uncontrolled tipping of the canine was decreased. Lingual tipping model showed very similar results compared with the Base model. Conclusion: The results of this study showed that height of the hook and compensating curve on the main archwire can influence the axis of anterior teeth. These data can be used as guidelines for clinical application.

Characteristics of Resistance to Chlorpyrifos in Diamondback-moth (Plutella xylostella L.) (Chlorpyrifos 저항성 배추좀나방(Plutella xylostella L.)의 살충제 저항성 특성)

  • To determine the mechanism of the resistance to organophosphorus insecticide, chlorpyrifos, in diamondback-moth (Plutella xylostella L.), activities of esterases, glutathione-S-transferase (GST) and AChE insensitivity which were known for causing factor of resistance were measured. Also, the relationship between AChE insensitivity and the resistant ratio was investigated to inquiry the cross-resistance. The resistant ratio of chlorpyrifos-resistant strain (CRS) of diamondback-moth at the 6th generation was developed 160 fold compared to susceptible strain (SS) one. Activity of GST that are extracted from CRS was 1.7-fold higher than that from SS. However, activity of total esterases from CRS was similar to that from SS. In AChE insensitivity test, CRS was 11.8-fold less sensitive than that from SS. CRS was ranged from 17.6 to 33.6-fold less sensitive than SS to other insecticides having same target site with chlorpyrifos such as dichlorvos, dimethylvinphos and carbofuran. Insensitivity of AChE to phenthoate-oxon, however, was 1.7-fold. Resistance of CRS was 82-fold, 47-fold and 42-fold higher than SS to dichlorvos, dimethylvinphos and carbofuran, respectively, but 2.3-fold to phenthoate and then we could identify that the resistance development of insecticide might have a lot of difference among the chemicals with the same target site. The relationship between the AChE insensitivity and the resistant ratio was significantly correlated$(r=0.9951^{**},\;p^{(0.01)}$. This result indicates that AChE insensitivity was associated with insecticide resistance. Overall, these results suggest that insensitivity of AChE was an important factors to chlorpyrifos resistance in diamondback-moth, and the slightly increased activity of GST may also have contributed to that.

Development of Analytical Method and Monitoring of Organophosphorus Pesticides in the Raw Water and Clean Water by Liquid Chromatography-Tandem Mass Spectrometry (LC/MS/MS를 이용한 유기인계 농약류의 최적 분석법 정립과 원·정수에서의 모니터링)

  • The analytical method for 16 organophosphorus pesticides was developed in this study. The 16 organophosphorus pesticides were analyzed by liquid chromatography-tandem mass spectrometry (LC/MS/MS) using on-line solid phase extraction (on-line SPE) with PLRP- S cartridge. Analysis of all analytes in the MS/MS was processed in the electrospray ioni-zation (ESI) positive mode. They are Azinphos ethyl, Chlorfenvinphos, Ethion, Famphur, Phosmet, Phosphamidon, Terbufos, Aspon, Chlorpyrifos-methyl, Crotoxyphos, Dichlofenthi-on, Dicrotophos, Fonofos, Thionazin, Dimethoate and Iprobenfos. Limits of detection (LODs) and Limits of quantification(LOQs) were obtained as 0.8~2.0 ng/L and 2.6~6.4 ng/L, respectively. All compounds were not detected at the 8 sampling points of the raw water and clean water.

Adsorption and Leaching of Organophosphorus Pesticides in the Soil of Cheju Island (제주도 토양에서 유기인계 농약의 흡착 및 용탈)

  • The adsorption and leaching of organophosphorus pesticides (phenthoate, diazinon, methidathion) were investigated in Namwon soli(black volcanic soil), Aewol soil(very dark brown volcanic soil) and Mureung soil(dark brown nonvolcanic soil) sampled in Cheju Island. The Freundlich constant, K value, was 52.4, 31.3 and 27.7 for phenthoate, diazinon and methidathion in Namwonsoil, respectively and decreased in the order of phenthoate, diazinon and methidathion among the pesticides. The K value of phenthoate was 52.4, 15.9 and 5.9 for Namwon, Aewol and Mureung soil, respectively and was the highest for Namwon soil with very high organic matter content and cation exchange capacity(CEC). The Freundlich constant, 1/n, showed a high correlation with organic matter content, i.e., its value was less than unity for organic matter rich soil(Namwon soil) and greater than unity for organic matter poor soil(Mureung soil). Total recoveries of pesticides in soil and leachate with leaching in soil column, were in the range of about 74~86%. The leaching of pesticides was less for phenthate with high K values, and more for methidathion with low K values among the pesticides. It was slower for Namwon soil with high K values, and more for methidathion with low K values among the pesticides. It was slower for Namwon soil with high K values, and faster for Mureung soil with low K values among the soils.
