월국환(越鞠丸) 메탄올 추출물이 산화적 간손상에 미치는 효과 (Effects of Wolgukwhan Methanol Extract on Oxidative Liver Injury)

  • Objectives: In traditional medicine, Wolgukwhan has been used for the treatment of digestive system disease, such as indigestion, brash, ructation, nausea and vomiting. This study was purposed to investigate the effects of Wolgukwhan methnol extract (WGWM) on oxidative liver cell injury. Methods: In vivo assay, we administerated acetaminophen(500mg/kg, i.p.) to starved mice 24hrs after pretreatment of WGWM for 6days. In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GPX), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results: In vivo administeration of WGWM showed effective inhibition of acetaminophen induced lipid peroxidation and elevations of glutathione level. The acetaminophen treatment resulted in a decrease of catalase, GPX and GST activities. By contrast, WGWM pretreatment increased compare to those of untreated groups. Conclusions: These results suggested that WGWM might protect against lipid peroxidation by free radicals, destruction of hepatic cell membranes.

월국환(越鞠丸) 물 추출물이 Acetaminophen으로 유도된 마우스의 급성 간손상에 미치는 효과 (Effects of Wolguk-whan Water Extract on Acute Oxidative Liver Injury Induced by Acetaminophen)

  • Objectives : Wolguk-whan has been used as a prescription of natural drug for the treatment of stress digestive system disease. Recently, we reported that Wolguk-whan methnol extract (WGWM) exerted a significant protective effect against oxidative damage to the liver of ICR mice. This study was purposed to investigate the effects of Wolguk-whan water extract (WGWW) on liver injury induced by oxidative stress. Methods : In order to investigate the effects of WGWW on acute liver injury, ICR mice were pretreated with WGWW for 6days, starved for 24hrs, and administerated acetamirtophen(500mg/kg, i.p.). In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results : In vivo administeration of WGWW showed effective inhibition of acetaminophen induced lipid peroxidation, and showed elevations of GSH level, catalase, GSH-Px, GST activities. Conclusions : These results suggested that WGWW might suppress the formation of oxidative metabolites, and prevent acetaminophen induced hepatotoxicity.

월국환(越鞠丸) 및 월국환가미방(越鞠丸加味方)의 항(抗)스트레스 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究) (Experiment Study on the Anti-Stress of Walgukhwan, Walgukhwan+Ejintang and Walgukhwan+Samultang)

  • This study aimed to evaluate the anti-stress effects of Walgukhwan, Walgukhwan+Ejintang and Wal gukhwan+Samultang. The experimental animals treated with each extracts during 15 days before the immobilization stress. And each experimental animals were endowed with 12 hours immobilization stress. Then the brain, blood and urine catecholamine contents and serum total cholesterol, triglyceride and free fatty acid levels were studied. The result were as followed; 1. Walgukhwan, Walgukhwan+Ejintang and Walgukhwan+Samultang were not statistically effective to the changes of the body weight. 2. Walgukhwan was statistically effective to the increase of the blood norepinephrine content as compared with control. 3. Walgukhwan+Ejintang was statistically effective to the increase of the blood and urine norepinephrine contents as compared with control. 4. Walgukhwan+Samultang was statisticallv effective to the increase of the brain, blood and urine norepinephrine contents and also to the increase of the blood and urine dopamine contents as compared with control. 5. Walgukhwan was statistically effective to the increase of the serum free fatty acid level as compared with control. 6. Walgukhwan+Ejintang was statistically effective to the increase of the serum triglyceride and free fatty acid levels as compared with control. 7. Walgukhwan+Samultang was statistically effective to the increase of the serum total cholesterol, triglyceride and free fatty acid levels as compared with control.

월국환합보중익기탕(越鞠丸合補中益氣湯)이 전기자극(電氣刺戟)스트레스를 받은 mouse의 면역기능(免疫機能)에 미치는 영향(影響) (Effects of Walgookwhanhab-Bojoongikgitang administration on immune-function in Balb/c mice stressed by electric footshock)

  • 본 실험은 월국환합보중익기탕(越鞠丸合補中益氣湯)의 항스트레스효과를 조사하기 위하여 시행되었다. 월국환합보중익기탕(越鞠丸合補中益氣湯)이 전기자극으로 스트레스 상황을 유발시킨 mouse에게 끼친 B 임파구 면역계 변화를 mouse 혈청의 면양적혈구(綿羊赤血球)에 대한 적혈구 응집소가와 비장세포에서 B 세포 및 T 세포가 차지하는 백분율의 변화를 통하여 측정하였으며 이를 대조군과 비교 분석하였다. 혈청 항체가의 측정에서 스트레스 자극하에서 월국환합보중익기탕(越鞠丸合補中益氣湯)을 투여받은 그룹에서 약제를 투여받지 않은 그룹에 비하여 유의성 있는 차이를 보여(p<0.01) 면역 기능을 정상군의 수준으로 유지하는 결과를 얻었다. T 세포와 B 세포의 백분율의 측정에서도 스트레스 자극하에서 월국환합보중익기탕(越鞠丸合補中益氣湯)을 투여 받은 그룹에서 약제를 투여 받지 않은 그룹에 비하여 유의성 있는 차이를 보여(p<0.01) 면역 기능을 정상군의 수준으로 유지하는 결과를 얻었다. 실험결과들로 보아 월국환합보중익기탕(越鞠丸合補中益氣湯)이 체액성 면역 반응과 비장 내 면역 세포의 백분율을 정상의 수준으로 유지하는 효과를 보여 면역 기능의 이상에 응용할 수 있는 가능성을 제시한다고 사료된다.
