그래핀(Graphene)은 탄소 원자가 6각 구조로 이루진 2차원 알려진 물질 중 가장 얇은(0.34 nm) 두께의 물질이며 그 밴드구로조 인해 우수한 전자 이동도($200000cmV^{-1}s^{-1}$)를 가지고 있며, 이외에도 기계적, 화학적으로 뛰어난 특성을 가진다. 대면적화 된 그래핀을 성장시키기 위한 방법으로는 화학적 기상 증착법(Chemical Vapor Deposition)이 있다. 하지만 실제 여러 전이금속에서 합성되는 그래핀은 다결정으로, 서로 다른 면 방향을 가진 계면에서 전자의 산란이 일어나며, 고유의 우수한 특성이 저하되게 된다. 따라서 전자소재로 사용되기 위해서는 단결정의 대면적화 된 그래핀에 대한 연구가 지속적으로 이루어지고 있다. 앞서의 두 문제점 중, 단결정의 그래핀 합성에 크게 영향을 미치는 요인으로는 크게 합성 온도, 촉매 기판의 탄소 용해도, 촉매 표면에서의 탄소 원자의 확산성이 있다. 본 연구에서는 구리, 니켈, 실리콘에 비해 탄소 용해도가 낮으며, 탄소 원자의 높은 확산성으로 인해 단결정의 단층 그래핀을 합성에 적합하다고 보고된 저마늄(Germanium) 기판을 사용하여 그래핀을 합성하였다. 단결정의 그래핀을 성장시키기 위해 메탄(Methane; $CH_4$)가스의 주입량과 수소 가스의 주입량을 제어하여 성장 속도를 조절 하였으며, 성장하는 그래핀의 면방향을 제어하고자 하였다. 표면의 산화층(Oxidized layer)을 제거하기 위하여 불산(Hydrofluoric acid)를 사용하였다. 불산 처리 후 표면의 변화는 원자간력현미경(Atomic force microscopipe)을 통하여 분석하였다. 합성된 그래핀의 특성을 저 에너지 전자현미경(Low energy electron microscopy), 광전자 현미경(Photo emission electron microscopy), 라만 분광법(Raman spectroscopy), 원자간력현미경(Atomic force microscopy)와 투과전자현미경 (transmission electron microscopy)을 이용하여 기판 표면의 구조와 결정성을 분석하였다.
We applied a new analyzing technique for the polyurethane acrylate hybrid emulsion sample composed of polyurethane resin and acrylate resin using the phase-lag mapping techniques of atomic force microscopy. For the analysis, we synthesized similarly sized pure polyurethane dispersion and acrylate emulsion particles, which were used for measuring the standard phase-lag intensities for each material. Based on these signal intensity, we could discriminate acryl particle in the polyurethane dispersion matrix with the resolution of a few tens of nanometers. Thus, the techniques show a new possibility in the analysis of the organic two-phase particles, and we believe the techniques are helpful to design organic particles.

This paper presents the vibration analysis and the performance evaluation of cantilevers with probing tips for non-contact scanning probe microscopy. One of the current issues of the scanning probe microscopy technology is to increase the measurement speed, which is closely tied with the dynamic characteristics of cantilevers. The primary concern in this research is to investigate the relation between the maximum possible speed of non-contact scanning probe microscopy and the dynamic characteristics of cantilevers. First, the finite element analysis is made for the vibration characteristics of various cantilevers in use. The computed natural frequencies of the cantilevers are in good agreement with measured ones. Then, each cantilever is tested with topographic measurement for a standard sample with the scanning speed changed. The performances of cantilevers are analyzed along with the natural frequencies of cantilevers. Experiments are also performed to test the effects of how to attach cantilevers in the piezo-electric actuator. Finally, measurement sensitivity has been analyzed to enhance the performance of scanning probe microscopy.

We describe a novel method of scanning probe nanofabrication using a AFM(atomic force microscopy) tip with assistance of Femtosecond laser pulses to enhance fabrication capability. Illumination of the AFM tip with ultra-short light pulses induces a strong electric field between the tip and the metal surface, which allows removing metal atoms from the surface by means of field evaporation. Quantum simulation reveals that the field evaporation is triggered even en air when the induced electric field reaches the level of a few volts per angstrom, which is low enough to avoid unwanted thermal damages on most metal surfaces. For experimental validation, a Ti: sapphire Femtosecond pulse laser with 10 fs pulse duration at 800 nm center wavelength was used with a tip coated with gold to fabricate nanostructures on a thin film gold surface. Experimental results demonstrate that fine structures with critical dimensions less than ${\sim}10nm$ can be successfully made with precise control of the repetition rate of Femtosecond laser pulses.

최근의 원자간력현미경(AFM)은 생체물질을 대상으로 여러 구조적 형상뿐만 아니라 물리적 특성 측정이 가능하여 바이오분야에 다양이 활용되고 있다. 줄기세포의 신경세포로 분화 인지에 대한 연구와 관련하여 본 연구에서는 AFM의 한 기능인 Force-Distance curve 측정법을 활용하여 신경암세포주라 불리는 SH-SY5Y를 대상으로 분화 전과 후의 세포막의 stiffness 변화를 측정하였다. 세포막의 stiffness값은 시료표면과 맞닿은 AFM 탐침에 계속적으로 수직방향의 힘이 가해질 시 AFM 캔티레버의 구부러짐 정도로 측정된다. SH-SY5Y는 RA (retinoic acid) 처리에 의해 분화유도 되었으며, 생물학적 방법인 western blotting법을 통해 분화여부를 확인하였다. 측정영역은 AFM topography 이미지 상에서 roughness가 가장 낮은 분화 전과 후 SH-SY5Y의 핵 주변영역으로 선정하였다. 선정된 영역 내에 여러 부분의 분화 전후 세포막의 stiffness 값을 측정하여 통계화한 결과, 분화 전과 후 세포막의 stiffness 차이를 확인할 수 있었다. 분화 전 SH-SY5Y 세포막의 stiffness는 0.79445 N/m인 반면, 분화 후 SH-SY5Y 세포막의stiffness는 0.60324 N/m로 확인되었다. 이는 분화 전에 비하여 분화 후 SH-SY5Y 세포막의 stiffness가 약 24.07% 감소된 것으로 판단할 수 있다. 본 연구는 생물학적 복잡한 방법이 아닌 간단한 방법으로 세포의 stiffness의 변화 측정을 통한 세포의 분화를 판별할 수 있는 방법을 개발한 것으로 여러 줄기세포의 특정세포로 분화여부 판단에 활용할 수 있을 것으로 사료된다.

최근의 원자간력현미경(AFM)은 soft한 생체물질을 비파괴적 방법 및 나노크기의 분해능으로 여러 구조적, 물리적 특성 측정이 가능하여 bio분야에 다양이 활용되고 있다. 본 연구에서는 AFM을 이용하여 줄기세포인 BM MSC(bone marrow mesenchymal stem cell)가 신경세포로 분화 여부를 측정하는 방법을 보고하고자 한다. 신경세포의 신호전달은 시냅스에서 신경전달물질을 매개로 하여 이루어지는데, 신경전달물질 중에 D-Glutamic acid는 시냅스후세포에서 흥분성 전위 크기를 증가시킨 상태를 장기간 유지시켜주는 물질로, 특정물질인 Glutamate와 항원-항체 결합을 한다. 본 연구에서는 이 두 물질간의 항원-항체 반응을 활용하여 줄기세포의 신경세포로 분화 여부를 AFM으로 측정하였다. 먼저, 수용성 시료인 두 물질을 증류수에 용해시켜 Mica 기판에 그 용액을 떨어뜨려 자연건조로 시료를 준비한 후, AFM으로 형태 및 크기를 측정하였다. D-Glutamic acid와 Glutamate는 구형 입자 형태를 보였으며, Glutamate의 너비는 ~100 nm이고, D-Glutamic acid는 ~50 nm였다. 두 물질이 든 용액을 섞었을 때, 항원-항체 반응에 의해 다른 크기의 두 구형입자가 붙어 있는 형태가 관찰되었다. 이 반응을 활용하여, 신경세포에서 분비되는 신경전달물질인 D-Glutamic acid를 선별하였다. DMEM 배지에 신경암세포주인 SH-SY5Y 를 접종한 후 $37.6^{\circ}C$의 incubator에서 24시간 배양하고, 화학적 자극(60~70 mM의 KCl 용액을 주입함)을 주어 신경전달물질 분비를 유도하였다. 그 배지에 항체 Glutamate 를 주입하여 자연건조 시킨 후 항원-항체 결합특성을 AFM으로 측정하여, 항원-항체 결합된 이미지와 동일함을 확인하였다. 결과적으로 AFM을 이용한 신경전달물질의 항원-항체 결합여부 측정을 통해, BM MSC 줄기세포의 신경세포로 분화를 판단할 수 있으며, 이 방법은 줄기세포의 특정 세포로의 분화 여부 판단에 활용될 것으로 기대된다.
The purpose of this study was analyzed to compare on the physical characters of two companies of color contact lenses. Artificial tear solution was used for measuring the rate of protein deposits and wettability. The surface roughness of lenses was measured by SEM(scanning electron microscope, Japan) and AFM(atomic force microscope, MultimodeTM, USA). As a results, The color contact lenses was not different from general soft contact lenses in a respect of other properties. However, the colored contact lenses showed a severe crack on the surface under SEM observation. There was no irregularity on the surface of the colored contact lenses in AFM photograph. The dyes were deposited in inside of lenses by microscope observation.
