  • 싸이클론은 대기오염물질의 샘플링과 여러 산업공정에서 분진을 제어, 분리하는데 널리 사용되고 있다. 이는 싸이클론의 구조 및 형태가 간단하여, 제작과 운전이 쉽고 비용이 적게 들기 때문이다 하지만 작은 입자에 대한 분리효율이 낮기 때문에 싸이클론의 구조 및 형태를 변화시키거나 싸이클론내에 운전 조건을 변화시켜 높은 분리효율을 얻으려는 연구가 수행되어 왔다. 특히, 싸이클론내에 이중으로 원심력이 작용하게 한 double cyclone(Zhu et al., 2001)과 입자 하전 및 전기장 형성을 위해 고전압을 인가하는 전기싸이클론이(Lim et al., 2001) 최근에 연구가 되어지고 있다.   • Analysis of extra- and intra- cellular metabolites is very important to study cell metabolish. Intracellular amino acids in yeast are of great interest as precursors of desired products in biomass production of the feed industries and the production of glutamate. In this study, the fermentations of P. pastoris X-33 and KM71H were carried out in shake flasks. After centrifugation, the harvested cells were mechanically disrupted by using glass bead. The supernatants were used to analyze intracellular amino acid by HPLC system. For HPLC analysis, Resolve C18 column was used with Fluorescence detector. The OPA(o-phthalaldehyde) derivation reaction was employed for analysing amino acids at 30 $^{\circ}C$, 1 ml/min and gradient mode. The concentration of intracellular protein was measured by spectrophotometer.

  • Vie investigated the clarification effects of foxtail millet wine, a traditional wine in Cheju island, by centrifugation, protease treatment and ultrafiltration(UF). It was difficult to remove completely cloudy substances in foxtail millet wine only by centrifugation. With protease concentration of $5{\times}10^{-4}%(w/v)\;at\;35^{\circ}C$, for 2 hrs, foxtail millet wine was clarified effectively. Papain and bromelain appeared similar effects to the clarification of wine, but ficin was inferior to those. Ultrafiltration with pore size $100K\;and\;0.22{\mu}m$ membranes had appeared better clarification effects than the best result of enzyme treatment, and then was considered as a simple and economic method.

  • The effects of enzyme combination, pH, acid washing and enzyme treatment sequence were investigated in the hydrolysis of soy protein. Comparing Alcalase vs. Neutrase/Alcalase, it appeared that Neutrase/Alcalase was more efficient than Alcalase alone, as the highest degree of hydrolysis (DH) was seen in Neutrase/Alcalase. A surprisingly high DH (more than 60%) was observed with Flavourzyme in the second hydrolysis. The separation of insolubles from the first hydrolysis had little effect on the second hydrolysis. When the washing water from the first hydrolysis was reused in the next hydrolysis, the DH and protein recovery were increased. The addition of calcium ion showed not so much positive effects by the stabilization of Neutrase on the Protein hydrolysis. The use of carbohydrase and repeated acid washing gave positive effects on DH. The simultaneous treatment using endoprotease and exoprotease with pH adjustment improved DH significantly.

Quality Changes of Orange Juice as Influenced by Clarification Methods (청징방법에 따른 오렌지주스의 품질특성)

  • Clarified orange juice was prepared using different clarification methods including centrifugation, enzyme treatment, ultrafiltration (UF), and combined treatment(CT). Effect of clarification methods as well as other quality parameters were investigated. Clarification was improved with increase in centrifugation speed and by lowering operating temperature. The optimum condition for centrifugation process was $5^{\circ}C$ and 10,000 rpm. UF and CT processes were very effective to produce clarified orange juice. The optimum condition of UF process was $45^{\circ}C$ and 150 kPa considering flux and turbidity CT did not significantly improve the clarification efficiency since most of the clarification was already achieved during UF process. $L^*$-values increased while $a^*$-values decreased significantly after clarification regardless of methods (p<0.05) Considering color, and recovering rate of vitamin C and soluble solids, UF process was superior than the other methods in producing clarified orange juice.

  • The present work is to introduce the preliminary experimental results for a primary hydrocyclones process in lake sediment thickening. A few cyclones based on the Rietema standard geometry were prepared. The test particles were sediments from a local lake and waste coal fly ash for a reference test. As a result of the chemical analysis, organic contaminants were abundantly found in smaller particles in overflow. Experimental results showed that the physical characteristics of particles, configuration of the cyclone and operating variables including feed solids concentration and volumetric flow rate could affect the separation efficiency. The limiting feeding velocity for the separation and enrichment of particles was 1.5 m/s, higher separation efficiency, in general, was obtained under the high velocity with the small cyclones.

Physicochemical Properties and Formulation of Citrus Juice Extracted with Different Methods (유자의 착즙방법별 착즙액의 특성 및 관능검사)

  • The effects of different extraction methods on quality of citron juices were investigated in terms of chemical composition and sensory evaluation. Extraction methods employed were press-filtration with 400 mesh sieve, centrifugation, and enzyme pretreatment using pectinase. Enzyme pretreatment method showed the highest yield of juice with 68%, while press-filtration method, the lowest yield of juice with 26.3%. The pH of the juices was relatively similar at 3.29~3.34. Filter-pressed juice showed the highest concentration of soluble solid at 6.0$^{\circ}$Brix, while the juice produced using centrifugation showed the lowest concentration at 4.5$^{\circ}$Brix. In all the juices, fructose was the main soluble sugar. Concentration of organic acid was the highest with the juice produced with centrifugation so that supplementation with higher amount with sugar was needed for overall acceptability.

Separation of Tofu-residue (biji) into Dietary Fiber and Protein Fractions (콩비지의 식이섬유와 단백질 분리)

  • Tofu-residue (biji) which was made on a laboratory scale from the three U.S. and three Korean soybean varieties contained approximately 57% dietary fiber, 20% protein, while the commercial residue contained 59% dietary fiber and 17% protein. The percent soluble fiber in total dietary fiber were 3% and 46% for residue and tofu, respectively. The tofu-residue was wet milled by blade grinding once or twice, followed by sieving and centrifugation of the liquid fraction. For twice-ground residue, the dietary fiber content increased from 58.70 to 80.6% in the sieved residue, with a fiber recovery of 90.4%. On the other hand, twice-ground centrifuged solids contained 46.8% protein, representing 42.4% of the total protein. Lipid levels in the sieved residue were much lower than in the original residue.
