Protective Effects of Ukyium(우귀음, Yougui-yin) in Zinc-induced Apoptosis of C6 Glial Cells (우귀음이 Zinc에 의한 신경교세포의 고사(Apoptosis)에 미치는 영향)

  • Objectives : The objective of the current study is to determine the protective effect of Ukyium(Yougui-yin) on the apoptosis induced by zinc. Methods : Zinc is known to generate reactive oxygen species (ROS), including superoxide anion ($O_2$) and hydrogen peroxide ($H_2O_2$), which eventually contribute to cytotoxicity in a variety of cell types. We investigated the viablity of cells, $H_2O_2$ generation, chromatin condensation and nuclear fragmentation in Hoechst dye staining and $IkB-{\alpha}$ degradation in C6 glial cells of $ZnCl_2$ between pretreatment- and not pretreatment-group with Ukyium. The former methods were researched by Time- and Dose-dependent manners. Results : We demonstrated that pretreatment with Ukyium prevented zinc-induced cell death of C6 glial cells and apoptotic characteristics including chromatin condensation and nuclear fragmentation. Ukyium also prevented $H_2O_2-induced$ cell death. We further confirmed that Ukyium decreased zinc-induced generation of $H_2O_2$ and inhibited degradation of $IkB-{\alpha}$ by zinc in C6 glial ceHs. Conclusions : These data indicated that Ukyium (Yougui-yin) prevents zinc-induced apoptotic death of C6 glial cells via inhibition of ROS generation, such as $H_2O_2$ as well as inhibition of $IkB-{\alpha}$ degradation.

Effects of Jwagyuyeum and Woogyuyeum on Free Radical Generating Enzyme Activities and Lipid Peroxidation in Rat's Kidney (흰쥐의 신(腎)에서 과산화지질(過酸化脂質) 생성(生成) 및 Oxygen Free radical 생성계(生成系) 효소(酵素) 활성(活性)에 미치는 좌귀음(左歸飮)과 우귀음(右歸飮)의 영향(影響))

  • Jwagyuyeum and Woogyuyeum, being known to reinforce Kidney-yin and -yang, were tested for the effects of on free radical generating enzyme and lipid peroxidation in rat's kidney. In vitro, levels of lipid peroxide in tissues of brain were proportionally decreased to concentration of extracts prepared from Jwagyuyeum and Woogyuyeum. They were much more decreased, when lipid peroxidation was induced with ferrous iron (Fe+2), In vivo, after both herbs were administered to the rat, levels of lipid peroxide in kidney were decreased only in Jwagyuyeum. The enzyme activities and the ratio of type conversion of xanthine oxidase were decreased only in Jwagyuyeum. But, We can't see special changes in Woogyuyeum. The enzyme activities of aldehyde oxidase was lowered in both herbs, especially Jwagyuyeum was much more done. These results suggest that Jwagyuyeum and Woogyuyeum decrease the activities of free radical generating enzymes such as xanthine oxidase and aldehyde oxidase which form lipid peroxide.

  • Jwagyuyeum and Woogyuyeum, being known to reinforce Kidney-yin and -yang, were tested for the effects of on free-radical generating enzyme and lipid peroxidation. In vitro, levels of lipid peroxide in tissues of liver were proportionally decreased to concentration of extracts prepared from Jwagyuyeum and Woogyuyeum. They were much more decreased, when lipid peroxidation was induced with ferrous iron (Fe+2). In vivo, after both herbs were administered to the rat, levels of lipid peroxide in liver were decreased only in Jwagyuyeum. And, enzyme activities of xanthine oxidase and aldehyde oxidase in liver were not changed. It was guessed that Jwagyuyeum and Woogyuyeum inhibited lipid peroxidation directly, or acted on free radical resolving enzymes which decrease lipid peroxide. Consequently both herbs, particularly in Jwagyuyeum might delay aging.

The ratio of type conversion of the brain xanthine oxidase was lowered in both, especially Jwagyuyeum was much more done. These results suggest that Jwagyuyeum and Woogyuyeum decrease the activities of free radical generating enzymes such as xanthine oxidase and aldehyde oxidase which form lipid peroxide, Consequently both herbs might delay aging.

  • This study was undertaken to examine the effect of Jwagyuyeum and Woogyuycum, being known to reinforce Kidney-yin and yang, on the activities of endogenous antioxidant enzymes and the production of oxygen free radicals in the liver and kidney tissues, Alterations in enzyme activities were observed after in vivo treatment in rats, Jwagyuyeum and Woogyuyeum caused a significant increase in the activities of superoxide dismutase(SOD) and catajase Jwagyuyeum significantly increased the activity of glutathione peroxidase in both liver and kidney, but the enzyme activity was not significantly altered by Woogyuyeum. Treatment in vitro of Jwagyuyeum and Woogyuyeum decreased the production of oxygen free radicals in a dose-dependent fashion. These results suggest that Jwagyuyeum and Woogyuyeum stimulate the activities of antioxidant enzymes and inhibit directly the production of oxygen free radicals. These effects of both herbs may contribute to prevent the oxygen free radical-induced impairment of cell function.

  • Purpose : This study is to examine what are the effects of the Woogwiyeum(WGY) on the ovariectomized rat model of postmenopausal osteoporosis. Methods : We devide mice to 3 Group(Sham operated group, control group, WGY treated group) and analyze each serum component. Results : 1. Body weight in control group showed significant increase in comparison with sham, but that in WGY-treated showed no change in comparison with control. 2. The level of serum albumin, ALP in control group showed significant decrease in comparison with sham. That in WGY-treated was decreased in comparison with control. 3. Trabecular bone area as well as trabecular thickness in control group showed significant decrease in comparison with sham. Those in WGY-treated showed significant increase in comparison with control. 4. Osteoclast number and oseoblast surface in control group showed significant increase in comparison with sham. Those in WGY-treated showed significant decrease in comparison with control. Conclusion : WGY has shown to be capable of preventing and curing osteoporosis caused by old-aged and postmenopause.

  • The purpose of this study is to observe the effect of Woogyuyem on serum reaction and hepatic tissue in Cyclosporin A treated rats. In this study, the experimental rats divided two group(Control group and Sample group). Under the same condition, both control group and sample group were injected to abdomen with Cyclosporin A for 7days. And then, control group were administered water, sample group were administered Woogy-uyeum for 300days. The change of transaminase and bile acid and SOD activity in blood serum, hepatic tissue were measured at 3rd, 7th, 15th, 30th day. The obtained results are summarised as follows : 1. In the change of SGOT contents, as compared with control group, sample group was significantly decreased at 7th and 15th day. 2. In the. change of SGPT contents, as compared with control group, sample group was significantly decreased at 15th day. 3. In the change of serum bile acid contents, as compared with control group, sample group was significantly decreased at 7th and 15th day. 4. In the change of serum SOD activity, as compared with control group, sample group was significantly increased at 15th and 30th day. 5. In the change of hepatic tissue, as compared with control group, the vacuolation of hepatocytes in the sample group was reduced, the liver plates were reconstructed, and the epitheriai cell wasn't destructed.
