The synthesis of N-benzyloxycarbonyl-L-aspartyl-L-phenylalanine methyl ester(ZAPM), a precursor of aspartame, from N-benzyloxycarbonyl-L-aspartic acid(Z-Asp) and L-phenylalanine methyl ester hydrochloride(L-PM-HCl) was investigated in ethylacetate-MES buffer two-phase system using thermolysin. In organic two-phase system, the degree of spontaneous hydrolysis of L-PM. HCl was significantly reduced with increasing the volume ratio of organic to aqueous phase. Stability of thermolysin in organic two-phase system was found to be higher than that in MES buffer solution. More than 90% of initial enzyme activity was maintained after 10 days of incubation in case that the volume of organic phase was equal to that of buffer phase, while the half life of thermolysin was about 2 days in aqueous buffer solution. The results of partitioning of substrates and product in organic two-phase system showed that the difference in partition coefficients between substrates and product was maximum at pH 5.5. The optimal pH for 2-APM synthesis in organic two-phase system was found to be 5.5-5.8, which is consistent with the value expected from the partition experiments. As the concentration of substrates was increased the conversion yield of Z-APM was increased with concomitant reduction of L-PMqHC1 hydrolysis. In case that the concentration of L-PM-HCl and Z-Asp were 160 mM and 80 mM respectively, the conversion yield of Z-APM reached 90% after 28 hrs of reaction. The yield obtained at different volume ratio of organic phase compares well with the predicted equilibrium constant in biphasic system.
In acidic media such as aqueous acetic acid, phthalimide is reduced with zinc dust to give 3-hydroxyphthalimidine while the 2,2,2-trichloroethyl esters or glycosides are reductively cleaved. However, it has been discovered that, by employing a mixture of THF and pH 4.5 buffer solution as a solvent, 2,2,2-trichloroethyl group can be selectively removed with activated zinc dust in the presence of phthalimido function, provided that the reactant or the product does not have any free carboxylic acid function. By applying the above methods, reaction of $2,2,2-trichloroethyl 3,4,6-tri-O-acetyl-2-deoxy-2-phthalimido-{\beta}-D-glucopyranoside$ (1) with activated zinc dust gave a good yield of $3,4, 6-tri-O-acetyl-2-deoxy-2-phthalimido-{\beta}-D-glucopyranose$ (5) in THF-buffer solution, and $3,4,6-tri-O-acetyl-2-deoxy-2-(3-hydroxyphthalimidino)-{\beta}-D-glucopyranose$ (6) in aqueous acetic acid.
International interests in chemical warfare agents (CWAs) have been increased recently because of the use of sarin (GB) in Syrian civil war which caused around 1,300 casualties in 2013. After exposing to natural environments, CWAs undergo hydrolysis or photodegrade to non-toxic degradation byproducts. Generally, CWAs and their degradation byproducts are present at very low concentration (e.g. several ppb), thus pretreatment processes including separation, extraction and concentration are required prior to any analyses. Liquid-liquid extraction and solid-phase extraction (SPE) are common techniques to pretreat environmental samples. Recently, a novel pretreatment method, liquid phase miecoextraction (LPME), has been applied to CWAs analysis, which could reduce amounts of solvent used but promote analytical efficiencies. Fundamental backgrounds of LPME and its application to CWAs analysis were reviewed.
Lignin is usable as fuels and heavy oil additives if depolymerized to monomer unit, because the chemical structures are similar to high octane materials found in gasoline. In this study, the solvent-phase thermal cracking(solvolysis) of lignin was performed at the various temperature and time in a laboratory tubular reactor. Conversion yield was measured for the properties of thermal cracking and liquefaction reaction of lignin. Highest conversion yield when acetone was used as thermal cracking solvent was 55.5% at $350^{\circ}C$, 50minutes and highest tar generation were $260{\sim}350mg/g\;{\cdot}\;lignin$ at $250^{\circ}C$, and highest conversion yield after tar removal was 76.88% at $300^{\circ}C$, 30minutes. Conversion yield, product compositions and amounts were determined by tar degradation yield.
BCNU 106, 154 and 171 was isolated in the areas contaminated by high concentrations of organic solvents. These strains had the tolerance to almost all kinds of organic solvents used in this study. Especially, it was interested that BCNU 106, and 171 had the tolerance to meta-, para- and ortho. xylene. And 3 strains had high MIC (minimal inhibition concentrations).
The effect of the extraction solvent on the physiological properties of the peel of the Korean pear (Pyrus pyrifolia cv. Niitaka) was evaluated. The total phenol content was highest in the 80%(wt) methanol extract, whereas flavonoid content was highest in the 80% ethanol extract. The antioxidant activities of the extracts were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging abilities, and their reducing power. The water and 80% methanol extracts of the pear peel had highest DPPH and ABTS radical scavenging activities, and reducing power, respectively. The inhibition of ${\alpha}$-glucosidase was highest in the 80% methanol extract, and alcohol dehydrogenase activity was highest in the water extract. All three extracts had similar antimicrobial activity. Because water, 80% ethanol, and 80% methanol extracts exhibited high activities in different assays of physiological properties, each solvent could be used for specific purposes.
Styrene dimers and trimers from polystyrene cup noodle containers were analyzed by GC and GC/MS extracted with various simulants. For the quantitation of styrene dimers and trimers, 1,3-diphenylpropane (DP) and benzyln-butyl phthalate (BBP) were chosen as the standards. The results showed that the average of the styrene dimers in the containers was 603 ppm, and that of trimers was 5731 ppm. Four styrene dimers, including 1,2-diphenyl-cyclobutane, were identified as well as seven trimers such as 2,4,6-triphenyl-l-hexene. The migration of the styrene dimers and trimers, from the cup noodle containers of polystyrene into foods, was conducted using simulants including boiling water as well as soybean oil and n-heptane. In addition to, the analysis of each migrated styrene was also performed filled with boiling water into noodle and soup after certain time (5, 10, 20, 30 min). The results showed that the migration of styrene dimers and trimers from cup noodle containers was not detected in the case of using boiled water or soybean oil as a simulant, while styrene dimers and trimers were detected 1.18 ppm and 14.21 ppm, respectively, when heptane was adopted as a simulant. In the case of filling with boiled water into noodle and soup, both styrenes were not detected at 5 min and 10 min, however, some samples standing for 20 min released styrene dimers and trimers as much as 0.009 ppm, and 0.019 ppm for 30 min.
This study was performed to investigate effective extract conditions in fronds of the Ostrich fern (Matteuccia struthiopteris) to increase antioxidant compound contents and antioxidant capacity. Powder (1 g) of lyophilizated fronds were mixed with 3 kinds of solvents (MeOH, 80% EtOH and water). Extractions were carried out using not only immersion (room temp.), heating ($60^{\circ}C$) and stirring (200rpm) for 6 hr, but also through sonication in a 42 kHz ultrasonic bath for 15, 30 and 45 min. Extracts were filtrated and measured for contents of soluble solids (SS), total polyphenols (TP; tannic acid as a standard) and total flavonoids (TF; Naringin as a standard). Antioxidant activity was expressed as $RC_{50}$ for DPPH and ABTS radical scavenging. SS (0.317 $g{\cdot}g^{-1}$ db), TP (70.90 $mg{\cdot}g^{-1}$ db) and TF (41.53 $mg{\cdot}g^{-1}$ db) contents reached their highest levels when 30 minute sonication extraction with 80% EtOH was performed, and the highest DPPH and ABTS scavenging activity was observed in the same extraction conditions ($RC_{50}$=0.14 $mg{\cdot}ml^{-1}$ and 0.09 $mg{\cdot}ml^{-1}$, respectively). From the present investigation, it can be concluded that fronds of the ostrich fern can be used as a natural material for antioxidants, and sonication for 15-30 min with 80% EtOH is an ideal extraction method for increasing their antioxidant effects and saving extraction time.
